Photoprotection mechanism in the 'Fuji' apple peel at different levels of photooxidative sunburn.

The xanthophyll cycle, flavonoid metabolism, the antioxidant system and the production of active oxygen species were analyzed in the peel of 'Fuji' apples re-exposed to sunlight after extended periods of fruit bagging treatment, resulting in different levels of photooxidative sunburn. After re-exposing bagged fruits to sunlight, the production of active oxygen species and the photoprotective capacity in apple peels were both significantly enhanced. As sunburn severity increased, the concentration of hydrogen peroxide increased, while xanthophyll cycle pool size decreased. For the key genes inv

The effect of cactus spines on light interception and Photosystem II for three sympatric species of Opuntia from the Mojave Desert.

Cactus spines reduce herbivory, direct water toward roots and reduce the impacts of high- and low-temperature extremes. Yet, shading of stems by spines reduces incident photosynthetic photon flux density (PFD), photosynthesis and growth. This study compared spinescence, PFD interception, stem temperature, Photosystem II (PSII) photochemistry and xanthophyll pigment composition for three species of cacti from the Mojave Desert, CA. The species vary in spinescence: Opuntia basilaris, which has no central or radial spines; Opuntia erinacea, which is densely covered with spines; and Opuntia phaeac

How do Mediterranean shrub species cope with shade? Ecophysiological response to different light intensities.

Under natural conditions, light exposure for Mediterranean shrubs can be highly variable, especially during cloudy days or under a canopy, and can interfere with other environmental factors such as temperature and water availability. With the aim of decoupling the effect of radiation and temperature from water availability, we conducted an experiment where two perennial and three summer semi-deciduous shrub species were subjected to different levels of irradiation. In order to follow plant responses to light exposure, we measured gas exchange, photosystem II photochemical efficiency, photosynt

Slowly reversible de-epoxidation of lutein-epoxide in deep shade leaves of a tropical tree legume may 'lock-in' lutein-based photoprotection during acclimation to strong light.

The kinetics of response to strong light have been examined in deeply shaded leaves of the tropical tree legume (Inga sp.) which have extraordinarily high levels of the alpha-xanthophyll lutein-epoxide that are co-located in pigment-protein complexes of the photosynthetic apparatus with the beta-xanthophyll violaxanthin. As in other species, rapidly reversible photoprotection (measured as non-photochemical chlorophyll fluorescence quenching) is initiated within the time frame of sun-flecks (minutes), before detectable conversion of violaxanthin to antheraxanthin or zeaxanthin. Photoprotection

Blood plasma investigations by resonance Raman spectroscopy: detection of carotenoid pigments.

Using Raman spectroscopy, we demonstrate that low levels of beta-carotene, lycopene, and xanthophyll give rise to resonance enhanced bands in blood plasma. These results explain the significance of previously unidentified spectral maxima which have been related to the state of health of the blood donor.

Regional photoprotection strategies in Indian Ocean phytoplankton: Arabian Sea vs South Tropical Indian Ocean responses to PAR/UVR stress.

Phytoplankton photoprotection is a key yet understudied determinant of marine ecosystem resilience to climate-driven environmental stressors, particularly in the Indian Ocean (IO), where observational constraints limit projections of productivity under warming and stratification. While photophysiological adaptation has been extensively characterised in Atlantic and Pacific systems, IO phytoplankton communities remain poorly resolved. We assessed the photophysiological responses of natural phytoplankton assemblages from two contrasting IO bioregions-the productive, monsoon-influenced Arabian Se

The protective functions of carotenoid and flavonoid pigments against excess visible radiation at chilling temperature investigated in Arabidopsis npq and tt mutants.

The npq1 mutant of Arabidopsis thaliana (L.) Heynh. has no xanthophyll cycle due to a lack of functional violaxanthin de-epoxidase. Short-term exposure (<2 days) of detached leaves or whole plants to the combination of high photon flux density (1,000 micromol m(-2) s(-1)) and low temperature (10 degrees C) resulted in PSII photoinhibition which was more acute in npq1 than in the wild type. This increased photosensitivity of npql at chilling temperature was attributable to the inhibition of nonphotochemical energy quenching (NPQ) and not to the absence of zeaxanthin itself. In contrast to PSII,

Relaxation of the non-photochemical chlorophyll fluorescence quenching in diatoms: kinetics, components and mechanisms.

Diatoms are especially important microorganisms because they constitute the larger group of microalgae. To survive the constant variations of the light environment, diatoms have developed mechanisms aiming at the dissipation of excess energy, such as the xanthophyll cycle and the non-photochemical chlorophyll (Chl) fluorescence quenching. This contribution is dedicated to the relaxation of the latter process when the adverse conditions cease. An original nonlinear regression analysis of the relaxation of non-photochemical Chl fluorescence quenching, qN, in diatoms is presented. It was used to

Light stress is not effective to enhanced crassulacean acid metabolism.

Clusia minor L., a C3-CAM intermediate, and Clusia multiflora H. B. K., a C3 obligate, present two physiotypes of a similar morphotype occurring sympatrically in the field. Both species, exposed 2 days to high light, show similar responses to this kind of stress: (i) the level of xanthophyll pigments in tested plants during the daycourse adapts to stress, (ii) the levels of antheraxanthin and zeaxanthin clearly increase during the afternoon showing increased de-epoxidation, (iii) the changes in the xanthophyll cycle are similar. Exposure to high light increases the malate levels in C. minor du

Plasticity of photoprotective mechanisms of Buxus sempervirens L. leaves in response to extreme temperatures.

