Highly selective chemosensor for reactive carbonyl species based on simple 1,8-diaminonaphthalene.

Reactive carbonyl species (RCSs) including one carbon formaldehyde (FA) and dicarbonyl compounds such as methylglyoxal (MGO) and glyoxal (GO) are produced during demethylase reactions and various glucose metabolic pathways respectively. Elevation of the RCSs concentrations in cells is due to abnormal DNA damage, glycation adducts with macromolecules that lead to various neurotoxic diseases. Hence, regular monitoring of these RCSs with an easy tool is of utmost interest. However, conventional methods such as chromatography and mass spectrometry for the detection of these species are not so econ

A Bioorthogonal Ratiometric Fluorescence Nanoprobe for Imaging the Regulation of O-GlcNAcylation in Atherosclerosis.

Protein post-translational modification (PTM) plays a significant role in the progression of atherosclerosis (AS). However, the role of O-GlcNAcylation, as an important O-glycosylation modification, in the formation of foam cells remains unclear, limiting early diagnosis and mechanistic studies. Here, utilizing two bioorthogonal reactions, we developed a ratiometric fluorescent nanoprobe-based tracker for O-GlcNAcylation. This tracker (BORFI) enables dynamic tracing, relative quantification, and multi-scale in situ visualization of protein O-GlcNAcylation during cellular events, and images the

Potent Caspase-1 Antagonists From Dihydropyridine Derivatives: In Vitro Fluorescence Assays and Cell Imaging.

Caspase-1 is a key marker in pyroptosis and inflammation, playing a central role in the pathogenesis of various acute and chronic inflammatory diseases. While some caspase-1 sensing and inhibition studies have been reported, the use of DHPs for these applications remains unexplored. To address this challenge, a novel fluorescent compound, zWEHD-NH-DHP, was synthesized and subsequently developed as a real-time caspase-1 detection in vitro and in cells. Furthermore, a new class of potential caspase-1 inhibitors was investigated through the screening of a series of nonfluorogenic DHPs, utilizing

A mitochondria-targeted H2O2/viscosity dual-responsive fluorescent probe for visualizing redox-biophysical remodeling in LPS-induced acute kidney injury.

Renal tubular epithelial cells are highly susceptible to mitochondrial dysfunction during acute kidney injury (AKI), in which oxidative stress and microenvironmental remodeling occur before overt functional deterioration. Hydrogen peroxide (H2O2) is an important but nonspecific redox mediator, whereas mitochondrial viscosity provides a complementary biophysical readout associated with organelle stress, protein aggregation, membrane damage, and impaired molecular diffusion. Simultaneous imaging of mitochondrial H2O2 and viscosity may therefore provide a dual-parameter strategy for interrogating

Colorimetric/fluorescent dual-mode detection of norfloxacin in food samples using Fe/Mn-MOF/CeO2 nanozyme with high peroxidase-like activity.

A dual-metal metal-organic framework (MOF)-based nanozyme, Fe/Mn-MOF/CeO2, with high peroxidase-like activity was successfully synthesized via a stepwise solvothermal-hydrothermal combined liquid-phase composite strategy. A colorimetric/fluorescence dual-mode sensing platform was fabricated for the detection of norfloxacin (NOR) in milk and honey. The material takes advantage of the synergistic electronic effect of Fe/Mn bimetallic active sites and the reversible Ce4+/Ce3+ valence cycle to remarkably enhance its peroxidase-like activity. The nanozyme can trigger the decomposition of hydrogen p

Reactive nitrogen species-driven oxidative and nitrative damage underlying cadmium/mercury co-exposure: A multimodal fluorescent visualization study.

Heavy metal contamination severely threatens ecological environment and human health. Few studies have established an integrated mechanistic framework linking heavy metal co-exposure, reactive nitrogen species (RNS) burst, and progressive cellular injury, especially for multi-metal combined pollution that better reflects real environmental scenarios. As downstream substance of nitric oxide (NO) and representative RNS, peroxynitrite (ONOO-) possess stronger oxidation/nitration capacity compared with hydrogen peroxide (H2O2). The oxidative/nitrifying stress ignited by RNS outburst under cadmium

Biofunctional alginate hydrogel enabling Fe³⁺ detection and therapeutic modulation of hepatic fibrosis.

