Lutein and zeaxanthin attenuates VEGF-induced neovascularisation in human retinal microvascular endothelial cells through a Nox4-dependent pathway.

Age-related macular degeneration (AMD) and proliferative diabetic retinopathy (DR) are two of the most common and severe causes of vision loss in the population. Both conditions are associated with excessive levels of vascular endothelial growth factor (VEGF) in the eye which results in an increase in the formation of new blood vessels through a process called neovascularisation. As such, anti-VEGF therapies are currently utilised as a treatment for patients with AMD however they are associated with painful administration of injections and potential degeneration of healthy endothelium. Zeaxanthin dipalmitate therapeutically improves hepatic functions in an alcoholic fatty liver disease model through modulating MAPK pathway.

In the current study, the therapeutic effects of zeaxanthin dipalmitate (ZD) on a rat alcoholic fatty liver disease (AFLD) model were evaluated. After-treatment with ZD from the 5th week to the 10th week in a 10-week ethanol intragastric administration in rats significantly alleviated the typical AFLD symptoms, including reduction in rat body weight, accumulation of hepatic fat droplets, occurrence of oxidative stress, inflammation, chemoattractive responses and hepatic apoptosis in the liver. The reduction of liver function abnormalities by ZD was partly through lower expression level of cyto

The Relative Bioavailability of Lutein and Zeaxanthin in the Presence of Omega-3 Supplements and Their Effect on Oxidative Stress Levels in Humans: A Pilot Study.

Introduction: Lutein+Zeaxanthin (L+Z) are the major constituents of macular pigments of the retina. There is a lack of information on the bioavailability of the two compounds in the presence and absence of omega-3 fatty acids in L+Z supplements which are commonly prescribed to treat macular degeneration. Despite growing interest in L+Z supplementation, there remains a limited understanding of their short-term bioavailability dynamics and the potential added value of omega-3 co-supplementation. This pilot study reports on the bioavailability of serum responses to L+Z supplements in the presence

Lutein and zeaxanthin reduce A2E and iso-A2E levels and improve visual performance in Abca4-/-/Bco2-/- double knockout mice.

Accumulation of bisretinoids such as A2E and its isomer iso-A2E is thought to mediate blue light-induced oxidative damage associated with age-related macular degeneration (AMD) and autosomal recessive Stargardt disease (STGD1). We hypothesize that increasing dietary intake of the macular carotenoids lutein and zeaxanthin in individuals at risk of AMD and STGD1 can inhibit the formation of bisretinoids A2E and iso-A2E, which can potentially ameliorate macular degenerative diseases. To study the beneficial effect of macular carotenoids in a retinal degenerative diseases model, we used ATP-bindin

Zeaxanthin pretreatment alleviates low-temperature stress-induced injury in flue-cured tobacco seedlings by modulating the xanthophyll cycle and photoprotection.

Low temperature is a major abiotic stress that affects flue-cured tobacco (Nicotiana tabacum L.) seedlings during the early transplanting stage. Zeaxanthin (Zea), a xanthophyll pigment closely associated with the photosynthetic apparatus has been reported to alleviate chilling injury in crops. However, its physiological roles in flue-cured tobacco under alternating chilling and rewarming conditions remain unclear. In this study, seedlings of the flue-cured tobacco cultivar Yunyan 87 were pretreated with Zea at 0, 25, 50, 75, or 100 mg L-1 and subsequently exposed to low-temperature stress at 1

Interaction between serum levels of Porphyromonas gingivalis immunoglobulin G and lutein/zeaxanthin is associated with risk for age-related macular degeneration.

Porphyromonas gingivalis (P. gingivalis) functions as a catalyst bacterium in the development of periodontitis, and the serum antibody level against P. gingivalis is considered a surrogate marker for the activity level of periodontopathic microbiota. The chronic systemic inflammation induced by P. gingivalis elevates the risk of various systemic and neurodegenerative disorders, including atherosclerosis, diabetes, and Alzheimer's disease. Although the connection between human microbiota and age-related macular degeneration (AMD) remains relatively unexplored, it is noteworthy that AMD shares r

Zeaxanthin inhibits endothelial pyroptosis and alleviates atherosclerotic plaque formation by suppressing Ang2-mediated activation of TLR4/NLRP3 signaling pathway.

