Performance of seven carbapenemase detection assays in Pseudomonas aeruginosa across different epidemiological settings: a multicenter cross-sectional study.

The detection of carbapenemases in Pseudomonas aeruginosa remains challenging due to a great variety of other resistance mechanisms, and most laboratories, therefore, do not test for them. This study aimed to comparatively evaluate seven phenotypic carbapenemase detection assays across three epidemiological settings. A total of 320 P. aeruginosa isolates from three German centers with varying carbapenemase prevalences (5.8%-51.4%), including 113 carbapenemase-producing isolates carrying VIM-2 (n = 58), NDM-1 (n = 19), and GIM-1 (n = 16), underwent whole-genome sequencing as reference to assess

Antibiotic Resistance Profile of Wound Infection Isolates: High Resistance to Cephalosporins and Penicillin Among Ghanaian Patients in a Cross-Sectional Study.

BACKGROUND AND AIM: Antibiotics remain among the most frequently prescribed medications in clinical practice. However, antimicrobial resistance (AMR) is an inevitable and irreversible consequence of bacterial adaptation to antimicrobial exposure. Despite its clinical importance, data on antibiotic resistance patterns of bacteria from wound infections in Ghana are limited. This study evaluated the prevalence and magnitude of antibiotic resistance among bacterial isolates from infected wounds in a Ghanaian population. METHODS: This experimental cross-sectional study enrolled 300 patients with s

Isolation, identification, antibiotic resistance profile and molecular analysis of Ornithobacterium rhinotracheal isolates from turkeys.

BACKGROUND: Ornithobacterium rhinotracheal (ORT) infects numerous birds, particularly chickens and turkeys. ORT is an emerging bacterial pathogen of global concern in the poultry industry. As ORT is rapidly spreading throughout commercial poultry, it requires intensive studies of its epidemiology, diagnostic procedures, molecular typing, virulence genes and antimicrobial resistance. OBJECTIVES: The present study was conducted in isolation and identification of ORT from slaughtered turkeys. METHODS: Cleft palate swabs of 200 were collected from slaughtered turkeys and cultured on blood agar.

Molecularly imprinted polymers as antibody mimics in automated on-line fluorescent competitive assays.

An automated molecularly imprinted sorbent based assay (MIA) for the rapid and sensitive analysis of penicillin-type beta-lactam antibiotics (BLAs) has been developed and optimized. The polymers were prepared using penicillin G procaine salt as template (PENGp) and a stoichiometric quantity of a urea-based functional monomer to target the single oxyanionic species in the template molecule. Highly fluorescent competitors (emission quantum yields of 0.4-0.95), molecularly engineered to contain pyrene labels while keeping intact the 6-aminopenicillanic acid moiety for efficient recognition by the

Development of a novel and automated fluorescent immunoassay for the analysis of beta-lactam antibiotics.

An automated immunosensor for the rapid and sensitive analysis of penicillin type beta-lactam antibiotics has been developed and optimized. An immunogen was prepared by coupling the common structure of the penicillanic beta-lactam antibiotics, i.e., 6-aminopenicillanic acid to keyhole limpet hemocyanin. Polyclonal antibodies raised in rabbits after immunization with this conjugate have been applied for the development of a competitive fluoroimmunoassay (FIA), using a novel fluorescent penicillin {[2S,5R,6R]-3,3-dimethyl-7-oxo-6-[(pyren-1ylacetyl)amino]-4-thia-1-azabicyclo[3.2.0]heptane-2-carbo

Occurrence and Spatial Distribution of Antibiotic-Resistant Bacteria and Antibiotic Resistance Genes in Surface and Groundwater of the Kelani River Basin, Sri Lanka.

Antimicrobial resistance (AMR) in aquatic environments represents a critical global public health threat, yet comprehensive environmental surveillance data remain scarce in low- and middle-income countries. The Kelani River Basin, supplying approximately 80% of Colombo's potable water and Sri Lanka's most industrially impacted waterway, lacks a basin-wide characterization of antibiotic-resistant bacteria (ARB) and antibiotic resistance genes (ARGs) across spatial and seasonal gradients. This study provides the first comprehensive basin-wide assessment of ARB distribution, ARG occurrence, minim

Quantitation of 10 antibiotics in plasma: Sulfosalicylic acid combined with 2D-LC-MS/MS is a robust assay for beta-lactam therapeutic drug monitoring.

Therapeutic drug monitoring (TDM) of antibiotics is particularly important in populations with high pharmacokinetic variabilities, such as critically ill patients, leading to unpredictable plasma concentrations and clinical outcomes. Here, we i) describe an original method for the simultaneous quantification of ten antibiotics (cefepime, ceftazidime, ampicillin, piperacillin/tazobactam, cefotaxime, amoxicillin, cloxacillin, oxacillin, linezolid) using 5-sulfosalicylic acid dihydrate (SSA) solution for protein precipitation together with 2D-LC-MS/MS, and ii) evaluate its impact in a one-year re
