In vivo and in vitro assays of immunocompetence in bronchogenic carcinoma.

To determine the degree of correlation among the various in vivo and in vitro assays that could be used to assess immunocompetence in bronchogenic carcinoma, the response to common recall skin antigens, primary sensitization to DNCB and lymphocyte function tests based on blastogenic response to mitogens and in mixed lymphocyte culture (MLC) were tested in 48 patients with bronchogenic carcinoma. The values were compared to responses in 94 age-matched healthy control subjects. There were impaired skin tests reactions among squamous cell carcinoma patients, with little impairment of their lympho

The effects of dexamethasone, cyclosporine, and vitamin D(3) on the activation of dendritic cells stimulated by haptens.

By their potent antigen-presenting function, dendritic cells (DCs) play a crucial role in the initiation of T cell-mediated immunity, including allergic contact hypersensitivity. To acquire such potent antigen-presenting ability, DCs in tissue must be activated, with increased expression of costimulatory molecules. Recent progress in DC biology has demonstrated that DCs can be activated via a variety of substances, e.g. various cytokines, CD40 ligand, bacterial products, and haptens, to increase their antigen-presenting ability, probably by different mechanisms. Therefore, in this study, to el

Chronic lymphadenopathic toxoplasmosis. A case with marked hyperglobulinemia and impaired delayed hypersensitivity responses during active infection.

A patient with lymphadenopathic toxoplasmosis characterized by prolonged symptoms and repeated relapses with isolation of toxoplasma from lymph nodes is described. As the disease persisted and progressed, striking immunologic changes occurred that ultimately resulted in a state of extreme hyperglobulinemia associated with impaired delayed hypersensitivity responses. The case in question illustrates that progressive infection may occur in the face of high antibody levels of all immunoglobulin types whereas the only demonstrable immunologic impairment was of delayed hypersensitivity.

[Development in the immunotherapy of malignant melanoma].

Recent progress in the investigation of malignant melanoma has encouraged the possibilities of using immunological methods in the treatment of cancer. Under investigation are especially attempts to influence both the humoral and cellular immunity with cytotoxic effect on tumor cells. In perspective of these studies schedules for greater effectiveness of immunotherapy by combination of active, passive, eliminative or adoptive methods are conceivable. They provide a rational basis particulary because essential questions of alternation between the immune reactions are not yet understood.

Antiatopic dermatitis activity of Lissachatina fulica mucin: Network pharmacology analysis and experimental validation.

BACKGROUND/AIM: Lissachatina fulica mucin (LFM) has been used as a nutraceutical, therapeutic agent, and cosmetic ingredient. The bioactivity of mucin depends on bioactive compounds whose composition is strongly influenced by the snail's diet. This study investigated the bioactive compounds in L. fulica mucin as potential therapeutic agents for atopic dermatitis (AD). MATERIALS AND METHODS: LFM was collected from snails fed long beans and melon rinds for 4 weeks, freeze-dried, and analyzed using liquid chromatography-mass spectrometry followed by network pharmacology (NP) analysis to identify

Isolation and Characterization of a Novel Marine Peptide, WPN-15, from Walleye Pollock (Gadus chalcogrammus) Tail By-Products and Its Therapeutic Effects Against Atopic Dermatitis.

Background/Objectives: Atopic dermatitis (AD) is a multifactorial inflammatory skin disorder in which epidermal barrier disruption and dysregulated immune responses drive persistent cutaneous inflammation. Owing to their broad spectrum of biological activities, marine-derived peptides have attracted increasing attention as potential therapeutic agents capable of modulating inflammatory and immune pathways. Methods: In this study, a novel peptide, WPN-15 (NGAIADQQPQRPNIV), was isolated from enzymatic hydrolysates of walleye pollock (Gadus chalcogrammus) tail by-products using an activity-guided

Increases in human epidermal DR+CD1+, DR+CD1-CD36+, and DR-CD3+ cells in allergic versus irritant patch test responses.

In an attempt to differentiate an allergic patch test response from an irritant response, we evaluated by flow cytometry the percentages of various epidermal cell populations isolated from allergen and irritant-treated patch test sites. Nine allergic individuals were patch tested with various allergens (Rhus, dinitrochlorobenzene [DNCB], or nickel chloride) and a vehicle control for 48 h. Eight additional individuals were patch tested with irritating chemicals (sodium lauryl sulfate or nonanoic acid) and with a vehicle control for 48 h. Epidermal cells, isolated from suction blisters, were dou

Allergic contact dermatitis releases soluble factors that stimulate melanogenesis through activation of protein kinase C-related signal-transduction pathway.

Phenylazo-naphthol (PAN) allergy induces visibly well-defined and late-appearing hyperpigmentation of brownish yellow guinea pig skin in clear contrast to dinitrochlorobenzene (DNCB) allergy, which has very low incidence of hyperpigmentation. Skin extract from PAN allergy at 20-29 d post-challenge exhibited marked melanogenic stimulatory effects (3H2O release and 14C-thiouracil incorporation) when added to cultured guinea pig melanocytes. The time course in the appearance of melanogenic factor was definitely consistent with the induction pattern of visible pigmentation. By contrast, the additi

Topical Anti-Inflammatory Effects of Quercetin Glycosides on Atopic Dermatitis-Like Lesions: Influence of the Glycone Type on Efficacy and Skin Absorption.

