BACKGROUND: To alleviate the shortage of human donor organs or tissues for the treatment of organ and tissue failure including diabetes, pigs are considered suitable donor animals. As organs from conventional pigs are usually too large, those from minipigs may be better suited. We recently characterized the Göttingen Minipigs, a breed well characterized concerning the presence of zoonotic microorganisms and found hepatitis E virus (HEV) and porcine cytomegalovirus (PCMV) in some animals. Here, we characterize another minipig, the Aachen Minipig (AaMP), a pig breed recently established close to

BACKGROUND: Porcine circovirus type 2 (PCV2)-associated diseases are a major problem for the swine industry worldwide. In addition to improved management and husbandry practices, the availability of several anti-PCV2 vaccines provides an efficient immunological option for reducing the impact of these diseases. Most anti-PCV2 vaccines are marketed as injectable formulations. Although these are effective, there are problems associated with the use of injectable products, including laborious and time-consuming procedures, the induction of inflammatory responses at the injection site, and treatmen

Differentiation of infected from vaccinated hosts (DIVH) is a critical step in virus eradication programs. DIVH-compatible vaccines, however, take years to develop, and are therefore unavailable for fighting the sudden outbreaks that typically drive pandemics. Here, we establish a protocol for the swift and efficient development of DIVH assays, and show that this approach is compatible with any type of vaccines. Using porcine circovirus 2 (PCV2) as the experimental model, the first step is to use Immunoglobin G (IgG) sero-dynamics (IsD) curves to aid epitope discovery (IsDAED): PCV2 Cap peptid

Risk factors for Post-weaning Multisystemic Wasting Syndrome (PMWS) at the pig level were identified using data from a longitudinal study in seven PMWS-affected farms in France. In each farm, a representative sample of 120 pigs (180 in one farm) was randomly selected after farrowing and followed from birth to slaughter. Individual information included serological status for Porcine Circovirus type 2 (PCV-2), Porcine Reproductive and Respiratory Syndrome (PRRS) virus, and Porcine Parvovirus (PPV), individual weight, rearing conditions, and clinical observations recorded at 7, 13, 16 and 21 week

INTRODUCTION: Porcine circoviruses (PCVs), belonging to the genus Circovirus in the family Circoviridae, are prevalent worldwide and pose a serious threat to the pig industry. Although vaccines against PCV2 are available, PCV3 vaccine development, prevention strategies, and surveillance for co-infections remain limited, leading to delayed outbreak responses. Therefore, the aim of this study was to develop a bivalent subunit vaccine targeting the prevalent strains of PCV2 and PCV3.
METHODS: Recombinant baculoviruses, Baculovirus-PCV2-Cap and Baculovirus-PCV3-Cap, were constructed to express th

Inappropriate municipal solid waste disposal areas lack waterproof base liners, drainage systems, treatment of waste degradation by-products, mass coverages, and vector control. Leachate is a by-product derived from water percolation through solid waste disposal sites, comprising an environmental and health contamination source when not adequately collected and treated. This study aimed to describe surface and groundwater viral diversity in a dumpsite area non-operational for over a decade and its surrounding neighborhood in the metropolitan Rio de Janeiro region employing shotgun metagenomic

Porcine circovirus type 2 (PCV2) infection causes significant economic losses in the global swine industry. The capsid (Cap) protein serves as the core antigen in PCV2 subunit vaccines, but its weak immunogenicity necessitates adjuvants to enhance the induced immune responses. Bacterial flagellin proteins are ideal candidates for next-generation vaccine adjuvants. In this study, we designed a novel flagellin-based adjuvant by co-expressing bacterial flagellin with nanobodies (Nb) specific to the PCV2 Cap protein (Nbcap-flagellin). This recombinant fusion protein leverages the high-affinity bin

Porcine circovirus type 2 (PCV-2) is the agent of one of the most important diseases in the swine industry. Although it has been controlled through vaccination, viremic piglets at birth may represent a risk by reducing vaccination efficacy. Since there are few reports on the viremic status of pre-suckling piglets regarding PCV-2 infection, we assessed the PCV-2 frequency in sows housed in 18 breeding farms with no history of clinical PCVAD in Brazil, using placental umbilical cord serum (PUCS). The selection criteria were: breeding farms with more than 1,000 sows; sows not vaccinated for PCV-2

