Fonsecaea pedrosoi is a fungal pathogen that produces melanin. The functions of melanin and its possible influence in the protective immunological response during infection by F. pedrosoi are not known. In this work, treatment of F. pedrosoi mycelia with proteases and glycosidases followed by a denaturing agent and hot concentrated acid left a black residue. Scanning electron microscopy demonstrated that this processed melanized residue resembled very closely the intact mycelium in shape and size. Melanin particles were also isolated from culture fluids of conidia or sclerotic forms of F. pedr

Mouldy hay was produced in the laboratory by sterilising good hay, inoculating with aqueous suspensions of microorganisms, and incubating at 40 degrees or 60 degrees C. Extracts were tested for presence of farmer's lung hay (F.L.H.) antigen by agar-gel double-diffusion and immunoelectrophoresis tests against sixteen to twenty sera from patients with farmer's lung. F.L.H. antigen developed in hay after: (1) inoculating with mixed microbial suspensions from antigenically active hay; (2) inoculation with mixed suspensions of pure cultures of thermophilic actinomycetes, after raising the pH of the

At present the main part of the mycelial waste of antibiotic production is collected in sludge lagoones, discarded to the sewage treatment system or burnt. The mycelial waste is one of the causes of the environmental pollution. It contains a significant quantity of valuable substances, such as lipids, carbohydrates, proteins and amino acids. The inorganic portion of the waste includes the compounds of calcium, magnesium, sodium, iron, silicium and phosphorus. The levels of the above substances in the mycelial waste allow using it as organic mineral additives in making expanded clay aggregate g

In the face of the global obesity epidemic (globesity), we present the first comprehensive investigation of fermented Sanghuangporus sanghuang mycelia extract (SS-IM1) and its novel bioactive compound hypholomine B in brown adipose tissue activation. Using HEK293 cells with FNDC5 promoter-EGFP constructs and differentiated 3T3-L1 adipocytes, we demonstrate that both compounds significantly enhance FNDC5 expression and irisin secretion. We reveal distinct mechanistic profiles: SS-IM1 showed superior efficacy in irisin induction and thermogenesis activation, while isolated hypholomine B demonstr

Ophiocordyceps sinensis is a natural fungus that has been valued as a health food and traditional Chinese medicine for centuries. The fungus is parasitic and colonizes insect larva. Naturally occurring O. sinensis thrives at high altitude in cold and grassy alpine meadows on the Himalayan mountain ranges. Wild O. sinensis is becoming increasingly rare in its natural habitats, and its price is out of reach for clinical practice. For these reasons, development of a standardized alternative is a great focus of research to allow the use of O. sinensis as a medicine. To develop an alternative for w

Paracoccidioides, a complex of several phylogenetic species, is the causative agent of paracoccidioidomycosis. The ability of pathogenic fungi to develop a multifaceted response to the wide variety of stressors found in the host environment is important for virulence and pathogenesis. Extracellular proteins represent key mediators of the host-parasite interaction. To analyze the expression profile of the proteins secreted by Paracoccidioides, Pb01 mycelia and yeast cells, we used a proteomics approach combining two-dimensional electrophoresis with matrix-assisted laser desorption ionization qu

BACKGROUND: Studies carried out during the 1990's demonstrated the presence of fungal glycoinositol phosphorylceramides (GIPCs) with unique structures, some of them showed reactivity with sera of patients with histoplasmosis, paracoccidioidomycosis or aspergillosis. It was also observed that fungal GIPCs were able to inhibit T lymphocyte proliferation "in vitro", and studies regarding the importance of these molecules to fungal survival showed that many species of fungi are vulnerable to inhibitors of sphingolipid biosynthesis.
RESULTS: In this paper, we describe a detailed characterization o

The 'Cuimi' kumquat (Citrus japonica), a geographical indication product in Rong'an County, China, is highly susceptible to post-harvest pathogenic infections, leading to soft decay. In spring 2024, fruits demonstrated symptoms of soft decay, displaying white mycelium and blue conidia on surfaces, with lesions later transitioning to a blue hue. The soft decay incidence averaged approximately 18.3%, based on assessments involving ten trees per orchard and fifty fruits per tree, across four orchards in Rongan County. To isolate the pathogen, six partially rotted fruits from each orchard, represe

Stillage is an acidic residue from ethanol production that has a high carbon load. Here, Ganoderma isolates were assessed for the treatment of rum stillage while producing biomass and associated metabolites. Isolates grew in 25% raw stillage, removing up to 73% of soluble organic carbon, 77% soluble nitrogen, and 74% phenolic compounds. Isolate G2 demonstrated faster removal of organic carbon and nitrogen. Biomass and metabolite production were benchmarked against a nutrient medium. In stillage, maximum values of the following were obtained: 8.2 g·L-1 biomass; 52.8% crude protein; 22.1 mg·g-1

Fungi possess an inherent ability to accumulate trace elements such as iron. In this study, we employed the Generally Recognized As Safe (GRAS) fungus Aspergillus oryzae (A. oryzae) A02 to biosynthesize an iron-enriched biomass (Fe-AA) and utilized a murine model to assess its physiological effects. Systematic optimization of fermentation parameters facilitaed pilot-scale production yielding mycelium with an organic iron content of 6.88 % (w/w), which represents the highest reported level for an A. oryzae-based iron supplement to date. Advanced characterization techniques (FTIR, SEM, TEM-EDS)

Post-harvest diseases of mango reduce fruit quality and cause severe yield losses with completely unmarketable fruits. The most common diseases of mangos are anthracnose (Colletotrichum gloeosporioides). In this study, the antibacterial activities of essential oils from Artemisia scoparia, Artemisia lavandulaefolia, and Artemisia annua against C. gloeosporioides were tested. The results showed that the essential oil of A. scoparia was more effective by the agar diffusion method; the EC50 value was 9.32 µL/mL. The inhibition rate was 100%, at a concentration of 10 μL/mL, through the spore germi

Attempts to bio-enrich fungal biomass with an essential trace elements to produce dietary supplements have some tradition and an example is selenium. Lithium salts have medical applications, but safer forms are sought after, and lithiated foods and food supplements may be an alternative. This study evaluated the lithiation of white Agaricus bisporus mushrooms using commercial compost fortified with LiNO3 and investigated the effects on co-accumulation of trace elements. The fortifications at levels of 1.0, 5.0, 10, 50 and 100 mg·kg-1 dw, resulted in corresponding median increases in mushroom L

Small cysteine-rich (SCR) proteins, including fungal avirulence proteins, play important roles in pathogen-plant interactions. SCR protein-encoding genes have been discovered in the genomes of Phytophthora pathogens but their functions during pathogenesis remain obscure. Here, we report the characterization of one Phytophthora capsici SCR protein (namely, SCR82) with similarity to Phytophthora cactorum phytotoxic protein PcF. The scr82 gene has 10 allelic sequences in the P. capsici population. Homologs of SCR82 were not identified in fungi or other organisms but in Phytophthora relative speci
