Exploring the abomasal lymph node transcriptome for genes associated with resistance to the sheep nematode Teladorsagia circumcincta.

This study exploited Blackface lambs that varied in their resistance to the abomasal nematode parasite, Teladorsagia circumcincta. Infection of these lambs over 3 months identified susceptible (high adult worm count, high faecal egg count and low IgA antibody) and resistant animals that had excluded all parasites. Previous work had shown that susceptibility and resistance is dependent on the differential immune response to the parasite, which occurs within the abomasal (gastric) lymph node (ALN) that drains the site of infection. The Affymetrix ovine gene array was used to interrogate the tran

Hexamethonium: a probe to assess autonomic nervous system involvement in upper gastrointestinal functions in conscious sheep.

Hexamethonium, which inhibits cholinergic transmission by preventing acetylcholine release, has been considered an ideal reference drug for the blockade of autonomic ganglia, Auerbach plexus and reflex gastrointestinal secretions. The degree of inhibition of ruminant gastrointestinal functions with this reference drug were as follows: cyclical contractions of the reticulo-rumen and abomasal motility greater than gastric acid secretion and duodenal migrating myoelectrical complexes. Although reduced at high dosages, the initiation of migrating myoelectric complexes was enhanced at clinically us

The Development of Ovine Gastric and Intestinal Organoids for Studying Ruminant Host-Pathogen Interactions.

Gastrointestinal (GI) infections in sheep have significant implications for animal health, welfare and productivity, as well as being a source of zoonotic pathogens. Interactions between pathogens and epithelial cells at the mucosal surface play a key role in determining the outcome of GI infections; however, the inaccessibility of the GI tract in vivo significantly limits the ability to study such interactions in detail. We therefore developed ovine epithelial organoids representing physiologically important gastric and intestinal sites of infection, specifically the abomasum (analogous to th

Effect of clinical mastitis on the lactation curve: a mixed model estimation using daily milk weights.

The objective of this study was to estimate the milk production losses associated with clinical mastitis using mixed linear models and correlation structures that have not been available previously. Data used included computer-recorded daily milk yields and detailed and accurate recordings of clinical mastitis cases. Two commercial Holstein dairy farms in New York State participated in the study, one with 650 lactating cows and another that began the study with 830 lactating cows and increased to 1120 cows by the end of the study. Cows on both farms were housed in free stall barns and milked 3

Bacterial communities in the gastrointestinal tract segments of helminth-resistant and helminth-susceptible sheep.

BACKGROUND: Helminth parasitism is a world-wide problem in livestock industries, with major impacts on health, welfare and productivity. The role of the gut microbiota in host-helminth interactions in ruminants has been extensively examined and the present study added to this body of knowledge by assessing the effects of resistance and susceptibility to helminth infection in the gastro-intestinal tract (GIT). Australian Sheep Breeding Values (ASBVs) for faecal egg count (FEC) were used to select the 10 highly helminth-susceptible (High-FEC) and 10 highly helminth-resistant (Low-FEC) sheep. FEC

Can orange essential oil reduce the severity of parasitic infection in sheep?

In this study, a three-step experiment was conducted to evaluate the effectiveness of orange essential oil (OEO) to reduce the severity of parasitic infection in lambs naturally infected with gastrointestinal parasites. In the first and second stages of the experiment, the lambs were distributed in groups (n = 10): without treatment (control), treated with levamisole (7.5 mg/kg), and treated either with 200, 300 or 400 mg/kg OEO in the first stage and with two doses of 300 or 350 mg/kg each of OEO with a six-hour interval in the second stage. In the third stage of the experiment, 20 lambs were

The bacterial community associated with the sheep gastrointestinal nematode parasite Haemonchus contortus.

Culture-independent methods were used to study the microbiota of adult worms, third-stage larvae and eggs, both in faeces and laid in vitro, of Haemonchus contortus, a nematode parasite of the abomasa of ruminants which is a major cause of production losses and ill-health. Bacteria were identified in eggs, the female reproductive tract and the gut of adult and third-stage larvae (L3). PCR amplification of 16S rRNA sequences, denaturing gradient gel electrophoresis (DGGE) and clone libraries were used to compare the composition of the microbial communities of the different life-cycle stages of

Decreasing the pH of milk replacer containing soy flour affects nutrient digestibility, digesta pH, and gastrointestinal development of preweaned calves.

