Enhanced α-ketoglutaric acid production and recovery in Yarrowia lipolytica yeast by effective pH controlling.

The replacement of chemical synthesis by environmentally friendly energy-efficient technologies for production of valuable metabolites is a principal strategy of developing biotechnological industry all over the world. In the present study, we develop a method for α-ketoglutaric acid (KGA) production from rapeseed oil with the use of Yarrowia lipolytica yeast. Sixty strains of Y. lipolytica yeasts were tested for their ability to produce KGA, and the strain Y. lipolytica 212 (Y. lipolytica VKM Y-2412) was selected as a promising KGA producer. Using a three-stage pH controlling, in which pH was

The Effect of Plasma-Treated Water on Microbial Growth and Biosynthesis of Gamma-Decalactones by Yarrowia lipolytica Yeast.

In recent years, the production of plasma-treated water (PTW) by low-temperature low-pressure glow plasma (LPGP) has been increasingly gaining in popularity. LPGP-treated water changes its physical and physiochemical properties compared to standard distilled water. In this study, a non-conventional lipolytic yeast species Yarrowia lipolytica was cultivated in culture media based on Nantes plasma water with heightened singlet oxygen content (Nantes PW) or in water treated with low-temperature, low-pressure glow plasma while in contact with air (PWTA) or nitrogen (PWTN). The research aimed to as

First Isolation of Yarrowia lipolytica in a Granulomatous Pneumonia of a Spectacled Caiman, Caiman crocodilus Linnaeus, 1758.

Contrary to humans, candidiasis is a rare infection in animals. However, in reptiles, candidiasis can cause gastrointestinal, cutaneous, or rarely systemic infections in stressed animals. The infections due to Yarrowia lipolytica have been increasingly described in human medicine, and hundreds of cases are reported, comprised of granulomatous lung lesions. Herein, granulomatous pneumonia of a spectacled caiman, Caiman crocodilus, was described, and the presence of Y. lipolytica in the lesion was confirmed through histopathology, microbiologic cultures, and molecular methods. The cause of death

Yarrowia lipolytica NCIM 3589, a tropical marine yeast, degrades bromoalkanes by an initial hydrolytic dehalogenation step.

The widespread industrial use of organobromines which are known persistent organic pollutants has led to their accumulation in sediments and water bodies causing harm to animals and humans. While degradation of organochlorines by bacteria is well documented, information regarding degradation pathways of these recalcitrant organobromines is scarce. Hence, their fates and effects on the environment are of concern. The present study shows that a tropical marine yeast, Yarrowia lipolytica NCIM 3589 aerobically degrades bromoalkanes differing in carbon chain length and position of halogen substitut

Waste Sunflower Oil as a Feedstock for Efficient Single-Cell Oil and Biomass Production by Yarrowia lipolytica.

In this study, single-cell oil (SCO) production from waste sunflower oil was optimized using Yarrowia lipolytica IFP29 (ATCC 20460). Optimizations were performed via a multi-response approach based on the Taguchi orthogonal array design (L16), targeting maximum biomass concentration and lipid content (based on dry cell weight). A total of 16 experimental conditions were tested with five key parameters: nitrogen concentration (0, 1, 2, and 4 g/L), WCO concentration (20, 40, 60, and 80 g/L), Tween 80 content (0, 0.5, 1, and 2%) as well as the application of sonication and sterilization. Carbon Dioxide Fixation Cascade for the Production of Single-Cell Protein by Clostridium ljungdahlii and Yarrowia lipolytica.

Clostridium species and yeast single-cell protein (SCP), produced via industrial fermentation, offer a sustainable, high-protein alternative to conventional sources while advancing circular economy goals through CO2 valorization. Here, we developed an integrated two-stage bioprocess for SCP production from CO2. First, Clostridium ljungdahlii converted CO2 and H2 into SCP and acetate via gas fermentation. Optimizing nitrogen and sulfur feeding based on acetate synthesis rate increased acetate titer from 6 to 53.2 g/L and biomass from 1.1 to 2.8 (OD600). The fermentation broth was then directly

Mitochondrial and plasma‑membrane transport‑based engineering enables industrial‑level isocitric acid production in Yarrowia lipolytica.

