Despite the vast availability of antibiotics, bacterial infections remain a leading cause of death worldwide. In an effort to enhance the armamentarium against resistant bacterial strains, 1,2,3-triazole (5a-x) and sulfonate (7a-j) analogues of natural bioactive precursors were designed and synthesized. Preliminary screening against two Gram-positive (Streptococcus pneumoniae and Enterococcus faecalis) and four Gram-negative bacterial strains (Pseudomonas aeruginosa, Salmonella enterica, Klebsiella pneumoniae, and Escherichia coli) was performed to assess the potency of these analogues as anti

Sleep is viewed typically through a brain-centric lens, with little known about the role of the periphery1,2. Here we identify a sleep function for peripheral macrophage-like cells (haemocytes) in the Drosophila circulation, showing that haemocytes track to the brain during sleep and take up lipids accumulated in cortex glia due to wake-associated oxidative damage. Through a screen of phagocytic receptors expressed in haemocytes, we discovered that knockdown of eater-a member of the Nimrod receptor family-reduces sleep. Loss of eater also disrupts haemocyte localization to the brain and lipid

The orphan cytokine receptor-like factor 3 (CRLF3) was identified as a neuroprotective erythropoietin receptor in locust neurons and emerged with the evolution of the eumetazoan nervous system. Human CRLF3 belongs to class I helical cytokine receptors that mediate pleiotropic cellular reactions to injury and diverse physiological challenges. It is expressed in various tissues including the central nervous system but its ligand remains unidentified. A CRLF3 ortholog in the holometabolous beetle Tribolium castaneum was recently shown to induce anti-apoptotic mechanisms upon stimulation with huma

Antioxidants produced by the parasite Schistosoma mansoni are believed to be involved in the maintenance of cellular redox balance, thus contributing to larval survival in their intermediate snail host, Biomphalaria glabrata. Here, we focused on specific antioxidant enzymes, including glutathione-S-transferases 26 and 28 (GST26 and 28), glutathione peroxidase (GPx), peroxiredoxin 1 and 2 (Prx1 and 2) and Cu/Zn superoxide dismutase (SOD), known to be involved in cellular redox reactions, in an attempt to evaluate their endogenous antioxidant function in the early-developing primary sporocyst st

This study investigates the prevalence and characteristics of disseminated neoplasia (DN) in Mytilus chilensis in the Los Lagos Region of southern Chile. DN is a malignant proliferative disease that has been described in several species of the genus Mytilus. More than 4.000 adult mussels, both cultured and natural, collected between 2016 and 2022 in different geographical areas (Calbuco-Hueihue, Castro, Quellón) and seasons (winter-spring, summer-autumn) were histologically analysed. The prevalence was assessed and three levels of severity of DN were defined, based on number of neoplastic cell

Apoptosis is an evolutionarily conserved mechanism that removes damaged or unwanted cells, effectively maintaining cellular homeostasis. It has long been suggested that a deficiency in this type of naturally occurring cell death could potentially lead to necrosis, resulting in the release of endogenous immunogenic molecules such as damage-associated molecular patterns (DAMPs) and a noninfectious inflammatory response. However, the details about how danger signals from apoptosis-deficient cells are detected and translated to an immune response are largely unknown. In this study, we found that D

Insect endoparasitoids are capable of suppressing the immune reaction of their habitual hosts in a specific way. Salt (1968) characterized some of the implications: This seeming contradiction--that defence reactions against all kinds of foreign bodies are available to insects and that endophagous parasitoids are nevertheless able to develop in insect hosts--is resolved by recourse to one of the principles of host specificity. Although insects as a group react to every foreign body in the sense that any organism or substance evokes a reaction in most insects, each species of insect fails to mak

The Epstein-Barr virus (EBV) commonly infects humans and is highly associated with different types of cancers and autoimmune diseases. EBV has also been detected in inflamed gastrointestinal mucosa of patients suffering from prolonged inflammation of the digestive tract such as inflammatory bowel disease (IBD) with no clear role identified yet for EBV in the pathology of such diseases. Since we have previously reported immune-stimulating capabilities of EBV DNA in various models, in this study we investigated whether EBV DNA may play a role in exacerbating intestinal inflammation through innat

Bee venom, a complex mixture of bioactive peptides and enzymes, is a promising natural immunostimulant for aquaculture applications, particularly as escalating antibiotic resistance necessitates alternative disease-prevention strategies. This study evaluated the dose-dependent effects of dietary lyophilized bee venom (LBV) from Apis mellifera on growth performance, antioxidant capacity, innate immunity, and resistance to Vibrio harveyi in juvenile Pacific white shrimp (Litopenaeus vannamei). LBV was chemically characterized by LC-DAD-ESI/MSⁿ, with melittin identified as the predominant compone

