A novel animal model for pseudoxanthoma elasticum: the KK/HlJ mouse.

Pseudoxanthoma elasticum is a multisystem ectopic mineralization disorder caused by mutations in the ABCC6 gene. A mouse model with targeted ablation of the corresponding gene (Abcc6(tm1JfK)) develops ectopic mineralization on the dermal sheath of vibrissae as biomarker of the progressive mineralization disorder. Survey of 31 mouse strains in a longitudinal aging study has identified three mouse strains with similar ectopic mineralization of the vibrissae, particularly the KK/HlJ strain. We report here that this mouse strain depicts, in addition to ectopic mineralization of the dermal sheath o

Representation of the body by single neurons in the dorsolateral striatum of the awake, unrestrained rat.

Single cell recordings in awake monkeys and cats have demonstrated that individual body parts are represented within striatal subregions receiving projections from somatic sensorimotor cortex. Literature indicating that the lateral striatum of the rat receives similar cortical inputs and subserves sensorimotor functions prompted a study of whether this subregion contains similar representations of the body. Single cell recordings were obtained from 923 neurons of 24 awake, unrestrained rats. Of 788 neurons categorized according to body part, 264 (34%) discharged in relation to active movement,

An example of neural plasticity evoked by putative behavioral demand and early use of vibrissal hairs after facial nerve transection.

Abnormally associated movements inevitably occur after surgical repair of the facial nerve. The reason for this postparalytic syndrome is poor navigation of regrowing axons. Despite the valuable functional advantage provided by the easily detected movement of vibrissae in rats, the major investigative tools for establishing the degree of misdirected reinnervation are still electrophysiologic recordings and retrograde tracing. In the present study we complemented data from pre- and postoperative retrograde labeling (FluoroGold, Fast Blue, DiI) of facial motoneurons with an evaluation of whisker

Coordination of Orofacial Motor Actions into Exploratory Behavior by Rat.

The delineation of sensorimotor circuits that guide exploration begins with an understanding of the pattern of motor outputs [1]. These motor patterns provide a clue to the form of the underlying circuits [2-4] (but see [5]). We focus on the behaviors that rodents use to explore their peripersonal space through goal-directed positioning of their nose, head, and vibrissae. Rodents sniff in response to novel odors, reward expectation, and as part of social interactions [6-12]. Sniffing serves olfaction [13, 14], while whisking synchronized to sniffing serves vibrissa-based touch [6, 15, 16]. We

A Descending Circuit Derived From the Superior Colliculus Modulates Vibrissal Movements.

The superior colliculus (SC) is an essential structure for the control of eye movements. In rodents, the SC is also considered to play an important role in whisking behavior, in which animals actively move their vibrissae (mechanosensors) to gather tactile information about the space around them during exploration. We investigated how the SC contributes to vibrissal movement control. We found that when the SC was unilaterally lesioned, the resting position of the vibrissae shifted backward on the side contralateral to the lesion. The unilateral SC lesion also induced an increase in the whiskin

Reorganization of vibrissal motor representation following severing and repair of the facial nerve in adult rats.

This study examined the ability of adult rat motor cortex to reorganize its relationship with the somatic musculature following the severing and regeneration of a motor nerve. For this purpose experiments were performed on ten male albino rats where the facial nerve on one side was severed, sutured and allowed to regenerate for 6 months. Cortical motor output organization was assessed by mapping the vibrissal movement area extension and thresholds evoked by intracortical electrical stimulation in anesthetized rats. In all ten animals, the cortical output pattern of the motor cortex contralater

Use of muscle fibrillation for tracking nerve regeneration.

Individual muscle fibers in denervated muscle demonstrate repetitive, spontaneous contraction. Such fibrillation activity disappears in denervated muscle if reinnervation occurs, but this relationship has not been formally studied. To test whether the disappearance of fibrillation can be used to track nerve regeneration, we quantified the presence and subsequent disappearance of electromyographic (EMG) fibrillation potentials and fibrillation-related movement in the rat tongue after unilateral hypoglossal nerve crush at two locations. In mice, fibrillation movement of vibrissae were monitored

Treatment with docosahexaenoic acid after hypoxia-ischemia improves forepaw placing in a rat model of perinatal hypoxia-ischemia.

OBJECTIVE: Docosahexaenoic acid (DHA) is a dietary fatty acid with neuroprotective properties. We hypothesized that DHA treatment after hypoxia-ischemia would improve function and reduce brain volume loss in a perinatal rat model. STUDY DESIGN: Seven-day-old Wistar rat pups from 7 litters (n = 84) underwent right carotid ligation, followed by 8% O(2) for 90 minutes. Fifteen minutes after hypoxia-ischemia, pups were divided into 3 treatment groups (intraperitoneal injections of DHA 1, 2.5, or 5 mg/kg) and 2 control groups (25% albumin or saline). At 14 days, rats underwent vibrissae-stimulated

Electrical stimulation facilitates rat facial nerve recovery from a crush injury.

OBJECTIVE: To study the effect of electrical stimulation on accelerating facial nerve functional recovery from a crush injury in the rat model. STUDY DESIGN: Experimental. METHOD: The main trunk of the right facial nerve was crushed just distal to the stylomastoid foramen, causing right-sided facial paralysis in 17 Sprague-Dawley rats. An electrode apparatus was implanted in all rats. Nine rats underwent electrical stimulation and eight were sham stimulated until complete facial nerve recovery. Facial nerve function was assessed daily by grading eyeblink reflex, vibrissae orientation, and vi

New modules are added to vibrissal premotor circuitry with the emergence of exploratory whisking.