BACKGROUND: Isocitric acid (ICA) has recently attracted increasing interest because of its nutraceutical relevance and emerging biomedical potential. However, despite its value, industrial exploitation of ICA is still limited owing to the lack of cost‑effective and selective microbial production processes. Mitochondrial transporters play a pivotal role in controlling the pool sizes and fluxes of tricarboxylic acid (TCA) cycle intermediates by mediating their selective exchange across the inner mitochondrial membrane. Recently, we identified the mitochondrial carriers YlYhm2 and YlSfc1 in Yarro

Systematic Metabolic Engineering of Oleaginous Yeast Yarrowia lipolytica for Efficient Synthesis of Polyketide 6-Methylsalicylic Acid.

The polyketides 6-methylsalicylic acid (6-MSA) exhibits good resistance to plant pathogens and considerable application potential. However, conventional methods, such as plant extraction, have limited its industrialization and green synthesis. Microbial synthesis of 6-MSA is a novel and efficient preparation method. In this study, Yarrowia lipolytica was engineered by introducing the 6-MSA synthase gene (6-MSAS) and its activating partner 4'-phosphopantetheinyl transferase, along with the enhancement of precursor synthesis and substrate utilization genes, combined with genome multicopy integra

Interaction of Yarrowia lipolytica lipase with dithiocarbamate modified magnetic carbon Fe3O4@C-NHCS2H core-shell nanoparticles.

Fe3O4@C core-shell nanoparticles were modified by (3-aminopropyl)triethoxysilane (APTES) to generate amine functionality in the surface. Then, the amine functional groups were converted to dithiocarbamate via post-modification with carbon disulfide. This nanostructure with new functional property was used to immobilize lipase (obtained from Yarrowia lipolytica U6). Biocatalytic activity of the Fe3O4@C-NHCS-LIP was studied in this project. The interaction of lipase and support though dithiocarbamate binder was examined in the hydrolysis of p-nitrophenyl laurate. In this paper, support showed a

Engineered yeast Yarrowia lipolytica as a chassis for biosynthesis of fatty acids from mannitol and macroalgal biomass extracts.

BACKGROUND: Yarrowia lipolytica possesses the capability to utilize many unconventional carbon sources, such as crude glycerol, alkanes and fatty acids. Despite producing polyols, such as erythritol, arabitol and mannitol, the re-utilization of mannitol is not as efficient as erythritol utilization. Genes involved in mannitol uptake and metabolism in Y. lipolytica remain undescribed. However, deletion of the EYD1 gene (YALI0F01650g), believed to encode erythritol dehydrogenase, has been found to result in a high rate of growth on media containing mannitol as the sole carbon source. Therefore t

Impact of Ethanol Stress on Yarrowia lipolytica for Sustainable Bioconversion of Agro-Food Oil Wastes into Lipases and Lipids.

Ethanol stress profoundly affects yeast metabolism, yet its integrated impact on lipase activity and lipid remodeling in Yarrowia lipolytica remains unexplored. Here, we investigated, for the first time, the combined effects of ethanol-induced stress on lipase production and fatty acid profiles in Y. lipolytica cultivated on two hydrophobic substrates: olive mill wastewater (OMW) and Waste Frying Oil (WFO). Ethanol was applied at increasing concentrations (3%, 5%, and 7% v/v), and the physiological responses were monitored over time (48, 72, and 96 h). Our results reveal a substrate-dependent

Biotransformation of ethylene glycol to glycolic acid by Yarrowia lipolytica: A route for poly(ethylene terephthalate) (PET) upcycling.

Biological recycling of PET waste has been extensively investigated recently to tackle plastic waste pollution, and ethylene glycol (EG) is one of the main building blocks recovered from this process. Wild-type Yarrowia lipolytica IMUFRJ 50682 can be a biocatalyst to biodepolymerize PET. Herein, we report its ability to perform oxidative biotransformation of EG into glycolic acid (GA): a higher value-added chemical with varied industrial applications. We found that this yeast tolerates high EG concentrations (up to 2 M) based on maximum non-inhibitory concentration (MNIC) tests. Whole-cell bio

Safety evaluation of the food additive steviol glycosides, predominantly Rebaudioside M, produced by fermentation using Yarrowia lipolyticaVRM.

