The γ-proteobacterium Xanthomonas campestris pv. campestris B100 is the causal agent of black rot disease in a wide range of economically important crops. In addition, X. campestris pv. campestris is commercially relevant due to the synthesis of the exopolysaccharide xanthan. In this work, we first introduce a novel transcriptional regulator, termed Crt1, and present the effect that the deletion of the crt1 gene exerts on growth, xanthan production, virulence, and transcriptome and proteome profiles. Differential transcriptome analysis of the deletion mutant X. campestris pv. campestris B100 Δ

UNLABELLED: Bacterial spot of tomato (BST), caused by Xanthomonas species, is a devastating disease in tomato production, leading to severe yield losses and reduced crop quality. Current control strategies, including cultural management and chemical applications, are increasingly challenged by the emergence of copper- and antibiotic-resistant strains. To address this issue, we are exploring the development of novel small molecule antibacterial as a promising alternative control strategy. Through structural expansion and modification of previously identified anti-Xanthomonas leads (X2, X5, and

The GH10 endoxylanase XccXynB from Xanthomonas campestris pv. campestris (Xcc) functions as a PAMP, triggering PTI responses and enhancing disease resistance in plants, in addition to its polysaccharide degradation capability. Plant pathogens of the Xanthomonas genus are capable of infecting a wide array of economically important plant species. Among their virulence factors, the GH10 family xylanase, XynB, is typically responsible for the predominant extracellular xylanase activity. However, the detailed biochemical characteristics of this enzyme in most Xanthomonas pathogens remain insufficie

The Gram-negative bacterium Xanthomonas citri subsp. citri (X. citri) is a phytopathogen causal agent of citrus canker, a severe disease affecting citrus crops worldwide. Disease control relies primarily on copper biocides, which pose toxicity risks to humans and the environment. Endolysins are enzymes that catalyze bacterial cell wall degradation and could serve as environmentally sustainable alternatives for the control of X. citri. Here, we conducted structural in silico studies with the endolysin CP2-07 encoded by the X. citri lytic phage CP2, followed by its heterologous expression and pu

OsICS1 but not OsICS1-L mediates the rice response to Xoo inoculation, with its overexpression increasing resistance against this pathogen. OsICS1 but not OsICS-L is directly upregulated by OsWRKY6. Rice (Oryza sativa) is a staple crop for about half of the global population and is particularly important in the diets of people living in Asia, Latin America, and Africa. This crop is continually threatened by bacterial leaf blight disease caused by Xanthomonas oryzae pv. oryzae (Xoo), which drastically reduces yields; therefore, it is needed to elucidate the plant's resistance mechanisms against

The magnesium chelatase subunit CHLI is well-characterized in chlorophyll biosynthesis, but its role in plant immunity remains unclear. In this study, we generated chimeric Fvchli mutant strawberries via CRISPR/Cas9 and obtained T1-T3 progeny with a wide range of editing efficiencies through self-pollination. Mutants with higher editing efficiency exhibited more leaf spots. Following inoculation with Xanthomonas fragariae, Fvchli mutants showed increased susceptibility, with both lesion area and disease incidence positively correlating with editing efficiency. We further demonstrated that stom

Transcription activator-like effector (TALE) proteins of the plant pathogenic bacterial genus Xanthomonas bind to and transcriptionally activate host susceptibility genes, promoting disease. Plant immune systems have taken advantage of this mechanism by evolving TALE binding sites upstream of resistance (R) genes. For example, the pepper Bs3 and rice Xa27 genes are hypersensitive reaction plant R genes that are transcriptionally activated by corresponding TALEs. Both R genes have a hallmark expression pattern in which their transcripts are detectable only in the presence and not the absence of

Characterization of strains of Xanthomonas axonopodis pv. citri by using DNA fingerprints that were generated from primers for enterobacterial repetitive intergenic consensus (ERIC) elements led to the discovery of differential sequences for a leucine-responsive regulatory protein (lrp) gene in two subgroups of strains with different host ranges on Citrus spp. DNA hybridization and PCR-based assays that used different sets of primers were designed to detect the core sequence, as well as to obtain the entire sequence of the lrp gene for several Xanthomonas species and pathovars. Bacterial leaf spot (BLS) is one of the most destructive diseases of pepper (Capsicum spp.) worldwide, causing significant yield and quality losses under warm and humid conditions. The disease is caused by multiple Xanthomonas species, whose rapid evolution continues to challenge effective management. This review synthesizes current knowledge on BLS in pepper, including pathogen taxonomy, global distribution, and the historical progression of disease research and resistance breeding. We summarize advances in the identification and deployment of resistance loci, including major race-specific ge

BACKGROUND: Xanthomonas oryzae pv. oryzae (Xoo), the causal agent of rice bacterial blight, poses a significant threat to global crop security by means of systemic vascular colonization. In order to address emerging resistance challenges, a rational drug design strategy was employed, which involved structurally optimizing the natural alkaloid berberine (BBR). Through a C-9/C-13 dual-site modification approach, we developed a novel berberine derivative, 9-benzoyloxy-13-(4-(tert-butyl)benzyl)-berberine bromide (HBBR-F4). This compound demonstrated exceptional anti-Xoo efficacy, achieving protect

Xanthomonas translucens, the causal agent of bacterial leaf streak disease (BLS) in cereals, is a re-emerging pathogen that is becoming increasingly destructive across the world. While BLS has caused yield losses in the past, there is anecdotal evidence that newer isolates may be more virulent. We observed that two X. translucens isolates collected from two sites in Colorado, USA, are more aggressive on current wheat and barley varieties compared to older isolates, and we hypothesize that genetic changes between recent and older isolates contribute to the differences in isolate aggressiveness.

