TRPA1 is differentially modulated by the amphipathic molecules trinitrophenol and chlorpromazine.

TRPA1, a poorly selective Ca(2+)-permeable cation channel, is expressed in peripheral sensory neurons, where it is considered to contribute to a variety of sensory processes such as the detection of painful stimuli. Furthermore, TRPA1 was also identified in hair cells of the inner ear, but its involvement in sensing mechanical forces is still being controversially discussed. Amphipathic molecules such as trinitrophenol and chlorpromazine have been shown to provide useful tools to study mechanosensitive channels. Depending on their charge, they partition in the inner or outer sheets of the lipi

Volume-activated chloride currents in pancreatic duct cells.

We have used the patch clamp technique to study volume-activated Cl- currents in the bicarbonate-secreting pancreatic duct cell. These currents could be elicited by a hypertonic pipette solution (osmotic gradient 20 mOsm/l), developed over about 8 min to a peak value of 91 +/- 5.8 pA/pF at 60 mV (n = 123), and were inhibited by a hypertonic bath solution. The proportion of cells which developed currents increased from 15% in freshly isolated ducts to 93% if the ducts were cultured for 2 days. The currents were ATP-dependent, had an outwardly rectifying current/voltage (I-V) plot, and displayed

ZIP14-VDAC2 mediates the transport of extracellular iron into mitochondria in hepatocytes under PFOS exposure.

Perfluorooctane sulfonate (PFOS), classified as a persistent organic pollutant, promotes mitochondrial iron overload through voltage-dependent anion channel 2 (VDAC2). However, the source of iron transported through VDAC2 remains unclear. Here, we reported that PFOS enhanced extracellular iron influx into human hepatocytes HepG2, which peaked at 6 h. Under PFOS exposure, total VDAC2 protein levels remained unchanged, whereas its expression increased in mitochondrial lysates but decreased in cytoplasmic lysates, which were observed both in HepG2 cells and mice liver. Meanwhile, the total protei

Multi-Allelic Mitochondrial DNA Deletions in an Adult Dog with Chronic Weakness, Exercise Intolerance and Lactic Acidemia.

(1) Background: An adult dog was presented to a board-certified veterinary neurologist for evaluation of chronic weakness, exercise intolerance and lactic acidemia. (2) Methods: A mitochondrial myopathy was diagnosed based on the histological and histochemical phenotype of numerous COX-negative muscle fibers. Whole-genome sequencing established the presence of multiple extended deletions in the mitochondrial DNA (mtDNA), with the highest prevalence between the 1-11 kb positions of the approximately 16 kb mitochondrial chromosome. Such findings are typically suggestive of an underlying nuclear

Predictive biomarkers for latent Mycobacterium tuberculosis infection.

Tuberculosis is a leading cause of infectious death worldwide, with almost a fourth of the world's population latently infected with its causative agent, Mycobacterium tuberculosis. Current diagnostic methods are insufficient to differentiate between healthy and latently infected populations. Here, we used a machine learning approach to analyze publicly available proteomic data from saliva and serum in Ethiopia's healthy, latent TB (LTBI) and active TB (ATBI) people. Our analysis discovered a profile of six proteins, Mast Cell Expressed Membrane Protein-1, Hemopexin, Lamin A/C, Small Proline R

A novel inhibitory mechanism of mitochondrion-dependent apoptosis by a herpesviral protein.

Upon viral infection, cells undergo apoptosis as a defense against viral replication. Viruses, in turn, have evolved elaborate mechanisms to subvert apoptotic processes. Here, we report that a novel viral mitochondrial anti-apoptotic protein (vMAP) of murine gamma-herpesvirus 68 (gammaHV-68) interacts with Bcl-2 and voltage-dependent anion channel 1 (VDAC1) in a genetically separable manner. The N-terminal region of vMAP interacted with Bcl-2, and this interaction markedly increased not only Bcl-2 recruitment to mitochondria but also its avidity for BH3-only pro-apoptotic proteins, thereby sup

Severe defects in absorptive ion transport in distal colons of mice that lack ClC-2 channels.

BACKGROUND & AIMS: The fluid secretion model predicts that intestinal obstruction disorders can be alleviated by promoting epithelial Cl(-) secretion. The adenosine 3',5'-cyclic monophosphate (cAMP)-activated anion channel CFTR mediates Cl(-)-dependent fluid secretion in the intestine. Although the role of the ClC-2 channel has not been determined in the intestine, this voltage-gated Cl(-) channel might compensate for the secretory defects observed in patients with cystic fibrosis and other chronic constipation disorders. We investigated whether mice that lack ClC-2 channels (Clcn2(-/-)) have

TMEM16A/ANO1 channels contribute to the myogenic response in cerebral arteries.

