A hydrogen peroxide-responsive near-infrared fluorescent probe for visualizing redox-associated signals in acute colitis and gastrointestinal tumor models and monitoring treatment-related changes.

Inflammatory bowel disease and gastrointestinal tumors are accompanied by redox imbalance, yet noninvasive tools for visualizing hydrogen peroxide-associated redox changes and following responses to intervention remain limited. This study aimed to develop and preclinically evaluate HD-YSY, a boronate-masked hemicyanine compound activated by hydrogen peroxide. High-performance liquid chromatography supported formation of the deprotected fluorophore HD. Under the stated solution conditions, the probe produced first-window near-infrared emission at 760 nm under 670 nm excitation, showed a rapid f

A rapid and selective phenothiazine-derived fluorescent probe for hypochlorous acid monitoring in living systems: from cellular imaging to in vivo systemic inflammatory response syndrome visualization.

Hypochlorous acid (HClO) is a pivotal constituent of the reactive oxygen species (ROS) family, playing a pivotal role in physiological processes such as immune defence and cellular signalling. However, it is important to note that excessive hypochlorous acid production is closely associated with various pathological conditions, including inflammation, neurodegenerative diseases, and cancer. Consequently, the development of tools capable of real-time, highly selective, and sensitive detection of HClO in biological systems is imperative for enhancing our comprehension of its mechanisms in life p

High-Throughput Screening of Natural Products Alleviating Acute Liver Injury Using a Polarity-Responsive NIR Ratiometric Fluorescent Probe.

Acute liver injury (ALI) is a rapidly progressive inflammatory disease lacking precise noninvasive early diagnostic tools, as traditional enzyme-based tests frequently yield false-positive results. Polarity is a crucial physiological indicator, and its abnormal fluctuations are deeply involved in ALI pathogenesis, necessitating accurate in vivo monitoring. We rationally designed BDP-JL, a new near-infrared ratiometric fluorescent probe featuring a D-π-A architecture for highly sensitive polarity detection. It possesses excellent vesicle-targeting capabilities, enabling precise tracking of pola

The First NIR Fluorescent Probe for Bioimaging of Cystathionine-γ-lyase Activity in Living Cells and Xenograft Tumor Models.

Cystathionine γ-lyase (CSE) is a pivotal enzyme in the transsulfuration pathway and the biosynthesis of hydrogen sulfide, with strong implications for regulating cellular redox homeostasis and tumor progression. Real-time and in situ detection of CSE activity in living systems remains a significant challenge. To address this, we herein report a novel near-infrared (NIR) fluorescent probe, NIR-CSE, for the selective and sensitive monitoring of CSE in living cells and xenograft tumor models. The probe features an NIR fluorophore Cy-NH2 tethered to l-homocysteine (recognition unit) methyl ester v

Multifunctional Fluorescent Schiff Base Probe Derived From Chlorosalicylaldehyde for Selective Detection of Proteins, Nitroexplosives, Metal Ions and Antibiotics.

Multifunctional fluorescent chemosensors capable of detecting chemically diverse analytes using a single molecular platform are increasingly valuable for environmental monitoring and biomedical analysis due to reducing analytical cost, time, and complexity. However, most Schiff base probes are limited to single-analyte detection or operate through a single sensing mechanism. In this study, a new Schiff base probe, ClSNP, synthesized from 3,5-dichlorosalicylaldehyde and N-phenyl-o-phenylenediamine, exhibited a fluorescence "turn-off" response toward Cu2 + and Co2 + ions in a CH3CN/H2O medium wi

An Environment-Sensitive Turn-On Fluorescent Probe Enables Detection of PAD4 Activation in Response to Distinct NETosis Stimuli.

Protein arginine deiminase 4 (PAD4) is a crucial regulator of human neutrophils, playing a central role in the innate immune response as well as the initiation and progression of several immune diseases, including rheumatoid arthritis, cancer, and neurodegeneration. Current approaches for detecting PAD4 activity employ methods that cannot be used in living cells, such as immunodetection or chemical probes that need fixed material or harsh experimental conditions. As a result, they provide limited information on the real-time dynamics of PAD4 activation and localization during neutrophil activa

A near-infrared fluorescent probe with rapid-response hypochlorite for imaging rheumatoid arthritis and wound healing in vivo.

Real-time monitoring of hypochlorite (ClO-) fluctuations in inflamed tissues is crucial for evaluating the onset, progression, and prognosis of inflammatory diseases such as rheumatoid arthritis (RA). In this study, we rationally developed a small-molecule fluorescent probe (SX-ClO) based on a malononitrile-isophorone fluorophore skeleton coupled with a diaminomaleonitrile moiety as the ClO--responsive unit. The probe operated through an intramolecular ICT modulation mechanism upon specific reaction with ClO-, enabling a remarkable fluorescence turn-on response with high selectivity, excellent

Super-resolution imaging with a mitochondria-targeted fluorescent probe: Evaluation of chemotherapy-induced hepatorenal injury.

With the rising global incidence of cancer, anticancer drugs including cisplatin (DDP) and sunitinib (SU) are being widely used in clinical practice. However, severe nephrotoxicity of DDP and prominent hepatotoxicity of SU drastically restrict their clinical application. Therefore, real-time monitoring of liver and kidney function, along with early intervention of tissue damage progression, is essential during treatment. To address this challenge, we have herein developed a mitochondria-targeted super-resolution fluorescent probe ARP for hypochlorous acid (HClO). ARP's dual recognition sites a

H₂S-Activatable dual-responsive fluorescent probe for bicolor visualization of Ferroptosis and drug-induced liver injury models.