Clinical studies showed that zeaxanthin reduces oxidized lipid products and inflammation, and alleviates the formation of atherosclerotic plaques. However, the molecular mechanism by which zeaxanthin alleviates atherosclerosis (AS) progression remains unclear. This study aimed to explore the molecular mechanism of zeaxanthin alleviating AS progression. Human umbilical vein endothelial cells (HUVECs) and human arterial endothelial cells (HAECs) were treated with 100 μg/mL oxidized low density lipoprotein (ox-LDL) to construct AS cell model in vitro, then treated with 50 or 150 μM zeaxanthin for

Turnip Z-ISO gene promotes zeaxanthin accumulation by modulating carotenoid metabolic flux and is involoved in negative responses to low-temperature stress.

Turnip, Brassica rapa ssp. rapa, is a biennial herbaceous root-type subspecies that is widely cultivated in East Asia. Carotenoids, important plastidial isoprenoid pigments and antioxidants, have important coloring functions in plants and contribute to health maintenance in humans. Here, the last key gene isolated in the carotenoid metabolic pathway, 15-cis-ζ-carotene isomerase (Z-ISO), which encodes 356 amino acids, has identified. It was expressed significantly more in the swollen root of yellow-turnip W25 than in white-turnip W21, and the BrrZ-ISO protein was located in the nucleus and cell

Zeaxanthin and Lutein Ameliorate Alzheimer's Disease-like Pathology: Modulation of Insulin Resistance, Neuroinflammation, and Acetylcholinesterase Activity in an Amyloid-β Rat Model.

Alzheimer's disease (AD) is characterized by impaired insulin/insulin-like growth factor-1 signaling in the hippocampus. Zeaxanthin and lutein, known for their antioxidant and anti-inflammatory properties, have been reported to protect against brain damage and cognitive decline. However, their mechanisms related to insulin signaling in AD remain unclear. This study investigated the efficacy and mechanisms of zeaxanthin, lutein, and resveratrol in modulating an AD-like pathology in an amyloid-β rat model. Rats were administered hippocampal infusions of 3.6 nmol/day amyloid-β (Aβ)(25-35) for 14

Lutein and Zeaxanthin Isomers Protect against Light-Induced Retinopathy via Decreasing Oxidative and Endoplasmic Reticulum Stress in BALB/cJ Mice.

Oxidative stress (OS) and endoplasmic reticulum stress (ERS) are the major factors underlying photoreceptor degeneration. Lutein, RR-zeaxanthin (3R,3’R-zeaxanthin) and RS (meso)-zeaxanthin (3R,3’S-RS- zeaxanthin) (L/Zi) could protect against cell damage by ameliorating OS in retina. In this study, we examined the effect of L/Zi supplementation in a mouse model of photoreceptor degeneration and investigated whether the treatment of L/Zi ameliorated OS and ERS. BALB/cJ mice after light exposure were used as the animal model. The protective effects of L/Zi were observed by electroreti

Lutein and zeaxanthin supplementation in preterm very low-birth-weight neonates in neonatal intensive care units: a multicenter randomized controlled trial.

BACKGROUND: Human milk feeding protects against oxidative stress-induced damage in preterm neonates, including severe multifactorial diseases such as retinopathy of prematurity (ROP), necrotizing enterocolitis (NEC), and bronchopulmonary dysplasia (BPD). The carotenoids, which are not found in formula milk, might play a key role in these actions. METHODS: A multicenter, double-blind, randomized controlled trial was conducted in three tertiary Italian neonatal intensive care units. All preterm infants < 32(+6) weeks' gestational age were eligible and were randomized to a single, oral, daily 0.

Determination of Lutein and Zeaxanthin Esters and Their Geometric Isomers in Carotenoid Ester Concentrates Used as Ingredients in Nutritional Supplements: Validation of a Combined Spectrophotometric-HPLC Method.

A combined spectrophotometric-LC method is described for the determination of total lutein and zeaxanthin ester content in carotenoid ester concentrates, including their main geometrical isomers. The concept of composite-specific absorbance is introduced for this purpose. The method is applicable to carotenoid ester concentrates used as ingredients in oil suspensions and dosage forms. The sample is dissolved in a hexane-2-propanol mixture (95 + 5, v/v) for spectrophotometric measurement at a maximum absorption of ~445 nm. Subsequently, in parallel, a sample is saponified and chromatographed on
