Biomanufacturing is being explored as a potential solution for improved space logistics in austere environments. One potentially useful tool is electroporation-the delivery of exogenous DNA into cells using pulsed electric fields to increase cell membrane permeability. Electroporation has the potential to enable tunable bioreactors in a space environment through the targeted delivery of nucleic acids encoding proteins of interest and may also present a solution for disinfection, purification, and biomolecule extraction without requiring disposable reagents. Unfortunately, electroporation has n

Astronauts in microgravity experience multi-system deconditioning, impacting their inflight efficiency and inducing dysfunctions upon return to Earth gravity. To fill the sex gap of knowledge in the health impact of spaceflights, we simulate microgravity with a 5-day dry immersion in 18 healthy women (ClinicalTrials.gov Identifier: NCT05043974). Here we show that dry immersion rapidly induces a sedentarily-like metabolism shift mimicking the beginning of a metabolic syndrome with a drop in glucose tolerance, an increase in the atherogenic index of plasma, and an impaired lipid profile. Weightlessness induces a cephalad shift of blood and cerebrospinal fluid that may increase intracranial pressure (ICP) during spaceflight, whereas lower body negative pressure (LBNP) may provide an opportunity to caudally redistribute fluids and lower ICP. To investigate the effects of spaceflight and LBNP on noninvasive indicators of ICP (nICP), we studied 13 crewmembers before and after spaceflight in seated, supine, and 15° head-down tilt postures, and at ∼45 and ∼150 days of spaceflight with and without 25 mmHg LBNP. We used four techniques to quantify nICP: cerebral and cochlear fluid pre

To investigate the effect of simulated weightlessness on the pharmacokinetics of orally administered moxifloxacin and the antacid Maalox or the antidiarrheal Pepto-Bismol using a tail-suspended (TS) rat model of microgravity. Fasted control and TS, jugular-vein-cannulated, male Sprague-Dawley rats received either a single 5 mg/kg intravenous dose or a single 10 mg/kg oral dose of moxifloxacin alone or with a 0.625 mL/kg oral dose of Maalox or a 1.43 mL/kg oral dose of Pepto-Bismol. Plasma concentrations of moxifloxacin were measured by HPLC. Pharmacokinetic data were analyzed using WinNonlin.

This paper summarizes what has been learned from studies of the effects of artificial gravity generated by centrifugation in actual and simulated weightless conditions. The experience of artificial gravity during actual space flight in animals and humans are discussed. Studies using intermittent centrifugation during bed rest and water immersion, as a way to maintain orthostatic tolerance and exercise capacity, are reviewed; their results indicate that intermittent centrifugation is a potential countermeasure for maintaining the integrity of these physiological functions in extended space miss

Microgravity exposure during spaceflight has been linked to cognitive impairments, including deficits in attention, executive function, and spatial memory. Both space missions and ground-based analogs-such as head-down bed rest, dry immersion, and hindlimb unloading-consistently demonstrate that altered gravity disrupts brain structure and neural plasticity. Neuroimaging data reveal significant changes in brain morphology, functional connectivity, and cerebrospinal fluid dynamics. At the cellular level, simulated microgravity impairs synaptic plasticity, alters dendritic spine architecture, an

With long-term space flights being planned for the Moon and Mars, proper countermeasures must be taken to facilitate human health in microgravity environments. Exercise is a vital countermeasure used to prevent bone and muscle loss, among other health interests. Future exploration missions encourage creating an exercise device that is both compact and can be used to properly execute exercise by the astronauts. Current design considerations include interfacing an exercise device with a vibration isolation and stabilization (VIS) system, which is necessary for protecting the spacecraft and sensi

Spaceflight is known to cause ophthalmic changes in a condition known as spaceflight-associated neuro-ocular syndrome (SANS). It is hypothesized that SANS is caused by cephalad fluid shifts and potentially mild elevation of intracranial pressure (ICP) in microgravity. Head-down tilt (HDT) studies are a ground-based spaceflight analogue to create cephalad fluid shifts. Here, we developed non-invasive magnetic resonance imaging (MRI)-based techniques to quantify ophthalmic structural changes under acute 15° HDT. We specifically quantified: (i) change in optic nerve sheath (ONS) and optic nerve (

BACKGROUND: The harsh environmental conditions during space travel, particularly weightlessness, impose a major burden on the human body including the cardiovascular system. Given its importance in adjusting the cardiovascular system to environmental challenges, the autonomic nervous system has been in the focus of scientists and clinicians involved in human space flight. This review provides an overview on human autonomic research under real and simulated space conditions with a focus on orthostatic intolerance.
METHODS: The authors conducted a targeted literature search using Pubmed.

