The hallmark of aerobic glycolysis in cancer progression is well-established, yet its specific role in pancreatic cancer (PC) remains elusive. Here, we reported that TFAP2A is upregulated in PC tissues, and elevated TFAP2A expression correlates with poor prognosis in patients. Functionally, we found that TFAP2A boosted the viability, invasion, and migration of PC cells in vitro, whereas TFAP2A depletion restrained tumor growth in vivo. Moreover, TFAP2A knockdown hindered aerobic glycolysis of PC cells. Further investigation revealed that TFAP2A deletion decreased LDHA expression in PC cells. L

Hepatocellular carcinoma (HCC) is a highly heterogeneous malignancy characterized by high incidence and mortality rates, representing a major global health challenge. Mounting evidence indicates that metabolic reprogramming contributes substantially to tumor development by promoting cancer cell proliferation and metastasis. However, the underlying mechanisms remain incompletely understood. Acetyl-CoA acyltransferase 2 (ACAA2), a mitochondrial enzyme that catalyzes the final step of fatty acid β-oxidation, plays a key role in cellular acetyl-CoA homeostasis. While acetyl-CoA acts as a pivotal m

Breast cancer has become the most common malignant tumor among women worldwide, among which triple-negative breast cancer (TNBC) is a subtype with the worst prognosis. Although the function of long non-coding RNA (lncRNA) RUNDC3A-AS1 in other cancers has been revealed, it has not been reported in TNBC. To explore the functional mechanism of RUNDC3A-AS1 in TNBC by regulating miR-224-3p/COL5A2, the predictive value of RUNDC3A-AS1 for disease prognosis was evaluated by using its serum level in TNBC patients. miR-224-3p/COL5A2, as a potential downstream of RUNDC3A-AS1, was predicted by the lncRNAS

Cancer is a redox disease. Low levels of reactive oxygen species (ROS) are beneficial for cells and have anti-cancer effects. ROS are produced in the mitochondria during ATP production by oxidative phosphorylation (OXPHOS). In the present review, we describe ATP production in primary brain tumors, glioblastoma, in relation to ROS production. Differentiated glioblastoma cells mainly use glycolysis for ATP production (aerobic glycolysis) without ROS production, whereas glioblastoma stem cells (GSCs) in hypoxic periarteriolar niches use OXPHOS for ATP and ROS production, which is modest because o

This review systematically explores the dynamic regulatory roles of lysine lactylation (Kla) in the tumor microenvironment (TME) and its clinical translational potential. As an emerging post-translational modification, Kla modifies histones and non-histone proteins via lactate generated by the Warburg effect, thereby reshaping tumor metabolism and immune landscapes. Mechanistically, Kla orchestrates metabolic reprogramming and immunosuppression through key signaling pathways such as HIF-1α, mTOR, and NF-κB. Specifically, it promotes the activation of immunosuppressive cells while inhibiting cy

As the foremost endocrine system cancer, thyroid carcinoma exhibits an accelerating epidemiological trend across populations. Despite advances in treatment modalities, recurrence and metastasis remain challenges. The long non-coding RNA PVT1 (lncPVT1) has emerged as a conserved oncogenic regulator in multiple cancer types, yet its specific function in TC requires further exploration. Our research focused on examining how lncPVT1 affects glycolysis under both normoxic and hypoxic conditions using TC cell lines TPC-1 and K1. Overexpression of lncPVT1 significantly enhanced glucose consumption an

Environmental exposure to formaldehyde is known to be associated with cancers and many other diseases. Although formaldehyde has been classified as a group I carcinogen, the molecular mechanisms of its carcinogenicity are still not fully understood. Formaldehyde is also involved in the folate-driven one‑carbon metabolism, and excess amount of formaldehyde was found to interfere with other metabolic pathways including glycolysis, which can enhance Warburg effect and induce immunosuppression in tumor microenvironment. Therefore, different tumor cells and THP-1 derived macrophages were utilized t

The harsh microenvironment of ductal carcinoma in situ (DCIS) exerts strong evolutionary selection pressures on cancer cells. We hypothesize that the poor metabolic conditions near the ductal center foment the emergence of a Warburg Effect (WE) phenotype, wherein cells rapidly ferment glucose to lactic acid, even in normoxia. To test this hypothesis, we subjected low-glycolytic breast cancer cells to different microenvironmental selection pressures using combinations of hypoxia, acidosis, low glucose, and starvation for many months and isolated single clones for metabolic and transcriptomic pr

Pancreatic ductal adenocarcinoma (PDAC) is one of the most lethal human cancers. It thrives in a malnourished environment; however, little is known about the mechanisms by which PDAC cells actively promote aerobic glycolysis to maintain their metabolic needs. Gene Expression Omnibus (GEO) was used to identify differentially expressed miRNAs. The expression pattern of miR-30d in normal and PDAC tissues was studied by in situ hybridization. The role of miR-30d/RUNX1 in vitro and in vivo was evaluated by CCK8 assay and clonogenic formation as well as transwell experiment, subcutaneous xenograft m

BACKGROUND: The tumor microenvironment (TME) is characterized by a reversed pH gradient-acidic extracellular and alkaline intracellular conditions-arising from mitochondrial dysfunction, metabolic reprogramming, and dysregulated proton transport. These alterations establish a permissive niche for tumor progression, immune evasion, and resistance to therapy. Although the TME is increasingly recognized as a key determinant of cancer behavior, effective and targeted strategies for its bioenergetic reprogramming remain scarce.
OBJECTIVES: This study introduces and evaluates Eigen/Zundel Complexes