SYAUP-491 is a novel alkyl sulfonamide. In this study, in vivo and in vitro tests were performed along with a proteomic analysis to determine the effects and underlying mechanisms of the antibacterial activity of SYAUP-491 against the causative agent of bacterial leaf blight in rice. The antibacterial test results suggested that SYAUP-491 exhibited significant activities against Xanthomonas oryzae pv. oryzae (Xoo) in vitro and in vivo. The minimal EC50 values reached 6.96 μg/mL and the curative activity reached 74.1%. Detailed studies demonstrated that SYAUP-491 altered membrane permeability a

African rice (Oryza glaberrima) was independently domesticated in West Africa around 3000 years ago, and has long been intertwined in the history of the region. Asian rice (Oryza sativa), which was introduced in Africa when European settlers arrived, gradually replaced African rice and has since dominated rice cultivation in the continent. Domesticated rice species are affected by bacterial leaf blight (BLB), which is caused by the pathogen Xanthomonas oryzae pv. oryzae (Xoo). Here we show that the bacterial leaf blight pathogen in Africa (AfXoo) belongs to a distinct phylogroup from the one c

Although several Pseudomonas spp. have recognised biocontrol properties, the ecological and functional potential of Pseudomonas soli remains largely unexplored. Here, the 5.6 Mb genome of the strain VMAP1 was sequenced and annotated to confirm its taxonomic identity and investigate genes associated with biocontrol traits. Genome mining and targeted in vitro and in planta assays were combined to assess its biocontrol potential. The genome harbours genes linked to the synthesis of bioactive compounds, as well as to regulatory and secretion systems homologous to those involved in biocontrol in ot

Indole-3-acetic acid (IAA) is an important plant hormone that regulates a variety of physiological processes in plants, and it is also produced by some microbes. However, the biosynthesis and roles of IAA in microorganisms, particularly in plant pathogens, remain to be determined. In this study, the plant pathogen Xanthomonas campestris pv. campestris (Xcc) strain XC1 was shown to produce IAA via an l-tryptophan ( l-Trp)-dependent pathway. The intermediate metabolite indole-3-ethanol and Xcc1569 encoding aromatic amino acid aminotransferase were shown to be partially involved in the uncharact

Phenazine-1-carboxylic acid (PCA), a member of phenazines secreted by microorganisms, inhibits the growth of many bacteria and fungi. Xanthomonas campestris pv. campestris is the causal agent of black rot, the most important disease of cruciferous crops worldwide, and is more tolerant to PCA than other Xanthomonas species. Previous studies reported that reactive oxygen species (ROS) scavenging ability is involved in regulating the PCA tolerance of Xanthomonas species. Additionally, the cytochrome c maturation (CCM) system has been found to play a more important role in tolerance to phenazines

XopDXcc8004, a type III effector of Xanthomonas campestris pv. campestris (Xcc) 8004, is considered a shorter version of the XopD, which lacks the N-terminal domain. To understand the functions of XopDXcc8004, in planta, a transgenic approach combined with inducible promoter to analyze the effects of XopDXcc8004 in Arabidopsis was done. Here, the expression of XopDXcc8004, in Arabidopsis elicited the accumulation of host defense-response genes. These molecular changes were dependent on salicylic acid and correlated with lesion-mimic phenotypes observed in XVE::XopDXcc8004 transgenic plants. Mo

In Xanthomonas campestris pv. campestris (Xcc), which is the causative agent of black rot in crucifers, the virulence factor level is substantially decreased in the mutant deficient in RpfG, a phosphodiesterase that degrades the second messenger cyclic di-GMP. The rpfG mutant also grew in an aggregated state. It is indicated that expression of Pseudomonas GGDEF domain protein WspR (a diguanylate cyclase that synthesizes cyclic di-GMP) in wild-type Xcc can produce a phenocopy of the rpfG mutant. In this study, we showed that over-expression of GGDEF domain protein XCC2731 in wild-type Xcc cause

Exopolysaccharides (EPS) represent an important tool for application in bio- and phytoremediation technologies due to their ability to enhance water and nutrient retention, support microclimate stability, and protect plants from environmental stress. In the present study, xanthan-like EPS produced by Xanthomonas translucens TRK8 was precipitated by ethanol and isopropanol, with the former yielding 9.2 g L-1 compared with 6.7 g L-1 obtained with the latter. The monosaccharide profile of the TRK8-derived EPS indicated a branched structure composed of rhamnose, mannose, glucose, and galactose res

Heroin is a highly addictive drug that exerts its primary effects through activation of μ-opioid receptors. Its principal active metabolite, 6-monoacetylmorphine (6-MAM), significantly contributes to heroin's neurological effects and acute toxicity. Current pharmacotherapies for heroin use disorder, employing opioid receptor agonist or antagonist, are often limited by risks of dependence, tolerance, and/or adverse side effects. In this context, enzyme-based therapy emerges as a promising alternative by rapidly converting drugs into inactive or less harmful metabolites in the blood. As a macrom
