Cloning, characterization, and transformation of the phosphoethanolamine N-methyltransferase gene (ZmPEAMT1) in maize (Zea mays L.).

N-methylation of phosphoethanolamine, the committing step in choline (Cho) biosynthesis in plants, is catalyzed by S-adenosyl-L-methionine: phosphoethanolamine N-methyltransferase (PEAMT, EC 2.1.1.103). Herein we report the cloning and characterization of the novel maize phosphoethanolamine N-methyltransferase gene (ZmPEAMT1) using a combination of bioinformatics and a PCR-based allele mining strategy. The cDNA sequence of ZmPEAMT1 gene is 1,806 bp in length and translates a 495 amino acids peptide. The upstream promoter sequence of ZmPEAMT1 were obtained by TAIL-PCR, and contained four kinds

Biochemical Adaptations of Zea mays L. to Abiotic Stress: A Focus on Drought, Salinity, and Heavy Metals.

Maize (Zea mays L.), one of the most significant cereal crops in the world, is very susceptible to abiotic stresses that include drought, salinity, and heavy metal toxicity, among others, all of which pose a risk to food and nutrition security in the face of climate change. These stresses disrupt cellular homeostasis, cause oxidative damage, inhibit photosynthesis, and eventually lower yield. This work is a synthesis of recent progress in knowledge of the biochemical, physiological, and molecular adaptation of maize to these conditions. The key processes involve reactive oxygen species (ROS) a

Fine mapping and candidate gene analysis of tassel glume closure1 (tgc1) gene in maize (Zea mays L.).

The tassel glume closure1 (tgc1) mutant was identified and mapped, and ZmKAO1, which encodes an ent-kaurenoic acid oxidase, is the key candidate gene. Tassel glume timely opening is a key trait that influences cross-pollination and hybrid breeding. However, in maize (Zea mays L.), the related genes regulating this process remain largely unexplored. Here, we identified tassel glume closure1 (tgc1), a recessive mutant that exhibits tassel glume closure, anther non-extrusion, and no pollen shedding at the adult-plant stage. Anatomical and scanning electron microscopy (SEM) observations revealed t

Optimization of Plant Oxalate Quantification and Generation of Low-Oxalate Maize (Zea mays L.) through O7 Overexpression.

Oxalate, the simplest dicarboxylic acid, is a prevalent antinutrient that chelates with various metals and can lead to the formation of kidney stones in humans. The accurate detection of the oxalate concentration in food and the cultivation of low-oxalate crops are important for enhancing public health. In this study, we established a high-throughput and highly sensitive technique for oxalate detection using ultra-high-performance liquid chromatographic-triple quadrupole tandem mass spectrometry (UPLC-QqQ-MS/MS). Additionally, we overexpressed the gene O7, which encodes oxalyl-CoA synthetase i

Phytotoxic effects of polyethylene microplastics combined with cadmium on the photosynthetic performance of maize (Zea mays L.).

Microplastics (MPs) and cadmium (Cd) has attracted increasing attention due to their combined toxicity to terrestrial vegetation. Photosynthesis which utilizes light energy to synthesize organic substances is crucial for crop production. However, the plant photosynthetic response to the joint toxicity of MPs and Cd is still unknown. Here, we studied the effects of polyethylene (PE) MPs on the photosynthetic performance of two maize cultivars Xianyu 335 (XY) and Zhengdan 958 (ZD) grown in a Cd contaminated soil. Results showed that the leaf Cd concentration in XY and ZD reached 26.1 and 31.9 μg

Cell wall: An important medium regulating the aggregation of quantum dots in maize (Zea mays L.) seedlings.

Quantum dots (QDs) find various applications in many fields, leading to increasing concerns regarding their uptake and subsequent interaction with plant body. Cell wall (CW), serving as a first target place that interacts with xenobiotic substances into plant body, its role in regulating the QDs cellular uptake needs to be explored. In the present study, maize (Zea mays L.) seedlings were hydroponically exposed to PEG-COOH-CdS/ZnS QDs (QDs-PEG-COOH) and MPA-CdS/ZnS QDs (QDs-MPA) functionalized with negatively charged and neutral coatings, respectively. Uptake rate of QDs-PEG-COOH was approxima

Correlations between photosynthetic enzymes, CO2-fixation and plastid structure in an albino mutant of Zea mays L.

An albino seedling of Zea mays L. was investigated for its potential for CO2-assimilation. In the mesophyll the number, dimensions and fine structure of chloroplasts are drastically reduced but to a lesser extent in the bundle sheath. Chlorophyll concentration is zero and carotenoid concentration almost zero. Albinism also exerts a strong influence on the stroma of bundle sheath chloroplasts; ribulose-1.5-biphosphate carboxylase (EC 4.1.1.39) activity and glyceraldehyde-3-phosphate dehydrogenase (NADP) (EC 1.2.1.13) activity is not detectable. The C4-enzymes phosphoenolpyruvate carboxylase (EC

Functional characterization of a chloroplast-localized Mn2+(Ca2+)/H+ antiporter, ZmmCCHA1 from Zea mays ssp. mexicana L.

