BACKGROUND: Proteoglycans (PG) are known to be involved in the organization and assembly of the extracellular matrix (ECM) prior to mineral deposition. Osteoadherin (OSAD), a keratan sulphate PG is a member of the small leucine-rich (SLRP) family of PGs and unlike other SLRPs, OSAD expression is restricted to mineralized tissues. It is proposed to have a high affinity for hydroxyapatite and has been shown to be expressed by mature osteoblasts but its exact role remains to be elucidated.
METHODOLOGY/PRINCIPAL FINDINGS: We investigated the protein distribution of OSAD in the developing mouse to

Bone and dentin share similar biochemical compositions and physiological properties. Dentin, a major tooth component, is formed by odontoblasts; in contrast, bone is produced by osteoblasts. Osterix (Osx), a zinc finger-containing transcription factor, has been identified as an essential regulator of osteoblast differentiation and bone formation. However, it has been difficult to establish whether Osx functions in odontoblast differentiation and dentin formation. To understand the role of Osx in dentin formation, we analyzed mice in which Osx was subjected to tissue-specific ablation under the

INTRODUCTION: Although human dental pulp stem cells isolated from healthy teeth have been extensively characterized, it is unknown whether stem cells also exist in clinically compromised teeth with irreversible pulpitis. Here we explored whether cells retrieved from clinically compromised dental pulp have stem cell-like properties.
METHODS: Pulp cells were isolated from healthy teeth (control group) and from teeth with clinically diagnosed irreversible pulpitis (diseased group). Cell proliferation, stem cell marker STRO-1 expression, and cell odonto-osteogenic differentiation competence were

Background/Objectives: Osteogenesis imperfecta (OI) is a rare hereditary connective tissue disorder characterized by bone fragility, recurrent fractures, and variable extra-skeletal manifestations. There are scarce epidemiological and clinical data from Eastern Europe, including Romania. This study aimed to characterize the phenotypic spectrum and diagnostic features of pediatric OI in a Romanian multicenter cohort. Methods: A retrospective, multicenter observational study was conducted over a 10-year period (January 2014-December 2024) in three tertiary pediatric referral centers in Bucharest

PURPOSE: To evaluate the long-term clinical and radiographical results of using gray mineral trioxide aggregate (MTA) in pulpotomy treatments in primary molars.
METHODS: A total of 69 primary molars were treated with pulpotomy procedures using MTA and follow-up every 6 months up to 42 months.
RESULTS: Clinical success was 100% as none of the molars showed clinical pathologic signs; radiographical success was 98.5% as one case of internal resorption was recorded. Reparative dentin deposition was found as stenosis of the pulp canals in 84% of the cases 42 months after treatment and as dentin b

The structure of reparative tertiary dentin in human deciduous teeth has been studied. Reparative dentin is secreted by a new generation of odontoblast-like cells which have been subject to strong stimuli, e.g., trauma or deep active caries lesions with associated pulp inflammation. Ground sections of 25 teeth were prepared, and contact microradiographs were produced. Another 30 teeth were demineralized, embedded in paraffin, sectioned, and stained with hematoxylin and eosin. Some demineralized sections from each tooth were also studied in the scanning electron microscope. Most of the teeth sh

AIM: Punicalagin (PCG), a natural polyphenol derived from pomegranate peel, possesses well-established antibacterial and anti-inflammatory properties. However, whether PCG promotes odontogenic differentiation of human dental pulp stem cells (DPSCs) remains unclear. This study investigated the effects of PCG on odontogenic differentiation and reparative dentin formation and further explored the underlying molecular mechanism.
METHODS: A rat pulpitis model was established to evaluate the effects of a PCG-loaded collagen sponge on pulpal repair. Pulpal inflammation and reparative dentin formatio

INTRODUCTION: The aim of this study was to evaluate pulpal responses after experimental direct pulp capping of mechanically exposed teeth with a new calcium silicate-based dentin replacement material.
METHODS: Thirty-four anterior and posterior teeth of 3 miniature swine were used. Class V or I cavities were prepared on the buccal or occlusal surfaces, respectively. Pulpal exposures were further performed using a round carbide bur 0.8 mm in diameter. Exposures were treated with white MTA Angelus (Angelus, Londrina, PR, Brazil) or Biodentine (Septodont, Saint Maur des Fosses, France), and the

