There have been relatively few studies which support a link between Ganoderma boninense, a phytopathogenic fungus that is particularly cytotoxic and pathogenic to plant tissues and roots, and antimicrobial compounds. We previously observed that liquid-liquid extraction (LLE) using chloroformmethanol-water at a ratio (1:1:1) was superior at detecting antibacterial activities and significant quantities of antibacterial compounds. Herein, we demonstrate that antibacterial secondary metabolites are produced from G. boninense mycelia. Antibacterial compounds were monitored in concurrent biochemical

Many species of edible mushrooms are known to contain a wide array of compounds with high nutritional and medicinal values. However, these values vary widely among mushroom species because of the wide diversity of compounds with different solubilities to solvents used in extraction. We report here the comparison of antioxidant activity and cytotoxicity against cancer cells in extracts of Pleurotus ostreatus, P. sajor-caju, Agaricus campestris, and A. bisporus from 7 different solvents, including water, ethanol, ethyl acetate, acetone, chloroform, hexane, and petroleum ether. The extracts were

Many short cationic peptides have been identified as potent antimicrobial agents, but their modes of action are not well understood. Peptide synthesis on cellulose membranes has resulted in the generation of peptide libraries, while high-throughput assays have been developed to test their antibacterial activities. In this paper a microtiter plate-based screening method for fungi has been developed and used to test nine antibacterial peptides against the model fungus Aspergillus nidulans. Microscopical studies using sublethal peptide concentrations caused defects in polarized growth, including

Pulmonary coccidioidomycosis shares characteristics with other pulmonary pathologies. In tissue, spherules containing endospores are markers of Coccidioides immitis and C. posadasii infection. Mycelial forms presenting without classical parasitic structures are often misdiagnosed. The study was performed at the National Institute of Respiratory Diseases (INER) of Mexico between September 1991 and June 2005 and analyzed the association between cases, controls, and risk factors, including co-morbidity. A case was defined as any patient who presented mycelial forms and a control as any patient wh

Previous studies in Penicillium chrysogenum and Aspergillus nidulans suggested that self-processing of the isopenicillin N acyltransferase (IAT) is an important differential factor in these fungi. Expression of a mutant penDE(C103S) gene in P. chrysogenum gave rise to an unprocessed inactive variant of IAT (IAT(C103S)) located inside peroxisomes, which indicates that transport of the proIAT inside these organelles is not dependent on the processing state of the protein. Co-expression of the penDE(C103S) and wild-type penDE genes in P. chrysogenum (Wis54-DE(C103S) strain) led to a decrease in b

A Gram-positive actinomycete, designated strain C-140ᵀ, was isolated from the crustose lichen Arthonia leioplacella collected in Thailand. A polyphasic taxonomic study demonstrated that the strain represents a member of the genus Actinomycetospora. The 16S rRNA gene sequence analysis showed that strain C-140ᵀ exhibited the highest sequence similarity to Actinomycetospora lutea TT00-04ᵀ (98.62%), followed by Actinomycetospora lemnae DW7H6ᵀ (98.55%). Phylogenomic analysis indicated that Actinomycetospora chlora JCM 17979ᵀ is the closest phylogenetic relative of strain C-140ᵀ. Morphologically, th

Ganoderma species have been studied for their pharmacological approaches, such as anticancer, antitumor, antiproliferative, and antioxidant activity. Elicitors are used to increase Ganoderma bioactive composite production. This study aims to evaluate the antiproliferative activity of ethanolic extracts from mycelium of Ganoderma tuberculosum (G. tuberculosum) grown in a liquid medium with vineyard pruning waste (VPW) extracts as elicitors. Ethanolic and aqueous VPW extracts contain resveratrol dimer 4, resveratrol tetramer 1, and naringenin, while toluene and chloroform extracts contain tetrad

INTRODUCTION: Glycogen is a universal carbon and energy reserve that plays crucial roles in fungal stress adaptation, cellular homeostasis, and predatory activity. However, the role of glycogen metabolism in nematode-trapping fungi has largely remained unexplored. Here, we performed a systematic functional characterization of AfGsy2, the glycogen synthase-encoding gene in the biocontrol fungus Arthrobotrys flagrans.
METHODS: Using a CRISPR-Cas9-mediated deletion mutant, together with overexpression and complementation strains, we investigated the involvement of AfGsy2 in mycelial growth, stre

Alternaria alternata is a destructive postharvest pathogen that causes fruit decay and produces multiple mycotoxins, including alternariol (AOH), alternariol monomethyl ether (AME), and tenuazonic acid (TeA), posing a serious threat to crop quality and food safety. Biocontrol stands out as a promising strategy to mitigate both fungal infection and mycotoxin production. This study investigated that Bacillus sp. Z26 effectively inhibits the mycelial growth and mycotoxin accumulation of A. alternata. Comparative transcriptomic analysis revealed that strain Z26 did not directly repress canonical m

