Carvacrol attenuates mechanical hypernociception and inflammatory response.

Carvacrol is a phenolic monoterpene present in the essential oil of the family Lamiaceae, as in the genera Origanum and Thymus. We previously reported that carvacrol is effective as an analgesic compound in various nociceptive models, probably by inhibition of peripheral mediators that could be related with its strong antioxidant effect observed in vitro. In this study, the anti-hypernociceptive activity of carvacrol was tested in mice through models of mechanical hypernociception induced by carrageenan, and the involvement of important mediators of its signaling cascade, as tumor necrosis fac

A novel Ru(II) complex with pyrene-1-carbaldehyde hydrazone: structural, electronic, theoretical, and biological studies.

A new Ru(II) half-sandwich complex, 1, containing an η6-p-cymene ligand and (E)-1-(pyren-1-ylmethylidene)hydrazine, was synthesized in good yield and characterized by FTIR, NMR, elemental analysis, and density functional theory calculations. The close agreement between experimental and theoretical FTIR, 1H NMR, and 13C NMR data confirmed the proposed structure, while geometry optimization revealed the expected pseudo-octahedral piano-stool arrangement around the Ru(II) center. QTAIM analysis indicated that the metal-ligand interactions, particularly Ru-N and Ru-arene bonds, possess partial cov

Improving the lethal effect of cpl-7, a pneumococcal phage lysozyme with broad bactericidal activity, by inverting the net charge of its cell wall-binding module.

Phage endolysins are murein hydrolases that break the bacterial cell wall to provoke lysis and release of phage progeny. Recently, these enzymes have also been recognized as powerful and specific antibacterial agents when added exogenously. In the pneumococcal system, most cell wall associated murein hydrolases reported so far depend on choline for activity, and Cpl-7 lysozyme constitutes a remarkable exception. Here, we report the improvement of the killing activity of the Cpl-7 endolysin by inversion of the sign of the charge of the cell wall-binding module (from -14.93 to +3.0 at neutral pH

Inhibition by menthol and its related chemicals of compound action potentials in frog sciatic nerves.

AIMS: Transient receptor potential (TRP) vanilloid-1 (TRPV1) and melastatin-8 (TRPM8) channels play a role in transmitting sensory information in primary-afferent neurons. TRPV1 agonists at high concentrations inhibit action potential conduction in the neurons and thus have a local anesthetic effect. The purpose of the present study was to know whether TRPM8 agonist menthol at high concentrations has a similar action and if so whether there is a structure-activity relationship among menthol-related chemicals. MAIN METHODS: Compound action potentials (CAPs) were recorded from the frog sciatic

Optimization of clofibrate by carvacrol results in a new hypolipidemic compound with low hepatic injury.

The hepatic injury caused by clofibrate (CF) is strongly associated with oxidative stress and inflammation. This study aims to develop a hypolipidemic compound that possesses antioxidant, anti-inflammatory properties and reduces liver injury. Carvacrol-clofibrate (CF-Carvacrol) was synthesized by optimizing the structure of CF by carvacrol. CF-Carvacrol showed significant lipid-lowering effects in hyperlipidemic mouse models induced by Triton WR 1339. The molecular docking results showed that CF-Carvacrol has a good affinity for PPAR-α. The liver injury study showed that CF-Carvacrol had signi
