Laboratory-derived females of Culex quinquefasciatus and Aedes aegypti were experimentally infected with Wuchereria bancrofti by allowing the insects to feed on infected volunteers of known microfilariae density between 24.00 and 2.00 hours. Of the 240 mosquitoes used for the experiments only 67 (55.8%) of C. quinquefasciatus and 83 (69.2%) of the Ae.aegypti actually fed on the volunteers' blood. After 16 days post-infection period, 50 (74.62%) of the engorged C. quinquefasciatus harboured infective L3 larval stages of W. bancrofti, while 16 (23.9%) and 1 (1.49%) harboured L2 and L1 pre-infect

Human lymphatic filariasis (LF) is primarily caused by the helminth parasites Wuchereria bancrofti (Wb) and Brugia malayi (Bm). Detecting parasitic circulating cell-free nucleic acids (ccfDNA/ccfRNA) in plasma is a promising approach for detection of active infections. Plasma RNA sequencing analyses from individuals with active Wb infection and uninfected controls identified 6 RNA targets that were specific to Wb and/or Bm. Assays developed to detect either ccfDNA and ccfRNA were successfully used in Wb-infected individuals, but DNA-based assays targeting WbTR1 were found to be the most sensit

Galectins regulate growth and differentiation of immune cells and inflammation through their carbohydrate-binding function in humans, while also play a role in parasite survival. This study focused on the galectin of lymphatic filarial parasite Wuchereria bancrofti (Wb-Gal). The multiple sequence alignment with other galectins showed that the Wb-Gal belonged to galactoside binding lectin family, particularly tandem repeat type galectin-9. A homology model of Wb-Gal was developed in the I-TASser server using high similarity 3D structures with a quality score of 89.5. Molecular docking and dynam

In 1992, a study on microfilaremia periodicity was carried out on 12 Wuchereria bancrofti carriers in the Marquesas islands. Blood samples were collected simultaneously every 4 hours during a 48 hour period by finger-prick and venipuncture for determination of microfilaremia by both blood film and membrane filtration technique methods, and for determination of antigenemia. The membrane filtration results showed no significant nycthemeral variations between the microfilaria densities at hours 16:00, 20:00, 24:00, 04:00, 08:00 and 12:00. Conversely, the blood film method showed a significant dif

The filarial-specific humoral immune response of adult residents of two areas of Papua New Guinea, differing in transmission of Wuchereria bancrofti infection was compared. The majority of residents of the village of Bonahoi, in an area where transmission of filariasis had been interrupted by a 20-year insecticide spray program to control malaria, showed no parasitologic signs of active W. bancrofti infection and were negative for both circulating phosphorylcholine Ag and peripheral blood microfilariae. In contrast, adult residents of the village of Nanaha were in an area exposed to infection,

Diurnally subperiodic filariasis due to Wuchereria bancrofti has been reported from tribal populations in the Nancowry group of islands in the Andaman and Nicobar Islands, India. This was confirmed in a survey carried out during the monsoon season in 1993. Microfilariae were detected in the peripheral blood throughout a 24 h period with a peak at 18:00. The microfilaria (mf) rate ranged from 1.2% to 18.7%, with a low disease rate (mean = 1.9%). Incidence of mf was low in children less than 10 years old (3.5%), but increased with increasing age. Culex (Culex) quinquefasciatus was present at a v

Although Wuchereria bancrofti (Wb), the causative agent of lymphatic filariasis, is endemic throughout Mali, the prevalence of Wb microfilaremia (Mf) can vary widely between villages despite similar prevalence of infection as assessed by circulating antigen. To examine this variation, cross-sectional data obtained during screening prior to an interventional study in two neighboring villages in Mali were analyzed. The overall prevalence of Wb, as assessed by Wb CAg (circulating antigen), was 50.3% among 373 participants, aged 14-65. Wuchereria bancrofti Mf-positive and negative individuals appe

BACKGROUND: The current antibody tests used for monitoring in lymphatic filariasis (LF) elimination programs suffer from poor specificity because of the considerable geographical overlap with other filarial infections such as Loa loa (Ll), Onchocerca volvulus (Ov), and Mansonella perstans (Mp).
METHODS: Using bioinformatics to assemble into contigs 2048 expressed sequence tags (ESTs) from the L3 infective larvae of W. bancrofti (Wb), these were next assessed for homology to known proteins and nucleotides and to similar assemblies of L3 larval ESTs of B. malayi (Bm - n = 5068), Ov (n = 4166),