The annual Zea mays ssp. mexicana L. is a member of the teosinte group and a close wild relative of maize. Thus, Zea mays ssp. mexicana L. can be effectively used in maize breeding. AtCCHA1 is a Mn2+ and/or Ca2+/H+ antiporter localized in chloroplasts in Arabidopsis. In this study, its homolog from Zea mays ssp. mexicana L., ZmmCCHA1, was isolated and characterized. The transcriptional level of ZmmCCHA1 in Zea mays ssp. mexicana L. was repressed in response to excessive Ca2+ or Mn2+. Heterologous functional complementation assays using yeast mutants showed that ZmmCCHA1 mediates Ca2+ and Mn2+

Vittatidera zeaphila (Nematoda: Heteroderidae), a new genus and species of cyst nematode parasitic on corn (Zea mays).

A new genus and species of cyst nematode, Vittatidera zeaphila, is described from Tennessee. The new genus is superficially similar to Cactodera but is distinguished from other cyst-forming taxa in having a persistent lateral field in females and cysts, persistent vulval lips covering a circumfenestrate vulva, and subventral gland nuclei of the female contained in a separate small lobe. Infective juveniles (J2) are distinguished from all previously described Cactodera spp. by the short stylet in the second-stage juvenile (14-17 μm); J2 of Cactodera spp. have stylets at least 18 μm long. Retention of provitamin A carotenoids in high beta-carotene maize (Zea mays) during traditional African household processing.

High beta-carotene maize, biofortified with beta-carotene through plant breeding, is being developed as a cost-effective, sustainable agronomic approach to alleviating the problem of vitamin A deficiency in Africa. We used high beta-carotene maize (10.49+/-0.16 microg beta-carotene/g) to prepare traditional maize porridges and compared the carotenoid contents in the following: (1) whole kernels; (2) wet milled flour; (3) wet milled flour, fermented; (4) wet milled flour, cooked; (5) wet milled flour, fermented and cooked. The cumulative losses of beta-carotene in the final, cooked products wer

Cadmium Toxicity to Zea mays and Its Implications for the Uptake of Other Heavy Metals by the Plant.

Cadmium is an element that is unnecessary for the functioning of plant and animal organisms, and its widespread presence in the environment poses a serious threat to human and animal health. Therefore, effective methods are being sought to remediate soils contaminated with this element, including through the enrichment of degraded soils with organic matter. To this end, the effectiveness of selected organic sorbents, including starch, fermented bark, compost and humic acids, in mitigating the transfer of cadmium and other heavy metals from soil to plants was assessed. Model studies compared th

Zein-Induced Enhancement of the Thermal Stability of Anthocyanins Extracted from Agricultural Residues of Red Corn (Zea mays).

The stability of anthocyanins is a key factor limiting their use as functional natural colorants. This study evaluated the protective effect of zein on anthocyanins extracted from red corn (Zea mays) chaff, which is an underutilized agro-industrial byproduct. The extraction of anthocyanins was optimized using a Box-Behnken design, employing a 55.5% (v/v) aqueous ethanol solution at 45.21 °C and a solid-to-solvent ratio of 1.86 mg/mL (w/v). The resulting extract was rich in acylated anthocyanins derived from cyanidin, pelargonidin, and malvidin, which were identified using electrospray ionizati

Screening of salt tolerance of maize (Zea mays L.) lines using membership function value and GGE biplot analysis.

Soil salinization is a widely recognized global environmental concern that has a significant impact on the sustainable development of agriculture at a global scale. Maize, a major crop that contributes to the global agricultural economy, is particularly vulnerable to the adverse effects of salt stress, which can hinder its growth and development from germination to the seedling stage. This study aimed to screen highly salt-tolerant maize varieties by using four NaCl concentrations of 0, 60, 120, and 180 mMol/L. Various agronomic traits and physiological and biochemical indices associated with

Hydrogen sulfide alleviates chromium toxicity by promoting chromium sequestration and re-establishing redox homeostasis in Zea mays L.

Hydrogen sulfide (H2S) is a multifunctional gaseous signaling molecule involved in the regulation of Cr stress responses. In the present study, we combined transcriptomic and physiological analyses to elucidate the mechanism underlying the mitigation of Cr toxicity by H2S in maize (Zea mays L.). We showed that treatment with sodium hydrosulfide (NaHS, a donor of H2S) partially alleviated Cr-induced growth inhibition. However, Cr uptake was not affected. RNA sequencing suggested that H2S regulates the expression of many genes involved in pectin biosynthesis, glutathione metabolism, and redox ho