The soil-borne fungal pathogen Rhizoctonia solani remains a primary constraint to global crop productivity, causing significant damping-off and fruit rot across diverse cropping environments. In response to the ecological demand for sustainable biopesticides, this study explores the potential of bacterial Volatile Organic Compounds (VOCs) as a contact-independent, fumigant-based biocontrol approach. B. siamensis strain NKIT9 was evaluated for its antagonistic activity against R. solani. In vitro sealed-plate bioassays demonstrated that NKIT9-derived volatiles effectively suppress mycelial grow

The tangerine pathotype of A. alternata, which produces a unique host-selective ACT toxin causes brown spots on citrus leaves and fruits. In this study, we report a methionine biosynthesis regulator (MetR), which belong to bZIP transcription factor, is required for methionine metabolism, oxidative stress tolerance and pathogenicity. We generated two ΔAaMetR mutants in the tangerine pathotype of Alternaria alternata and investigated the resulting mutant phenotypes. The ΔAaMetR disruption mutant grew poorly in the absence of methionine and unable to produce conidia. Furthermore, pathogenicity te

Botrytis cinerea is the causal agent of gray mold disease in various plant species and produces grayish macroconidia and/or black sclerotia at the end of the infection cycle. It has been suggested that the pigmentation is due to the accumulation of 1,8-dihydroxynaphthalene (DHN) melanin. To unravel its basis and regulation, the putative melanogenic and regulatory genes were identified and functionally characterized. Unlike other DHN melanin-producing fungi, B. cinerea and other Leotiomycetes contain two key enzyme (PKS)-encoding enzymes. Bcpks12 and bcpks13 are developmentally regulated and ar

BACKGROUND: Resistance to carbendazim and other benzimidazole fungicides in Botrytis cinerea and most other fungi is usually conferred by one or several allelic mutations in the β-tubulin. Carbendazim resistance in Fusarium graminearum, however, differs from that in B. cinerea and other fungi in that F. graminearum has two β-tubulins (Fgtub1 and Fgtub2) rather than one, and the resistance is conferred by mutations in the β2 -tubulin. In a previous study, the β1 -tubulin of F. graminearum (Fgtub1) was replaced with the β-tubulin of B. cinerea conferring carbendazim resistance (BctubE198A). The

Fourteen strains of Pseudomonas fluorescens isolated from rhizosphere soil of rice were tested for their antagonistic effect towards Rhizoctonia solani, the rice sheath blight fungus. Among them, PfMDU2 was the most effective in inhibiting mycelial growth of R. solani in vitro. Production of chitinase, beta-1,3-glucanase, siderophores, salicylic acid (SA) and hydrogen cyanide (HCN) by P. fluorescens strains was evaluated. The highest beta-1,3-glucanase activity, siderophore production, SA production and HCN production were recorded with PfMDU2. A significant relationship between the antagonist

Tetramycin, a novel polyene macrolide antibiotic, has strong activity against a broad spectrum of fungi and may have potential uses in future agricultural applications. Thus, the antifungal activity and biochemical action of tetramycin on Colletotrichum scovillei were investigated in this study. The experimental results indicated that tetramycin had strong inhibitory activity against the mycelial growth, spore germination and germ tube elongation of C. scovillei. The baseline sensitivity curves were unimodal, with mean EC50 values of 1.98 ± 0.078 μg/mL and 0.003 ± 0.005 μg/mL for mycelial grow

Many factors must be considered in order to develop and implement treatment systems to improve the microbial quality of surface water and prevent the accidental introduction of plant and human pathogens into vegetable crops. The efficacy of chlorine gas (Cl2(g)) and chlorine dioxide (ClO2) injection systems in combination with rapid sand filtration (RSF) was evaluated in killing fecal indicator microorganisms in irrigation water in a vegetable-intensive production area. The efficacy of ClO2 and Cl2(g) was variable throughout the distribution systems and coliform bacteria never dropped below le

Biomaterials are increasingly being explored as sustainable alternatives to petroleum-derived materials, but many systems that perform well at the laboratory scale remain difficult to manufacture reliably at the industrial scale. One reason for this disconnect is that biomaterial structure and function are strongly influenced by the physical conditions present during assembly, including flow, transport, dewatering, and drying conditions that differ between laboratory and industrial environments. Here, I propose that scalable biomaterials should be designed with formation conditions and manufac

Dematophora necatrix (syn. Rosellinia necatrix), a fungal pathogen causing white root rot in trees, dies when exposed to 35 °C for more than 2 days. This study aimed to elucidate the mechanisms underlying fungal death under non-lethal heat conditions in the presence of soil microbes, testing the hypothesis that heat stress in D. necatrix is a trigger. After treatment at 35 °C for 2-12 h, correlative analysis was conducted using one mycelium of D. necatrix (W562) for gene expression, growth recovery, and competition ability against the hyphosphere fungus Talaromyces pinophilus (TAi6s), and anot