Atopic dermatitis (AD) is a multifaceted inflammatory skin condition characterized by the involvement of various cell types, such as keratinocytes, macrophages, neutrophils, and mast cells. Research indicates that flavonoids possess anti-inflammatory properties that may be beneficial in the management of AD. However, the investigation of the glycoside forms for anti-AD therapy is limited. We aimed to assess the ability of quercetin-3-O-glycosides in treating AD-like lesions through in silico-, cell-, and animal-based platforms. The glycosylated flavonols of quercitrin, isoquercitrin, and rutin

Oral and topical administration of a geranyl acetophenone attenuates DNCB-induced atopic dermatitis-like skin lesions in BALB/c mice.

Atopic dermatitis (AD) is a chronic, allergic inflammatory skin disorder that lacks a definite cure. Using a mouse DNCB-induced AD-like skin lesions model, this study evaluated the potential therapeutic utility of tHGA as an oral and topical treatment for AD. Male BALB/c mice were sensitised and challenged with 1% and 0.5% DNCB on their shaved dorsal skin. Mice in the treatment group were administered tHGA (20, 40, and 80 mg/kg) orally three times per week for 2 weeks, or tHGA (0.2%, 1%, and 5%) topically once daily for 12 days. On day 34, the mice were euthanized, and blood and dorsal skin sa

Fermented Maesil (Prunus mume) with probiotics inhibits development of atopic dermatitis-like skin lesions in NC/Nga mice.

Maesil (Prunus mume Siebold & Zucc.), a potential source of free radical scavengers and inhibitor of pro-inflammatory mediators, is used in traditional Korean medical preparations as a remedy for skin disorders as have probiotics. The action of a probiotic fermented Maesil preparation on the development of atopic dermatitis (AD)-like skin lesions was determined in a NC/Nga mouse model as an initial step towards the development of a therapeutic feed supplement for use in dogs. Continuous ingestion of the experimental feed markedly inhibited the development of the AD-like skin lesions, as eviden

Band 3 tyr-phosphorylation in normal and glucose-6-phospate dehydrogenase-deficient human erythrocytes.

Haemolysis is usually episodic in glucose-6-phosphate dehydrogenase (G6PD) deficiency, often triggered by a period of oxidative stress. In the present work, we investigate a possible biochemical mechanism underlying the enhanced susceptibility of G6PD deficient red blood cells (RBC) to oxidative stress. We analysed eight male subjects with Mediterranean glucose-6P-dehydrogenase deficiency (G6PDd), class II, for their ability in phosphorylating erythrocyte membrane band 3 following oxidative and osmotic stress. Our findings show that this sensitivity is connected to an early membrane band 3 Tyr

PN-CeO2 as a potential therapeutic agent for atopic dermatitis: Antioxidant and anti-inflammatory effects.

Atopic dermatitis (AD) is a common inflammatory skin disorder, and oxidative stress plays a central role in its pathogenesis. Nanoceria, owing to its redox-regulating properties, hold promise as a treatment option for AD. Porous ceria nanorods (PN-CeO2) were synthesized to scavenge reactive oxygen species (ROS). PN-CeO2 was characterized by transmission electron microscopy, nitrogen adsorption/desorption tests, energy dispersive spectroscopy, X-ray diffraction, and X-ray photoelectron spectroscopy. Cytotoxicity was assessed using light microscopy and a Cell Counting Kit-8 assay. ROS-scavenging

Prevention of type 1 diabetes by inducing subclinical dermatitis on a small area.

OBJECTIVE: We have previously epidemiologically shown that type 1 diabetes is inversely associated with contact allergy. This finding is intriguing as type 1 diabetes and contact allergy are two completely different diseases, although T cells are involved in both diseases. The objective of this study was therefore to experimentally study the effect of contact allergens on the development of diabetes in non-obese diabetic mice. METHODS: Non-obese diabetic mice 4 weeks of age were separated into seven groups. One group was exposed to tapped water every 14th day, whereas the remaining six groups

Sensitivity to sunburn is associated with susceptibility to ultraviolet radiation-induced suppression of cutaneous cell-mediated immunity.

Skin cancer incidence is highest in white-skinned people. Within this group, skin types I/II (sun sensitive/tan poorly) are at greater risk than skin types III/IV (sun tolerant/tan well). Studies in mice demonstrate that ultraviolet radiation (UVR)-induced suppression of cell-mediated immune function plays an important role in the development of skin cancer and induces a susceptibility to infectious disease. A similar role is suspected in humans, but we lack quantitative human data to make risk assessments of ambient solar exposure on human health. This study demonstrates that ambient levels o
