A revised excretion factor for estimating ketamine consumption by wastewater-based epidemiology - Utilising wastewater and seizure data.

The rate of drug excretion (excretion factor) is a critical parameter for monitoring drug consumption in the population by wastewater-based epidemiology (WBE). Previous studies have refined excretion factors for common illicit drugs, such as cocaine, amphetamine, methamphetamine, heroin, to improve the accuracy and reduce uncertainty in back-calculating consumption. Nevertheless, for ketamine, one of the most prevalent psychoactive substances, a careful review of its excretion factors has not been performed due to limited pharmacokinetic data. Here we review WBE studies and seizure data to ref

Evaluation of the Enterovirus serotype monitoring approach for wastewater surveillance of hand foot and mouth disease using secondary epidemiological surveillance data.

The serotypes of Enterovirus A (EVA) causing hand, foot, and mouth disease (HFMD) exhibit cyclical dominance, with the dominant serotypes being EVA71, Coxsackievirus A type 6 (CVA6), CVA10, and CVA16. In this study, a quadruplex digital polymerase chain reaction (dPCR) assay was developed for serotype-level monitoring in wastewater surveillance for HFMD. The quadruplex dPCR assay performance was comparable to that of the singleplex assays. Grab influent samples (n = 122) were collected weekly from a wastewater treatment plant in Yamanashi Prefecture, Japan, from March 2022 to July 2024. The dP

The effect of the modifiable areal unit problem (MAUP) on spatial aggregation of COVID-19 wastewater surveillance data.

Large wastewater-based epidemiology (WBE) projects often have wide coverage and multiple sampling sites, necessitating spatial aggregation for data reporting and interpretation. However, the outcome may be impacted by a type of statistical bias called the modifiable areal unit problem (MAUP). In this study, we examined the presence and extent of the MAUP scaling effect on a New York State COVID-19 wastewater surveillance project. Specifically, we investigated three metrics: 1) the difference in wastewater SARS-CoV-2 concentrations between sampling at city-level site (i.e., city's primary waste

Implementation and Early Outcomes of Laboratory Proficiency Testing Program for the National Wastewater Surveillance System, United States, 2024.

Underlying the core goals of the Centers for Disease Control and Prevention National Wastewater Surveillance System (NWSS) in the United States is the need for robust and reliable pathogen data. The rapid build-out of the NWSS network and participation of many laboratories with varying expertise in environmental molecular methods has resulted in a lack of method standardization and poor data comparability. Proficiency testing is a powerful strategy to address this challenge. We describe preliminary outcomes from 2 proficiency testing rounds with 38 total participating laboratories, where repor

Tracking SARS-CoV-2 variants in wastewater in San Pedro de la Paz, Chile.

Studies have shown the presence of SARS-CoV-2 in the stool of both symptomatic and asymptomatic COVID-19 patients, enabling wastewater-based surveillance (WBS) to complement clinical monitoring. The emergence of variants can enhance viral transmissibility, highlighting the need for ongoing surveillance to detect and control infectious diseases. This study aimed to detect SARS-CoV-2 variants in wastewater from a treatment plant in San Pedro de la Paz, Chile, between January and November 2021. Wastewater samples were concentrated using the polyethylene glycol method, and RT-qPCR assays were perf

Predictive values of time-dense SARS-CoV-2 wastewater analysis in university campus buildings.

Wastewater-based SARS-CoV-2 surveillance on college campuses has the ability to detect individual clinical COVID-19 cases at the building-level. High concordance of wastewater results and clinical cases has been observed when calculated over a time window of four days or longer and in settings with high incidence of infection. At Duke University, twice a week clinical surveillance of all resident undergraduates was carried out in the spring 2021 semester. We conducted simultaneous wastewater surveillance with daily frequency on selected residence halls to assess wastewater as an early warning

Wastewater-based epidemiology approach to assess population exposure to pesticides: a review of a pesticide pharmacokinetic dataset.

Wastewater-based epidemiology is an innovative approach to estimate a population's intentional and unintentional consumption of chemicals based on biomarker assays found in wastewater. This method can provide real-time objective information on the xenobiotics to which a population is directly or indirectly exposed. This approach has already been used to assess the population exposure to four classes of pesticides: organochlorines (chlordecone), triazines, organophosphates, and pyrethroids. This review aims to obtain the data (excretion rates) and characteristics (pesticide and metabolites stab

Evaluation of wastewater SARS-CoV-2 surveillance for monitoring COVID-19 trends and early warning in Sweden, 2023-2024.