Porcine reproductive and respiratory syndrome virus (PRRSV) and Porcine circovirus (PCV) are two important pathogens, which caused respiratory disease in pigs. PRRSV and PCV2 had caused great economic losses to the pig industry. Pigs coinfection with PCV2 and PRRSV were common in the clinic, PCV2 antibodies can be detected in most of the pigs. PCV2d and HP-PRRSV(JXA1-like) were two major viruses circulating in the pigs in China. In this study, HP-PRRSV (JXA1-like) and PCV2d were used to coinfect and (or) sequential infect 5-week-old weaned PCV2-antibody positive pigs and the clinical indicatio

BACKGROUND: Granulomatous enteritis in pigs is mostly associated with porcine circovirus 2 systemic disease (PCV2-SD) but less often caused by fungi, Salmonella spp., foreign bodies, Mycobacterium spp. or Lawsonia intracellularis. This unusual and rare presentation of Lawsonia intracellularis infection can only be diagnosed by histopathological and immunohistochemical confirmation.
CASE PRESENTATION: Postmortem examination was performed in three 9-10-week-old post-weaned growing pigs from an outdoor farm submitted with a clinical history of increased mortality and wasting over the last two ba

Cloacal swabs were collected from 12 Eurasian goshawk chicks during routine veterinary inspections to evaluate the gut-associated virome of goshawks used for falconry. The material was used for metagenomic screening with high-throughput sequencing. Sequences belonging to several viral families (Poxviridae, Circoviridae, Anelloviridae, Retroviridae, Parvoviridae and Astroviridae) were detected and their occurrence differed across samples G1-G12. Most of the identified sequences were highly similar to those of viruses that infect poultry, suggesting a probable alimentary transmission route. Two

Pigs are affected by a variety of pathogenic agents that need to be identified correctly and diagnosed even when co-infections may complicate the application of specific and targeted assays. Next-generation sequencing can provide new perspective to monitor viruses infecting or co-infecting diseased pigs. In this study, we tested, for the first time for diagnostic purposes in a livestock species, a new method based on whole-genome sequencing of all the DNAs extracted from the blood of nine pigs sampled from a farm where there was a suspected outbreak of Post-weaning Multisystemic Wasting Syndro

BACKGROUND: The objective of this study was to assess the efficacy of a trivalent vaccine mixture and compare it to the respective monovalent vaccines against Mycoplasma hyopneumoniae, porcine circovirus type 2 (PCV2), and porcine reproductive and respiratory syndrome virus (PRRSV).
RESULTS: Pigs that were triple challenged with M. hyopneumoniae, PCV2, and PRRSV following vaccination with the trivalent vaccine mixture exhibited a significantly better growth performance when compared to unvaccinated and challenged pigs. A statistical difference was not found when comparing pig populations whic

BACKGROUND: Porcine parvovirus 2 (PPV2) was detected in swine serum without showing any relationship with disease. The emergence of the virus seemed to be a unique event until other genetically highly similar parvoviruses were identified in China and, later in 2012, the presence of the virus was also described in Europe. PPV2 is widely distributed in pig populations where it is suspected to be involved in respiratory conditions, based on its frequent detection in lung samples. In order to investigate the potential pathogenic involvement of PPV2, 60 dead pigs were examined from two farms. They

Anatipestifer disease is a contagious disease caused by Riemerella anatipestifer, affecting primarily ducks, geese and turkeys, and characterised by listlessness, diarrhoea, sneezing, nasal discharge, and nervous signs. Sporadically, it occurs in a wide range of other domesticated and wild birds as well. The incidence and characteristics of the disease seen in the three main host species are summarised based on birds submitted for routine laboratory investigation in Hungary over the period 2010-2014. The infection was diagnosed in a higher percentage in geese (9.9%) and ducks (7.5%). It occurr

The aim of this work was to evaluate whether acute phase protein (APP) determinations could assist Official Veterinarians carrying out work in slaughterhouses. To test this hypothesis, the diagnostic accuracy of APP determinations in meat juice of pigs was analysed to differentiate between healthy and diseased pigs. One hundred and one pigs of two different origins were classified into two groups according to their health status (healthy and diseased pigs), which was determined by a veterinary clinical examination on the farm. To assess the pigs' immune status, against the main porcine disease

The emergence of severe porcine circoviral disease in North America is associated with Porcine circovirus-2 genotype b (PCV-2b), which has led to speculation that PCV-2b is more virulent than PCV-2a. The objectives of this study were to 1) correlate the PCV-2 DNA concentration and lesions in wasting (WST) and age-matched healthy (HLTH) pigs from 2 clinically affected farms, and unaffected (UNFCT) pigs from a farm with no prior clinical or diagnostic history of PCVD; and 2) to determine the initial estimates of sensitivity and specificity of PCV-2 quantitative polymerase chain reaction (qPCR).