RATIONALE: Pressure-induced arterial depolarization and constriction (the myogenic response) is a smooth muscle cell (myocyte)-specific mechanism that controls regional organ blood flow and systemic blood pressure. Several different nonselective cation channels contribute to pressure-induced depolarization, but signaling mechanisms involved are unclear. Similarly uncertain is the contribution of anion channels to the myogenic response and physiological functions and mechanisms of regulation of recently discovered transmembrane 16A (TMEM16A), also termed Anoctamin 1, chloride (Cl(-)) channels i

State-dependent inhibition of LRRC8 volume-regulated anion channels by DCPIB.

DCPIB is the prototypical inhibitor of leucine-rich repeat containing protein 8 volume-regulated anion channels (VRACs), yet the structural basis and state dependence of inhibition remain unclear. Here, we used chimeric channels, targeted mutagenesis, and electrophysiology to define the determinants of DCPIB inhibition and elucidate how drug binding is coupled to channel gating. Effective inhibition required coordinated contributions from extracellular loop 1 (EL1) and transmembrane domain 2 (TM2), which together form a functional module governing access to and stabilization of DCPIB-bound sta

Mitochondrial binding of α-enolase stabilizes mitochondrial membrane: its role in doxorubicin-induced cardiomyocyte apoptosis.

α-Enolase is a metabolic enzyme in the catabolic glycolytic pathway. In eukaryotic cells, the subcellular compartmentalization of α-enolase as well as its multifaceted functions has been identified. Here, we report that α-enolase is a regulator of cardiac mitochondria; it partially located in the mitochondria of rat cardiomyocytes. Doxorubicin treatment displaced α-enolase from mitochondria, accompanied by activation of mitochondrial cell death pathway. Furthermore, in isolated mitochondria, recombinant α-enolase significantly alleviated Ca(2+)-induced loss of membrane potential, swelling of m

Bicarbonate and chloride secretion in Calu-3 human airway epithelial cells.

Serous cells are the predominant site of cystic fibrosis transmembrane conductance regulator expression in the airways, and they make a significant contribution to the volume, composition, and consistency of the submucosal gland secretions. We have employed the human airway serous cell line Calu-3 as a model system to investigate the mechanisms of serous cell anion secretion. Forskolin-stimulated Calu-3 cells secrete HCO-3 by a Cl-offdependent, serosal Na+-dependent, serosal bumetanide-insensitive, and serosal 4,4'-dinitrostilben-2,2'-disulfonic acid (DNDS)-sensitive, electrogenic mechanism as

Lymphocystis Disease Virus (Iridoviridae) Enters Flounder (Paralichthys olivaceus) Gill Cells via a Caveolae-Mediated Endocytosis Mechanism Facilitated by Viral Receptors.

In previous research, voltage-dependent anion channel protein 2 (VDAC2) and the receptor of activated protein C kinase 1 (RACK1) in flounder (Paralichthys olivaceus) were confirmed as functional receptors for lymphocystis disease virus (LCDV) entry; however, the underlying mechanism of VDAC2- and RACK1-mediated LCDV entry remains unclear. In this study, we elucidated the endocytosis pathway of LCDV entry into flounder gill (FG) cells by treatment with specific inhibitory agents, siRNAs, and co-localization analysis. LCDV entry was significantly inhibited by the disruption of caveolae-mediated

Citrullination of VDAC3 by PAD2 restricts mitochondrial metabolism and suppresses macrophage reprogramming in sepsis.

Sepsis is a life-threatening inflammatory disorder characterized by immune dysregulation and organ failure, and while peptidylarginine deiminase-2 (PAD2) regulates immune responses via citrullination, its role in immunometabolism remains unclear. Here we identify a PAD2-dependent mechanism that impairs mitochondrial glutamine metabolism and macrophage polarization during sepsis. Through single-cell RNA sequencing of bronchoalveolar lavage fluid from septic mice, we found that Pad2 -/- promotes M2 macrophage polarization, reduces lung injury, and improves survival. Using mass spectrometry-based

Proteomic investigation of effects of hydroxysafflor yellow A in oxidized low-density lipoprotein-induced endothelial injury.

