Topical delivery of bioactive compounds from Cortex Dictamni alleviates atopic dermatitis-like lesion by inhibiting the activation of keratinocytes, macrophages, and basophils: Dictamnine versus fraxinellone.

Dictamnine and fraxinellone constitute the primary alkaloid and limonoid components in Cortex Dictamni, respectively. Both compounds exhibit anti-inflammatory properties. This study aims to assess the ability of dictamnine and fraxinellone in treating atopic dermatitis (AD) through in silico-, cell-, and animal-based experiments. The effects of these compounds on the coordinated activation of keratinocytes, macrophages, and basophils in AD development were investigated. A dinitrochlorobenzene (DNCB)-sensitized AD model in mice was employed to examine the in vivo anti-AD effects. Dictamnine and

Bakuchicin attenuates atopic skin inflammation.

Psoralea corylifolia is a medicinal herb that provides advantageous pharmacological effects against vitiligo and skin rash. Former studies have shown that bakuchicin, a furanocoumarin compound from the fruits of P. corylifolia, has therapeutic effects against inflammation, and infection. This study aimed to define the pharmacological effects of bakuchicin on inflammatory responses and lichenification, the major symptoms of atopic dermatitis (AD). To induce AD-like skin inflammation, we exposed the ears of female BALB/c mice to 2, 4-dinitrochlorobenzene (DNCB) and Dermatophagoides farinae (hous

UV protective effects of DNA repair enzymes and RNA lotion.

Solar UV radiation is known to cause immune suppression, believed to be a critical factor in cutaneous carcinogenesis. Although the mechanism is not entirely understood, DNA damage is clearly involved. Sunscreens function by attenuating the UV radiation that reaches the epidermis. However, once DNA damage ensues, repair mechanisms become essential for prevention of malignant transformation. DNA repair enzymes have shown efficacy in reducing cutaneous neoplasms among xeroderma pigmentosum patients. In vitro studies suggest that RNA fragments increase the resistance of human keratinocytes to UVB

Topical application of silymarin reduces chemical-induced irritant contact dermatitis in BALB/c mice.

Irritant contact dermatitis (ICD) is a non-allergic local inflammatory reaction of a skin and one of the most frequent occupational health problems. Silymarin has been clinically used in Europe for a long time to treat liver diseases and also known to have anti-cancer and anti-inflammatory activities. In the present study, we report that topical application of silymarin reduces chemical-induced ICD. Topical application of 2,4-dinitrochlorobenzene (DNCB) induced an ear swelling in BALB/c mice and silymarin suppressed DNCB-induced increase in ear thickness. Prophylactic and therapeutic applicati

Assessment of contact allergen cross-reactivity by retesting.

At former allergic contact dermatitis reaction sites retesting causes augmented hyper-reactivity, characterized by an accelerated onset within a few hours. This expression of 'local skin memory' has been ascribed to locally persisting allergen-specific effector/memory T cells. To verify this hypothesis, we investigated whether accelerated retest reactivity also occurs with cross-reactive allergens. Guinea pigs were immunized with either or both 2,4-dinitrochlorobenzene (DNCB) and 2-hydroxyethyl methacrylate (HEMA), and primary skin tests to these and cross-reactive methacrylic compounds were p

Profile of patients with chemical injury and sensitivity.

Patients reporting sensitivity to multiple chemicals at levels usually tolerated by the healthy population were administered standardized questionnaires to evaluate their symptoms and the exposures that aggravated these symptoms. Many patients were referred for medical tests. It is thought that patients with chemical sensitivity have organ abnormalities involving the liver, nervous system (brain, including limbic, peripheral, autonomic), immune system, and porphyrin metabolism, probably reflecting chemical injury to these systems. Laboratory results are not consistent with a psychologic origin

Mesenchymal stem cell-derived extracellular vesicles exert Th1-mediated anti-inflammatory effects via miR-146a/NF-κB pathway: comparison with dupilumab in a mouse model of atopic dermatitis.