Wastewater surveillance (WWS) of SARS-CoV-2 RNA levels offers a way to monitor virus circulation independently of clinical testing. With COVID-19 now endemic, clarifying the role of WWS within routine surveillance is essential. The aim was to evaluate the potential of WWS as an early warning indicator for COVID-19 in Sweden, based on its correlation with reported case data. The study includes data from July 2023 to December 2024, from 19 wastewater treatment plants (WWTPs), covering 43% of Sweden's population, and surveillance data of reported COVID-19 cases. Analyses were performed by WWTP, s

Tracking SARS-CoV-2 transmission: evaluation of the Québec wastewater surveillance program.

Québec's COVID-19 wastewater surveillance program, initiated in March 2022, was evaluated after its first year to determine its effectiveness in tracking SARS-CoV-2 transmission. The evaluation assessed the program's validity, completeness, and timeliness. As per visual analysis, results showed temporal coherence between wastewater and clinical data, although this was period dependent. Cross-correlation function analysis failed to generate consistent results and interpretable patterns between wastewater signal and clinical indicators. The percentage of unplanned missing wastewater results was

Challenges on the implementation of wastewater-based epidemiology as a prediction tool: the paradigm of SARS-CoV-2.

Wastewater Based Epidemiology (WBE) has been identified as a tool for monitoring and predicting patterns of SARS-CoV-2 in communities. Several factors may lead to a day-to-day variation in the measurement of viral genetic material. Wastewater samples are systematically collected from the two major wastewater treatment plants in Crete, Greece. Physico-chemical factors were tested, viral concentration was determined by RT-real time PCR and the results were normalized. The influence of restriction measures, rain and physico-chemical agents was addressed. Statistics together with machine learning

Effective method to mitigate impact of rain or snowmelt sewer flushing events on wastewater-based surveillance measurements.

Wastewater-based surveillance (WBS) is increasingly used for monitoring disease targets in wastewaters around the world. This study, performed in Ottawa, Canada, identifies a decrease in SARS-CoV-2 wastewater measurements during snowmelt-induced sewer flushing events. Observations first revealed a correlation between suppressed viral measurements and periods of increased sewage flowrates, air temperatures above 0 °C during winter months, and solids mass flux increases. These correlations suggest that high sewage flowrates from snowmelt events or intense precipitation events lead to the scourin

Wastewater-Based Epidemiology of Hepatitis A in Mexico: Temporal Association with Hospital Admissions and Clinical-Environmental Phylogenetic Relatedness.

Hepatitis A (HepA) is a viral hepatitis caused by the hepatitis A virus (HAV), and currently Mexico is considered to have an intermediate-HAV endemicity. Wastewater-based epidemiology (WBE) is a tool for monitoring infectious diseases but has been poorly evaluated in intermediate-HAV-endemicity settings. From June 2024 to June 2025, 24-hour composite wastewater samples were collected twice a week from three wastewater treatment plants in the metropolitan area of San Luis Potosí, Mexico. Wastewater samples were ultracentrifuged, RNA was extracted, and HAV and crAssphage (for normalization) were

A Systematic Review of Methodological Approaches to SARS-CoV-2 Wastewater Surveillance.

Following the COVID-19 pandemic, researchers have increasingly focused on monitoring the spread of the virus and improving methods to detect changes in the SARS-CoV-2 genome. Although clinical surveillance provides direct and reliable results, it has limited applicability. Wastewater-based epidemiology (WBE) has therefore emerged as a valuable, non-invasive complementary tool for disease surveillance. It provides a comprehensive picture of virus circulation in a population, including asymptomatic individuals and those who do not seek healthcare. In addition, it facilitates early detection of o

Genotype-Specific Detection of Measles Virus Using Wastewater Surveillance - Wisconsin, February 2026.

In February 2026, two travel-associated cases of measles were reported to the Wisconsin Department of Health Services (WDHS). Public health investigations for each case included wastewater (sewage) surveillance for wild-type measles, which was first implemented by the WDHS Wastewater Monitoring Program in June 2025. The virus from one case, caused by measles genotype D8, was detected by a wastewater surveillance assay before the case was identified by local health authorities, leading to public health notifications and action. Measles virus from the second case, caused by measles genotype B3,

Assessment of the clinical and analytical performance of three Seegene Allplex SARS-CoV-2 assays within the VALCOR framework.

The coronavirus disease 2019 (COVID-19) pandemic demonstrated the need for accurate diagnostic testing for the early detection of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Although the pandemic has ended, accurate assays are still needed to monitor viral spread at national levels and beyond through population and wastewater surveillance. To enhance early detection, SARS-CoV-2 assays should have high diagnostic accuracy and should be validated to assure accurate results. Three distinct SARS-CoV-2 assays were evaluated with clinical samples using the VALCOR (VALidation of SAR
