Bioactive cell-derived matrices combined with polymer mesh scaffold for osteogenesis and bone healing.

Successful bone tissue engineering generally requires an osteoconductive scaffold that consists of extracellular matrix (ECM) to mimic the natural environment. In this study, we developed a PLGA/PLA-based mesh scaffold coated with cell-derived extracellular matrix (CDM) for the delivery of bone morphogenic protein (BMP-2), and assessed the capacity of this system to provide an osteogenic microenvironment. Decellularized ECM from human lung fibroblasts (hFDM) was coated onto the surface of the polymer mesh scaffolds, upon which heparin was then conjugated onto hFDM via EDC chemistry. BMP-2 was

Restoring Blood Pressure in Hypertensive Mice Fails to Fully Reverse Vascular Stiffness.

BACKGROUND: Hypertension is a well-established driver of vascular remodeling and stiffening. The goal of this study was to evaluate whether restoring normal blood pressure (BP) fully restores vascular stiffness toward that of normotensive controls. METHODS: C57Bl6/J male mice received angiotensin II (angII; 1 μg/kg/min) via infusion pump for 8 weeks (hypertension group: HH), angII for 4 weeks (hypertension group: H4), angII for 4 weeks followed by 4 weeks of recovery (reversal group: HN), or sham treatment (normotensive group: NN). BP, heart rate, and pulse wave velocity (PWV) were measured l

Volumetric printed biomimetic scaffolds support in vitro lactation of human milk-derived mammary epithelial cells.

The human breast is remarkably plastic and remodels with each birth to produce milk optimally suited for the changing demands of the newborn. This dynamic nature of lactation makes it challenging to study under controlled conditions. Given the health benefits of human milk, models of secretory mammary tissue would offer opportunities to study factors that influence this important food source. First, 3D models of the mammary duct/alveoli (D/A) were designed inspired by shapes found in vivo. Photoresins based on mammary decellularized extracellular matrix (dECM) were optimized to match the mecha

Bioengineering of a human iPSC-derived vascularized endocrine pancreas for type 1 diabetes.

Intrahepatic islet transplantation in patients with type 1 diabetes is limited by donor availability and lack of engraftment. Alternative β cell sources and transplantation sites are needed. We demonstrate the feasibility to repurpose a decellularized lung as an endocrine pancreas for β cell replacement. We bioengineer an induced pluripotent stem cell (iPSC)-based version, fabricating a human iPSC-based vascularized endocrine pancreas (iVEP) using iPSC-derived β cells (iPSC-derived islets [SC-islets]) and endothelial cells (iECs). SC-islets and iECs are aggregated into vascularized iβ spheroid

Improved Models of Human Endometrial Organoids Based on Hydrogels from Decellularized Endometrium.

Organoids are three-dimensional (3D) multicellular tissue models that mimic their corresponding in vivo tissue. Successful efforts have derived organoids from primary tissues such as intestine, liver, and pancreas. For human uterine endometrium, the recent generation of 3D structures from primary endometrial cells is inspiring new studies of this important tissue using precise preclinical models. To improve on these 3D models, we decellularized pig endometrium containing tissue-specific extracellular matrix and generated a hydrogel (EndoECM). Next, we derived three lines of human endometrial o

Porcine Decellularized Diaphragm Hydrogel: A New Option for Skeletal Muscle Malformations.

Hydrogels are biomaterials that, thanks to their unique hydrophilic and biomimetic characteristics, are used to support cell growth and attachment and promote tissue regeneration. The use of decellularized extracellular matrix (dECM) from different tissues or organs significantly demonstrated to be far superior to other types of hydrogel since it recapitulates the native tissue's ECM composition and bioactivity. Different muscle injuries and malformations require the application of patches or fillers to replenish the defect and boost tissue regeneration. Herein, we develop, produce, and charac

ECM-based triple layered scaffolds for vascular tissue engineering.

