Pathological effects of dietary zearalenone and/or tamoxifen on female mink reproductive organs.

The efficacy of dietary tamoxifen (TAM) to alleviate the hyperestrogenic effects of the mycotoxin zearalenone (ZEN) was assessed by pathologic examination of the reproductive organs of female mink (Mustela vison). Mink were fed 20 mg/kg ZEN, 10 mg/kg TAM, or 20 mg/kg ZEN + 10 mg/kg TAM from about 2 mo prior to breeding until the kits reached 3 w of age. All female mink fed ZEN mated, but only 25% whelped. No mink fed TAM or TAM + ZEN mated. Postmortem examination revealed moderate to severely distended uteri filled with caseated necrotic substances in the TAM, ZEN and ZEN + TAM fed mink. Histo

Zearalenone suppresses pituitary follicle-stimulating hormone synthesis via ALKBH5-YTHDF2-SMAD4 axis.

Zearalenone (ZEA) is a mycotoxin derived from contaminated feed and causes endocrine disruption, posing a significant environmental threat to reproductive health. In this study, we investigated the molecular mechanism through which ZEA regulates pituitary gonadotropin synthesis via N6‑methyladenosine (m6A) modification. We demonstrated that ZEA downregulates alkB homolog 5, RNA demethylase (ALKBH5) in the pituitary by promoting its ubiquitination and subsequent protein degradation. This downregulation increases m6A modification of SMAD family member 4 (SMAD4) mRNA, which in turn recruits the m

Molecular confinement-engineered electrochemiluminescence aptasensor for ultrasensitive monitoring of zearalenone in cereals.

Zearalenone (ZEN), a prevalent estrogenic mycotoxin in cereals, poses significant risks to food safety and public health. Herein, an ultrasensitive electrochemiluminescence (ECL) aptasensor was developed for ZEN detection in cereal-based foods, integrating molecular confinement engineering and catalytic amplification strategies. 5,10,15,20-tetrakis(4-aminophenyl)-21H,23H-porphyrin (TCPP) was encapsulated into zirconium-based metal-organic frameworks (Zr-TCPP) through molecular confinement, which addressed the inherent issues of luminophores such as poor water solubility and structural instabil

Isolation and Characterization of Sphingopyxis sp. Q841: A New Zearalenone- Degrading Strain.

Zearalenone (ZEN) is a mycotoxin exhibiting estrogenic activity that poses a considerable risk to human health. In this study, a ZEN-degrading bacterium isolated from soil was identified as Sphingopyxis sp. and designated strain Q841. Q841 completely degraded 5 mg/L of ZEN within 4 h in mineral medium. It showed the highest activity within the tested ranges (25-37°C, pH 4.0-9.0), with optimal activity at pH 9.0 and 37°C. Assays of degradation efficiency in different fractions indicated that the ZEN-degrading activity was mainly associated with intracellular components rather than with the extr

New Insights into the Deposition of Zearalenone in Minipigs: A Suitable Bioindicator for Internal Exposure.

The detrimental footprint of mycotoxins in agriculture and on animal production has been widely recognized, especially in swine. Despite an increased number of research evaluating the toxicokinetics of mycotoxins in animal organisms, the absorption, distribution, metabolization, and excretion (ADME) patterns of zearalenone (ZEN) need further understanding. Furthermore, in vivo bioindicator for ZEN exposure in individual pigs has yet to be characterized. This study explored the ADME of ZEN in Bama Aroma pigs, a Chinese miniature pig breed, that has been used herein as a swine model. The finding

Zearalenone affects the growth of endometriosis via estrogen signaling and inflammatory pathways.

Endometriosis is a chronic, inflammatory, estrogen-dependent gynecological disease characterized by the growth of endometrial stromal cells and glands outside the uterine cavity in response to hormones, which commonly occurs in reproductive-age women. Zearalenone (ZEA) is a toxic metabolite produced by Fusarium, which acts as estrogen activity because of the similarity of its structure to estrogen. In this study, we used an endometriosis mouse model: 15 days after ovariectomy, endometrial fragments were sutured on the pelvic wall, and exogenous estrogen was supplied using an estrogen-releasing

Changes in intestinal barrier functions and gut microbiota in rats exposed to zearalenone.

Zearalenone (ZEN) is a mycotoxin that causes serious health problems in humans and animals. However, few studies have focused on the destruction of the intestinal barrier caused by ZEN. In this study, rats were exposed to different dosages of ZEN (0, 0.2, 1.0 and 5.0 mg/kg bw) by gavage for 4 weeks. The results showed that 1.0 and 5.0 mg/kg ZEN impaired gut morphology, induced the inflammatory response, reduced mucin expression, increased intestinal permeability, decreased the expression of TJ proteins and activated the RhoA/ROCK pathway. However, 0.2 mg/kg ZEN had no significant effect on int

Zearalenone in the Intestinal Tissues of Immature Gilts Exposed per os to Mycotoxins.

