In vitro bacterial adherence onto pellicle-coated aesthetic restorative materials.

Adhesion of oral bacteria to teeth and restorative materials plays an important role in the pathogenesis of oral diseases. This study investigated the initial adhesion of Streptococcus mutans to enamel and restorative materials. Three types of composites and two types of glass ionomer cements were used. The specimens were coated with freshly collected human parotid saliva. The salivary coated samples were incubated with cell-free glucosyltransferase, and further incubated with sucrose solution. Finally, the specimens were incubated with 3H-thymidine labelled bacteria. Adhesion of the bacteria

Craniofacial Evolution and Alveolar Bone Loss: A Lesson From Hominins.

BACKGROUND: In hominins, the reduction of prognathism during craniofacial evolution was a significant derived trait differentiating Homo from earlier hominins and other apes and might have contributed significantly to calvarial expansion and encephalization. Gnathic remains of Australopithecus africanus , A. robustus , Homo habilis, and H. erectus from the Plio-Pleistocene boundary 5.3-2.6 MYA were studied for evidence of alveolar bone (AB) loss indicative of periodontal disease(s). AB loss may provide critical insights into craniofacial evolution and the divergence of Homo from the Australopi

Effects of HCl concentration and immersion time on physicochemical properties, BMP-2 detection, degradation, and osteoblast response of human demineralized tooth matrix.

OBJECTIVE: Human demineralized tooth matrix (hDTM) is a promising tooth-derived graft material; however, demineralization protocols vary widely and may alter matrix structure, degradation behavior, and growth factor detectability. This study investigates the combined effects of hydrochloric acid (HCl) concentration (0.5 vs 1.0M) and immersion time (10 vs 20 min) on the physicochemical characteristics, Bone Morphogenetic Protein-2 (BMP-2) detection, degradation behavior and osteoblast response of hDTM. METHODS: Caries-free permanent teeth (particle size 500-1000 μm) were demineralized using fo

Comparative in vitro evaluation of sonic, passive ultrasonic, and Er: YAG laser-activated irrigation on Enterococcus faecalis reduction and smear layer/debris removal in root canals.

BACKGROUND: Persistent intraradicular infection remains a major cause of endodontic treatment failure because conventional syringe irrigation may not adequately reach the apical third, dentinal tubules, and canal irregularities. The aim of this in vitro study was to compare the antimicrobial efficacy and smear layer/debris removal ability of sonic activation, passive ultrasonic irrigation, and Er: YAG laser-activated irrigation using photon-induced photoacoustic streaming (PIPS), relative to conventional syringe irrigation, in prepared root canals infected with a mono-species Enterococcus faec

On-demand caries prevention: on-site generated slightly acidic electrolyzed water as a potent and biocompatible anti- Streptococcus mutans agent.

BACKGROUND: Dental caries remains a prevalent public health disorder dominated by Streptococcus mutans. Novel biocompatible antibacterial agents are urgently required for caries control. Slightly acidic electrolyzed water (SAEW), rich in endogenous hypochlorous acid, boasts broad bactericidal effects and minimal cellular toxicity, yet rapid degradation during storage limits its reproducibility. Thus, this study adopted freshly prepared SAEW to systematically assess its anti-cariogenic performance for translational reference. METHODS: In vitro, the minimum inhibitory concentration (MIC) and mi

Efficacy confirmation of traditional Chinese medicine Yuchi powder in preventing dental caries and active ingredients screening.

ETHNOPHARMACOLOGICAL RELEVANCE: Yuchi Powder (YCP) is a classical Traditional Chinese Medicine formula traditionally used for oral and dental disorders. Despite its long history of clinical application, its anti-caries efficacy and active constituents remain insufficiently understood. This study provides scientific evidence supporting the traditional use of YCP for dental caries prevention and identifies key bioactive constituents associated with its anti-cariogenic activity. AIM OF STUDY: We aimed to confirm the anti-caries efficacy of YCP and to screen its pharmacodynamic constituents throu

Inflammation-Regeneration Axis of Dental Pulp Stem Cells: Wnt/NF-κB Crosstalk.