Previous studies have shown decreased microbial diversity in astronaut gut microbiomes during spaceflight, raising potential health concerns. Enterobacter cloacae, a commensal member of the human gastrointestinal tract microbiota, has the potential to cause opportunistic infections in immunocompromised patients. However, little is known about how E. cloacae cells adapt to extreme environments such as microgravity, and whether this bacterium is of medical concern for astronauts during long-term spaceflight missions, as astronaut immune systems are compromised by microgravity. To this end, E. cl

BACKGROUND: As a medical emergency that can affect even well-screened, healthy individuals, peritonitis developing during a long-duration space exploration mission may dictate deviation from traditional clinical practice due to the absence of otherwise indicated surgical capabilities. Medical management can treat many intra-abdominal processes, but treatment failures are inevitable. In these circumstances, percutaneous aspiration under sonographic guidance could provide a "rescue" strategy.
HYPOTHESIS: Sonographically guided percutaneous aspiration of intra-peritoneal fluid can be performed i

BACKGROUND: Astronauts in Earth's orbit experience microgravity, resulting in a decline of skeletal muscle mass and function. On Earth, models simulating microgravity have shown that the extent of the loss in muscle force is greater than the loss in muscle mass. The reasons behind this disproportionate loss of muscle force are still poorly understood. In the present study, we hypothesize that alongside the loss in skeletal muscle mass, modifications in the expression profile of genes encoding critical determinants of resting membrane potential, excitation-contraction coupling and Ca2+ handling

Electrochemically active bacteria (EAB) characterized by extracellular respiration is promising for bioregenerative life support systems and elemental cycling at extraterrestrial planetary bases. However, it remains unclear how microgravity modulates genes related to electricity generation and reshapes intracellular energy metabolism. In this study, the impact of simulated microgravity on EAB was systematically analyzed through electricity generation, gene enrichment, energy metabolism, and central carbon metabolic reprogramming. The results demonstrate that simulated microgravity increases th

Microphysiological systems (MPS) are valuable in vitro models that recreate the structure and function of human tissues. Specifically, tissue- or organ-on-chips that incorporate microfluidic technology replicate the in vivo mechanical and biochemical environment while enabling real-time monitoring of cellular growth and development. These systems can be particularly useful for spaceflight studies, as they can be sent into space, such as to the International Space Station (ISS), to investigate the impact of space travel on the human body. A major impediment to the success of the MPS is the form

Despite exercise countermeasures to sustain health and performance in spaceflight, complete maintenance of muscle mass and functions in microgravity is still not possible for most astronauts. The principal cause of the limited effectiveness of existing exercise countermeasures is the difficulty in achieving full loading forces in space. The implementation of countermeasures which require small devices and simulate Earth-like loading forces to maintain muscle mass, strength and endurance is therefore highly desirable. At present, the cellular mechanisms that induce muscle atrophy in weightlessn

Cryptococcus neoformans is a fungal pathogen that causes cryptococcal meningitis, mainly in immunocompromised individuals, such as those with HIV. Its recent detection on the International Space Station raises concerns about potential health risks in space, where immune systems may be compromised. However, its behavior in space-like conditions remains unclear. In this study, we examined the effects of simulated microgravity on C. neoformans. We found that the condition enhanced the fungus's resistance to membrane and osmotic stress and increased key virulence factors, including capsule formati

Mechanical unloading during head movements in weightlessness may be an etiological factor in space-motion sickness. We simulated altered head loading on Earth without affecting vestibular stimulation by having subjects wear a weighted helmet. Eight subjects were exposed to constant velocity rotation about a vertical axis with direction reversals every 60 sec. for eight reversals with the head loaded and eight with the head unloaded. The severity of motion sickness elicited was significantly higher when the head was loaded. This suggests that altered sensory-motor control of the head is also an
