Therapeutic potential of ELABELA in alleviating hereditary hypertrophic cardiomyopathy.

INTRODUCTION: Hereditary hypertrophic cardiomyopathy (HCM) is a common genetic heart disease associated with heart failure and sudden cardiac death. Current therapies are limited, and the myosin inhibitor mavacamten is constrained by safety concerns and restricted applicability. ELABELA (ELA), an endogenous ligand of the apelin receptor (APJ), plays an important role in cardiovascular regulation, but its involvement in HCM remains unclear. OBJECTIVES: This study aimed to determine the role of ELA in hereditary HCM and to assess the therapeutic potential of restoring ELA signaling. METHODS: E

Adipose Mesenchymal Stem Cells Overexpressing Chemokine (CXCL6) Showed Strong Angiogenesis and Anti-Inflammatory Ability in the Treatment of Pulmonary Hypertension.

Pulmonary arterial hypertension (PAH) is a progressive disease characterized by vascular remodeling and right ventricular failure, with limited effective treatments. This study aimed to evaluate therapeutic effects of adipose-derived mesenchymal stem (ADMS) cells engineered to overexpress chemokine CXCL6 in monocrotaline-induced PAH rats. ADMS cells were transduced with a CXCL6-expressing lentivirus and characterized by flow cytometry, migration assays, and tube formation tests. Following establishment of PAH model, rats were administered intravenous injections of CXCL6-ADMS or control cells.

Beta-sitosterol alleviates inflammatory culprits and disease severity in arthritic irradiated rat's model: role of brain-spleen axis.

BACKGROUND: The complex interaction between the adaptive immune system and innate immunological components causes rheumatoid arthritis (RA), a systemic autoimmune disorder that involves synovitis and joint deformity. Brain- spleen axis is a reciprocal cross-communicative circuit serves a crucial role in maintaining immunomodulation and homeostasis. RA complicates extra-articular manifestations in spleen and brain. Exposure to ionizing radiation might exacerbate the severity of RA. Recently, beta- sitosterol (BS), which is a promising phytosterols, has shown efficacy against various inflammator

A Morphological and Behavioral Study of Demyelination and Remyelination in the Cuprizone Model: Insights into APLNR and NG2+ Cell Dynamics.

Multiple sclerosis (MS) is a chronic neurodegenerative disorder involving demyelination. The cuprizone model is commonly used to study MS by inducing oligodendrocyte stress and demyelination. The subventricular zone (SVZ) plays a key role in neurogenesis, while the neuronal/glial antigen 2 (NG2) is a marker for immature glial cells, involved in oligodendrocyte differentiation. The apelin receptor (APLNR) is linked to neurogenesis and behavior modulation. This study explores the role of APLNR in NG2-positive cells during de- and remyelination phases in the experimental cuprizone mouse model. Th

PEGylated and Acylated Elabela Analogues Show Enhanced Receptor Binding, Prolonged Stability, and Remedy of Acute Kidney Injury.

Acute kidney injury (AKI), mostly caused by renal ischemia-reperfusion (I/R) injury and nephrotoxins, is characterized by rapid deterioration in renal-functions without effective drug treatment available. Through activation of a G protein-coupled receptor APJ, a furin-cleaved fragment of Elabela (ELA[22-32], E11), an endogenous APJ ligand, protects against renal I/R injury. However, the poor plasma stability and relatively weak APJ-binding ability of E11 limit its application. To address these issues, we rationally designed and synthesized a set of E11 analogues modified by palmitic acid (Pal)

Differential Biventricular Strain Recovery With Selective Fibrosis Biomarker Regression After Sleeve Gastrectomy: A Prospective CMR Feature-Tracking Study.

PURPOSE: Obesity injures the heart through hemodynamic, fibrotic, and adipokine pathways, but whether these reverse together or on different timelines after bariatric surgery is unknown. We performed, to our knowledge, the first integrated assessment of biventricular and biatrial cardiovascular magnetic resonance feature-tracking (CMR-FT), fibrosis and adipokine biomarkers, cardiovascular risk, and quality of life after laparoscopic sleeve gastrectomy (LSG), testing whether recovery is uniform or uneven. MATERIALS AND METHODS: In this single-center prospective cohort, 40 patients underwent LS

[Drug therapy of pulmonary arterial hypertension in 2014].

Pulmonary arterial hypertension (PAH) is a primary pulmonary arteriolar disease, characterized by a progressive increase in pulmonary vascular resistance and pressure in the pulmonary circulation. It progressively leads to hypertrophy of the right ventricle and with no treatment to its failure and patient´s death. Etiology of pulmonary hypertension (PH) has been reclassified repeatedly, most recently during the 4th World Symposium on Pulmonary Hypertension held in 2008 [1]. Currently, the first group contains PAH with either unknown or known cause (systemic connective tissue disease, liver dis

Influence of Dietary Fiber and Polyphenols During Pre-Gestation, Gestation, or Lactation on Intestinal Gene Expression.

