CAD/CAM-guided vascularized fibula graft for anterior maxillary reconstruction: a case report.

INTRODUCTION AND IMPORTANCE: The reconstruction of large anterior maxillary defects poses a significant challenge due to the complex three-dimensional anatomy and functional demands of the midface. While autogenous bone grafts are the gold standard, vascularized free flaps, particularly the fibula, offer superior outcomes for large defects. This case report highlights the application of computer-aided design and computer-aided manufacturing (CAD/CAM) technology to enhance the precision and predictability of such reconstructions. CASE PRESENTATION: A 22-year-old male presented with a substanti

The role of FGF9-mediated TGF-β1/Smad signaling in enamel hypoplasia induced by exposure to fluoride and SO2 in rats.

Many geographical areas of the world are polluted by both fluoride and sulfur dioxide (SO2). However, the effects of simultaneous exposure to fluoride and SO2 on teeth are unknown. Fibroblast growth factor-9 (FGF9) and transforming growth factor-β1 (TGF-β1) are key signaling molecules in enamel development. The purpose of the study was to explore the effects of co-exposure to fluoride and sulfur dioxide on enamel and to investigate the role and mechanism of FGF9 and TGF-β1. First, sodium fluoride (NaF) and SO2 derivatives were used to construct rat models and evaluate the enamel development of

Prospective in (Primate) dental analysis through tooth 3D topographical quantification.

The occlusal morphology of the teeth is mostly determined by the enamel-dentine junction morphology; the enamel-dentine junction plays the role of a primer and conditions the formation of the occlusal enamel reliefs. However, the accretion of the enamel cap yields thickness variations that alter the morphology and the topography of the enamel-dentine junction (i.e., the differential deposition of enamel by the ameloblasts create an external surface that does not necessarily perfectly parallel the enamel-dentine junction). This self-reliant influence of the enamel on tooth morphology is poorly

Splicing mutation in DSPP causes dentinogenesis imperfecta and amelogenesis imperfecta.

BACKGROUND: Dentin sialophosphoprotein (DSPP) is an extracellular matrix protein, which is highly expressed in odontoblasts and transiently expressed in presecretory ameloblasts. DSPP mutations were related to dentinogenesis imperfecta (DGI), some of which can be accompanied by amelogenesis imperfecta (AI). However, the mechanism underlying DGI and AI caused by DSPP mutations is still unclarified. This study aimed to reveal the molecular pathogenesis in a Chinese family with both DGI and AI caused by the DSPP splicing mutation. METHODS: One Chinese family with DGI and AI was recruited. SLC4A9 deficiency is involved in Sjögren's syndrome and enamel defects through RPS27-triggered ribosomal stress.

Xerostomia, a salivary gland disorder affecting up to 30% of adults, compromises oral homeostasis by reducing salivary flow and buffering capacity. Amelogenesis imperfecta is a hereditary condition characterized by defective enamel formation and mineralization, with a prevalence ranging from 1 in 700 to 1 in 14,000 children worldwide. Although xerostomia has been associated with increased susceptibility to dental caries and subsequent enamel damage, its relationship with amelogenesis imperfecta remains unclear, and no shared molecular pathway has been causally linked to both hyposalivation and

Novel genetic linkage of rat Sp6 mutation to Amelogenesis imperfecta.

BACKGROUND: Amelogenesis imperfecta (AI) is an inherited disorder characterized by abnormal formation of tooth enamel. Although several genes responsible for AI have been reported, not all causative genes for human AI have been identified to date. AMI rat has been reported as an autosomal recessive mutant with hypoplastic AI isolated from a colony of stroke-prone spontaneously hypertensive rat strain, but the causative gene has not yet been clarified. Through a genetic screen, we identified the causative gene of autosomal recessive AI in AMI and analyzed its role in amelogenesis. METHODS: cDN

Retinoic Acid Excess Impairs Amelogenesis Inducing Enamel Defects.

