[Effects of combined CaO-CO2 detoxification treatment on the conversion of sugarcane bagasse enzymatic hydrolysate to xanthan gum].

Replacing grain-derived carbon sources with non-grain biomass carbon sources for fermentation is a core research direction in the field of biomanufacturing. However, inhibitory byproducts present in the enzymatic hydrolysate of non-grain biomass severely restrict its practical application. We aim to reduce the dependence of xanthan gum fermentation on grain-based carbon sources, clarify the impacts of typical inhibitors from non-grain biomass enzymatic hydrolysate on xanthan gum synthesis, break through the existing application bottlenecks, and develop a green, low-loss and high-efficiency det

Repurposing onion peel waste as a cost-effective and sustainable substrate for xanthan production.

This study investigates the potential of onion peel waste as a sustainable and cost-effective alternative substrate for xanthan gum production using Xanthomonas campestris NCIM 2961. The results demonstrate that onion peel-based medium consistently produced higher xanthan gum yields compared to the standard medium, achieving a maximum yield of 5.21 g L-1 at optimal conditions (pH 7.0, 30°C, 100 rpm agitation, and a 48 h incubation period), without supplements or pretreatments. FT-IR confirmed the chemical identity and purity of xanthan gum derived from the onion peel-based medium, closely matc

Effect of Chain Length on the Antimicrobial Properties of Dialkylresorcinols.

Dialkylresorcinols (DARs) are naturally occurring compounds produced in several species of the Photorhabdus and Pseudomonas bacterial genera, which are involved in modulating quorum-sensing in the producing organisms, as well as possessing significant antimicrobial properties. A range of dialkylresorcinols were synthesized, systematically varying the chain-lengths of the C-2 and C-5 alkyl chains, and the antimicrobial properties of the products assessed. Against two Staphylococcus test strains, medium-length alkyl chains (5-7 carbons) in both positions proved optimum for high antibiotic activi

Root zone warming represses foliar diseases in tomato by inducing systemic immunity.

Plants employ systemic-induced resistance as part of their defence arsenal against pathogens. In recent years, the application of mild heating has been found to induce resistance against several pathogens. In the present study, we investigated the effect of root zone warming (RZW) in promoting tomato's resistance against the necrotrophic fungus Botrytis cinerea (Bc), the hemibiotrophic bacterium Xanthomonas campestris pv. vesicatoria (Xcv) and the biotrophic fungus Oidium neolycopersici (On). We demonstrate that RZW enhances tomato's resistance to Bc, On and Xcv through a process that is depen

Antibacterial Radicicol Analogues from Pochonia chlamydosporia and Their Biosynthetic Gene Cluster.

Chemical investigation of fungus Pochonia chlamydosporia strain 170, derived from rice fermentation sediment samples, afforded seven radicicol analogues, including two new compounds, monocillin VI (1) and monocillin VII (2), and five known compounds, monocillin II (3), monorden D (4), monocillin IV (5), monocillin V (6), and pochonin M (7). The structures of compounds 1-7 were established primarily by analysis of nuclear magnetic resonance data, and the absolute configurations of the secondary alcohol in compounds 1 and 2 were assigned by the modified Mosher method. All seven compounds have mo

The structure and inhibition of a GGDEF diguanylate cyclase complexed with (c-di-GMP)(2) at the active site.

Cyclic diguanosine monophosphate (c-di-GMP) is a key signalling molecule involved in regulating many important biological functions in bacteria. The synthesis of c-di-GMP is catalyzed by the GGDEF-domain-containing diguanylate cyclase (DGC), the activity of which is regulated by the binding of product at the allosteric inhibitory (I) site. However, a significant number of GGDEF domains lack the RxxD motif characteristic of the allosteric I site. Here, the structure of XCC4471(GGDEF), the GGDEF domain of a DGC from Xanthomonas campestris, in complex with c-di-GMP has been solved. Unexpectedly,

New perspectives of purple starthistle (Centaurea calcitrapa) leaf extracts: phytochemical analysis, cytotoxicity and antimicrobial activity.

Ethnobotanical and ethnopharmacological studies of many Centaurea species indicated their potential in folk medicine so far. However, investigations of different Centaurea calcitrapa L. extracts in terms of cytotoxicity and antimicrobial activity against phytopathogens are generally scarce. The phenolic profile and broad antimicrobial activity (especially towards bacterial phytopathogens) of methanol (MeOH), 70% ethanol (EtOH), ethyl-acetate (EtOAc), 50% acetone (Me2CO) and dichloromethane: methanol (DCM: MeOH, 1: 1) extracts of C. calcitrapa leaves and their potential toxicity on MRC-5 cell l

Differential effectiveness of salicylate-dependent and jasmonate/ethylene-dependent induced resistance in Arabidopsis.

Salicylic acid (SA), jasmonic acid (JA), and ethylene (ET) are each involved in the regulation of basal resistance against different pathogens. These three signals play important roles in induced resistance as well. SA is a key regulator of pathogen-induced systemic acquired resistance (SAR), whereas JA and ET are required for rhizobacteria-mediated induced systemic resistance (ISR). Both types of induced resistance are effective against a broad spectrum of pathogens. In this study, we compared the spectrum of effectiveness of SAR and ISR using an oomycete, a fungal, a bacterial, and a viral p

One pot synthesis of N-7 alkylated xanthine derivatives and their exploration as antioxidant, antimicrobial and nematicidal agents.