Previously, we reported the molecular cloning of cDNA for the prophenoloxidase activating factor-I (PPAF-I) that encoded a member of the serine proteinase group with a disulfide-knotted motif at the N-terminus and a trypsin-like catalytic domain at the C-terminus [Lee, S.Y., Cho, M.Y., Hyun, J.H., Lee, K.M., Homma, K.I., Natori, S. , Kawabata, S.I., Iwanaga, S. & Lee, B.L. (1998) Eur. J. Biochem. 257, 615-621]. PPAF-I is directly involved in the activation of pro-phenoloxidase (pro-PO) by limited proteolysis and the overall structure is highly similar to that of Drosophila easter serine protea

Shrimp aquaculture is a primary supplier of high-quality aquatic protein worldwide. Incorporating natural feed additives can strengthen health and disease resilience in cultured species. This study evaluated the dietary effects of Hylocereus undatus (dragon fruit, HU) peel powder on growth performance, metabolic profiles, digestive capacity, immune response, antioxidant capacity, tissue integrity, and disease resistance against Vibrio parahaemolyticus in Pacific white shrimp (Litopenaeus vannamei). Shrimp with an initial weight of 4.23±0.03 (n = 60 per treatment) were randomly distributed into

Hemolymph coagulation is a vital component of insect immunity, rapidly reducing fluid loss and limiting pathogen spread after injury. In Manduca sexta larvae, paralytic peptide (PP) is a fast-acting cytokine activated by an unknown serine protease, and its influence on hemolymph mechanics remains largely unexplored. Here, we examine how PP exposure alters the extensional and surface rheological properties of larval hemolymph, providing insight into the physical manifestation of the immune activation. Hemolymph collected from fifth-instar larvae exhibited weak viscoelastic behavior in 84% of ex

The present study was undertaken to elucidate the cellular mechanisms, which govern metal sequestration and detoxification in gastropods. For this purpose the pond snail, Lymnaea stagnalis was exposed to environmentally relevant concentrations of three species of metals (Al, Zn and Cd) for 30 days and the localization and fate of these metals were followed in different tissues of the snails. The measurement of relative distribution of metals between tissues revealed that the digestive gland and kidney account for most of the accumulated metals. Al and Cd (non-essential metals) were redistribut

The outcome of infection in the host snail Biomphalaria glabrata with the digenean parasite Schistosoma mansoni is determined by the initial molecular interplay occurring between them. The mechanisms by which schistosomes evade snail immune recognition to ensure survival are not fully understood, but one possibility is that the snail internal defence system is manipulated by the schistosome enabling the parasite to establish infection. This study provides novel insights into the nature of schistosome resistance and susceptibility in B. glabrata at the transcriptomic level by simultaneously com

Despite the known anti-inflammatory, antibacterial, and antioxidant properties of bee venom (apitoxin), its application for promoting growth and health in shrimp (Litopenaeus vannamei), remains largely unexplored. This study explored the effects of bee venom (BV) encapsulated nano-chitosan (BV-CSNPs) on the health and disease resistance of white shrimp. Two hundred forty shrimp (mean weight 6.0 ± 0.02 g) were randomly assigned to four groups (n = 60 per group), each consisting of four replicate hapa (1 m³ each) stocked with 20 shrimp. Shrimp were fed either a basal diet (control) or diets adde

Hexokinase (HK) is generally recognized as a crucial enzyme participating in glycolysis. In the present study, a HK (named CgHK) was identified as a potential pattern recognition receptor (PRR) from the Pacific oyster Crassostrea gigas. The open reading frame (ORF) of CgHK was of 1395 bp, encoding a peptide of 464 amino acids with one Hexokinase_1 domain and one Hexokinase_2 domain. The predicted amino acid sequence of CgHK shared 17%-29% similarities with that of other known HKs. The mRNA transcripts of CgHK were constitutively detected in all the examined tissues, with relative high expressi

Following infection with Schistosoma mansoni larvae, haemocytes of resistant Biomphalaria glabrata snails execute a rapid defence during which they migrate towards and encapsulate the parasites. Such immediate and precise responses are thought to depend on signal transduction cascades though the signalling components involved remain largely unknown. It is proposed that mitogen-activated protein kinases may play a role in B. glabrata immune signalling, in particular p38 mitogen-activated protein kinases, which are known to be associated with stress and inflammatory signalling. Using degenerate

Extracellular vesicles (EVs) are nano-sized lipid bilayer structures released by diverse cell types. For pathogenic fungi, EVs play critical roles by modulating immune responses and influencing disease progression. Previously, we demonstrated that vaccination with EVs from Candida albicans promoted protection in immunosuppressed mice subsequently infected with lethal doses of this organism. However, the impact of EVs in the presence of fungi is poorly investigated in vivo. Here, we demonstrated that co-injection of C. albicans yeasts and their EVs into Galleria mellonella larvae led to a rapid

Because parasite virulence factors target host immune responses, identification and functional characterization of these factors can provide insight into poorly understood host immune mechanisms. The fruit fly Drosophila melanogaster is a model system for understanding humoral innate immunity, but Drosophila cellular innate immune responses remain incompletely characterized. Fruit flies are regularly infected by parasitoid wasps in nature and, following infection, flies mount a cellular immune response culminating in the cellular encapsulation of the wasp egg. The mechanistic basis of this res
