Endothelial cells release soluble factors that support the long-term survival of filarial worms in vitro.

The inability to maintain filarial nematodes in long-term in vitro culture greatly limits research into the basic biology of these parasites and hinders in vitro screening of novel anti-filarial agents. In this study, we sought to characterize nutrients that promote the long-term survival of filarial worms in vitro. Using microfilariae (MF) obtained from gerbils infected with Litomosoides sigmodontis, a filarial parasite of rodents, we found that Dulbecco's Modified Eagle Medium (DMEM) supplemented with 10% fetal bovine serum (FBS) resulted in MF survival of only 5 days. However, co-culturing

Trans-spliced leader RNA, 5S-rRNA genes and novel variant orphan spliced-leader of the lymphatic filarial nematode Wuchereria bancrofti, and a sensitive polymerase chain reaction based detection assay.

A genomic library of Wuchereria bancrofti was examined for the presence of the 22 nucleotide spliced leader (SL) which plays a vital role in the maturation of the 5' end of certain mRNAs through the addition of a small spliced leader (SL) exon and also in the generation of monocistronic mRNA from initial polycistronic transcripts in nematodes. Here, we report the characterization of three SL RNA genes (SLG1, SLG2 and SLG3), an internal copy of a novel variant SL1 sequence (SL1v) with 23 nucleotides within an open reading frame of 75 amino acid residues of an unknown gene and two 5S-rRNA genes

[Acanthamoebae, Naegleria and other free-living Amoebae in cooling and rinsing water of dental treatment units].

215 water samples were taken from 49 dental treatment units and investigated for the existence of free-living amoebae. In all water-carrying systems of the dental treatment units it was possible to verify the incidence of one or more amoeba species. In 8.2 per cent of the units Naegleria species was found and in 12.2 per cent Acanthamoeba species was present. Seven Naegleria and six Acanthamoeba strains (2 A. castellanii and 4 A. polyphaga) were isolated. From samples originating from 12 dental treatment units (DTU) another 42 amoeba strains were isolated which consisted of 14 different specie

[The usefulness of ELISA test for early serological detection of Trichinella spp. infection in pigs].

Trichinellosis is a parasitic zoonosis transmitted to humans through consumption of raw or undercooked meat from animals infected with nematodes of the Trichinella genus. Every year seropositive cases are found among the human population and thus trichinellosis still remains an epidemiologically important disease. The first step of the study was the optimization of a new ELISA method enabling an early and specific serological diagnosis of trichinellosis in pigs and wild boars using excretory-secretory (ES) antigens obtained from in vitro cultures of L1 T. spiralis. Serum samples were assayed f

Prevalence of Onchocerca lupi in shelter dogs from an endemic region of the Southwestern USA.

BACKGROUND: Onchocerca lupi is a zoonotic, vector-borne filarioid nematode that mainly infects wild and domestic canids in the Southwestern USA, Europe, Asia, and Africa. Clinical canine infections are associated with ocular disease, characterized by the presence of nodules and conjunctivitis. Subclinical cases can be challenging to diagnose, even with evaluation of cutaneous tissues for microfilariae. Current diagnostic tests include conventional polymerase chain reaction (cPCR) to detect O. lupi DNA, and, alternatively, real-time PCR (qPCR), which provides more rapid results and higher throu

Reduction in the number of infective Trichinella spiralis larvae in mice by use of homeopathic drugs.

BACKGROUND: Trichinellosis caused by the gastrointestinal nematode Trichinella spiralis occurs in humans, domestic animals and wild animals. It is difficult to control the muscle phase of the parasite. Homeopathic drugs such as Cina and Santoninum have anthelmintic properties. We have observed that in material doses, the homeopathic drug Podophyllum also has nematotoxic properties. We have also observed that homeopathic potency can influence the water permeability of cells. OBJECTIVE: The purpose of this study was to investigate whether potentized homeopathic drugs such as Cina 30, Santoninum

Whole-genome resequencing of the Kazakh coarse-wool fat-tailed sheep identifies functional variants in disease-associated genes.

The Kazakh coarse-wool fat-tailed sheep is an indigenous breed of Central Asia and represents a valuable genetic resource adapted to harsh continental environmental conditions. Despite its economic importance, its genomic architecture, population structure, and the genetic basis of disease resistance remain largely unexplored. Whole-genome sequencing was performed on 20 Kazakh coarse-wool fat-tailed sheep using the Illumina NovaSeq 6000 platform. Population structure was assessed by principal component analysis (PCA) using comparative genomic data from six domestic sheep breeds and the wild an

Pulmonary toxocariasis presenting as migratory pulmonary infiltrates and mediastinal lymphadenopathy: a case report and literature review.

BACKGROUND: Pulmonary toxocariasis, caused by the nematode Toxocara canis or T. cati, is an underdiagnosed cause of eosinophilic lung disease with highly variable radiological presentation that often mimics malignancy or other eosinophilic conditions. CASE PRESENTATION: A 53-year-old female smoker presented with progressive dyspnea and cough. Initial chest CT revealed right lower lobe consolidation with mediastinal lymphadenopathy (station 4R with short-axis diameters 12 mm), raising concern for lung cancer. Bronchoscopy with EBUS-TBNA was non-diagnostic. The patient was empirically started o

A UAV-based semi-supervised segmentation framework for pine wilt disease via vegetation Index-RGB synergy.