Abnormalities of enamel matrix proteins deposition, mineralization, or degradation during tooth development are responsible for a spectrum of either genetic diseases termed Amelogenesis imperfecta or acquired enamel defects. To assess if environmental/nutritional factors can exacerbate enamel defects, we investigated the role of the active form of vitamin A, retinoic acid (RA). Robust expression of RA-degrading enzymes Cyp26b1 and Cyp26c1 in developing murine teeth suggested RA excess would reduce tooth hard tissue mineralization, adversely affecting enamel. We employed a protocol where RA was

Preameloblast-Derived Factors Mediate Osteoblast Differentiation of Human Bone Marrow Mesenchymal Stem Cells by Runx2-Osterix-BSP Signaling.

Epithelial-mesenchymal interaction occurs during development of various tissues, including teeth and bone. Recently, a preameloblast-conditioned medium (PA-CM) from mouse apical bud cells (ABCs), a type of dental epithelial cell, was found to induce odontogenic differentiation of dental pulp stem cells and promote dentin formation. The aims of the present study were to investigate the effects of PA-CM on human bone marrow mesenchymal stem cells (hBMSCs) in vitro, and to investigate the bone regenerative capacity in vivo through epithelial-mesenchymal interactions of developmental osteogenesis.

The distribution of cellular retinoic acid-binding protein I (CRABPI) and cellular retinol-binding protein I (CRBPI) during molar tooth development and eruption in the rat.

The distribution of cellular retinoic acid-binding protein (CRABPI) and cellular retinol binding protein (CRBPI) was studied in a series of prenatal and early postnatal rats, covering the main stages of development and eruption of the molar teeth. CRABPI positive cells were found in the mesenchymal cells of the dental follicle from the cap stage and in the dental papilla from the early bell stage. In the dental papilla, CRABPI positive cells were situated adjacent to the enamel organ in the cervical loop region and in the subodontoblastic region. Newly formed odontoblasts were CRABPI positive

Leucine rich amelogenin peptide prevents ovariectomy-induced bone loss in mice.

Amelogenins, major extra cellular matrix proteins of developing tooth enamel, are predominantly expressed by ameloblasts and play significant roles in the formation of enamel. Recently, amelogenin has been detected in various epithelial and mesenchymal tissues, implicating that it might have distinct functions in various tissues. We have previously reported that leucine rich amelogenin peptide (LRAP), one of the alternate splice forms of amelogenin, regulates receptor activator of NF-kappa B ligand (RANKL) expression in cementoblast/periodontal ligament cells, suggesting that the amelogenins,

Mutations Causing X-Linked Amelogenesis Imperfecta Alter miRNA Formation from Amelogenin Exon4.

Amelogenin plays a crucial role in tooth enamel formation, and mutations on X-chromosomal amelogenin cause X-linked amelogenesis imperfecta (AI). Amelogenin pre-messenger RNA (mRNA) is highly alternatively spliced, and during alternative splicing, exon4 is mostly skipped, leading to the formation of a microRNA (miR-exon4) that has been suggested to function in enamel and bone formation. While delivering the functional variation of amelogenin proteins, alternative splicing of exon4 is the decisive first step to producing miR-exon4. However, the factors that regulate the splicing of exon4 are no

Trends in Research of Odontogenic Keratocyst and Ameloblastoma.

Odontogenic keratocyst (OKC) and ameloblastoma (AM) are common jaw lesions with high bone-destructive potential and recurrence rates. Recent advancements in technology led to significant progress in understanding these conditions. Single-cell and spatial omics have improved insights into the tumor microenvironment and cellular heterogeneity in OKC and AM. Fibroblast subsets in OKC and tumor cell subsets in AM have been analyzed, revealing mechanisms behind their biological behaviors, including OKC's osteolytic features and AM's recurrence tendencies. Spatial transcriptomics studies of AM have
