Strontium-driven physiological to pathological transition of bone-like architecture: A dose-dependent investigation.

Whilst strontium (Sr2+) is widely investigated for treating osteoporosis, it is also related to mineralization disorders such as rickets and osteomalacia. In order to clarify the physiological and pathological effects of Sr2+ on bone biomineralization , we performed a dose-dependent investigation in bone components using a 3D scaffold that displays the hallmark features of bone tissue in terms of composition (osteoblast, collagen, carbonated apatite) and architecture (mineralized collagen fibrils hierarchically assembled into a twisted plywood geometry). As the level of Sr2+ is increased from

A Bioinspired Ultratough Composite Produced by Integration of Inorganic Ionic Oligomers within Polymer Networks.

The nacre-inspired laminates are promising materials for their excellent mechanics. However, the interfacial defects between organic-inorganic phases commonly lead to the crack propagation and fracture failure of these materials under stress. A natural biomineral, bone, has much higher bending toughness than the nacre. The small size of inorganic building units in bone improves the organic-inorganic interaction, which optimizes the material toughness. Inspired by these biological structures, here, an ultratough nanocomposite laminate is prepared by the integration of ultrasmall calcium phospha

Specifiable biomimetic microsponges for timed release of crystal entrapped biomolecules useful in bone repair.

Most marine materials, by nature, contain crystals of inorganic matter with specific structures that allow the loading, release, and delivery of biomolecules that can be utilized in clinical applications. These structures can be biomimetically synthesized. Aggregates of inorganic particles generated by biomimetic microsponges may provide surfaces and structures for cell attachment, organization, and promotion of matrix synthesis. Biomimetic microsponges have been developed with tunable release profiles differing by the rate (speed over distance), velocity (rate of change in direction), and the

Pyronaridine: An update of its pharmacological activities and mechanisms of action.

Pyronaridine (PYR) is an erythrocytic schizonticide with a potent antimalarial activity against multidrug-resistant Plasmodium. The drug is used in combination with artesunate for the treatment of uncomplicated P. falciparum malaria, in adults and children. The present review briefly retraces the discovery of PYR and recent antimalarial studies which has led to the approval of PYR/artesunate combination (Pyramax) by the European Medicines Agency to treat uncomplicated malaria worldwide. PYR also presents a marked antitumor activity and has revealed efficacy for the treatment of other parasitic

Mechanical design of the highly porous cuttlebone: A bioceramic hard buoyancy tank for cuttlefish.

Cuttlefish, a unique group of marine mollusks, produces an internal biomineralized shell, known as cuttlebone, which is an ultra-lightweight cellular structure (porosity, ∼93 vol%) used as the animal's hard buoyancy tank. Although cuttlebone is primarily composed of a brittle mineral, aragonite, the structure is highly damage tolerant and can withstand water pressure of about 20 atmospheres (atm) for the species Sepia officinalis Currently, our knowledge on the structural origins for cuttlebone's remarkable mechanical performance is limited. Combining quantitative three-dimensional (3D) struct

Cloning and Expression Analysis of Human Amelogenin in Nicotiana benthamiana Plants by Means of a Transient Expression System.

Enamel is the covering tissue of teeth, made of regularly arranged hydroxyapatite crystals deposited on an organic matrix composed of 90% amelogenin that is completely degraded at the end of the enamel formation process. Amelogenin has a biomineralizing activity, forming nanoparticles or nanoribbons that guide hydroxyapatite deposit, and regenerative functions in bone and vascular tissue and in wound healing. Biotechnological products containing amelogenin seem to facilitate these processes. Here, we describe the production of human amelogenin in plants by transient transformation of Nicotiana

Biochemical characterization of the native α-carbonic anhydrase purified from the mantle of the Mediterranean mussel, Mytilus galloprovincialis.

A α-carbonic anhydrase (CA, EC 4.2.1.1) has been purified and characterized biochemically from the mollusk Mytilus galloprovincialis. As in most mollusks, this α-CA is involved in the biomineralization processes leading to the precipitation of calcium carbonate in the mussel shell. The new enzyme had a molecular weight of 50 kDa, which is roughly two times higher than that of a monomeric α-class enzyme. Thus, Mytilus galloprovincialis α-CA is either a dimer, or similar to the Tridacna gigas CA described earlier, may have two different CA domains in its polypeptide chain. The Mytilus galloprovi

Microbial mineralization of struvite: a promising process to overcome phosphate sequestering crisis.