Mitochondria play a central role in cellular energetics, metabolism, and apoptosis, and their dysfunction is increasingly recognized as a hallmark of cancer. The mitochondrial genome encodes essential subunits of oxidative phosphorylation (OXPHOS) complexes, tRNAs, and rRNAs, which are critical for mitochondrial functions. However, mitochondrial DNA (mtDNA) lacks protective histones and efficient repair mechanisms, rendering it highly susceptible to mutations and epigenetic changes. This review examines the role of mitochondrial-encoded genes (MEGs) in cancer progression, emphasizing the molec

Nasopharyngeal carcinoma (NPC) is a prevalent malignancy with a distinct geographical distribution. This study aimed to identify core glycolysis-related genes and investigate their functional roles in NPC pathogenesis. Integrated transcriptomic analysis was performed on multiple Gene Expression Omnibus datasets. Screening of core genes was performed computationally via machine learning algorithms on the pool of glycolysis-related differentially expressed genes (Gly-DEGs). The tumor-suppressive function of insulin-like growth factor 1 (IGF1) was demonstrated through a combination of in vitro as

The Warburg effect is a shift from oxidative phosphorylation to anaerobic glycolysis, accompanied by an enormous increase in glucose uptake into cancer cells. We have utilized this effect to design a new group of targeted 1,4-naphthoquinone-glucose derivatives conjugated with a novel thiomethylene linker that are cytotoxic to prostate cancer cells. Compound PeS-9 revealed the highest efficacy and selectivity, which was conditioned by a GLUT-1-mediated uptake. PeS-9 induced androgen receptor degradation followed by downregulation of its signaling. In addition, it increased reactive oxygen speci

Alternative splicing of pyruvate kinase gene (PKM) results in a higher PKM2/PKM1 ratio that contributes to the Warburg effect and reversing the Warburg effect has opened novel avenues for cancer treatment. miR-206 functions as a tumor suppressor in several types of cancer. However, the effect and underlying mechanisms of miR-206 on the Warburg effect are not yet elucidated. Here, we showed that miR-206 expression was obviously decreased in CRC tissues based on LinkedOmics. A significant decrease in miR-206 expression was negatively correlated with advanced tumor stage, while inversely correlat

The metabolic reprogramming of tumor cells plays a critical role in cancer progression, contributing to drug resistance and tumor survival. Tyrosine kinase inhibitors (TKIs) have shown promising clinical results by targeting specific signaling pathways in cancer cell proliferation, survival, and metastasis and are now standard of care for NSCLC with actionable mutations. However, secondary resistance to TKIs remains a significant challenge. Here, we explored the rationale behind combining TKIs with an inhibitor of glucose metabolism (dichloroacetate, DCA), focusing on the synergistic effects f

Infection triggers expansion and effector differentiation of T cells specific for microbial antigens in association with metabolic reprograming. We found that the glycolytic enzyme lactate dehydrogenase A (LDHA) is induced in CD8+ T effector cells through phosphoinositide 3-kinase (PI3K) signaling. In turn, ablation of LDHA inhibits PI3K-dependent phosphorylation of Akt and its transcription factor target Foxo1, causing defective antimicrobial immunity. LDHA deficiency cripples cellular redox control and diminishes adenosine triphosphate (ATP) production in effector T cells, resulting in atten

The immunosuppressive tumor microenvironment (TME) remains a major therapeutic challenge in oncology. This immunosuppression arises through two synergistic mechanisms: impaired cytotoxic function of T cells and NK cells, and differentiation of immune cells into immunosuppressive subtypes. Metabolic reprogramming, particularly the Warburg effect (aerobic glycolysis in tumor cells), plays a pivotal role in shaping this immunosuppressive TME. While lactate was traditionally viewed as a metabolic byproduct, emerging evidence reveals its profound immunomodulatory functions. As the end-product of gl

BACKGROUND AND AIMS: Intrahepatic cholangiocarcinoma (ICC) is an aggressive malignancy with high heterogeneity and poor prognosis. Long noncoding RNAs (lncRNAs) play critical roles in tumorigenesis through dysregulated expression. We explored the effects and mechanisms of nuclear transcription factor NF-κB interacting lncRNA (NKILA) in ICC.
METHODS: Bioinformatic analysis was performed to determine the expression and relationship of NKILA with metaxin 1 (MTX1), and translocase of outer mitochondrial membrane 40 (TOMM40) expression in ICC tissue samples. Cholangiocarcinoma cell lines were cult

Carbonic anhydrase IX (CA9), a pH-regulating transmembrane protein, is highly expressed in solid tumors, and particularly in clear cell renal cell carcinoma (ccRCC). The catalytic mechanisms of CA9 are well defined, but its roles in mediating cell migration/invasion and survival in ccRCC remain to be determined. Here, we confirmed that the mRNA expression of CA9 in ccRCC was significantly higher than that in para-carcinoma tissues from analysis of the datasets in The Cancer Genome Atlas. CA9 knockdown upregulated oxidative phosphorylation-associated proteins and increased mitochondrial biogene
