Accumulating evidence highlights the importance of the endocannabinoid anandamide (AEA) as a key mediator in reproductive physiology. Current data suggest potential roles for AEA in gametogenesis, fertilization, and parturition. AEA exerts its actions through two G protein-coupled receptors, termed cannabinoid receptor 1 (CB1), and 2 (CB2), and the ligand-gated transient receptor potential vanilloid receptor type 1 (TRPV1) ion channel. At present, the cellular mechanism(s) and consequences of AEA signaling in reproductive tissues, especially the myometrium, are poorly understood. Here, we exam

Dietary exposure is a recognized route of metal toxicity, yet its mechanistic impacts on aquatic organisms remain poorly understood. In this study, adult zebrafish (Danio rerio, five months old, both sexes) were exposed to dietary Cu (2.4 (Control), 4.4 (Cu-L), 9.8 (Cu-M), and 54.4 (Cu-H) μg/g ww) for 28 days to elucidate tissue-specific Cu accumulation, cuproptosis-related toxicity, intestinal disruption, reproductive effects, and their potential linkage through the gut-gonadal axis. Cu concentrations significantly increased in all examined tissues of both sexes, with tissue-specific fold cha

Moonfish (Mene maculata) has been recognized as a prospective commercial species in Taiwanese waters; however, its diet and trophic ecology remain poorly understood. It was hypothesized that diet composition and trophic position (TP) vary with body size and monsoonal environmental changes. This study presents the first investigation of its feeding ecology in southwest Taiwan (SWT), focusing on seasonal and ontogenetic variation. A total of 2329 individuals collected from September 2021 to May 2024 were analyzed using stomach content analysis (SCA) and stable isotope analysis (SIA). Eight zoopl

Serine/arginine-rich protein kinases (SRPKs) are cell cycle-regulated serine/threonine protein kinases and are important regulators of splicing factors. In this study, we functionally characterize SRPK1 of the human malaria parasite Plasmodium falciparum. P. falciparum SRPK1 (PfSRPK1) was expressed in asexual blood-stage and sexual-stage gametocytes. Pfsrpk1- parasites formed asexual schizonts that generated far fewer merozoites than wild-type parasites, causing reduced replication rates. Pfsrpk1- parasites also showed a severe defect in the differentiation of male gametes, causing a complete

BACKGROUND: Mammalian spermatogenesis is a process that involves a complex expression program in both somatic and germ cells present in the male gonad. A number of studies have attempted to define the transcriptome of male meiosis and gametogenesis in rodents and primates. Few human transcripts, however, have been associated with testicular somatic cells and germ cells at different post-natal developmental stages and little is known about their level of germline-specificity compared with non-testicular tissues.
METHODS: We quantified human transcripts using GeneChips and a total of 47 biopsie

BACKGROUND AND PURPOSE: The endocannabinoid plays vital roles in several aspects of reproduction, including gametogenesis, fertilization and parturition. However, little is known regarding the presence or role of the endocannabinoid system in myometrial function. Here the presence of the endocannabinoid system and signalling properties of cannabinoid receptors were characterized.
EXPERIMENTAL APPROACH: Components of the endocannabinoid system were identified using qRT-PCR, immunohistochemical, immunoblotting and radioligand binding experiments. Cannabinoid receptor signalling pathways were ch

Acyl-CoA binding proteins (ACBPs) are conserved intracellular lipid-binding proteins found across eukaryotes that play essential roles in fatty acid metabolism, membrane biogenesis, and cellular energy homeostasis. In this study, the gene ENH_00086110, which encodes a novel ACBP-family protein designated EnACBP, was cloned and characterized from Eimeria necatrix. The deduced mature protein contains 511 aa with a calculated molecular mass of 56.18 kDa. Bioinformatic analysis indicated that EnACBP contains a conserved acyl-CoA binding (ACB) domain in its C-terminal region (residues 424-506) and

Steroid hormones are indispensable for regulating the typical reproductive processes during the development of gonads. This study aimed to clarify the expression patterns of androgen receptors (Ars) and estrogen receptors (Ers) in Pampus argenteus to better understand gonadal development and improve artificial breeding. In this study, the androgen receptors (arα and arβ) and estrogen receptors (esr1, esr2a, and esr2β) of P. argenteus were cloned and the expression profiles were detected by qPCR. The results showed that the highest expression of arα was found in hypothalamus, while arβ was main

The ovarian follicle is the functional unit of the ovary that produces hormones and gametes needed to sustain female reproductive function and health. The ability to recapitulate folliculogenesis, ovulation, and luteinization in vitro has broad basic, translational, and clinical utility. The most advanced in vitro follicle growth systems maintain the follicle's three-dimensional (3D) architecture, which is crucial for the development of meiotically competent metaphase II oocytes in humans. Recently, a scaffold-free method for in vitro follicle growth of mouse multilayer secondary follicles was

BACKGROUND: Microbial communities are increasingly recognized for their essential roles in the reproductive system. However, the microbial communities in healthy gonads-neither in the ovary nor the testis-have not been extensively explored, particularly with respect to sex differentiation. Sex reversal is a unique mode of sex differentiation that is a well-documented phenomenon in various animal species, with the swamp eel (Monopterus albus) being a notable example of a hermaphroditic species that undergoes natural female-to-male sex reversal. Thus, swamp eel offers a robust system for explori

