Borrelia burgdorferi sensu lato and spotted fever group rickettsiae in small rodents and attached ticks in the Northern Apennines, Italy.

Ticks and ear biopsies were collected from wild small rodents in 2011 and 2012 in the northern Apennines (Italy), up to 1650 m above sea level. Apodemus spp. (n = 83) and Myodes glareolus (n = 22) were infested by Ixodes ricinus (192 larvae and two nymphs), Dermacentor marginatus (179 larvae and 29 nymphs), and Ixodes trianguliceps (three larvae and two nymphs). We detected several Borrelia burgdorferi sensu lato (s.l.) genospecies (B. afzelii, B. burgdorferi sensu stricto, B. garinii, B. lusitaniae, B. valaisiana) in I. ricinus and skin biopsies. The most common genospecies found in I. ricinu

A retrospective study of the characterization of Rickettsia species in ticks collected from humans.

Rickettsiae (family Rickettsiaceae, order Rickettsiales) are obligate intracellular bacteria transmitted by arthropod vectors. Several Rickettsia species causing vector-borne rickettsioses belong to the spotted fever group (SFG). Traditionally, Rickettsia conorii has been considered as the main etiologic agent of Mediterranean spotted fever. However, the molecular characterization of rickettsiae allowed identifying other species involved in spotted fever in the Mediterranean region. In this study, 42 ticks collected from humans were subjected to morphological identification and molecular chara

Neuropeptide signaling sequences identified by pyrosequencing of the American dog tick synganglion transcriptome during blood feeding and reproduction.

Ticks are important vectors of numerous pathogens that impact human and animal health. The tick central nervous system represents an understudied area in tick biology and no tick synganglion-specific transcriptome has been described to date. Here we characterize whole or partial cDNA sequences of fourteen putative neuropeptides (allatostatin, insulin-like peptide, ion-transport peptide, sulfakinin, bursicon alpha/beta, eclosion hormone, glycoprotein hormone alpha/beta, corazonin, four orcokinins) and five neuropeptide receptors (gonadotropin receptor, leucokinin-like receptor, sulfakinin recep

Genotypic identification of three new strains of spotted fever group rickettsiae isolated in China.

Polymerase chain reaction (PCR) and restriction endonuclease fragment length polymorphism (RFLP) analysis were used to characterize the genotypic diversity of three isolates of spotted fever group (SFG) rickettsiae isolated from ticks in China. A primer pair designed from DNA sequence encoding 190 K protein antigen of R. rickettsii and genomic DNAs obtained from the isolates were used in PCR. The PCR products were cleaved with restriction endonucleases PstI and RsaI, and the digestion patterns were analyzed by polyacrylamide gel electrophoresis (PAGE) and compared with those of all known speci

Rocky Mountain spotted fever (tick-borne typhus) in South Carolina: an educational program and tick/rickettsial survey in 1973 and 1974.

Because the incidence of spotted fever is increasing in South Carolina, campaigns were carried out in 1973 and 1974 to provide the public and medical practitioners, through pamphlets and news media, with information about spotted fever and the ticks which transmit the causative agent, Rickettsia rickettsii. People were also invited to save and submit live ticks removed from vegetation, animals and humans, for examination by the hemolymph test. A total of 1,186 ticks consisting of 987 Dermacentor variabilis, 103 Rhipicephalus sanguineus, and 96 Amblyomma americanum were examined. Rickettsiae id

Comparative mitogenomic and phylogenetic insights from four newly sequenced tick mitochondrial genomes.

Ticks, recognized as the second most significant vector of pathogens after mosquitoes, are of considerable interest in medical research. Although mitochondrial genomes are commonly employed in the phylogenetic studies of insects and arthropods, investigations into tick mitochondrial genomes are relatively scarce. The evolutionary and phylogenetic relationships at the family and genus levels remain unresolved. In this study, the mitochondrial genomes of Haemaphysalis warburtoni, Ixodes crenulatus, Rhipicephalus bursa, and Rhipicephalus pumilio were first sequenced and annotated based on the Ill

Genome sequencing and multi-stage, blood-feeding, and tissue-specific transcriptome atlas of the Rocky Mountain wood tick provide a critical resource for this vector.

