Vinyl chloride industry in the courtroom and corporate influences on the scientific literature.

Pressure from the vinyl chloride (VC) industry on researchers involved in industry-sponsored studies and on regulatory agencies has been documented since 1970s. This commentary describes the influence of a lawsuit pursued by workers of an Italian VC plant on the recent scientific debate on VC exposure and risk of hepatocellular carcinoma (HCC). Original studies carried out by consultants of the public prosecutors and by independent researchers supported the above association. VC-industry consultants published two reviews during the lawsuit, claiming that liver angiosarcoma was the only VC-rela

Vinyl chloride-induced hepatic lesions in man and rodents. A comparison.

Histologic sequences in the liver of rodents exposed by inhalation to gaseous vinyl chloride were compared to the lesions in man exposed to the same agent, mainly in vinyl chloride polymerization plants. An identical sequence, starting with circumscribed proliferation of hepatocytes, soon followed by proliferation of a variety of sinusoidal cells and frequently associated with sinusoidal dilatation, progresses to intralobular and more frequently to trabecular angiosarcoma. Predominantly in young animals and rarely in man, hepatocellular carcinoma develops, but never cirrhosis. The sequence rep

Clinical and morphologic features of hepatic angiosarcoma in vinyl chloride workers.

Fifteen male workers exposed to vinyl chloride developed angiosarcoma of the liver. Thirteen died of disease and two are currently living for short periods after diagnosis. Their ages ranged from 36 to 58 years (average 47.5 years). Their exposure time ranged from 4 to 27.8 years (average 16.9 years). The most common presenting symptoms were fatigue, weight loss, and abdominal pain. Hepatomegaly followed by splenomegaly were the most common physical findings. Biochemical profiles yielded variable results and proved to be of little value in the detection or diagnosis. Of eight patients autopsie

Alterations of liver and spleen among workers exposed to vinyl chloride.

Preliminary review of hepatic biopsy and autopsy specimens obtained from workers engaged for prolonged periods in the polymerization of vinyl chloride indicates a fibrotic precursor lesion in the liver. It is the only lesion in some instances but also was found in the uninvolved liver of patients with angiosarcoma and in two instances in liver biopsy specimens obtained before angiosarcoma developed. This precursor stage is characterized by a conspicuous subcapsular fibrosis, a nonpathognomonic progressive portal fibrosis, and a borderline increase of intralobular connective tissue, all associa

Do occupational exposures to vinyl chloride cause hepatocellular carcinoma and cirrhosis?

Controversy exists about the association between occupational exposures to vinyl chloride and hepatocellular carcinoma and cirrhosis. Two large multicentre mortality cohort studies, one American and another European, reported higher mortality for primary cancer of liver and biliary tract. However, the American study was not able to rule out misclassification, because based on death certificates and under the heading primary liver cancers, some angiosarcomas, the typical neoplasia associated with vinyl chloride, may have been included. The American study does not report on cirrhosis mortality.

Poly(vinyl chloride) formulations: acute toxicity to cultured human cell lines.

Two quantitative cytotoxicity assay methods (cytoplasmic retention of carboxyfluorescein and mitochondrial cleavage of 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT)) have been used to evaluate the response of two cultured human cell lines; HepG2 (hepatoma) and W138va13 (transformed lung fibroblasts) to extracts of a range of poly(vinyl chloride) (PVC) formulations. Two plasticizers; di(2-ethylhexyl)phthalate (DEHP) and di-isooctyl phthalate and a range of tin and non-tin stabilizers were incorporated in the study. Only those formulations containing both a plasticizer and

Respiratory Vinyl Chloride Reductive Dechlorination to Ethene in TceA-Expressing Dehalococcoides mccartyi.

Bioremediation of chlorinated ethenes in anoxic aquifers hinges on organohalide-respiring Dehalococcoidia expressing vinyl chloride (VC) reductive dehalogenase (RDase). The tceA gene encoding the trichloroethene-dechlorinating RDase TceA is frequently detected in contaminated groundwater but not recognized as a biomarker for VC detoxification. We demonstrate that tceA-carrying Dehalococcoides mccartyi (Dhc) strains FL2 and 195 grow with VC as an electron acceptor when sufficient vitamin B12 (B12) is provided. Strain FL2 cultures that received 50 μg L-1 B12 completely dechlorinated VC to ethene

p53 mutation pattern in hepatocellular carcinoma in workers exposed to vinyl chloride.

BACKGROUND: Vinyl chloride (VC) is a known animal and human carcinogen that is associated with liver angiosarcoma and most likely also with hepatocellular carcinoma (HCC) in humans. METHODS: The authors examined the presence of p53 gene mutations in 18 HCC specimens from patients with known exposure to VC (median, 8883 parts per million-years; median duration, 245 months). In all cases, other risk factors for the development of HCC (hepatitis B virus and hepatitis C virus infections, alcohol consumption, and metabolic or autoimmune disorders) were excluded. Three patients had concomitant cirr

Built-in tandem catalysis in hierarchical pores for efficient vinyl chloride production.

Tandem catalysis over disparate active sites is frequently designed to promote the catalytic efficiency for industrial reactions. Herein, a built-in tandem system in a single hierarchical porous catalyst is constructed for vinyl chloride production from ethylene dichloride and acetylene cascade. Using a pore-selective filling strategy, Ru, CuRu, and AuRu components are sequentially confined in the macro-, meso- and micropores of carbon material. Mechanism study shows that ethylene-dichloride dehydrochlorination occurs on macropore Ru sites, generating vinyl chloride. H and Cl radicals react wi

C1-Based Route for Vinyl Chloride Synthesis with Environmental and Economic Benefits.

Selective coupling of C1 platform molecules to C2 olefins is a cornerstone for establishing a sustainable chemical industry based on nonpetroleum sources. Vinyl chloride (C2H3Cl), one of the top commodity petrochemicals, is commercially produced from coal- or oil-derived C2 hydrocarbon (acetylene and ethylene) feedstocks with a high carbon footprint. Here, we report a C1-based route for vinyl chloride synthesis via the selective oxidative coupling of methyl chloride. This is enabled by a solid catalyst, featuring tungstate nanoclusters embedded in a zirconia matrix, which effectively captures

Preparation and antimicrobial activity of poly (vinyl chloride)/gelatin/montmorillonite biocomposite films.

The aim of this study was using a novel antimicrobial thermoplastic plasticizer based on aliphatic anhydride derivative dodecenyl succinic anhydride (DSA) for blending poly (vinyl chloride), PVC, with gelatin in presence of montmorillonite (MMT) using Brabender via polymer melting technique. This anhydride-based plasticizer blended the membrane ingredients homogenously under melting process. The used plasticizer exhibited high performance antimicrobial potency for some biomedical and industrial applications. The prepared biocomposite films were evaluated for antimicrobial activity using agar d

Reductive dechlorination of the vinyl chloride surrogate chlorofluoroethene in TCE-contaminated groundwater.

At many trichloroethene (TCE)-contaminated field sites, microbial transformation of TCE results in the accumulation of vinyl chloride (VC), a known carcinogen and neurotoxin. Quantitative tools are needed to determine the in situ rates of VC transformation to ethene in contaminated groundwater. For this study, E-/Z-chlorofluoroethene (E-/Z-CFE) was evaluated as a surrogate for VC in laboratory microcosm and field push-pull tests. Single-well push-pull tests were conducted at a TCE-contaminated field site by injecting E-/Z-CFE and monitoring for the formation of fluoroethene (FE) over a period
