Affinity-oriented online ligand screening with LC coupled to different detectors is widely popular to capture active compounds from herbal medicines (HMs). However, false-positive extensively occurs because insufficient information is recorded for the existence and stability of ligand-protein complex. Here, efforts were made to advance the hit confidences via configuring post-column infusion-LC-energy-resolved-affinity MS (PCI-LC-ER-AMS) to achieve "four-in-one" monitoring of: 1) response decrement of potential ligands; 2) response decrement of protein; 3) ions relating to ligand-protein compl

Non-alcoholic fatty liver disease (NAFLD) is the main cause of chronic liver disease, and its pathological development is closely related to the gut-liver axis. The intestinal barrier, an important component of the gut-liver axis, can prevent gut microbes and endotoxins from entering the liver. Intestinal barrier function is impaired in patients with NAFLD. Baicalein, which is the main flavonoid in Scutellariae Radix, can improve NAFLD. However, whether baicalein alleviates NAFLD by ameliorating intestinal barrier dysfunction remains unclear. In this study, a methionine-choline deficient (MCD)

OBJECTIVE: Baicalin, which is isolated from Scutellariae Radix, has been shown to possess therapeutic potential for different diseases. Cardiac microvessel injury in myocardial ischemia-reperfusion (IR) has been extensively explored. However, there have been no studies investigating the physiological regulatory mechanisms of baicalin on nitric oxide production and the necroptosis of cardiac microvascular endothelial cells (CMECs) in myocardial IR injury. This study was designed to investigate the contribution of baicalin to repressing necroptosis and preventing IR-mediated CMEC dysfunction.

Dried roots of the plants Acanthopanax senticosus, Angelica sinensis and Scutellaria baicalensis are used in traditional oriental medicine and reportedly possess anti-inflammatory properties. Using the murine air pouch model of inflammation, we investigated the efficacy and mode of action of an extract from these three plants in crystal-induced inflammation. Air pouches were raised on the backs of 8-week-old BALB/c mice. Mice were fed 100 mg/kg body weight of root extracts (A. senticosus:A. sinensis:S. baicalensis mixed in a ratio of 5:4:1 by weight) or vehicle only on days 3-6. Inflammation w

INTRODUCTION: Dysfunction of axoplasmic transport is closely linked to depression. Baicalin, a major flavonoid in Scutellaria baicalensis, a well-known traditional Chinese medicine used in depression treatment, has demonstrated antidepressant-like effects in previous studies. However, its potential role in regulating axoplasmic transport has not been explored. This study aims to investigate the antidepressant mechanisms of baicalin through modulation of axoplasmic transport in hippocampal neurons.
METHODS: Male C57BL/6N mice were exposed to chronic unpredictable mild stress (CUMS) and treated

Vascular calcification (VC) is associated with cardiovascular disease. Baicalein, a natural flavonoid extract of Scutellaria baicalensis rhizome has several biological properties which may inhibit VC. We investigated whether baicalein suppresses Runt-related transcription factor 2 (Runx2) and bone morphogenetic protein 2 (BMP-2) and upregulates smooth muscle 22-alpha (SM22-α) and alpha-smooth muscle actin (α-SMA). In an in vitro experiment, primary rat aortic vascular smooth muscle cells (VSMCs) were pretreated with 0.1, 1, and 5 μM baicalein, followed by β-glycerophosphate (β-GP) to induce ca

Vibrio alginolyticus is a major aquaculture pathogen whose virulence is largely mediated by the type II secretion system (T2SS). However, the functions of key T2SS components, including gspJ, gspL, and gspM, remain unclear. Earlier research indicated that a Scutellaria baicalensis extract at 8 mg/mL markedly suppressed the growth, motility, and biofilm-forming capacity of V. alginolyticus ND-01 (OD600 = 0.5) following 12 h of treatment, with the resulting stress substantially altering the expression of multiple genes, including gspJ, gspL, and gspM. The gspJ-RNAi, gspL-RNAi, and gspM-RNAi stra