BACKGROUND/OBJECTIVES: Diet composition is important for health, especially during critical periods such as pre-gestation (P), gestation (G), or lactation (S), due to its potential impact not only on the mother but on the offspring. The Mediterranean diet includes many healthy foods rich in fiber and/or polyphenols, such as whole grains, fruits, vegetables, beans, and nuts. The present preclinical study assesses the impact of a diet rich in fiber and polyphenols (HFP diet) during one of those three periods (P, G, or S, three weeks each) on the rat gene expression of the small intestine obtaine

Multi-omics analysis unveils dysregulation of the tumor immune microenvironment and development of a machine learning-based multi-gene classifier for predicting lateral lymph node metastasis in papillary thyroid carcinoma.

PURPOSE: Lateral lymph node metastasis (LNM) critically influences surgical decision-making in papillary thyroid carcinoma (PTC). However, the sensitivity of preoperative imageological examination in detecting LNM remains suboptimal, necessitating the development of more accurate diagnostic and predictive tools. This study aims to identify multi-omics biomarkers and construct a predictive model for LNM. METHODS: We performed a comprehensive multi-omics analysis of 50 PTCs presenting with (LNM group) or without lateral lymph node metastases (LNN group) using whole exome sequencing and whole tr

Single-cell Transcriptome Analysis Reveals the Potential Role of Hepatic Stellate Cells in Liver Fibrosis.

BACKGROUND: Upon activation, hepatic stellate cells (HSCs) can convert into fibroblasts and increase the production of extracellular matrix, a major cause of liver fibrosis (LF) and a growing health issue worldwide. Other mechanisms by which HSCs may induce fibrosis remain to be explored, and the role of cell dynamic gene expression in liver fibrogenesis is not well understood. In this study, analysis by single-cell transcriptome sequencing (scRNA-seq) was used to explore the potential effects of HSCs in a bile duct ligation (BDL)-induced mouse model of LF, followed by the identification of no

Adipokines and their potential impacts on susceptibility to myocardial ischemia/reperfusion injury in diabetes.

Coronary artery disease has a high mortality rate and is a striking public health concern, affecting a substantial portion of the global population. On the early onset of myocardial ischemia, thrombolytic therapy and coronary revascularization could promptly restore the bloodstream and nutrient supply to the ischemic tissue, efficiently preserving less severely injured myocardium. However, the abrupt re-establishment of blood flow triggers the significant discharge of previously accumulated oxidative substances and inflammatory cytokines, leading to further harm referred to as ischemia/reperfu

Association of AGTR1 with 18-month treatment outcome in late-life depression.

OBJECTIVE: Converging lines of evidence implicate vascular factors in late-life depression, and argue that late-life depression is a distinct entity among the mood disorders. The A1166C polymorphism in the angiotensin II receptor, vascular type 1 (AGTR1) gene has been associated with a range of vascular diseases. This study investigated the association of AGTR1 genotype on 18-month treatment outcome in late-life depression. METHODS: In a large, prospective cohort study, patients with late-life depression received individualized treatment using a standardized algorithm. The authors genotyped p

The pathophysiology of retinopathy of prematurity: an update of previous and recent knowledge.

Retinopathy of prematurity (ROP) is a disease that can cause blindness in very low birthweight infants. The incidence of ROP is closely correlated with the weight and the gestational age at birth. Despite current therapies, ROP continues to be a highly debilitating disease. Our advancing knowledge of the pathogenesis of ROP has encouraged investigations into new antivasculogenic therapies. The purpose of this article is to review the findings on the pathophysiological mechanisms that contribute to the transition between the first and second phases of ROP and to investigate new potential therap

Postnatal overfeeding causes early shifts in gene expression in the heart and long-term alterations in cardiometabolic and oxidative parameters.

BACKGROUND: Postnatal overfeeding (OF) in rodents induces a permanent moderate increase in body weight in adulthood. However, the repercussions of postnatal OF on cardiac gene expression, cardiac metabolism and nitro-oxidative stress are less well known. METHODOLOGY/PRINCIPAL FINDINGS: Immediately after birth, litters of C57BL/6 mice were either maintained at 10 (normal-fed group, NF), or reduced to 3 in order to induce OF. At weaning, mice of both groups received a standard diet. The cardiac gene expression profile was determined at weaning and cardiac metabolism and oxidative stress were as