We evaluated the effect of feeding soy flour-containing milk replacer (MR) with a lower than normal pH on growth, nutrient digestibility, digesta pH, and gastrointestinal development of preweaned calves. The study consisted of 48 neonatal calves, allocated to 8 treatments in a 2-factor 4×2 randomized design. The calves received 2 types of MR containing soy flour. The proportion of soy crude protein (CP) to total CP (SP/CP) in MR was 50% (MR50 treatment) or 80% (MR80 treatment), respectively. The pH of the reconstituted MR was decreased by 1 N HCl from to 6.2 to 5.5, 5.0, or 4.5. We measured bo

Effects of fatty acid supplements on feed intake, and feeding and chewing behavior of lactating dairy cows.

Saturated and unsaturated fatty acid supplements (FS) were evaluated for effects on feed intake, meal patterns, and chewing behavior. Eight ruminally and duodenally cannulated cows were used in a replicated 4 x 4 Latin square design experiment with 21-d periods. Treatments were control and a linear substitution of 2.5% fatty acids from supplemented saturated FS (SAT; prilled, hydrogenated free fatty acids) for partially unsaturated FS (UNS; calcium soaps of long-chain fatty acids). All rations contained identical forage and concentrate components including 37.2% forage and 13.5% cottonseed. Dr

The effect of simulated rainfall on the efficacy of doramectin pour-on against nematode parasites of cattle.

Two studies were conducted with doramectin topically administered at 500 microg/kg body weight to assess retention of therapeutic efficacy against nematode infections of cattle before, and after, simulated rainfall. In the first study, 50 heifers, with patent nematode infections, were allocated to one of five treatment groups. An untreated control group and one doramectin-treated group were not exposed to simulated rainfall. Simulated rainfall was applied at a rate of 25.4 mm of water in 35 min to three of the five groups: one group immediately before treatment, the second group 90 min after t

Plasma and tissue zinc enrichment altered by different sources of zinc using stable isotopes in weaned male Holstein calves facing dietary antagonism.

Trace minerals are an important component of ruminant nutrition. Furthermore, understanding the bioavailability of various trace mineral sources is essential for accurate dietary formulation. The objective of this trial is to determine the effect of a Zn antagonism, elevated S, on the resulting plasma and tissue Zn enrichment, while also determining if differences occur depending on Zn source, Zn glycinate, and Zn methionine hydroxy analog chelate, in Holstein calves. Thirty weaned male Holstein calves were used in a randomized complete block design. Calves were fed a texturized starter varyin

Mannheimia granulomatis-Infected Pyogranulomatous Oral Lesions with Concurrent Amyloidosis in Roe Deer (Capreolus capreolus) in France.

Mannheimia granulomatis-associated lesions have been poorly described compared with those associated with other members of the Pasteurellaceae family; they include pneumonia, panniculitis, and keratoconjunctivitis in domestic and wild ruminants and hares. We describe 15 cases of roe deer (Capreolus capreolus) showing M. granulomatis-infected oral lesions that were detected in France between 2016 and 2023. Comprehensive necropsies were performed, with additional analyses based on findings. All deer were >15 mo of age on the basis of tooth eruption. They were thin (1/15), cachectic (13/15), or o

Effects of oral calcium on reproduction and postpartum health in cattle: a meta-analysis and quality assessment.

Postpartum blood calcium (Ca) concentration is related to the reproduction and health of cattle. Oral calcium supplements were given to dairy cows after calving to increase blood Ca concentration and reduce the risk of hypocalcemia. However, studies have shown that oral Ca has different effects in preventing disease. The purposes of this study were (i) to conduct a meta-analysis to evaluate the expected effect of oral Ca on incidence of calving-related diseases, pregnancy risk and milk yield in dairy cows, and (ii) to make a quality assessment of these related studies. In total, 22 eligible st

Fetal organ growth in response to oesophageal infusion of amniotic fluid, colostrum, milk or gastrin-releasing peptide: a study in fetal sheep.

The hypothesis of the present study was that the infusion of the biological fluids to which the developing gut is normally exposed (i.e. amniotic fluid, colostrum, milk) and a single growth factor (gastrin-releasing peptide), which is found in high concentrations in fetal fluids and milk, could ameliorate the altered growth induced by the elimination of swallowed input secondary to ligation of the oesophagus. At 108-110 days of gestation the fetal oesophagus was ligated and a catheter inserted towards the stomach (32 fetuses). At 117-119 days of gestation saline (n = 5), amniotic fluid (n = 5)
