Predicted values for human total clearance of a variety of typical compounds with differently humanized-liver mouse plasma data.

Prediction of human pharmacokinetics is important in the preclinical stage. Values for total clearance of compounds from plasma should be one of the most important pharmacokinetic parameters for predictions. Although several physiological and empirical methods including single-species allometry for prediction of values for human clearance of compounds using humanized-liver mice have been reported, further improvement of prediction accuracies would be still expected. To optimize these approaches, we proposed methods for unbound intrinsic clearance in virtually 100% humanized-liver mouse by inco

[Irbesartan: a new possibility in the treatment of hypertension].

Hypertension means a basic public health problem in many countries in the world. The therapeutic attempts of the last years did not fulfill the hopes pinned on them, and most of the patients live with blood pressure above the goal value. This is why there is a need for new, more efficient antihypertensive drugs. On the 1st of July, 2001 irbesartan (Aprovel) was introduced in practice in Hungary. The drug belongs to the family of the angiotensin II receptor inhibitors. Several clinical studies were made with irbesartan in order to evaluate its efficiency, tolerability and safety. In other studi

Antihypertensive Effects of Diosmetin in Hypertension-Induced Cardiovascular Abnormalities in Rats.

INTRODUCTION: Hypertension is associated with cardiovascular dysfunction and remodeling. Diosmetin, a flavonoid isolated from citrus seeds, has various biological properties. This work investigated the effects of diosmetin on cardiovascular parameters and classical and non-classical renin-angiotensin systems (RAS) in two-kidney one-clip (2K-1C) rats. METHODS: 2K-1C hypertension was induced by placing a silver clip on the left renal artery. Three weeks after induction, rats were orally gavage with either vehicle, diosmetin (20 or 40 mg/kg), or telmisartan (5 mg/kg) for four weeks. Blood pressu

Medicine-Induced Acute Kidney Injury Findings from Spontaneous Reporting Systems, Sequence Symmetry Analysis and a Case-Control Study with a Focus on Medicines Used in Primary Care.

INTRODUCTION: Primary care provides an opportunity to prevent community acquired, medicine or drug-induced acute kidney injury. One of the barriers to proactive prevention of medicine-induced kidney injury in primary care is the lack of a list of nephrotoxic medicines that are most problematic in primary care, particularly one that provides a comparison of risks across medicines. OBJECTIVE: The aim of this study was to consolidate evidence on the risks associated with medicines and acute kidney injury, with a focus on medicines used in primary care. METHOD: We searched the MEDLINE and EMBASE

Effects of renin-angiotensin system blockade on the islet morphology and function in rats with long-term high-fat diet.

The renin-angiotensin system (RAS) has an important role in the endocrine pancreas. Multiple researches have shown that even in the insulin resistance phase, there are many abnormalities in islets morphology and function. This study aimed at investigating the effects of RAS blockade on the islets function in rats with long-term high-fat diet and its mechanisms. Wistar rats with 16-week high-fat diet were randomly divided into perindopril intervention group (FP, n = 15) and telmisartan intervention group (FT, n = 15). After 8-week intervention, insulin sensitivity and islets function were detec

Efficacy of Terminalia arjuna in essential hypertension: A randomized controlled study.

OBJECTIVE: Hypertension is still the most prevalent of all non-communicable chronic diseases worldwide. It is the most common risk factor for cardiovascular disease (CVD) burden and mortality. In 2019, hypertension was reported as the number one and two cause of death in women, and men respectively, accounting for almost 20% of deaths globally. In India, 57% of stroke and 24% of coronary heart disease (CHD) deaths are directly related to high blood pressure. To achieve true control over this disorder, novel antihypertensive therapies need to be designed. In light of the current situation, a cl

Factors associated with changes in retinal microcirculation after antihypertensive treatment.

We report the results of hypertensive treatment over retinal arteriole narrowing in a group of 189 hypertensive patients during a 6-month treatment programme for hypertension. These patients were included in an observational study under clinical practice conditions and analysed using digital photographs of the eye fundus with a previously described semiautomatic snakes method. We analysed the relation between blood pressure control and retinal microcirculation changes during the treatment. We included 189 hypertensive patients; 74.0% of them had BP under control after 6 months of follow-up. Th

Three Low-Dose Antihypertensive Agents in a Single Pill after Intracerebral Hemorrhage.

BACKGROUND: Blood-pressure reduction is the only proven treatment to prevent stroke. Whether a single pill that combines three antihypertensive drugs at low doses, in addition to standard antihypertensive treatment, can lower blood pressure more than standard care alone and reduce the risk of recurrent stroke after intracerebral hemorrhage is uncertain. METHODS: We conducted a multinational, double-blind, randomized, placebo-controlled trial involving patients with a history of intracerebral hemorrhage. Patients were eligible for the trial if they had a systolic blood pressure of 130 to 160 m

Angiotensin-II receptor antagonists: their place in therapy.

Angiotensin-II receptor antagonists (or blockers) are a newer class of antihypertensive agents. These drugs are selective for angiotensin II (type 1 receptor); unlike angiotensin-converting enzyme inhibitors, they do not inhibit bradykinin metabolism or enhance prostaglandin synthesis. Angiotensin-II receptor antagonists are well tolerated. Cough occurs much less often with these agents than with angiotensin-converting enzyme inhibitors, and they do not adversely affect lipid profiles or cause rebound hypertension after discontinuation. Clinical trials indicate that angiotensin-II receptor ant

Gene Expression Profiling to Unveil Novel Biomarkers for Early Diagnosis and Therapies for Breast Cancer.

INTRODUCTION: Breast cancer (BC) is a leading cause of cancer-related death worldwide. Early detection and accurate diagnosis can improve patient outcomes and survival rates. Thus, identifying drugs and biomarkers for early detection is necessary for diagnosis, prognosis, and treatment. MATERIALS AND METHODS: Using machine learning techniques, we identified 160 differentially expressed genes (DEGs) between BC and control samples from four microarray datasets (GSE26910, GSE3744, GSE29431, and GSE42568), which were adopted due to the frequent presence of outliers in microarray gene-expression d

The Effect of Commonly Used Fixed-Dose Single Pill Combinations of Renin-Angiotensin-System Blockers and Calcium Channel Blockers on HOMA-IR Index in Hypertensive Patients with Impaired Fasting Glucose: a 12-Week Randomized Open-Label Prospective Study.

Backround: The effect of antihypertensive drugs on glucose homeostasis and insulin resistance remains an issue under investigation. There is evidence that renin-angiotensin system (RAS) blockers may favorably affect glucose metabolism, while treatment with calcium channel blockers (CCBs) is considered to have an overall neutral metabolic effect. However, the effects on glycemic indices may differ among agents within the same class of antihypertensive drugs. Objective: To evaluate the effects of different fixed-dose single pill combinations of RAS blockers with CCBs on homeostatic model assessm

Triple pharmacological blockade of the renin-angiotensin-aldosterone system in nondiabetic CKD: an open-label crossover randomized controlled trial.

BACKGROUND: Agents inhibiting the renin-angiotensin-aldosterone (RAAS) system have an important role in slowing the progression of chronic kidney disease. We evaluated the hypothesis that the addition of an aldosterone receptor antagonist to an angiotensin-converting enzyme (ACE) inhibitor and angiotensin II type 1 (AT-1) receptor blocker (ARB) (triple RAAS blockade) may provide an additional benefit compared with an ACE inhibitor and ARB (double RAAS blockade). DESIGN: Randomized open controlled crossover study. SETTING & PARTICIPANTS: 18 whites (7 women, 11 men) from the Outpatient Departm