Summer 2003 was unusually hot in Western Europe, with local droughts and an intense heatwave, that led to a massive damage in vegetation. Since high temperatures are supposed to generate photooxidative stress, we analysed photoprotective responses in leaves of the evergreen boxtree (Buxus sempervirens L.) during summer 2003. All the photoprotective compounds analysed (alpha-tocopherol, beta-carotene, and xanthophylls cycle pigments) were simultaneously induced in parallel with a reduction in photochemical efficiency (Fv/Fm). To characterise these responses, we compared these data with other da

Antioxidants Differentially Regulate Non-Photochemical Quenching.

Non-photochemical quenching (NPQ) dissipates excess light energy as heat, limiting reactive oxygen species production during photosynthesis. Exogenous antioxidants are widely reported to improve plant performance under stress, yet their impacts on NPQ are inconsistent across studies, complicating mechanistic interpretation. Here, we systematically compared six antioxidants - ascorbate, chitosan, coenzyme M, reduced glutathione, melatonin, and N-acetyl cysteine - applied to Arabidopsis thaliana seedlings using individual concentration gradients, under standardised pH and illumination regimes. A genetic dissection of intestinal fat-soluble vitamin and carotenoid absorption.

Carotenoids are currently investigated regarding their potential to lower the risk of chronic disease and to combat vitamin A deficiency. Surprisingly, responses to dietary supplementation with these compounds are quite variable between individuals. Genome-wide studies have associated common genetic polymorphisms in the BCO1 gene with this variability. The BCO1 gene encodes an enzyme that is expressed in the intestine and converts provitamin A carotenoids to vitamin A-aldehyde. However, it is not clear how this enzyme can impact the bioavailability and metabolism of other carotenoids such as x

Zeaxanthin deficiency enhances the high light sensitivity of an ascorbate-deficient mutant of Arabidopsis.

The ascorbate content of plants is usually increased in high light (HL), implying a function for ascorbate in the acclimation of plants to HL. Nevertheless, the importance of ascorbate in HL acclimation has not yet been tested directly. Here, we report on the acclimation process of an ascorbate-deficient Arabidopsis mutant to HL. The mutant vtc2 has only 10% to 30% of wild-type levels of ascorbate, and it is also slightly deficient in feedback de-excitation (qE), a photoprotective mechanism that causes the dissipation of excess light as heat. The vtc2 mutant was unable to acclimate to HL, when

Fucoxanthin Bioproduction from Marine Diatoms: Regulatory Strategies and Industrial Potential of Phaeodactylum tricornutum and Odontella aurita-A Review.

Fucoxanthin is a high-value marine xanthophyll with a unique epoxy-allene structure, predominantly produced by brown macroalgae and marine diatoms, and additionally reported in other heterokont lineages including chrysophytes and certain haptophytes. As a core light-harvesting pigment, it exhibits multiple bioactivities including lipid-lowering, antioxidant, and anti-inflammatory effects, with broad applications in functional foods, dietary supplements, and pharmaceuticals. Currently, over 70% of commercial fucoxanthin is extracted from brown macroalgae such as Saccharina japonica and Undaria

Papaya (Carica papaya L.) Peel as a Sustainable Source of Vision-Related Carotenoids Through Green Extraction Optimization.

Agro-industrial by-products represent a sustainable and underutilized source of bioactive compounds with potential applications in human health. Among them, papaya (Carica papaya L.) peel, typically discarded during industrial processing, constitutes a promising and underexploited matrix for carotenoid recovery and valorization. In this study, different extraction strategies were evaluated and compared to identify the most efficient approach for carotenoid recovery. Total carotenoid contents of 7.13 ± 0.32, 5.35 ± 0.24, and 4.44 ± 0.38 µg β-carotene/100 g dry weight (DW) were obtained, respect

Lutein and Zeaxanthin and Their Roles in Age-Related Macular Degeneration-Neurodegenerative Disease.

Lutein and zeaxanthin belong to the xanthophyll family of carotenoids, which are pigments produced by plants. Structurally, they are very similar, differing only slightly in the arrangement of atoms. Key sources of these carotenoids include kale, savoy cabbage, spinach, broccoli, peas, parsley, corn, and egg yolks. The recommended daily intake of lutein is approximately 10.0 mg and that of zeaxanthin is 2 mg. Lutein intake in adults varies, with average intakes being 1-2 mg/day. Due to the lack of synthesis of consumption of these compounds in humans, these substances are extremely important f

Effects of high temperature coupled with high light on the balance between photooxidation and photoprotection in the sun-exposed peel of apple.

The sun-exposed peel of 'Gala' apple with or without sunburn was compared in terms of photooxidation and photoprotection, and a controlled experiment was conducted to probe the initial responses of PSII to high light and high temperature. The content of carotenoids, lutein and xanthophylls on a chlorophyll basis was higher in the sunburned peel although they were lower expressed on a peel area basis. Significant loss of beta-carotene and neoxanthin was observed relative to chlorophylls in the sunburned peel. O(2) evolution rates and the activity of key enzymes in the Calvin cycle were lower in

Peanut violaxanthin de-epoxidase alleviates the sensitivity of PSII photoinhibition to heat and high irradiance stress in transgenic tobacco.

This is the first study on peanut VDE, which led to multiple biochemical and physiological changes to heat and HI stress by improving de-epoxidation of the xanthophylls cycle. A peanut (Arachis hypogaea L.) violaxanthin de-epoxidase gene (AhVDE) was isolated by RT-PCR and RACE methods. The deduced amino acid sequence of AhVDE showed high identities with violaxanthin de-epoxidase of other plant species. The expression of AhVDE was obviously upregulated by 4, 40 °C and high light, NaCl, and abscisic acid. Sense and RNAi transgenic tobaccos were further used to investigate the physiological effec