Dermacentor andersoni, the Rocky Mountain wood tick, is an important vector for pathogens impacting human and animal health, including bovine anaplasmosis, Colorado tick fever, and Rocky Mountain spotted fever. A better understanding of the biology of this tick is needed for developing disease prevention and vector control strategies. A reference genome was assembled for D. andersoni using high-fidelity (HiFi) long-read PacBio sequences and Hi-C contact mapping, yielding a contiguous assembly in which most contigs matched one of 11 chromosomes. Characterization of Tick-Borne Encephalitis Virus Isolates from Ixodes persulcatus Ticks Collected During 2020 in Selenge, Mongolia.

Tick-borne encephalitis virus (TBEV) causes neurological disease in humans, with varied clinical severity influenced by the viral subtype. TBEV is endemic to Mongolia, where both Siberian and Far-Eastern subtypes are present. Ixodes persulcatus is considered the main vector of TBEV in Mongolia; although, the virus has also been detected in Dermacentor species. To further characterize the disease ecology of TBEV within the endemic Selenge province of Mongolia, 1300 Ixodes persulcatus ticks were collected in May 2020 from regions outside Ulaanbaatar. Pooled tick samples (n = 20-50) were homogeni

Mapping and monitoring tick (Acari, Ixodida) distribution, seasonality, and host associations in the United Kingdom between 2017 and 2020.

Tick-borne disease risk is intrinsically linked to the distribution of tick vector species. To assess risk and anticipate disease emergence, an understanding of tick distribution, host associations, and seasonality is needed. This can be achieved, to some extent, using passive surveillance supported by engagement with the public, animal health, and public health experts. The Tick Surveillance Scheme (TSS) collects data and maps tick distribution across the United Kingdom (UK). Between 2017 and 2020, 3720 tick records were received and 39 tick species were detected. Most records were acquired i

A Francisella tularensis Chitinase Contributes to Bacterial Persistence and Replication in Two Major U.S. Tick Vectors.

Nearly 100 years after the first report of tick-borne tularemia, questions remain about the tick vector(s) that pose the greatest risk for transmitting Francisella tularensis (Ft), the causative agent of tularemia. Additionally, few studies have identified genes/proteins required for Ft to infect, persist, and replicate in ticks. To answer questions about vector competence and Ft transmission by ticks, we infected Dermacentor variabilis (Dv),Amblyomma americanum (Aa), and Haemaphysalis longicornis (Hl; invasive species from Asia) ticks with Ft, finding that although Aa ticks initially become i

Molecular identification of vector-borne organisms in Ehrlichia seropositive Nicaraguan horses and first report of Rickettsia felis infection in the horse.

Certain vector-borne organisms serve as etiological agents of equine disease. After previously identifying a new Ehrlichia species in horses from Mérida, we aimed to determine the infection frequency and screen for a wide range of vector-borne organisms from 93 tick-exposed, Ehrlichia seropositive horses in this region. PCR assays were performed to identify infection by organisms within the following genera: Anaplasma, Babesia, Bartonella, Ehrlichia, Leishmania, Mycoplasma, Neorickettsia, Rickettsia and Theileria. Overall, 90/93 horses (96.8%) were infected with one or more vector-borne organi

Detection of a Borrelia sp. From Ixodes granulatus Ticks Collected From Rodents in Malaysia.

The Borrelia genus consists of spirochete bacteria known to cause Lyme disease (LD) and relapsing fever in humans. Borrelia pathogens are commonly transmitted via arthropod vectors such as ticks, mites, or lice. Here, we report the molecular screening of LD group Borrelia sp. from ticks (Acari: Ixodidae) collected from rodents trapped in recreational forests and a semiurban residential area in the Selangor state in Malaysia. Of 156 adult ticks surveyed, 72 ticks were determined as positive for Borrelia sp. by polymerase chain reaction (PCR). All Borrelia PCR-positive ticks belonged to the Ixod

Rickettsioses in Europe.

Bacteria of the genera Rickettsia and Orientia (family rickettsiaceae, order rickettsiales) cause rickettsioses worldwide, and are transmitted by lice, fleas, ticks and mites. In Europe, only Rickettsia spp. cause rickettsioses. With improvement of hygiene, the risk of louse-borne rickettsiosis (epidemic typhus) is low in Europe. Nevertheless, recrudescent form of Rickettsia prowazekii infection persists. There could be an epidemic typhus outbreak if a body lice epidemic occurs under unfavorable sanitary conditions. In Europe, endemic typhus or Rickettsia typhi infection, transmitted by rats a