The overall aim of this thesis was to study some aspects of the repair of the dentine barrier, especially in conjunction with dental pulp capping. Understanding the events leading to the healing of the dentine and pulp, and hence successfully preserving the vitality and functions of the tooth, would lead to a scientific basis for a less invasive treatment of pulp exposures than performing root canal treatments. The surfaces of the body have physiological barrier functions aimed at protecting the body from external noxious agents. In the tooth, the odontoblasts, which line the outermost part of

This case report presents a patient with Dentogenesis Imperfecta (DI) associated with Osteogenesis Imperfecta (OI) and its subsequent dental manifestations. The patient in this report (see Figure 1) has spent his life living with his disability type III OI (also known as brittle bone disease) and its degenerative affects. The patient is independent and enjoys his social life but felt his existing dentures were having an adverse effect on the quality of his life. The patient attended Dorset County Hospitals Special Care Dentistry and on clinical examination it was noted the patient was partiall

In a clinical, radiological and biochemical study of forty-two patients from Oxford with osteogenesis imperfecta, it was found that patients could be divided simply into mild, moderate and severe groups according to deformity of long bones. In the severe group (seventeen patients) a family history of affected members was uncommon and fractures began earlier and were more frequent than in the mild group (twenty-two patients); sixteen patients in the severe group had scoliosis and eleven had white sclerae; no patients in the mild group had white sclerae or scoliosis. Radiological examination of

Transforming growth factor beta, TGF-beta, is expressed during tooth formation and can induce pre-odontoblast differentiation and formation of functional odontoblast-like cells in vitro. In addition, exogenous TGF-beta can increase reparative dentin formation, presumably by acting on odontoblasts. In this study, we examined the tooth phenotype of transgenic mice, in which TGF-beta 2 expression is directed by the osteocalcin promoter. Previous studies have shown that these mice have a bone phenotype that resembles that of human osteoporosis, including the existence of spontaneous fractures. Mic

LIM mineralization protein 1 (LMP-1) is an essential positive regulator of osteoblast differentiation and maturation and bone formation. However, the expression and distribution of LMP-1 in human teeth are largely unknown. The aim of this study was to detect the expression of LMP-1 in normal healthy human teeth and human teeth with various pathologic conditions. LMP-1 expression was determined by immunohistochemistry in all of the samples including 12 healthy teeth, 10 teeth with caries, 12 teeth with pulpitis, and 4 teeth with pulp calcification. We found that LMP-1 was expressed primarily in

This article identifies certain syndromes of the head and neck, which a dentist may see in clinical practice, and relates these syndromes to their sites of mutation on involved genes. This paper is timely with the near completion of the Human Genome Project, the mapping of the entire human genetic material. Knowing the site of the genetic lesion is important in helping clinicians understand the genetic basis for these conditions, and may help in our future understanding of remedies and treatments.

Insulin-like growth factors (IGF-I and IGF-II) have been shown to play an important role in growth and differentiation in a number of tissues including mineralizing bone. Little is known about their role in tooth mineralization. Previous work in our laboratory has shown the presence of IGFs ligands, their receptors, and their binding proteins during mouse tooth morphogenesis. The expression of IGF I coincides with the expression of amelogenin, ameloblastin and enamelin at the late bell and secretory stage. The objective of this study is to determine the mechanisms by which IGFs modulate enamel

The overall aim of this series of studies was to determine whether the extensive narrowing of the dental pulp chamber seen in dental radiographs of patients with end stage renal disease was related to the method of treatment of the renal disease. The treatments studied were immunosuppression, renal transplantation and hemodialysis. A strong correlation was found between narrowing of the dental pulp chamber and renal transplantation. In the immunosuppression group and in the renal transplantation group, the patients received immunosuppressive drugs, mainly corticosteroids and azathioprine. Sign

Disturbances in dental development were studied with the use of radiography and histology in a patient with acute lymphoblastic leukemia who was treated with induction chemotherapy at 2.3 years of age and bone marrow transplantation at 4.3 years of age. The follow-up 9.5 years after bone marrow transplantation showed evidence of short tapered roots, enamel hypoplasia, microdontia, and aplasia. A histologic examination of two extracted permanent teeth showed that the crown of the maxillary lateral incisor exhibited numerous incremental lines that corresponded closely to the treatment periods wi
