Capsular polysaccharides of cultured phototrophic biofilms.

Phototrophic biofilm samples from an Italian wastewater treatment plant were studied in microcosm experiments under varying irradiances, temperatures and flow regimes to assess the effects of environmental variables and phototrophic biomass on capsular exopolysaccharides (CPS). The results, obtained from circular dichroism spectroscopy and High Performance Liquid Chromatography, suggest that CPS have a stable spatial conformation and a complex monosaccharide composition. The total amount present was positively correlated with the biomass of cyanobacteria and diatoms, and negatively with the bi

Incorporation of natural uncultivable Legionella pneumophila into potable water biofilms provides a protective niche against chlorination stress.

Legionella pneumophila is a waterborne pathogen that has been isolated sporadically from drinking water distribution systems (DWDS). Resistance to disinfectants is mainly attributed to the association of cells with amoebae, but biofilms are also thought to provide some degree of protection. In the present work, a two-stage chemostat was used to form heterotrophic biofilms from drinking water to study the influence of chlorine on the presence of naturally occurring L. pneumophila. The pathogen was tracked in planktonic and sessile biofilm phases using standard culture recovery techniques for cu

Influence of polymeric 3-alkylpyridinium salts from the marine sponge Reniera sarai on the growth of algae and wood decay fungi.

Polymeric alkylpyridinium salts (poly-APS) isolated from the marine sponge Reniera sarai act as antifouling and anticholinesterase agents. They also show moderate haemolytic and cytotoxic activities against different cell lines. The haemolytic activity of poly-APS is due to their detergent-like structure and behaviour in aqueous solutions. In this work, the lytic activity of poly-APS against freshwater and marine algae, and inhibitory effects on wood decay fungi, were investigated. The results show that poly-APS inhibit the proliferation and movements of susceptible algae. Effects of poly-APS

Pyrithiones as antifoulants: environmental fate and loss of toxicity.

The environmental fate and the loss of toxicity of two important antifouling actives, zinc pyrithione (ZnPT) and copper pyrithione (CuPT), were investigated using a bioassay study and an outdoor microcosm study. The bioassay used inhibition of the growth of a marine diatom (Amphora coffeaeformis) to measure the toxicity of ZnPT and CuPT over time in sterile, natural, and sediment-supplemented seawater. In natural seawater and sediment-supplemented seawater in the dark and in sterile seawater exposed to light, growth inhibition was reduced at rates corresponding to the rapid degradation rates f

Synthesis and anti-barnacle activities of novel isocyanocyclohexane compounds containing an ester or an ether functional group.

Ten novel isocyanocyclohexane compounds that possess an oxygenic functional group at the 4-position were synthesized and evaluated for their antifouling activity against the larvae of the barnacle Balanus amphitrite with the aim of exploring the structure-activity relationships further. The anti-barnacle effect of the synthesized compounds was in the EC50 range of 0.0096-17.0 microg ml(-1). Some ester derivatives exhibited extremely high antifouling activities, and none of the synthesized isocyanocyclohexane compounds showed significant toxicity. The results suggest that the ester function is

The development of a marine natural product-based antifouling paint.

Problems with tin and copper antifouling compounds have highlighted the need to develop new environmentally friendly antifouling coatings. Bacteria isolated from living surfaces in the marine environment are a promising source of natural antifouling compounds. Four isolates were used to produce extracts that were formulated into ten water-based paints. All but one of the paints showed activity against a test panel of fouling bacteria. Five of the paints were further tested for their ability to inhibit the settlement of barnacle larvae, Balanus amphitrite, and algal spores of Ulva lactuca, and

Inspection method for the identification of TBT-containing antifouling paints.

In order to ensure the effectiveness of the international convention which will prohibit the use of organotin compounds in antifouling paints applied to ships, it is essential to establish an inspection system to determine the presence of the prohibited compounds in the paint. In the present study, a method for the identification of organotin containing antifouling paints using a two-stage analysis process is investigated. Firstly, X-ray fluorescence analysis (XRF) is utilized, which could be used at the place of ship surveys or port state control. Using a portable XRF instrument customized fo

The influence of antimicrobial peptides and mucolytics on the integrity of biofilms consisting of bacteria and yeasts as affecting voice prosthetic air flow resistances.

The integrity of biofilms on voice prostheses used to rehabilitate speech in laryngectomized patients causes unwanted increases in airflow resistance, impeding speech. Biofilm integrity is ensured by extracellular polymeric substances (EPS). This study aimed to determine whether synthetic salivary peptides or mucolytics, including N-acetylcysteine and ascorbic acid, influence the integrity of voice prosthetic biofilms. Biofilms were grown on voice prostheses in an artificial throat model and exposed to synthetic salivary peptides, mucolytics and two different antiseptics (chlorhexidine and Tri

In vitro performance of eugenol and cinnamaldehyde on biofilms produced by Candidozyma auris.

Candidozyma auris has high rates of resistance to antifungals and hospital disinfectants. This yeast remains on surfaces for long periods, favoring the colonization of patients. Similarly Candida species, it produces persistent biofilms, which contribute to invasive infections, resulting in high mortality rates. In the search for new therapeutic alternatives of antifungal action, natural products have been widely studied due to their pharmacological potential and diverse biological activities. Eugenol and cinnamaldehyde are derived from the essential oils of cinnamon and clove, respectively, a

Development and evaluation of a dynamic in vitro biofilm model of dental unit waterlines incorporating simulated back-suction.