The present study focused on the development of three layered small-diameter (<6 mm) extracellular matrix (ECM)-based vessels. These were engineered artificially through the freeze-drying technique. A layer of decellularized bovine aorta (DAM) was deposited on a mandrel and, after lyophilization, it was dipped into a poly-L-lactide acid (PLLA)/polyethylene glycol (PEG) 2000 dichloromethane solution then quickly wrapped with a pre-prepared thin DAM sheet. Mechanical properties of three-layered scaffolds were evaluated by means of uniaxial tensile measurement. Furthermore, human endothelial and

3D-printed niche-matched GelMA-dECM bioinks rewire lipid metabolic reprogramming and chemosensitization in pancreatic ductal adenocarcinoma.

Primary pancreatic ductal adenocarcinoma (PDAC) and hepatic metastases grow in anatomically distinct niches defined by organ-specific extracellular matrix (ECM) cues related to matrix composition and biomechanics such as substrate stiffness. Metabolic remodeling is a fundamental mechanism by which PDAC cells adapt to environmental stress. However, whether ECM actively imposes site-dependent metabolic liabilities remains unclear. To this end, we developed a digital light processing-printable GelMA-dECM platform incorporating decellularized pancreas- or liver-derived ECM under controlled three-d

Development of a decellularized human amniotic membrane-based electrospun vascular graft capable of rapid remodeling for small-diameter vascular applications.

The performance of small-diameter vascular grafts adapted to vascular replacement is commonly hindered by stenosis. To address this issue, a graft featuring rapid remodeling with degradation is warranted. In this work, a 1.8-mm-diameter graft was constructed by fabricating a decellularized human amniotic membrane (HAM) with polycaprolactone (PCL)/silk fibroin (SF) around it through electrospinning, namely, an HPS graft, and applied in a rat aortic grafting model for comparison to a decellularized porcine small intestinal submucosa (SIS)-integrated PCL/SF (SPS) graft and an autologous aorta. Building a Total Bioartificial Heart: Harnessing Nature to Overcome the Current Hurdles.

Engineering a bioartificial heart has become a possibility in part because of the regenerative medicine approaches to repairing or replacing damaged organs that have evolved over the past two decades. With the advent of inducible pluripotent stem cell technology, it is now possible to generate personalized cells that make the concept of autologous tissue engineering imaginable. Scaffolds that provide form, function, and biological cues to cells likewise potentially enable the engineering of biocompatible vascularized solid organs. Decellularized organs or tissue matrices retain organ complexit

Laminin expression profiles of osteogenic-and chondrogenic-induced dECM sheets.

Decellularized extracellular matrix sheets (dECMSs) produced by stem cells have attracted attention because they preserve the natural biological activity of the ECM to direct lineage-specific differentiation with less immunogenicity. As a core ECM protein, laminin modulates cellular phenotype and differentiation. Nevertheless, no studies thus far have explored the distribution and abundance of laminins in diverse dECMSs. Herein, we first compared the differential expression of laminins among dECMSs in osteogenic-induced medium (OI-dECMS), chondrogenic-induced medium (CI-dECMS), and standard me

A 3D-Printed Biochemical-Mechano-Thermally Responsive Multicellular Tumor Organoid Platform Decodes Biochemical-Physical Regulation of Cancer-Immune Crosstalk and Chemoresistance.

Oral tumors are not shaped by biochemical signals alone; they also evolve within mechanically active and thermally fluctuating microenvironments that critically regulate malignant progression, immune remodeling, and therapeutic resistance. Here, we develop a dynamic bioorthogonal organoid platform that couples a 3D-printed photothermal elastic scaffold with an interpenetrating hydrogel network derived from decellularized oral mucosa extracellular matrix, enabling the reconstruction of in vivo-like structural support, matrix-derived biochemical cues, spatial cellular compartmentalization, and d

Multifunctional Skin Dermal Extracellular Matrix Enabling Skin-Relevant Bioactivity for Tissue Remodeling, Hydration, and Anti-Hyperpigmentation.

