Alterations in the noradrenergic projection to the cerebellum of the dystonic (dt) rat.

The genetically dystonic rat (dt) has elevated resting levels of cerebellar norepinephrine (NE) in comparison with phenotypically normal littermates. This difference is not secondary to cerebellar hypoplasia. Increased NE is observed as early as postnatal day 12, when clinical symptoms have become evident. The elevation in cerebellar NE levels in the dt rat involves all cerebellar areas, but is not generalized to all terminal fields of the locus coeruleus. Elevations in cerebellar NE are followed developmentally by a reduction in sensitivity to the NE-depleting effects of reserpine, a change w

Modulation of luteinizing hormone release and catecholamine activity by opiates in the female rat.

Previous studies have shown that the stimulation of LH release by the opiate receptor blocker, naloxone, can be prevented by catecholamine synthesis inhibitors, suggesting opiate regulation of catecholamine release. The present study tested whether an opiate agonist and antagonist would affect the depletion of hypothalamic catecholamines observed after synthesis inhibition, as a measure of catecholamine activity, concomitant with changes in LH secretion. Administration of naloxone to estradiol-primed rats increased LH release and potentiated the depletion of norepinephrine in the preoptic-ante

Cerebral dopamine and noradrenaline turnover and effects of morphine test dose in rats withdrawn from 20 days' morphine treatment.

Depletion of dopamine (DA) and noradrenaline (NA) induced by alpha-methyl-p-tyrosine (alpha MT) was studied in five different brain areas of rats withdrawn from 20 days' chronic treatment for 0.5, 1, 2, 3, 4 or 6 days. Some of the alpha MT-treated rats were given a challenge dose of morphine (10 mg/kg) 2.5 h before decapitation. The depletion of DA was clearly retarded in the limbic forebrain but not in the striatum of rats withdrawn from morphine for 1 and 2 days, whereas the alpha MT-induced NA depletion was slightly accelerated in the hemispheres but not in any other part of the brain of ra

[Effect of alpha-methyl-p-tyrosine and dihydroxyphenylalanine on plasma corticosterone levels in rats pretreated with prednisolone acetate].

The effects on hypothalamic-pituitary-adrenal function and on negative feed-back action of a synthetic adrenal steroid, prednisolone acetate, have been studied in female Wistar rats treated with alpha-methyl-p-tyrosine (alpha MpT) and L-dihydroxyphenylalanine (L-DOPA) administered for six consecutive days. The parameter for study was taken from the plasmatic changes of corticosterone levels. alpha-MpT injection determines an increase of plasmatic corticosterone values, while L-DOPA injection produces the opposite effect, decreasing corticosterone levels as compared to the saline control group.

Effects of a new TRH analogue, YM-14673, on spontaneous motor activity in rats.

The effects of a new TRH analogue, YM-14673 (N alpha-[[(S)-4-oxo-2-azetidinyl]carbonyl]-L-histidyl-L-prolinamide dihydrate) on spontaneous motor activity (SMA) were compared with those of TRH in rats. Intraperitoneal, oral and intracerebral administrations of YM-14673 caused a significant and dose-dependent increase in SMA. TRH also showed similar actions as YM-14673 did, except after oral dosing. These actions of YM-14673 were about 20-100 times more potent than those of TRH. Pretreatment with haloperidol, alpha-methyl-p-tyrosine (alpha-MT) and amino-oxyacetic acid (AOAA) but not atropine, in

Perioperative management of a patient with a malignant pheochromocytoma.

A case report is presented of a patient with a known malignant pheochromocytoma, who was to undergo an orthopaedic procedure using methylmethacrylate bone cement. Preoperative preparation of the patient included the use of prazosin and metyrosine. Epidural blockade was chosen as the anaesthetic technique for intraoperative management. Surgical fixation with methylmethacrylate did not produce profound or prolonged hypotension despite sudden massive blood loss prior to its use. Epidural morphine was administered for postoperative pain control.

Hypothalamic dopamine and catechol oestrogens in rats with spontaneous pituitary tumours.

Dopamine concentration within the hypothalamus and its depletion after the administration of alpha-methyl-para-tyrosine were measured in young rats and compared with values obtained in aged animals with and without spontaneously occurring pituitary tumours. Old rats had significantly reduced hypothalamic dopamine concentrations and there was less depletion of dopamine compared with young animals but there were no differences between tumorous and non-tumorous animals. Hyperprolactinaemia induced in young animals caused a much greater depletion of hypothalamic dopamine than in old tumorous rats

Hyperprolactinaemia and DNA synthesis in the pituitary gland of the rat.

Hyperprolactinaemia produced in rats by the transplanted prolactin-secreting tumours MtTW15 and 7315a significantly (P less than 0.01) inhibited by 70% the incorporation of [3H]thymidine into the pituitary DNA of the host animals. The weight and the DNA content of the glands were significantly (P less than 0.01) reduced by 30%. The administration of haloperidol, a dopamine receptor blocking agent, to the tumour-bearing rats increased the suppressed DNA replication in the anterior pituitary glands by approximately 560% in the MtTW15-bearing rat and by 100% in the 7315a-bearing animals. Angiotensin II increases norepinephrine turnover in the anteroventral third ventricle of spontaneously hypertensive rats.