BACKGROUND: Atopic dermatitis (AD) is a chronic inflammatory skin disease primarily treated with corticosteroids and dupilumab, a monoclonal antibody targeting interleukin (IL)-4 and IL-13. Mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) are promising alternatives owing to their anti-inflammatory properties. This study compared the therapeutic effects of dupilumab and MSC-EVs in a murine model of AD. We employed clinical, serological, and histological analyses to assess the efficacy of these treatments and investigated their mechanisms in vitro and in vivo. METHODS: We generate

Possible prenatal impact of sertraline on human placental glutathione S-transferase-π.

Sertraline (SER), a tricyclic antidepressant, is considered to belong to the group of selective amine reuptake inhibitors. Its ability to cross the blood-brain barrier and transplacental transport has been reported previously. It is widely distributed in the brain and is bound to human glutathione S-transferase-π (GST-π). If SER is taken during pregnancy, it gets accumulated in the embryo and fetus, and some studies have suggested it may cause congenital malformations, thus the study of the interaction of GST-π with antidepressants is crucial. In this study, the interaction of human placental

Identification of an enzymatically formed glutathione conjugate of the cyanobacterial hepatotoxin microcystin-LR: the first step of detoxication.

Cyanobacterial toxins have adverse effects on mammals, birds and fish and are being increasingly recognised as a potent stress factor and health hazard factor in aquatic ecosystems. Microcystins, cyclic heptapeptides and a main group of the cyanotoxins are mainly retained within the producer cells during cyanobacterial bloom development. However, these toxins are released into the surrounding medium by senescence and lysis of the blooms. Any toxin present could then come into contact with a wide range of aquatic organisms including phytoplankton grazers, invertebrates, fish and aquatic plants.

Topical application of artesunate on guinea pig allergic contact dermatitis.

Artesunate is derived from the Chinese medicinal herb qinghao. Many animal studies suggest that systemic artesunate has immunoregulatory activity. We investigated the effect of topical artesunate on contact sensitivity in guinea pigs sensitized with DNCB. Female hairless guinea pigs were used and the contact reaction graded by visual (5 point) score and erythema measured with a color analyzer. Topical artesunate inhibited the elicitation reaction of contact hypersensitivity when given at the 1st and 12th h after challenge (p < 0.05), but showed no effect when given from day 3 to day 1 before c

Decreased glutathione transferase activities in zinc-deficient rats.

Weanling rats fed a zinc-deficient diet for 56 days exhibit a decrease in the total activities of glutathione transferases with 1-chloro-2,4-dinitrobenzene (CDNB) as substrate in the liver, esophagus and stomach. The level of acid-soluble sulfhydryl group, on the other hand, is decreased in the liver, but elevated in the esophagus and kidney. Deficient rats fed a zinc-replenished diet for just 7 days have a fully restored glutathione transferase activity in all tissues except the liver. In view of our earlier findings that the carcinogenic effects of nitrosamines are enhanced in zinc-deficient

Thermochemiluminescent assay of porcine, rat, and human erythrocytes for antioxidative deficiencies.

The thermal induction of chemiluminescence of luminol-horseradish peroxidase-labeled erythrocytes from pigs, rats, and man was studied. The luminescent responses of rat, porcine, and human erythrocytes to heating were linear in respect to logs of counts per minute versus temperature. Landrace-Duroc crossbred pigs with a history of malignant hyperthermia (porcine stress syndrome) and Poland-China-miniature pigs inbred for malignant hyperthermia (MH) yielded erythrocytes with high-level thermochemiluminescence (TCL). Sprague-Dawley rat erythrocytes were intermediate in their production of TCL. N

[Cellular immunity indices in primary cancer and nontumorous diseases of the liver].

By means of a delayed hypersensitivity reaction to 2,4 DNCB, tuberculin and quantitation of T-RFC it was shown that cell immunity indices in primary hepatic cancer are reduced to a greater extent than in persons with nontumor lesions. Persistant opisthorchosis invasion is of great significance in impairing the cell immunity in man, which is an immunological support to the idea of opisthorchosis being precancer of the liver.