Dermal extracellular matrix (ECM) deterioration is a central driver of skin aging, contributing to impaired elasticity, decreased moisturization, and uneven pigmentation. However, commonly used single-component ingredients and cell-derived bioactives provide limited coordinated cues and may therefore be insufficient to address these multifactorial processes. Here, we propose decellularized skin-derived ECM (skin ECM) as a multifunctional cosmetic ingredient through comparison with various existing cosmetic ingredients. Proteomic analysis shows that skin ECM retains diverse collagen subtypes al

"Smart" Nerves Sprout and Assemble in an Extracellular Matrix-Based 3D Nerve-in-a-Chip Microfluidic Model.

Controlling directional neurite outgrowth and Schwann cell (SC) migration remains critical for nerve regeneration. While 2D anisotropic topographies guide nerve growth via contact guidance, these models fail to recapitulate the 3D extracellular matrix microenvironment of native nerves. Herein, a microfluidic "nerve-in-a-chip" platform is customized and filled with decellularized nerve matrix hydrogel (DNM-G) to investigate how microchannel dimensions regulate 3D neurite extension and SC dynamics. Mid-sized microchannels optimally enhanced longitudinal neurite growth and fasciculation, whereas

Organic and Synthetic Substitutes in Tracheal Reconstruction: A Scoping Review (2015-2025).

Tracheal defects have been the focus of research since the 19th century, but reconstructing this complex structure remains challenging. Identifying a safe, effective tracheal substitute is a key goal of surgery. This integrative review explores current tracheal substitutes and tissue engineering techniques. Data were collected from June 2024 to March 2025 from electronically available databases. Articles published between 2015 and 2025 were selected using the individualized protocol for each database. After screening 190 articles, 82 were excluded, and 108 were reviewed, with 100 meeting the f

Growth factors and mechano-regulated reciprocal crosstalk with extracellular matrix tune the keratocyte-fibroblast/myofibroblast transition.

Improper healing of the cornea after injury, infections or surgery can lead to corneal scar formation, which is associated with the transition of resident corneal keratocytes into activated fibroblasts and myofibroblasts (K-F/M). Myofibroblasts can create an extracellular matrix (ECM) niche in which fibrosis is promoted and perpetuated, resulting in progressive tissue opacification and vision loss. As a reversion back to quiescent keratocytes is essential to restore corneal transparency after injury, we characterized how growth factors with demonstrated profibrotic effects (PDGF, FGF, FBS, TGF

Decellularized nerve matrix hydrogel scaffolds with longitudinally oriented and size-tunable microchannels for peripheral nerve regeneration.

The scaffolding biomaterials and their internal structures are crucial in constructing growth-permissive microenvironment for tissue regeneration. A functional bioscaffold not only requires sufficient extracellular matrix components, but also provides topological guidance by mimicry of the ultrastructure of the native tissue. In our laboratory, a decellularized nerve matrix hydrogel derived from porcine sciatic nerve (pDNM-G) is successfully prepared, which shows great promise for peripheral nerve regeneration. Herein, longitudinally oriented microchannel structures were introduced into pDNM-G

Conserved and tissue-specific immune responses to biologic scaffold implantation.

Upon implantation into a patient, any biomaterial induces a cascade of immune responses that influences the outcome of that device. This cascade depends upon several factors, including the composition of the material itself and the location in which the material is implanted. There is still significant uncertainty around the role of different tissue microenvironments in the immune response to biomaterials and how that may alter downstream scaffold remodeling and integration. In this study, we present a study evaluating the immune response to decellularized extracellular matrix materials within

The Preparation of Decellularized Mouse Lung Matrix Scaffolds for Analysis of Lung Regenerative Cell Potential.

Lung transplantation is the only option for patients with end-stage lung disease, but there is a shortage of available lung donors. Furthermore, efficiency of lung transplantation has been limited due to primary graft dysfunction. Recent mouse models mimicking lung disease in humans have allowed for deepening our understanding of disease pathomechanisms. Moreover, new techniques such as decellularization and recellularization have opened up new possibilities to contribute to our understanding of the regenerative mechanisms involved in the lung. Stripping the lung of its native cells allows for