Hepatic fibrosis, driven by hepatic stellate cell (HSC) activation and excessive extracellular matrix (ECM) deposition, is closely associated with iron overload. To enable simultaneous diagnosis and treatment, a multifunctional hydrogel nanocomposite (SA-1@MG) was developed by modifying sodium alginate with a compound 1-based Fe³⁺-responsive fluorophore and loading mangiferin (MG). SA-1@MG exhibited strong, selective fluorescence quenching toward Fe³⁺ (R² = 0.9992) and high stability under physiological conditions. In vitro studies using TGF-β1-induced LX-2 cells confirmed that SA-1@MG effecti

Decoupling carboxyl position and annulation site effects on ICT, pH response, and serum albumin binding in dual-regioisomeric benzoindole fluorophores.

This study reports the synthesis of three donor-acceptor (D-A) fluorescent probes (BI-1, BI-2, and BI-3) based on a benzoindole scaffold. The effects of carboxyl substitution position and benzoindole fusion pattern on their photophysical properties and protein-binding behavior were systematically examined. Spectroscopic investigations combined with molecular docking analysis demonstrate that subtle structural variations lead to pronounced differences in absorption and emission characteristics, fluorescence quantum yields, pH responsiveness, and binding preferences toward serum albumin. Ultrasensitive low-density lipoprotein monitoring in early cardiovascular risk assessment via Cu-MOF-Apt/MoS₂ fluorescent nanosensor.

Cardiovascular disease (CVD) has emerged as the leading cause of mortality worldwide. Low-density lipoprotein (LDL), a critical serum biomarker for ASCVD diagnosis, plays a pivotal role in early cardiovascular disease detection and therapeutic monitoring. To address this need, we have developed a highly ultrasensitive fluorescent aptamer sensor based on a copper-based metal-organic framework (Cu-MOF) fluorescent probe. In this system, MoS₂ nanosheets can effectively quench the fluorescence of Cu-MOF via and fluorescence resonance energy transfer (FRET). Upon LDL binding, the aptamer interactio

Programmable two-dimensional CoOOH nanosheets with a hybridization chain reaction amplification system for label-free fluorescent detection of miRNA-205 in colorectal cancer.

MicroRNA-205 (miRNA-205) has been identified as a promising biomarker, yet its clinical application is hindered by the lack of detection strategies that combine simplicity, ultrahigh sensitivity, and high specificity. To address these obstacles, we introduced an innovative biosensor integrating hybridization chain reaction (HCR) with two-dimensional CoOOH nanosheets for precisely detecting the colorectal cancer biomarker miRNA-205 (HCR-CoOOH system). This system took advantage of the preferential binding of CoOOH nanosheets for single-stranded DNA over double-stranded DNA, along with HCR for s

A Dual-Functional Two-Photon PLIM Probe Based on Ruthenium(II) Complex: Sequence Selectivity and Aβ Aggregation Monitoring.

A key pathological feature of Alzheimer's disease (AD) is β-amyloid (Aβ) plaque formation from abnormal peptide aggregation. In addition to full-length Aβ1-42, various N/C-truncated Aβ fragments exist in the human brain and are found to be associated with AD etiology. However, the precise toxic Aβ species, aggregation states, and neurotoxic mechanisms remain unclear, highlighting the urgent need for probes with sequence selectivity and real-time aggregation monitoring capability─functions lacking in reported Aβ probes. Herein, we present Ru-dmpip, a two-photon phosphorescence lifetime imaging

Ratiometric Fluorescent Chemosensing for Predicting Response to Irinotecan-Based Therapies in Pancreatic Ductal Adenocarcinoma.

Pancreatic ductal adenocarcinoma (PDAC) is an aggressive and devastating disease, with most patients facing fatality due to resistance to chemotherapy. The chemotherapeutic cocktail FOLFIRINOX serves as a first line of therapy, and its success can be predicted by the activity of carboxylesterase 2 (CES2), an enzyme responsible for metabolically activating irinotecan (CPT-11), a component of FOLFIRINOX, where low CES2 activity forecasts resistance. Such predictive power enables clinicians to decide whether FOLFIRNOX is worth pursuing prior to administration. Despite this, current methods to mea

A Smart Dicyano-Based Fluorescent Probe for In Vivo Amyloid-β Visualization to Illuminate Alzheimer's Disease.