This study developed a dynamic in vitro dental unit waterlines (DUWLs) biofilm model incorporating simulated back-suction. The model consisted of 165-cm polyurethane tubing connected to a dynamic flow system and operated intermittently to simulate routine clinical use. Simulated back-suction was produced by injecting 0.1 mL of clinical oral fluid into the tubing after each operating cycle. Over 6 weeks, three model waterlines with simulated back-suction and three without simulated back-suction were compared with three clinical DUWLs from dental chair units. Myricetin as a potential therapeutic agent against Pseudomonas aeruginosa: inhibition of biofilm formation, quorum sensing, and virulence factor production.

Multidrug-resistant pathogenic bacteria, particularly Pseudomonas aeruginosa (P. aeruginosa), pose a significant threat to human health. Despite the huge persistence of antibiotics, there remains a lack of effective natural compounds capable of simultaneously disrupting quorum sensing (QS), biofilm formation, and virulence in this pathogen. This study aimed to investigate the inhibitory potential of myricetin against P. aeruginosa focusing on its ability to interfere with QS-regulated virulence traits. Antibacterial, antibiofilm, anti-QS, and virulence factor activities were evaluated using cr

Antifungal activity of chrysin against Candida albicans biofilms In vitro and In vivo.

Candida albicans (C. albicans) biofilms exhibit markedly enhanced tolerance to conventional antifungal agents, necessitating the identification of novel therapeutic alternatives. Chrysin, a naturally occurring flavonoid distributed in propolis and Passiflora species, possesses well-documented antioxidant and anti-inflammatory properties. However, its activity against C. albicans biofilms remains uncharacterized. This study comprehensively evaluated the antibiofilm efficacy of chrysin in vitro and in vivo. The sessile minimum inhibitory concentration causing 50% inhibition (SMIC50) of chrysin a

Murepavadin is a novel peptide antibiotic that exhibits potent antibiofilm activity and enhances the efficacy of conventional antibiotics against Pseudomonas aeruginosa.

Multidrug-resistant (MDR) Pseudomonas aeruginosa infections are difficult to treat due to biofilm formation. This study evaluates the antimicrobial and antibiofilm efficacy of murepavadin (MUR), a novel peptidomimetic targeting the LptD protein, alone and combined with conventional antibiotics against clinical isolates. The minimum inhibitory concentrations (MICs) of 50 isolates against MUR and five antibiotics were determined. Synergy in planktonic cultures was assessed by checkerboard assays. For biofilm, the Bliss independence model was applied using MBIC (minimum biofilm inhibitory concent

Anti-biofilm activity of essential oils from Salvia abrotanoides leaves and flowers against extensively drug-resistant and multidrug-resistant Acinetobacter baumannii.

Acinetobacter baumannii is a formidable nosocomial pathogen whose biofilm production is a key contributor to its multidrug resistance. This study reveals the significant antibacterial and antibiofilm potential of Salvia abrotanoides essential oils (EOs) against MDR and XDR A. baumannii. GC-MS analysis identified 19 novel compounds within the leaf (L-EO) and flower (F-EO) extracts. F-EO exhibited greater potency, inhibiting bacterial growth and biofilm formation at 1.75 mg/mL and 0.9 mg/mL, respectively. At sub-inhibitory concentrations, both EOs caused substantial downregulation (>84%) of the

Geraniol disrupts mature MRSA biofilms on venous catheters and shows synergistic activity with oxacillin against planktonic cells.

Infections caused by methicillin-resistant Staphylococcus aureus (MRSA), particularly those associated with biofilms on invasive medical devices such as venous catheters, represent a significant public health challenge due to increased antimicrobial resistance. This study evaluated the anti-biofilm potential of geraniol, a natural monoterpene, against MRSA strains. In a catheter-based in vitro model, geraniol substantially inhibited initial biofilm formation and effectively disrupted pre-formed mature MRSA biofilms on catheter surfaces. Scanning electron microscopy (SEM) corroborated structura

Corynebacterium striatum modulates the sessile lifestyle of wild-type and ΔsrtA mutant Staphylococcus aureus in multi-species biofilms.

Corynebacterium striatum and Staphylococcus aureus are opportunistic pathogens that share ecological niches and are frequently co-isolated in healthcare-associated infections. They commonly exist as sessile populations organized within multi-species biofilms associated with medical devices. Sortase A (SrtA) plays a critical role in anchoring surface proteins in S. aureus, thereby influencing adhesion, colonization, and biofilm formation. This study evaluated the influence of C. striatum on the formation and stability of biofilms produced by wild-type and isogenic ΔsrtA mutant S. aureus on hydr

Innovative microparticle strategies: harnessing essential oil-loaded lecithin/chitosan systems against Burkholderia pseudomallei.

Burkholderia pseudomallei, the causative agent of melioidosis, presents high antimicrobial resistance, largely due to its biofilm-forming ability. Here, innovative lecithin/chitosan microparticles loaded with lemongrass (LEO) and geranium (GEO) essential oils (LEOLCN and GEOLCN) were developed and characterized as a novel strategy to combat B. pseudomallei. Microparticles were characterized and evaluated for their effects on planktonic growth and biofilm inhibition, as well as for potential synergistic interactions with antibiotics. Microparticles exhibited a spherical morphology, high encapsu

Non-destructive monitoring of antibiotic action in Bacillus subtilis biofilms using dual-parameter swept-source optical coherence tomography.

Biofilms show an enhanced tolerance to antimicrobials compared to planktonic cells and conventional susceptibility tests provide limited insight into antibiotic action within intact biofilms. We assessed swept-source optical coherence tomography (SSOCT) as a non-destructive, label-free platform for biofilm monitoring, extracting two label-free parameters, the optical attenuation coefficient (μt) and biospeckle temporal contrast (γ). Biofilms cultured for 24 h, were treated with antibiotics at minimum biofilm eradication concentration (MBEC) determined by the Calgary biofilm device and imaged a