Pine wilt disease (PWD), caused by the pine wood nematode (Bursaphelenchus xylophilus), continues to threaten forest ecological security. Unmanned aerial vehicle (UAV) remote sensing makes large-scale screening feasible, but accurate canopy segmentation remains difficult in practice. The main obstacles are threefold: (i) canopy appearance changes noticeably from early to late infection stages, which can cause model representations to drift toward later-stage symptoms and weaken subtle early-stage cues; (ii) dense pixel-wise annotation is costly, making semi-supervised learning dependent on imp

Description of Heterorhabditis americana n. sp. (Rhabditida, Heterorhabditidae), a new entomopathogenic nematode species isolated in North America.

BACKGROUND: Heterorhabditis are important biological control agents in agriculture. Two Heterorhabditis populations, S8 and S10, were isolated from agricultural soils in the United States of America. Molecular analyses, based on mitochondrial and nuclear genes, showed that these populations are conspecific and represent a novel species of the "Bacteriophora" clade. This species was named Heterorhabditis americana n. sp. and is described in this study. METHODS: To describe H. americana n. sp., we carried out phylogenetic reconstructions using multiple genes, characterized their morphology, con

Knowledge, attitudes and practices of Australian dairy goat farmers towards the control of gastrointestinal parasites.

BACKGROUND: Gastrointestinal parasites such as nematodes and coccidia are responsible for significant economic losses in the goat industry globally. An indiscriminate use of antiparasitic drugs, primarily registered for use in sheep and cattle, in goats has resulted in drug-resistant gastrointestinal parasites. Very little is known about the gastrointestinal parasite control practices used by Australian dairy goat farmers that are pivotal for achieving sustainable control of economically important parasites. The study reported here provides insights into gastrointestinal parasite control pract

Reducing fecal egg count through selective breeding alters dorper lamb response to Haemonchus contortus in an artificial challenge trial.

Infection by gastrointestinal nematodes (GIN), particularly Haemonchus contortus, can be detrimental to sheep health and performance. Genetic susceptibility to GIN varies between breeds, with those lacking high levels of natural resistance often requiring frequent anthelmintic treatment when facing parasitic challenge. Genetic technology can serve as a tool to decrease GIN susceptibility via selection for sheep with reduced fecal egg count (FEC) estimated breeding values (EBVs). However, the physiological changes that result from implementation of this strategy are not well described. Addition

Prevalence of Strongyloides in Southeast Asia: a systematic review and meta-analysis with implications for public health and sustainable control strategies.

BACKGROUND: Strongyloidiasis, caused by the nematodes Strongyloides stercoralis and Strongyloides fuelleborni, is estimated to affect over 600 million individuals worldwide. The disease is endemic in Southeast Asia, where a warm-humid climate and socio-economic conditions maintain the parasite's life cycle and transmission. However, the current diagnostic methods may not be sufficiently sensitive, suggesting that the true prevalence of strongyloidiasis could be seriously underestimated in this. This study aims to determine the prevalence of strongyloidiasis in Southeast Asia through a systemat

Establishment of an in vitro culture system to study the developmental biology of Onchocerca volvulus with implications for anti-Onchocerca drug discovery and screening.

BACKGROUND: Infections with Onchocerca volvulus nematodes remain a threat in Sub-Saharan Africa after three decades of ivermectin mass drug administration. Despite this effort, there is still an urgent need for understanding the parasite biology especially the mating behaviour and nodule formation as well as the development of more potent drugs that can clear the developmental (L3, L4, L5) and adult stages of the parasite and inhibit parasite reproduction and behaviour. METHODOLOGY/PRINCIPAL FINDINGS: Prior to culture, freshly harvested O. volvulus L3 larvae from dissected Simulium damnosum f

Intra Vitam Diagnosis of Neglected Gurltia paralysans Infections in Domestic Cats (Felis catus) by a Commercial Serology Test for Canine Angiostrongylosis and Insights into Clinical and Histopathological Findings-Four-Case Report.

Gurltia paralysans is a metastrongyloid nematode which belongs to the Angiostrongylidae family and presents tropism for veins of the subarachnoid space in vivo of domestic and wild felids causing a progressive and chronic clinical manifestation of paraparesis/paraplegia. The geographic distribution of G. paralysans includes rural and periurban areas of South America and was recently reported in Europe. To date, a definitive diagnosis of feline gurltiosis is still conducted by post-mortem examination of the spinal cord in affected animals. A presumptive diagnosis of feline gurltiosis can also b

Extreme-QTL mapping of monepantel resistance in Haemonchus contortus.

BACKGROUND: Haemonchus contortus, a gastrointestinal nematode parasite of sheep, is mainly controlled by anthelmintics; the occurrence of anthelmintic resistance leads to treatment failures and increases economic burden. Because molecular mechanisms involved in drug resistance can be elucidated by genomic studies, an extreme quantitative trait locus (X-QTL) mapping approach was used to identify co-segregation of the resistance phenotype with genetic markers to detect the genome-wide variants associated with monepantel resistance in H. contortus. METHODS: A cross between H. contortus isolates