The EFSA Panel on Food Additive and Flavourings (FAF Panel) provides a scientific opinion on the safety of a new process to produce steviol glycosides by fermentation of simple sugars using a genetically modified strain of Yarrowia lipolytica (named Y. lipolytica VRM). The manufacturing process may result in impurities different from those that may be present in the other steviol glycosides E 960a-d, therefore the Panel concluded that separate specifications are required for the food additive produced as described in the current application. Viable cells and DNA from the production strain are

Yarrowia lipolytica L-asparaginase inhibits the growth and migration of lung (A549) and breast (MCF7) cancer cells.

L-asparaginase is an enzyme capable of hydrolyzing the asparagine to aspartic acid and ammonia. L-asparaginase is widely used in the treatment of acute lymphoblastic leukemia (ALL) and other cancers. Here, for the first time, the effects of a novel yeast L-asparaginase from Yarrowia lipolytica were studied on human lung (A549) and breast cancer (MCF7) cell lines as the solid cancer cell lines in terms of cell growth and metastasis inhibition. Functional analysis showed the L-asparagine deprivation mediated anti-proliferation effects by apoptosis induction and changes in the expression of targe

Optimization of medium composition and fermentation conditions for α-ketoglutaric acid production from biodiesel waste by Yarrowia lipolytica.

This work demonstrates the ability of the yeast Yarrowia lipolytica cultivated on biodiesel waste to synthesize α-ketoglutaric acid with a minimal content of pyruvic acid as the main byproduct. The key factor promoting the microbial production of α-ketoglutaric acid from the waste is a strong deficiency of thiamine in the cultivation medium. The production of α-ketoglutaric acid by the yeast can be regulated by changing the concentration of nitrogen, iron, zinc, copper, and manganese in the medium, as well as by pH medium and the aeration rate. The optimization of these parameters in flask exp

GlcNAc induces GlcNAc catabolic genes and inhibits filamentation via YlRep1-YlNgs1 signaling in the dimorphic yeast Yarrowia lipolytica.

Many fungal species can utilize N-acetylglucosamine (GlcNAc) as a carbon source. Studies in the pathogenic yeast Candida albicans have revealed that GlcNAc utilization and the induction of GlcNAc catabolic genes depend on the Ndt80 family transcription factor CaRep1 and the histone acetyltransferase CaNgs1. Additionally, GlcNAc induces filamentation via both alkalinization of the medium and CaNgs1 signaling. However, the roles of YlRep1 and YlNgs1 in GlcNAc catabolism and filamentous growth are not clear in the dimorphic yeast Yarrowia lipolytica. In this study, we demonstrate that YlRep1 and

Harnessing Solar Energy for the Photocatalytic Reduction of Hexavalent Chromium: A High-Performance Yarrowia lipolytica-CdS Biohybrid System.

Photosynthetic semiconductor biohybrids, which combine the light-harvesting capacity of semiconductors and catalytic activity of whole-cell microorganisms, show substantial potential for advancing bioremediation technology. However, few yeast-based biohybrid systems for pollutant removal were reported. In this study, we have constructed a whole-cell biohybrid system based on Yarrowia lipolytica featuring in situ synthesized biocompatible cadmium sulfide (CdS) nanoparticles (NPs) for the photocatalytic reduction of hexavalent chromium [Cr(VI)] under UV irradiation. Dissection of centromeric DNA from yeast Yarrowia lipolytica and identification of protein-binding site required for plasmid transmission.

In a dimorphic yeast, Yarrowia lipolytica, replicative plasmids can be established only in the coexistence of the replication origin (ORI) and centromere (CEN) from its own chromosomal DNA. Although six CEN sequences so far isolated from this yeast exhibit no similarity with conventional CEN DNA elements from other budding yeasts, they are confined within short regions (approximately 0.2 kb) and contain various conserved sequence blocks. We surveyed here a CEN1-1 sequence on an ORI-containing plasmid by deletion and site-directed mutagenesis, and found a partial palindrome, CCTAATTTGG designat