Zearalenone and its metabolites, α-zearalenol and β-zearalenol, demonstrate estradiol-like activity and disrupt physiological functions in animals. This article evaluates the carryover of zearalenone and its selected metabolites from the digesta to intestinal walls (along the entire intestines) in pre-pubertal gilts exposed to low doses of zearalenone over long periods of time. The term "carryover" describes the transfer of mycotoxins from feed to edible tissues, and it was used to assess the risk of mycotoxin exposure for consumers. The experimental gilts with body weight of up to 25 kg were

Hydrophobic Microenvironment Engineering Enhances Proton Transfer and Hydrolytic Activity of Zearalenone Lactonases.

Zearalenone (ZEN) lactonases are promising biocatalysts for ZEN detoxification, yet the catalytic mechanism underlying ZEN hydrolysis remains poorly understood. Here, we combined structural analysis, quantum-mechanical (QM) calculations, and molecular dynamics (MD) simulations to elucidate the catalytic mechanism and guide enzyme engineering. QM and MD analyses identified a near-attack conformation of the catalytic His245 as essential for proton transfer. Crystal structure analysis revealed that mutations within the active pocket enhanced the local hydrophobic microenvironment, thereby optimiz

Magnetic separation assisted NiTiO3/CdIn2S4Z-scheme heterojunction photoelectrochemical aptasensor for zearalenone detection.

Zearalenone (ZEN) is a harmful mycotoxin with estrogen-like activity that can cause reproductive and immune system disorders. In this study, a sandwich-structured photoelectrochemical (PEC) sensor was developed for the ultrasensitive detection of ZEN. The sensor utilizes a Z-scheme NiTiO3/CdIn2S4 heterojunction as the photoactive layer, which significantly enhances light absorption, charge separation efficiency, and photostability. NiFe2O4 nanoparticles modified with ZEN-specific aptamers serve as recognition probes, with ZEN molecules acting as a bridge to connect the probes to the PEC substr

In Vitro Evaluation of the Potential Interactions of Zearalenone-14-sulfate and Zearalenone-14-glucuronide with Human Cytochrome P450 Enzymes, Organic Anion Transporting Polypeptides, and ATP-Binding Cassette Multidrug Transporters.

Zearalenone (ZEN) is a mycotoxin that is typically produced by Fusarium strains. ZEN and its derivatives are common food contaminants and known xenoestrogens. Previous studies demonstrated the interactions of ZEN and zearalenols with certain cytochrome P450 (CYP) enzymes, organic anion transporting polypeptides (OATPs), and ATP-binding cassette (ABC) multidrug transporters. However, no data are available regarding the conjugated metabolites of the mycotoxin. Therefore, in the current study, we aimed to investigate the potential interactions of zearalenone-14-sulfate (Z14S) and zearalenone-14-g

Protective Effects of Taurine Against Zearalenone-Induced Quality Impairment During Porcine Oocyte Maturation.

Zearalenone (ZEN) is a common non-steroidal estrogenic mycotoxin that exists widely in contaminated feeds, and its toxicities target multiple organs, especially the reproductive system, since ZEN is shown to affect oocyte quality. In the present study, we reported that taurine (TAU), a sulfur-containing amino acid that exhibits antioxidant properties, protected the maturation quality of ZEN-exposed porcine oocytes. We showed that TAU significantly reversed the cumulus cell expansion and polar body extrusion defects of porcine oocytes under ZEN treatment. The rescue effect of TAU was attributed

Bacillus velezensis-derived laccase and superoxide dismutase detoxify aflatoxin B1, zearalenone, and alternariol.

Aflatoxin B1(AFB1), zearalenone(ZEN), and alternariol(AOH) are major mycotoxins that threaten food safety, yet enzymes capable of detoxifying these structurally diverse toxins remain limited. In this study, two enzymes from Bacillus velezensis HNGD-JQ06, laccase BVLac and superoxide dismutase BV2SOD, were identified as broad-spectrum detoxification candidates. BVLac degraded 93.33% of AFB1 at 60 °C and pH 7.0, 94.10% of ZEN at 60 °C and pH 8.0, and 96.87% of AOH at 50 °C and pH 8.0. BV2SOD degraded 92.59% of AFB1 and 91.99% of ZEN at 50 °C and pH 7.0, and 98.94% of AOH at 60 °C and pH 8.0. UHP