Hydrogen sulfide (H2S) is a gas with dual roles in both environmental and biological systems. Although it is a natural atmospheric constituent and an enzymatically produced signaling molecule, elevated levels of H2S are associated with air pollution, cytotoxicity, and various diseases. Current methods for detecting H₂S are often limited by complex sample preparation, low sensitivity, and inadequate selectivity. To address these challenges, we report the development of a novel dual-channel and dual-responsive fluorescent probe HAR-H₂S, which enables rapid, sensitive, and selective detection of

Simultaneous detection of H₂S and viscosity in pancreatic cancer with a lysosome-targeted NIR fluorescent probe.

Pancreatic cancer is one of the most aggressive malignancies and has one of the lowest survival rates among all major cancers. Accurate diagnosis at an early stage can improve patient survival. Near-infrared fluorescent (NIRF) probes have been emerged as promising tools for tumor imaging due to their superior optical properties. However, most probes suffer from significant limitations in the simultaneous detection of multiple tumor biomarkers, which may lead to false-positive signals. Herein, we constructed a novel lysosome-targeted NIRF probe (LNX-H2S) incorporating an azide moiety and an eth

A Solid-State Near-Infrared Fluorescent Probe by a Synergistic Extended Conjugated System for Detecting Cys with Long-Term Imaging in Orthotopic Bladder Cancer.

Bladder cancer (BC) is a highly prevalent malignant tumor of the urinary system with high recurrence and mortality rates. Currently, urine cytology and imaging techniques exhibit insufficient sensitivity for low-grade early tumors, while clinically used surgery navigation agents suffer from short tumor retention time. To address these limitations, this study develops a solid-state fluorescent probe, HPQ-TH-BI-Cys, enabling a specific response to cysteine (Cys) for precise identification and long-term imaging of bladder cancer. The solid-state fluorophore (HPQ-TH-BI) is designed by incorporatin

Recent advancements in ubiquitous colorimetric and fluorometric sensors for the detection of methanol.

Methanol, an aliphatic alcohol, is a widely recognized volatile organic compound (VOC) extensively used as a cost-effective solvent in various chemical, agricultural, and biological industries. However, the intentional adulteration of alcoholic beverages with inexpensive methanol for economic gain poses significant health hazards, including birth defects, respiratory illnesses, nervous system damage, and various chronic conditions. Prolonged exposure to methanol vapour can result in adverse effects such as eye inflammation, dizziness, headaches, insomnia, stomach problems, visual impairment, a

Fluorination-enhanced ESIPT probes based on 3-hydroxyflavone for cysteine detection in hydrophobic environments and living cells.

Excited-state intramolecular proton transfer (ESIPT) fluorophores, particularly 3-hydroxyflavone derivatives, are broadly utilized in fluorescent probe design due to their distinct photophysical properties. However, their practical applications are often hindered by inherent limitations. Herein, we report a strategy via site-specific fluorination at the 2-phenyl position of the 3-hydroxyflavone scaffold to improve their photophysical performance. Two fluorinated ESIPT fluorophores (PHF and PFF) were synthesized and further functionalized with an acrylate recognition moiety to afford the turn-o

Mild synthesis of biomass-derived fluorescent Fe,N-carbon dots for fluorometric and smartphone-assisted visual detection of aloin.

Herein, a mild one-pot synthesis of fluorescent Fe,N-doped carbon dots (Fe-NCDs) was developed from biomass waste in a choline chloride/urea deep eutectic solvent (DES) with FeCl3 as a promoter. The DES disrupts the lignocellulosic network while FeCl3 catalyzes carbonization and provides in situ Fe doping. The resulting Fe-NCDs exhibited bright blue emission with a high quantum yield of 38.5% owing to the introduction of Fe and N elements. The probe showed outstanding environmental stability but could be selectively fluorescence quenched toward aloin because of static coordination and the inne

Ultra-large Stokes-shifted NIR fluorescent probe for diagnosis of APAP-induced liver injury and its applications in food and bioimaging.

Cysteine (Cys), an essential biothiol in the human body, plays a significant role in physiological and pathological processes and is closely associated with various diseases. Therefore, the detection of Cys is very important for exploring the metabolic and physiological processes of Cys, which can be beneficial for early disease diagnosis and assessment of progression. In this study, we designed a fluorescent probe named PEN-Cys, which is a D1-π-A-π-D2-π-A-π-D1 system that specifically responds to Cys via an intramolecular charge transfer (ICT) pathway. The probe employs acrylate as the Cys re

Synthesis of fluorescent iron-doped carbon dots with Fenton activity for noninvasive dual-mode urinary uric acid monitoring toward point-of-care renal diagnostics.

Rapid, selective and point-of-care urinary uric acid (UA) monitoring is important for managing renal and metabolic disorders. This study employed solvothermal synthesis of fluorescent iron-doped carbon dots (Fe-CDs) exhibiting a fluorescence quantum yield of 44.5%. Pronounced fluorescence quenching of Fe-CDs was observed upon the introduction of hydrogen peroxide (H₂O₂), attributable to a Fenton reaction-based mechanism. In contrast, non-doped CDs exhibited no fluorescence quenching in the presence of H₂O₂, confirming the essential role of iron doping in the quenching process. The uricase-cata

Real-Time Detection of Colorectal Cancer Using Topical Application of a pH-Activatable Fluorophore.

INTRODUCTION: Standard treatment for locally advanced rectal cancer involves neoadjuvant chemoradiation therapy (nCRT) followed by total mesorectal excision, but this approach carries significant morbidity and often results in incomplete resections owing to poor intraoperative tumor visualization. For patients with complete response to nCRT, a watch-and-wait (W&W) strategy can spare surgery, but current imaging techniques inadequately identify complete responders, leading to regrowth in ~ 30% of cases. To improve nCRT response assessment and guide resections, we developed PH10, a topically app