BACKGROUND: Inflammation critically determines dental pulp regenerative outcomes, with dental pulp stem cells (DPSCs) orchestrating tissue homeostasis through differentiation, self-renewal and immunomodulation processes dynamically regulated by Wnt/β-catenin and NF-κB signaling crosstalk. Given the rising therapeutic potential of Wnt-targeted interventions in dental tissue engineering, elucidating these molecular interactions under pathological conditions is essential for developing regenerative therapeutics capable of simultaneously promoting reparative dentinogenesis while resolving inflamma

The action of different irrigant activation methods on engineered endodontic biofilm: an in vitro study.

INTRODUCTION: Endodontic infections are biofilm-mediated, demanding effective biofilm eradication from the root canal. Root canal complexities, coupled with bacterial biofilm resistance, pose challenges to thorough disinfection. Irrigation, particularly with sodium hypochlorite, is crucial in endodontics. Activation techniques, like sonic or ultrasonic oscillations, enhance irrigant penetration and biofilm disruption, improving decontamination and treatment outcomes. The aim of the present study was to evaluate the effectiveness of XP Finisher, EndoUltra, Eddy and Irriflex in the reduction of t

The dental cavities of equine cheek teeth: three-dimensional reconstructions based on high resolution micro-computed tomography.

BACKGROUND: Recent studies reported on the very complex morphology of the pulp system in equine cheek teeth. The continuous production of secondary dentine leads to distinct age-related changes of the endodontic cavity. Detailed anatomical knowledge of the dental cavities in all ages is required to explain the aetiopathology of typical equine endodontic diseases. Furthermore, data on mandibular and maxillary pulp systems is in high demand to provide a basis for the development of endodontic therapies. However, until now examination of the pulp cavity has been based on either sectioned teeth or

Development of a novel mouse osteoclast culture system including cells of mandibular body and erupting teeth.

Osteoclasts are multinucleated cells with the specialized function of resorbing calcified tissues. These cells develop from hemopoietic cells of the monocyte-macrophage lineage with the support of osteoblasts/stromal cells. Tooth eruption is a vertical movement of teeth via creation of an eruption pathway in and through the alveolar bone. The precise cellular and molecular determinants of tooth eruption are not yet clear, and a cell culture system that can reproduce the activity of osteoclast formation during tooth eruption is expected to be a useful tool to clarify the mechanism of eruption p

Differentiation and functions of osteoclasts and odontoclasts in mineralized tissue resorption.

The differentiation and functions of osteoclasts (OC) are regulated by osteoblast-derived factors such as receptor activator of NFKB ligand (RANKL) that stimulates OC formation, and a novel secreted member of the TNF receptor superfamily, osteoprotegerin (OPG), that negatively regulates osteoclastogenesis. In examination of the preosteoclast (pOC) culture, pOCs formed without any additives expressed tartrate-resistant acid phosphatase (TRAP), but showed little resorptive activity. pOC treated with RANKL became TRAP-positive OC, which expressed intense vacuolar-type H(+)-ATPase and exhibited pr

Endodontic Management Before, During and After Head and Neck Radiotherapy: Biological, Diagnostic and Clinical Considerations in Head and Neck Cancer Patients.

BACKGROUND: Head and neck radiotherapy creates a high-consequence dental environment in which tooth loss may precipitate osteoradionecrosis, particularly after extraction in high-dose mandibular regions. Tooth preservation through nonsurgical root canal treatment is therefore a risk reduction strategy. However, evidence relevant to endodontic decision-making remains dispersed among clinical series, laboratory research, pulp testing studies and oncology dental guidance. OBJECTIVE: To synthesise contemporary evidence relevant to endodontic management before, during and after head and neck radio

Dental Practitioners' Thresholds for Restorative Intervention in Carious Lesions: A Survey-Based Systematic Review Update.

UNLABELLED: <p>Introduction: Despite evidence supporting the clinical and cost-effectiveness of minimally invasive dentistry (MID), its adoption by the dental profession has been slow. A systematic review in 2016 found the majority of dentists intervene invasively earlier than necessary. The aim was to update this review of the assessment of dental practitioners' thresholds for providing restorative treatment for carious lesions given changes in evidence, teaching, and guidelines since 2016. The primary outcome was dental practitioners' restorative thresholds (the extent of the lesion when the