BACKGROUND: Lymphatic filarial parasites survive within the lymphatic vessels for years despite the complex immune environment surrounding them. Parasites possibly accomplish this by adopting various immunomodulatory strategies, which include release of glutathione-S-transferases (GSTs) that counteract the oxidative free radicals produced by the host. Since GSTs produced by parasites appear to be critical for the survival of parasites in the host, several studies evaluated the potential of parasite GSTs as vaccine candidates especially against schistosomiasis, fascioliasis and Seteria cervi. BACKGROUND: Anopheles gambiae s.l. and An. funestus are important vectors of malaria and bancroftian filariasis, which occur as co-endemic infections along the Kenyan Coast. However, little is known about the occurrence and prevalence of concomitant infections of the two diseases in mosquito and human populations in these areas. This study reports the prevalence of concomitant infections of Plasmodium falciparum and Wuchereria bancrofti in mosquito and human populations in Jilore and Shakahola villages in Malindi, Kenya.
METHODS: Mosquitoes were sampled inside houses by pyrethrum spray sheet

The relationship between infection and clinical disease in Wuchereria bancrofti infection was investigated in a community-based study in different endemic areas in Ghana. At the individual level, there was no association between acute adenolymphangitis and infection (microfilaraemia) status. There was a negative association between infection status and lymphoedema/elephantiasis, but a positive association with hydrocele; however, the intensity of infection was negatively associated with both elephantiasis and hydrocele. The community prevalence of infection was strongly associated with the pre

The present study was undertaken to assess the relationship between microfilarial clearance and clearance of circulating filarial antigen from the blood of Wuchereria bancrofti-infected persons following chemotherapy with either diethylcarbamazine or ivermectin. Patients received either 12 weekly doses of 6 mg/kg of diethylcarbamazine (DEC), a single dose of 6 mg/kg of DEC, a single dose of 420 microg/kg of ivermectin, or 20 microg/kg of ivermectin, followed by 6 mg/kg of DEC five days later. Microfilarial clearance was marked in all groups, but was significantly less in the single-dose DEC. I

This paper reports the susceptibility of Anopheles anthropophagus (Xu et Feng, 1975) to experimental infection with Wuchereria bancrofti (Cobbold, 1877), comparing with that of An. sinensis Wiedemann, 1828, Culex pipiens quinque fasciatus Say, 1823, and Aedes togoi (Theobald, 1907). Of 188 An. anthropophagus, 202 An. sinensis, 280 Cx. pipiens quinque fasciatus and 129 Ae. togoi infected by the blood of a bancroftian microfilaremia with mff density of 190 mff/20cmm, the infective rates were 35.64%, 9.41%, 30.00% and 65.89%, respectively; while of 188 An. lesteri anthropophagus, 134 An. sinensis

Microfilariae of Wuchereria bancrofti were detected by fine needle aspiration biopsy (FNAB) in an axillary lymph node from a 27-year-old female. The patient also had intestinal tuberculosis. A routine hemogram did not show any eosinophilia, and three consecutive night blood samples also were negative for microfilariae. This was an early case of filariasis without microfilaremia and was detected by FNAB of the lymph node. The association of filariasis and tuberculosis was purely coincidental.

The prevalence of transplacental transfer of microfilariae and structural lesions in the placentas of amicrofilaraemic (n = 10) and Wuchereria bancrofti microfilaraemic (n = 10) pregnant women from the prenatal care unit of the Hospital das Clinicas, Recife, Brazil, was investigated. Study participants were selected by filtration (3 microns/13 mm) of nocturnal (2300-0100 h) venous blood. A detailed histopathological examination was performed to detect abnormalities in the placenta, the cord and the foetal membranes. Both study groups had similar obstetric profiles. Parasitaemia was present dur

To elucidate the local release of immunomodulatory prostaglandins by intravascular filarial parasites, the formation of prostaglandin E2 (PGE2) was examined in individual microfilariae of Wuchereria bancrofti and Brugia malayi. Following incubation of living microfilariae immobilized in an agar matrix, prostaglandins released by the parasites were fixed by carbodiimide and localized by indirect immunofluorescence. Prostaglandin E2 was specifically detected around the entire surface of microfilariae with anti-PGE2 antiserum, but not with control nonimmune or PGE2 affinity-immunoadsorbed antiser

The occurrence of circulating Wuchereria bancrofti microfilariae was investigated in 304 pregnant women from the "Unidade Mista Pof. Barros Lima" and "Hospital das Clínicas/UFPE". The filtration technique was employed to detect microfilaraemia during pregnancy, and 13 women were found to be positive (4.2%). The investigation of microfilariae in the cord blood of their children was negative (6/13), and also in venous blood samples collected up to 72 hours after birth and with 6 months of life. Milk samples from these mothers also had no microfilariae. The authors suggest that the occurrence of