Rodents begin to use bilaterally coordinated, rhythmic sweeping of their vibrissae ("whisking") for environmental exploration around 2 weeks after birth. Whether (and how) the vibrissal control circuitry changes after birth is unknown, and the relevant premotor circuitry remains poorly characterized. Using a modified rabies virus transsynaptic tracing strategy, we labeled neurons synapsing directly onto vibrissa facial motor neurons (vFMNs). Sources of potential excitatory, inhibitory, and modulatory vFMN premotor neurons, and differences between the premotor circuitry for vFMNs innervating in

Topography of rodent whisking--I. Two-dimensional monitoring of whisker movements.

During 'active touch' the rodent whiskers scan the environment in a series of repetitive movements ('whisks') generating afferent signals which transform the spatial properties of objects into spatio-temporal patterns of neural activity. Based upon analyses carried out in a single movement plane, it has been generally assumed that these trajectories are essentially uni-dimensional, although more complex movements have been described in some rodents. The present study was designed to examine this assumption and to more precisely characterize whisking topography by monitoring whisking trajectori

Trigeminal structure-function relationships: a reevaluation based on long-range staining of a large sample of brainstem a beta fibers.

Prior studies suggest that some classes of thickly myelinated (A beta) afferents have distinct morphologies in the trigeminal (V) brainstem complex, and that single fibers have collaterals with different shapes in the four V subnuclei. However, these conclusions are based upon relatively few and incompletely stained fibers and limited statistical rigor. In the present study, 104 fibers were stained more completely with neurobiotin in rats to provide within-fiber intersubnucleus comparisons, and between-fiber intrasubnucleus comparisons, of collaterals associated with a vibrissa, guard hairs, h

Fructus Sophorae Extract Generated Using the Natural Deep Eutectic Solvent ChCl Urea Activates Wnt/β Catenin and cAMP/MITF Signaling to Stimulate Hair Growth and Pigmentation.

Androgenetic alopecia remains challenging to treat, with limited FDA-approved therapies. Natural products, such as the fruit of Sophora japonica L. (Fructus Sophorae), could be alternatives. This study developed a green extraction method using natural deep eutectic solvents (NADESs) coupled with ultrasonic assistance to recover active phytochemicals from Fructus Sophorae and evaluated the extract for promoting hair growth and melanogenic activities. Among ten NADES formulations, ChCl-Urea (1:2) with 40% water content, named CU-40, generated the highest yield of sophoricoside (8.79 mg/g), surpa

The injury of marginal mandibular branch unexpectedly promotes the repair of buccal branch of facial nerve in a rat model.

CONCLUSIONS: The results indicate that the injury of the marginal mandibular branch improved the recovery of the buccal branch in a rat model. OBJECTIVE: The aim of this study was to investigate whether the injury or intactness of the marginal mandibular branch affects the regeneration of the facial nerve buccal branch in a rat model. METHODS: This experiment was conducted on 30 adult rats, which were randomly and equally divided into two groups. The buccal branch of the facial nerve was transected and reconstructed, with the marginal mandibular branch damaged (group A) or intact (group B).

Aberrant mineralization of connective tissues in a mouse model of pseudoxanthoma elasticum: systemic and local regulatory factors.

Pseudoxanthoma elasticum (PXE) is caused by mutations in the ABCC6 gene, but the cellular and molecular events leading to aberrant mineralization of soft tissues are unknown. To characterize the mineralization process, we examined a PXE animal model, the Abcc6-/- mouse, with respect to specific proteins serving as inhibitors of mineralization. The levels of calcium and phosphate in serum of these mice were normal, but the Abcc6-/- serum had less ability to prevent the mineral deposition induced by inorganic phosphate in a cell culture system. Addition of fetuin-A to the culture system prevente

An automated whisker tracking tool for the rat facial nerve injury paradigm.

BACKGROUND: The two-dimensional videographic analysis of vibrissal movements in behaving rodents has become a standard method to estimate the degree of functional impairment and recovery after facial nerve injuries quantitatively. The main limitation of the method is the time consuming, uneconomic process of manually tracking the vibrissae in video sequences. NEW METHOD: We developed a novel tool allowing automated detection of untagged vibrissae (two on each side of the snout). To compare the new method with the standard manual tracking approach, we used videos of unrestrained rats with unil

Changes in neuronal activities in the two ventral posterior medial thalamic nuclei in an experimental model of trigeminal pain in the rat by constriction of one infraorbital nerve.

The present study analyzes the activity of 120 neurons recorded in the two ventro-postero-medial (VPM) nuclei of the thalamus in a rat model of trigeminal neuropathic pain. Twenty-eight rats were tested 2 weeks after a chronic constriction injury (CCI) of the infraorbital nerve (IoN). These animals exhibited violent pain-related reactions to extremely weak mechanical stimuli applied to the lesioned and, to a lesser extent, unlesioned IoN territories. The activities of neurons recorded in the VPMc (n = 80) and VPMi (n = 40), contralateral and ipsilateral to the injured nerve, respectively, were