Amyloid-β (Aβ) is a key biomarker for Alzheimer's disease (AD), with its abnormal aggregation closely linked to AD pathology. Fluorescence imaging shows promise for Aβ detection but faces challenges such as optical instability, a short blood half-life, and poor blood-brain barrier (BBB) permeability. Additionally, only a few probes could target Aβ oligomers with a higher neurotoxicity. In this study, we designed and synthesized 14 fluorescent probes using the dicyano group as an electron acceptor. Among them, Dicyano-18 emerged as the most promising probe, with a binding affinity for Aβ aggreg

Silent-To-Bright CTCs-Hitchhiking Peptide Nanoprobe for Activatable NIR Imaging of Lung Metastasis in Mice.

Metastasis remains the leading cause of cancer-related mortality, yet its early detection is hindered by the subtle and unpredictable nature of the metastatic niche. In this study, we developed a self-assembling peptide-based activatable nanoprobe, 1-CyBr(PBA)-NGR NPs, designed for the early detection of the metastatic niche through circulating tumor cells (CTCs)-mediated hitchhiking and hydrogen peroxide (H2O2)-induced fluorescence activation. The peptide probe spontaneously assembled into stable spherical nanoparticles in aqueous media and undergoes H2O2-triggered fluorescence recovery via o

NIR chemosensor for rapid tracking and evaluating hypochlorous acid level within biosystem and inflammatory DILI models.

Hypochlorous acid, one of the commonly available active ROS species that regulate metabolism and complex biosystem. It is also a crucial hazardous oxidative waste component in the environment linked to inflammation and hepatic damage. Thus, the strategic development of fluorescence chemosensor exhibiting elevated selectivity and sensitivity continues to be a primary research emphasis for hypochlorous acid detection. This paper presents the design and synthesis of a novel near-infrared (NIR) fluorescent probe, HY720, for the selective detection of hypochlorous acid. The probe demonstrates a rap

Azoreductase-Responsive NIR Fluorescent Self-Assembled Probe for Hypoxia Imaging of Tumors, Hindlimb Ischemia, and Ulcerative Colitis.

Azoreductase (AzoR) has emerged as a key player in reductive metabolism under hypoxia and a potential biomarker for hypoxia-associated pathologies, such as cancer and ulcerative colitis (UC). However, current clinical practice lacks a reliable method for real-time AzoR monitoring to effectively guide personalized therapies and assess treatment responses. We address this challenge by developing an AzoR-activatable near-infrared (NIR) fluorescent self-assembled probe, NFK-(Azo)-DCM, which combines self-assembly properties with hypoxia-specific activation through a rationally designed azobenzene

Tuning Lipophilicity to Develop an MMP-Independent Fluorescent Probe for Visualizing Mitochondrial Peroxynitrite in Drug-Induced Liver Injury.

Mitochondrial peroxynitrite (ONOO-), a highly reactive nitrogen species, is critically involved in oxidative-stress-induced cell damage, playing vital roles in the pathogenesis of numerous diseases. However, the real-time detection of ONOO- in mitochondria is still challenging due to the reliance of existing probes on mitochondrial membrane potential (MMP), often disrupted under disease conditions. Therefore, we attempted to develop and investigate an MMP-independent, near-infrared fluorescent probe (Mito-ONOO-). To achieve this, a series of fluorophores Flu-(1-5) was rationally constructed by

A Near-Infrared Fluorescent Probe for Monitoring Carboxylesterase Dynamics: Application to NAFLD Diagnosis and Therapeutic Assessment of Qiwei Gandanqing Granules.

As a prevalent metabolic dysfunction, non-alcoholic fatty liver disease (NAFLD) represents an escalating challenge to global healthcare systems. Hepatic carboxylesterases (CES), primarily represented by the CES1 and CES2 isoforms, are important regulators of lipid homeostasis. Emerging evidence suggests that dysregulation of CES1 and CES2 contributes to hepatic lipid accumulation in NAFLD and shows promise as potential biomarkers for NAFLD diagnosis and therapeutic evaluation. A NIR fluorescent probe for viscosity detection and its application to spinal cord imaging in mice with multiple sclerosis.

Cellular microenvironment viscosity, as a key biophysical parameter, is significantly associated with disease processes such as tumors, atherosclerosis, and multiple sclerosis (MS). The core pathological mechanism of MS involves neuroinflammation-mediated demyelination and axonal degeneration, which ultimately leads to multi-system neurological dysfunction. In this study, we developed a novel near-infrared fluorescent probe, CABT (based on a dimethylaminobenzene-benzothiazole dye), which achieves viscosity-responsive fluorescence enhancement through an intramolecular single-bond rotationally r