Oxidized low-density lipoprotein (ox-LDL)-induced vascular endothelial damage is a key event in early atherosclerosis. Safflower has been used to treat atherosclerotic heart disease in China for many years, but its molecular basis remains unclear. Hydroxysafflor yellow A (HSYA) is the main active ingredient of aqueous safflower extract. We identified the proteins involved in HSYA activity against ox-LDL-induced endothelial injury using isobaric tags for relative and absolute quantification-coupled two-dimensional liquid chromatography-tandem mass spectrometry. HSYA (1, 5, or 25 μM) alleviated

Is the Voltage-Dependent Anion Channel a Major Player in Neurodegenerative Diseases?

The family of voltage-dependent anion channels (VDACs) comprises three isoforms (VDAC-1, VDAC-2, VDAC-3). VDACs have been extensively described as localised in the outer mitochondrial membrane where they are involved in the exchange of ions, metabolites, and ATP/ADP between mitochondria and cytosol. The VDAC interacts with disease-specific proteins and thus regulates the mitochondrial function and controls the cellular energy resources, explaining its involvement in cell death and apoptosis. In addition, VDAC-1 and -2 can also be found at other cellular locations such as in the sarcoplasmic re

Regulation of chloride secretion across porcine endometrial epithelial cells by prostaglandin E2.

1. The objective of this study was to investigate the mechanism of PGE2 regulation of Cl- transport across glandular endometrial cells grown in primary culture. 2. Most of the basal short circuit current (Isc) was inhibited by luminal addition of 5-nitro-2-(3-phenylpropylamino)benzoic acid (NPPB) or glibenclamide, suggesting the presence of a basally active Cl- conductance in the apical membrane. 3. Basolateral addition of 10 microM PGE2 increased Isc by 41 +/- 3 microA. A similar response was observed when cells were treated with 8-(4-chlorophenylthio) adenosine 3',5'-cyclic monophosphate (CP

Activation of glycine and glutamate receptors increases intracellular calcium in cells derived from the endocrine pancreas.

We studied calcium signaling in a newly described pancreatic cell line, GK-P3, that expresses functional amino acid neurotransmitter receptors. GK-P3 cells express the first strychnine-sensitive glycine receptors reported in a permanent cell line. In addition, GK-P3 cells express alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA)-type glutamate receptors. Both types of amino acid receptors showed electrophysiological and pharmacological behavior similar to their neuronal counterparts. The glycine receptors were permeable to Cl- and blocked by the selective antagonist strychnine. A

[Effects and mechanisms of hesperetin in attenuating alcoholic liver injury by targeting mitochondria to regulate hepatocyte pyroptosis].

This study investigated the effects and potential mechanisms of hesperetin(HST) in targeting mitochondria to regulate hepatocyte pyroptosis and alleviate alcoholic liver injury through in vitro experiments. Human immortalized hepatocytes(THLE-2) were used to establish an in vitro alcoholic liver injury model induced by 200 mmol·L~(-1) ethanol. The effective concentrations of HST(40, 80, and 160 μmol·L~(-1)) were determined using the MTT assay. The effects of HST on hepatocyte pyroptosis were evaluated by optical microscopy, lactate dehydrogenase(LDH) release assay, and flow cytometry, and its

Nitric oxide cross-links calcium signals to enhance cold tolerance via inhibiting calmodulin expression in watermelon.

Nitric oxide (NO) is a pivotal gaseous signaling molecule that plays a critical role in regulating plant tolerance to cold stress; however, the underlying mechanisms of signal transduction remain poorly elucidated. In this study, knockout of nitrate reductase 1 (ClNR1), a crucial gene for NO biosynthesis, led to reduced cold tolerance in watermelon (Citrullus lanatus), accompanied by downregulation of cycle nucleotide-gated channel (ClCNGC) 20, a key Ca2+-permeable channel gene, decreased Ca2+ influx, and upregulation of calmodulin (ClCaM) 2/5/7. Conversely, application of the NO donor sodium

Ethanol influences on Bax associations with mitochondrial membrane proteins in neonatal rat cerebellum.

These studies investigated interactions taking place at the mitochondrial membrane in neonatal rat cerebellum following ethanol exposure and focused on interactions between proapoptotic Bax and proteins of the permeability transition pore (PTP), voltage-dependent anion channel (VDAC) and adenine nucleotide translocator (ANT) of the outer and inner mitochondrial membranes, respectively. Cultured cerebellar granule cells were used to assess the role of these interactions in ethanol neurotoxicity. Analyses were made at the age of maximal cerebellar ethanol vulnerability (P4), compared to the late