Due to extensive exploitation of non-renewable phosphate minerals, their natural reserves will exhaust very soon. This necessitates looking for alternatives and an efficient methodology through which indispensable phosphorus can be harvested back. The current study was undertaken to explore the potential of a metallophilic bacterium Enterobacter sp. EMB19 for the recovery of phosphorus as phosphate rich mineral. A very low phosphate concentration strategy was adopted. The process led to the mineralization of phosphorus as homogeneous struvite crystals. For each gram of Epsom salt added, the ce

The effect of aquatic plant abundance on shell crushing resistance in a freshwater snail.

Most of the shell material in snails is composed of calcium carbonate but the organic shell matrix determines the properties of calcium carbonate crystals. It has been shown that the deposition of calcium carbonate is affected by the ingestion of organic compounds. We hypothesize that organic compounds not synthesized by the snails are important for shell strength and must be obtained from the diet. We tested this idea indirectly by evaluating whether the abundance of the organic matter that snails eat is related to the strength of their shells. We measured shell crushing resistance in the sna

Using human neural crest-derived progenitor cells to investigate osteogenesis: an in vitro study.

Human tooth contains a distinct population of neural crest-derived progenitor cells (dNC-PCs) which are known to give rise to specialized daughter cells of an osteogenic lineage. We hypothesised that dNC-PCs could develop into neural crest-derived bone in a self-propagating and extracorporal culture system. Thus, we examined the three-dimensional structure obtained from osteogenic-stimulated dNC-PCs by morphological, biochemical and spectroscopic methods. After the onset of stimulation, cells formed a multilayer with outer cells covering the surface and inner cells secreting a hyaline matrix.

Bioactivity of polycrystalline silicon layers.

After oxygen, silicon is the second most abundant element in the environment and is present as an impurity in most materials. The widespread occurrence of siliceous biominerals as structural elements in lower plants and animals suggests that Si plays a role in the production and maintenance of connective tissue in higher organisms. It has been shown that the presence of Si is necessary in bones, cartilage and in the formation of connective tissue, as well as in some important metabolic processes. In this work, polycrystalline silicon layers are tested in terms of bioactivity, i.e., their abili

The organic preservation of fossil arthropods: an experimental study.

Modern arthropod cuticles consist of chitin fibres in a protein matrix, but those of fossil arthropods with an organic exoskeleton, particularly older than Tertiary, contain a dominant aliphatic component. This apparent contradiction was examined by subjecting modern cockroach, scorpion and shrimp cuticle to artificial maturation (350 degrees C/700 bars/24 h) following various chemical treatments, and analysing the products with pyrolysis-gas chromatography/mass spectrometry (Py-GC/MS). Analysis of artificially matured untreated cuticle yielded moieties related to phenols and alkylated substit

Expression of amelogenins in developing embryonic and neonatal rat teeth.

OBJECTIVE: Tooth enamel is formed by ameloblasts, which are derived from epitheliums and secrete an extracellular matrix containing a complex arrangement of protein components. The epithelial component, referred to as the enamel organ, contains a layer of cells that secrete an organic matrix that biomineralizes to become tooth enamel. Adjacent ectomesenchyme cells differentiate to become dentinproducing odontoblasts. These two mineralized matrices form the crown of the vertebrate tooth. Therefore, amelogenins play a critical role in tooth enamel formation. We have examined the expression patte

Fullerene C60 in dental materials: a comprehensive review of carbon nanotechnology applications and future prospects.

Teeth and dental materials are very noteworthy because of their important role in digestion and facial beauty. It is necessary to develop dental materials with suitable physical, chemical and biological properties to improve the quality and beauty of teeth. Fullerene, as a spherical allotrope of carbon, has potent properties for medical applications. In this combinatorial review article, we focus on the application of fullerene C60 in dentistry. By searching the database for suitable keywords ("fullerene", "dental" and "dentistry"), 12 related articles were found. The data extracted from these

Gradual Modification of Ferritin 4-Fold Pore Promotes Cage Instability, Fe2+ Exit, and Iron-Induced Protein Precipitation.