The widespread presence of economically dominant pathogens has significantly hindered agricultural growth and productivity by affecting the quality and yield of crops. The recent deregistration of essential bactericides and nematicides have underscored the urgency for the development of novel agrochemical candidates. In response, N-7 alkylated xanthine derivatives were synthesized using various alkylbromides and characterized with the help of various spectroscopic techniques. The synthesized derivatives were explored for antioxidant, antibacterial, antifungal and antinemic properties. A compre

Pepper heat shock protein 70a interacts with the type III effector AvrBsT and triggers plant cell death and immunity.

Heat shock proteins (HSPs) function as molecular chaperones and are essential for the maintenance and/or restoration of protein homeostasis. The genus Xanthomonas type III effector protein AvrBsT induces hypersensitive cell death in pepper (Capsicum annuum). Here, we report the identification of the pepper CaHSP70a as an AvrBsT-interacting protein. Bimolecular fluorescence complementation and coimmunoprecipitation assays confirm the specific interaction between CaHSP70a and AvrBsT in planta. The CaHSP70a peptide-binding domain is essential for its interaction with AvrBsT. Heat stress (37°C) an

Development of a xanthan gum film for the possible treatment of vaginal infections.

Bacterial vaginosis is a vaginal infection that affects 60% of women of reproductive age worldwide. It is mainly caused by the bacterium Gardnerella vaginalis and is a factor that increases the probability of getting sexually transmitted diseases. We aimed to develop a new pharmaceutical form for the treatment of vaginal infections. We employed the solving-casting method to fabricate a polymeric film with Xanthan gum, a natural polymer produced by the bacterium Xanthomonas campestris, and metronidazole, one of the most commonly used drugs for vaginal infections. In order to characterize the fi

Expanding Role of Type II Secretion in Bacterial Pathogenesis and Beyond.

Type II secretion (T2S) is one means by which Gram-negative pathogens secrete proteins into the extracellular milieu and/or host organisms. Based upon recent genome sequencing, it is clear that T2S is largely restricted to the Proteobacteria, occurring in many, but not all, genera in the Alphaproteobacteria, Betaproteobacteria, Gammaproteobacteria, and Deltaproteobacteria classes. Prominent human and/or animal pathogens that express a T2S system(s) include Acinetobacter baumannii, Burkholderia pseudomallei, Chlamydia trachomatis, Escherichia coli, Klebsiella pneumoniae, Legionella pneumophila,

cDNA-AFLP analysis of differential gene expression in the prokaryotic plant pathogen Erwinia carotovora.

For studies of differential gene expression in prokaryotes, methods for synthesizing representative cDNA populations are required. Here, a technique is described for the synthesis of cDNA from the potato pathogens Erwinia carotovora subsp. atroseptica (Eca) and Erwinia carotovora subsp. carotovora (Ecc) using a combination of short oligonucleotide (11-mer) primers that were known to anneal to conserved sequences in the 3' regions of enterobacterial genes. Specific PCR amplifications with primers designed to anneal to 14 known genes from either Eca or Ecc revealed the presence of the correspond

Two-Component Signaling System VgrRS Directly Senses Extracytoplasmic and Intracellular Iron to Control Bacterial Adaptation under Iron Depleted Stress.

Both iron starvation and excess are detrimental to cellular life, especially for animal and plant pathogens since they always live in iron-limited environments produced by host immune responses. However, how organisms sense and respond to iron is incompletely understood. Herein, we reveal that in the phytopathogenic bacterium Xanthomonas campestris pv. campestris, VgrS (also named ColS) is a membrane-bound receptor histidine kinase that senses extracytoplasmic iron limitation in the periplasm, while its cognate response regulator, VgrR (ColR), detects intracellular iron excess. Under iron-depl

Herbivore-induced resistance against microbial pathogens in Arabidopsis.

Caterpillars of the herbivore Pieris rapae stimulate the production of jasmonic acid (JA) and ethylene (ET) in Arabidopsis (Arabidopsis thaliana) and trigger a defense response that affects insect performance on systemic tissues. To investigate the spectrum of effectiveness of P. rapae-induced resistance, we examined the level of resistance against different pathogens. Although the necrotrophic fungus Alternaria brassicicola is sensitive to JA-dependent defenses, herbivore-induced resistance was not effective against this pathogen. By contrast, caterpillar feeding significantly reduced disease

Constitutive Metabolomic Profiles Linked to Seasonal Climate Niche Adaptation and Resilience to Black Rot in Cabbage.

Plants are dynamic chemical factories that produce diverse specialised metabolites essential for growth, defence, and environmental adaptation. Metabolomes, shaped by genetic architecture, enable plants to respond to abiotic and biotic pressures, including temperature fluctuations. In Brassica systems, warm and humid conditions favour black rot caused by Xanthomonas campestris pv. campestris (Xcc), whereas cooler temperatures reduce disease pressure, thus influencing seasonal cultivar deployment. In this study, we investigated whether the constitutive metabolomes of two commercially cultivated

Synthesis, Spectroscopic, and Biological Studies of Mixed Ligand Complexes of Gemifloxacin and Glycine with Zn(II), Sn(II), and Ce(III).

Three novel mixed ligand metal complexes have been synthesized by the reaction of Zn(II), Sn(II), and Ce(III) with gemifloxacin (GMFX) in the presence of glycine (Gly) (1:1:1 molar ratio). The coordination possibility of the two ligands toward metal ions has been proposed in the light of elemental analysis, molar conductance, spectral infrared (IR), ultraviolet-visible (UV-Vis) and proton-nuclear magnetic resonance (¹H NMR), and magnetic studies. Results suggest that GMFX and Gly interact with the metal ions as bidentate ligands. Electronic and magnetic data proposed the octahedral structure f
