The sodium cycle. I. Na+-dependent motility and modes of membrane energization in the marine alkalotolerant vibrio Alginolyticus.

Respiration, membrane potential generation and motility of the marine alkalotolerant Vibrio alginolyticus were studied. Subbacterial vesicles competent in NADH oxidation and delta psi generation were obtained. The rate of NADH oxidation by the vesicles was stimulated by Na+ in a fashion specifically sensitive to submicromolar HQNO (2-heptyl-4-hydroxyquinoline N-oxide) concentrations. The same amounts of HQNO completely suppressed the delta psi generation. Delta psi was also inhibited by cyanide, gramicidin D and by CCCP + monensin. CCCP (carbonyl cyanide m-chlorophenylhydrazone) added without

Opportunistic Vibrio alginolyticus and six other Vibrio species from ready-to-eat seafood associated with the Korean coast: Public health implications as emerging human pathogens.

Vibrio spp. are causative agents of gastrointestinal diseases, wound infection, and septicemia, primarily through consumption of or contact with contaminated seafood/water. However, little is known about the pathogenic traits of opportunistic Vibrio species: particularly in the context of their potential to cause human infections, rather than being considered solely as pathogens of aquatic animals. This study aimed to investigate the pathogenic potential and interspecies relationships of seven Vibrio spp. (V. cholerae, V. parahaemolyticus, V. vulnificus, V. alginolyticus, V. mimicus, V. fluvia

Sulfadiazine degrading bacteria Vibrio alginolyticus isolated from mariculture wastewater: Performance, degradation pathways, and mechanisms based on transcriptomic analysis.

In this study, two strains of novel sulfadiazine (SDZ)-degrading bacteria Vibrio alginolyticus SUL-3 and SUL-5 were isolated from the SDZ contaminated mariculture wastewater. The degradation rate of two strains reached 54.11% ± 0.15% and 63.57% ±0.07% of 1 mg/L SDZ within 4 d, respectively, and could also degrade other 3 sulfonamides (sulfamethoxazole, sulfamonomethoxine, and sulfamethazine). Optimal degradation conditions were identified as: neutral pH (7.0), marine salinity (35‰), and temperature of 30 °C. The addition of exogenous carbon and nitrogen sources was found to enhance bacterial d

Isoborneol as a natural food-grade antimicrobial: Targeting biofilm resilience of Vibrio alginolyticus on seafood and contact material surfaces.

Vibrio alginolyticus is an emerging seafood-borne pathogen with a high ability to form biofilms on seafood and food-contact surfaces, causing persistent contamination and higher tolerance to sanitation. Here, we evaluate the antibacterial, antibiofilm and anti-virulence potential of isoborneol, a natural food-grade monoterpenoid, against V. alginolyticus in this study. Isoborneol consistently demonstrated antibacterial activity against V. alginolyticus, with minimum inhibitory concentration and minimum bactericidal concentration values of 500 μg/mL. Membrane permeability, nucleic acid leakage,

Global climate change and wound care: case study of an off-season vibrio alginolyticus infection in a healthy man.

Vibrio alginolyticus is a halophilic Gram-negative bacterium normally present in seawater. Vibrios are not capable of cutaneous invasion through intact skin and their isolation from extraintestinal sites is uncommon. However, interruptions in skin integrity (cuts or abrasions) can allow these bacteria to cause complicated skin and soft tissues infections. This case study describes the clinical assessment and management of a nonhealing traumatic wound, sustained in a coastal area during the winter months, in a healthy 70-year-old man. Culture results were positive for V. alginolyticus. Appropri

Mass kills in hatchery-reared European seabass (Dicentrarchus labrax) triggered by concomitant infections of Amyloodinium ocellatum and Vibrio alginolyticus.

Amyloodiniosis and vibriosis are serious diseases in European seabass (Dicentrarchus labrax) hatcheries with noticeable high mortality. This study was conducted on tank-cultured D. labrax frys at a private marine hatchery near Mariout Lake (Alexandria, Egypt). Frys showed a high mortality rate (70%), lethargy, darkening, asphyxia, ascites, and velvety skin appearance. Both infectious agents were presumptively identified in all investigated frys. The identities of the two recovered agents were confirmed by molecular assay and phylogenetic analysis. On the tissue level, histopathological examina

Serine/threonine kinase PpkA coordinates the interplay between T6SS2 activation and quorum sensing in the marine pathogen Vibrio alginolyticus.

Type VI secretion systems (T6SS) are multiprotein secretion machines that can mediate killing of bacterial cells and thereby modify the composition of bacterial communities. The mechanisms that control the production of and secretion of these killing machines are incompletely understood, although quorum sensing (QS) and the PpkA kinase modulate T6SS activity in some organisms. Here we investigated control the T6S in the marine organism Vibrio alginolyticus EPGS, which encodes two T6SS systems (T6SS1 and T6SS2). We found that the organism principally relies on T6SS2 for interbacterial competiti

Molecular characterization and phylogenetic analysis of highly pathogenic Vibrio alginolyticus strains isolated during mortality outbreaks in cultured Ruditapes decussatus juvenile.

In the summer of 2008 and 2009, a series of mortalities in growing out seeds of R. decussatus juveniles were occurred in the eastern Tunisian littoral. Nine predominant bacterial strains were isolated from dead and moribund juveniles and characterized as Vibrio alginolyticus. These isolates were subjected to biochemical and molecular characterization. All the Vibrio strains were tested for their susceptibility against the most widely used antibiotic in aquaculture as well as, the assessment of the presence of erythromycin (emrB) and tetracycline (tetS) resistance genes among the tested bacteri

Uncommon Pathogens in Common Presentations: Genetic Profiling and Virulence Determinants of Vibrio alginolyticus Isolated from a Case of External Otitis.

Backgrunod/Objectives: Routine identification of common bacterial pathogens is typically efficient, utilizing standardized, cost-effective methods. However, the diagnostic process becomes significantly more complex when dealing with rare or unexpected microorganisms, especially as they can be considered colonizers in many cases. Methods: This study presents diagnostic details of an uncommon pathogen, Vibrio alginolyticus, isolated from auricular discharge in a patient with non-Hodgkin lymphoma diagnosed with persistent otitis externa and explores its identification through both conventional an

Hidden Resistances: How Routine Whole-Genome Sequencing Uncovered an Otherwise Undetected blaNDM-1 Gene in Vibrio alginolyticus from Imported Seafood.

Vibrio alginolyticus causes vibriosis of marine vertebrates, invertebrates, and humans, and while there have been several reports of multidrug resistance in V. alginolyticus, carbapenem resistance is rare. V. alginolyticus strain AUSMDU00064140 was isolated in Melbourne, Australia, from imported prawns. Routine genomic surveillance detected the presence of a full-length blaNDM-1 gene, subsequently shown to be collocated with additional acquired antimicrobial resistance genes on a resistance cassette on the largest chromosome, flanked by mobilization gene annotations. Roles of virulence regulator ToxR in viable but non-culturable formation by controlling reactive oxygen species resistance in pathogen Vibrio alginolyticus.

Under adverse circumstances, bacteria enter the viable but non-culturable (VBNC) state, a dormancy-like state for survival. The altered gene regulation underlying the entry of the VBNC state has not yet been well elucidated. Here, we reported that a subpopulation of cells (23.8 %) in Vibrio alginolyticus cultures enters the VBNC state in response to nutrient limitation at alkaline pH. The proteolysis of pivotal virulence regulator ToxR at these conditions is associated with VBNC formation. Meantime, ToxR abrogation impaired the mobility and the expression of virulence-associated genes, resulti