We evaluated the effect of angiotensin II (Ang II) administered by intracerebroventricular injection on norepinephrine turnover in the anteroventral third ventricle in adult spontaneously hypertensive rats (SHR, n = 35) and age-matched Wistar-Kyoto rats (WKY, n = 38). Ang II (100 ng) or saline (vehicle control) was administered into the cerebral ventricle 30 minutes after injection of alpha-methyl-p-tyrosine (250 mg/kg IP). Norepinephrine turnover was assessed by evaluation of the norepinephrine concentration before and 1 hour after such administration. The pressor response to Ang II administr

[The beta-adrenoreceptors of the cerebral cortex in rat pups after exposures altering the noradrenaline level].

Elevation of noradrenaline level in the brain cortex of 7-day old rats induced by prenatal corticosterone administration was accompanied by a decrease in beta-adrenoreceptor density determined by specific [3H]dihydroxyalprenolol binding. Affinity of receptors to the ligand remained unchanged. Injections to 4-6-day old rats of alpha-methyl-p-tyrosine that blocks catecholamine synthesis resulted in a decreased noradrenaline content and increased beta-adrenoreceptor density in the brain cortex of 7-day old rats. The extent of changes in beta-adrenoreceptor density in the brain cortex of 7-day old

Differential effects of hypoxia on the turnover of norepinephrine and epinephrine in the heart, adrenal gland, submaxillary gland and stomach.

The authors examined the effects of hypoxia (8% O2 in N2) on the turnover rates of norepinephrine (NE) and epinephrine (E) in the heart, adrenal gland, submaxillary gland and stomach. The turnover rates were estimated by measuring the decrease in the content of NE and E after an i.p. injection of alpha-methyl-p-tyrosine (250 mg/kg), an inhibitor of tyrosine hydroxylase. After a 4-h exposure to hypoxia, the turnover rate of NE in the heart and those of NE and E in the adrenal gland were increased, whereas that of NE in the submaxillary gland was decreased. The turnover rate of NE in the stomach

Catecholamines and pituitary function. IV. Effects of low-dose dopamine infusion and long-term bromocriptine treatment on the abnormal thyrotroph (TSH) dynamics in patients with pathological hyperprolactinaemia.

In order to gain further insight into the role of dopamine (DA) in the control of TSH release and to investigate whether an increased or defective DA inhibition on pituitary thyrotrophs may be considered responsible for the abnormal TSH dynamics in pathological hyperprolactinaemia, we examined the effect of low-dose DA infusion on TRH stimulated TSH secretion in normally cycling women and in patients with pathological hyperprolactinaemia. The effect of long-term bromocriptine therapy on TSH dynamics was also evaluated in a selected group of hyperprolactinaemic women. Fifty-two hyperprolactinae

Supersensitivity of the isolated guinea-pig vas deferens induced by a toxic fraction of scorpion venom (Tityus serrulatus).

Tityustoxin (TsTx), a toxic fraction of Tityus serrulatus venom, was studied on the isolated guinea-pig vas deferens. It increased significantly the maximal response of the preparation to both norepinephrine and acetylcholine and decreased the effective median dose of norepinephrine. The effect of TsTx on norepinephrine median dose was unchanged when atropinized or pharmacologically "denervated" preparations were used but was abolished when both procedures were associated. Atropinization of pharmacologically denervated muscles almost never modify the TsTx-induced increase in the maximal respon

Effect of manipulating central catecholamines on puberty and the surge of luteinizing hormone and gonadotropin releasing hormone induced by pregnant mare serum gonadotropin in female rats.

We have investigated the effect of manipulating central catecholamines on the timing of puberty (as assessed by vaginal opening) in female rats and the surge of luteinizing hormone (LH) and gonadotropin releasing hormone (GnRH) induced by pregnant mare serum gonadotropin (PMSG) in immature female rats. Manipulation of the catecholamines was carried out with either 6-hydroxydopamine (6-OHDA) administered with or without either desipramine (DMI) or pargyline, or alpha-methyl-p-tyrosine (alpha-MPT). The neonatal administration of 6-OHDA delayed puberty, an effect which was potentiated by pretreat

Noradrenaline-induced emesis. Alpha-2 adrenoceptor mediation in the area postrema.

The emetic action of noradrenaline was investigated in unanesthetized cats, after it was injected into the cerebral ventricles through chronically implanted cannulae. Intracerebroventricular injection of noradrenaline produced dose-dependent and shortlasting emesis, which was abolished after ablation of the area postrema. However, copper sulphate, given orally, evoked emesis in cats with an ablated area postrema. D-1 receptor-linked mechanism modulates cholinergic neurotransmission in rat striatum.

SCH 23390, a D-1 dopaminergic antagonist, was examined for its effects on the cholinergic system in rat brain. The compound raised the content of acetylcholine selectively in striatum and not in other brain areas including the hippocampus, nucleus accumbens, hemispheric residuum and midbrain-hindbrain, mirroring the action of dopaminomimetic drugs. That the increase in acetylcholine content reflected a depression of striatal cholinergic neuronal activity was substantiated by the drug's ability to inhibit sodium-dependent high affinity choline uptake, to reduce the electrically evoked release o

Effects of prenatal ethanol exposure on later sensitivity to the low-dose stimulant actions of ethanol in mouse offspring: possible role of catecholamines.

The purpose of this study was to examine whether prenatal ethanol (EtOH) exposure alters later sensitivity to the low-dose stimulant effects of EtOH. Because the locomotor stimulant effects of EtOH are thought to be mediated, at least in part, by activation of brain monoamine systems, and because prenatal EtOH exposure has been shown to alter brain monoamine activity, it was hypothesized that prenatal EtOH exposure may alter sensitivity to the stimulant actions of EtOH. To test this hypothesis, sensitivity to the locomotor stimulant effects of various challenge doses of EtOH was examined in ma