Seasonality considerably impacts on the life of organisms and leads to numerous evolutionary adaptations. Some species face seasonal changes by entering a diapause during different life stages. During adulthood, a diapause in the non-reproductive period can affect male gametogenesis as, for example, it occurs in insects. Spiders are distributed worldwide and show a variety of life cycles. However, data on spiders' life cycles and seasonal adaptations are limited. Here, we explored the effect of reproductive diapause in a seasonal spider for the first time. We used the South American sand-dwell

BACKGROUND: Hybridization events between Triatoma spp. have been observed under both natural and laboratory conditions. The ability to produce hybrids can influence different aspects of the parent species, and may even result in events of introgression, speciation and extinction. Hybrid sterility is caused by unviable gametes (due to errors in chromosomal pairing [meiosis]) or by gonadal dysgenesis (GD). All of the triatomine hybrids analyzed so far have not presented GD. We describe here for the first time GD events in triatomine hybrids and highlight these taxonomic and evolutionary implicat

Although the insulin-like peptide hormone INSL3 and its cognate receptor RXFP2 (relaxin-family peptide receptor 2) have existed throughout chordate evolution, their physiological diversification appears to be linked closely with mammalian emergence and radiation. In contrast, they have been lost in birds and reptiles. Both hormone and receptor are expressed from autosomal genes which have maintained their synteny across vertebrate evolution. Whereas the INSL3 gene comprises only two exons closely linked to the JAK3 gene, RXFP2 is normally encoded by 18 exons. Both genes, however, are subject t

Gametocyte proteins of Eimeria spp. are essential components of the oocyst wall, and some of these proteins have been analysed to identify targets of transmission-blocking vaccines against avian coccidiosis. In the present study, a cDNA from E. necatrix gametocytes was cloned and sequenced. The cDNA is 1473 bp in length and encodes a 490-amino-acid protein containing a tyrosine-serine (Tyr/Ser)-rich domain and a proline-methionine (Pro/Met)-rich domain. A quantitative real-time PCR (qPCR) analysis showed that the cDNA is expressed only during gametogenesis. A fragment containing the Tyr/Ser-ri

Mouse embryonic stem cells (ESCs) have the potential to differentiate into germ cells (GCs) in vivo and in vitro. Interestingly, XY ESCs can give rise to both male and female GCs in culture, irrespective of the genetic sex. Recent studies showed that ESC-derived primordial GCs contributed to functional gametogenesis in vivo; however, in vitro differentiation techniques have never succeeded in generating mature oocytes from ESCs due to cryptogenic growth arrest during the preantral follicle stages of development. To address this issue, a mouse ESC line, capable of producing follicle-like struct

An efficient animal cloning technology would provide many new opportunities for livestock agriculture, human medicine, and animal conservation. Nuclear cloning involves the production of animals that are genetically identical to the donor cells used in a technique known as nuclear transfer (NT). However, at present it is an inefficient process: in cattle, only around 6% of the embryos transferred to the reproductive tracts of recipient cows result in healthy, longterm surviving clones. Of concern are the high losses throughout gestation, during birth and in the post-natal period through to adu

Somatic portions of gonads in two phanerozonian sea-stars, Ctenodiscus crispatus and Hippasteria phrygiana, were similar in all aspects of gross structure and histology seen previously in both forcipulate and spinulosan asteroids. For the first time, detailed ultrastructural observations have been made of cells and tissues that reveal several features believed to be of universal occurrence in the gonads of asteroids. These include flagellated-collar cells in the visceral peritoneum and other coelomically derived epithelia, muscular-flagellated-collar cells in the visceral peritoneum and genita

The requirement for next-generation antimalarials to be both curative and transmission-blocking necessitates the identification of previously undiscovered druggable molecular pathways. We identified a selective inhibitor of the Plasmodium falciparum protein kinase PfCLK3, which we used in combination with chemogenetics to validate PfCLK3 as a drug target acting at multiple parasite life stages. Consistent with a role for PfCLK3 in RNA splicing, inhibition resulted in the down-regulation of more than 400 essential parasite genes. Inhibition of PfCLK3 mediated rapid killing of asexual liver- and

Fertility traits are pivotal to the productivity and sustainability of dairy production systems; however, their genetic improvement remains constrained due to complex polygenic control and low heritability. Genome-wide association studies (GWAS) offer a powerful approach to identify genomic regions underlying reproductive traits, particularly in tropically adapted indigenous breeds like Sahiwal cattle, which exhibit resilience to environmental stress. This study aimed to elucidate the genetic basis of 8 fertility traits in 303 Sahiwal animals using 50 K SNP genotyping. Animals were genotyped w

BACKGROUND: The golden pompano (Trachinotus ovatus) is an economically significant warm-water aquaculture species in China. The time required for sexual maturity of T. ovatus is relatively long. Consequently, it has prompted researchers to investigate gonadal development process of this fish. To gain further insight into the function of competing endogenous RNA (ceRNA) in the gonads of T. ovatus and the regulatory mechanism of the ceRNA network, whole transcriptome libraries were constructed from the testes and ovaries.
RESULTS: Overall, a total of 96 differentially expressed microRNAs (DE-mi