Ferritins are symmetrical, hollow nanocaged iron storage proteins, which sequester and concentrate iron as a hydrated ferric oxyhydroxide (ferrihydrite) biomineral. Spontaneous self-assembly of 24 subunits in eukaryotic ferritins leads to the formation of symmetrical pores, i.e., eight hydrophilic 3-fold pores and six hydrophobic 4-fold pores. However, unlike the relatively wider 3-fold pores, which drive Fe2+ inside, the functions of narrow 4-fold pores are relatively understudied. The current work investigates the role of 4-fold pores by gradual alterations of specific amino acids using site

Combining soft-bodied and three-dimensional fossils to reveal evolutionary modifications in early lingulellotretid brachiopods.

Living lingulide brachiopods are traditionally recognised as representatives of evolutionary conservatism, showing little change in general-morphology from their Cambrian ancestors. However, less attention has been given to their anatomical and ontogenetic modifications since their initial appearance. Among these, lingulellotretids are unique, characterized by their typical elongate pedicle foramen and large pseudointerarea. This study describes exquisitely preserved soft-tissue and phosphatic shells of Lingulellotreta from Cambrian Series 2 deposits in China and Kazakhstan. Biomineralized nov

Calcium Oxalate Crystals with a Bipyramidal Morphology and a High Tendency to Aggregate Attach Directly to Breast Cancer Cells and Affect Their Phenotype.

Various types of crystal can form in our bodies under normal and pathological conditions, with the more harmful of these crystals being studied intensively. However, much less is known about calcium oxalate dihydrate (COD) crystals, which are commonly associated with benign conditions. In general, calcium oxalates are pathological biominerals found in kidney stones and as microcalcifications (MCs) in breast and thyroid cancers. In breast cancer MCs, COD crystals are exclusively associated with benign lesions, and we have recently shown that they suppress breast precancer progression in vitro.

The Mechanism of Mineral Nucleation and Growth in a Mini-Ferritin.

Iron is an enigmatic element. While necessary for life, as Fe(II) it also catalyzes formation of reactive oxygen species. To mitigate this, cellular life has evolved the ferritin protein superfamily, which includes the 24 subunit ferritins and bacterioferritins, and 12 subunit mini-ferritins (DPS). Each catalyze the oxidation of Fe(II) to ferric oxyhydroxide, which is then sequestered within the hollow protein shell. While there is a wealth of structural information on unmineralized ferritins, high resolution information on iron loaded ferritins is lacking, and the mechanism of iron mineraliza

A multifunctional theranostic nanoagent for precise NIR-II FL imaging and synergistic chemodynamic/H2/SGB therapy of allergic rhinitis.

Allergic rhinitis (AR) is a chronic inflammatory disease of the nasal mucosa mediated by immunoglobulin E (IgE) and characterized by infiltration of inflammatory cells, such as eosinophils, and excessive production of reactive oxygen species (ROS). Alleviating oxidative stress and inflammation at AR sites is an effective approach for improving pathological conditions and modulating the microenvironment. Herein, we have developed novel multifunctional theranostic nanoagents (ACB NGs) to achieve precise and rapid second near-infrared fluorescence (NIR-II FL) imaging and a promising combination t

LCMS-Metabolomic Profiling and Genome Mining of Delftia lacustris DSM 21246 Revealed Lipophilic Delftibactin Metallophores.

Bacteria have evolved various strategies to combat heavy metal stress, including the secretion of small molecules, known as metallophores. These molecules hold a potential role in the mitigation of toxic metal contamination from the environment (bioremediation). Herein, we employed combined comparative metabolomic and genomic analyses to study the metallophores excreted by Delftia lacustris DSM 21246. LCMS-metabolomic analysis of this bacterium cultured under iron limitation led to a suite of lipophilic metallophores exclusively secreted in response to iron starvation. Additionally, we conduct
