[From conduct disorder in childhood to psychopathy in adult life].

Mental health professionals seldom recognize psychopathy in their daily practice. Usually forensic psychiatrists and psychologists are involved because individuals with psychopathic personality are involved in serious criminal behavior and implicated with the law. Most of the times the profiles of children who evolve in adult psychopaths have components from other disorders, especially conduct disorder. The term psychopathy originates from the Greek words "psyche" (soul) and "pathos" (passion) and was used to identify initially every mental illness. Although in the bibliography the terms Antis

Targeted next generation sequencing application in cardiac channelopathies: Analysis of a cohort of autopsy-negative sudden unexplained deaths.

Genetic testing for cardiac channelopathies in sudden unexplained death (SUD) has developed substantially over the last years. The Next Generation Sequencing (NGS) technology provides an unprecedented opportunity to screen for genetic variations underlying arrhythmogenic genes in a short period of time at a low cost. The present study aimed to perform genetic testing with NGS technologies on the Ion Torrent Personal Genome Machine™ (Ion PGM™) sequencer, in targeting a total of 23 genes reported to be associated with inherited cardiac channelopathies in order to identify the possible cause of d

mRNA expression patterns in human myocardial tissue, pericardial fluid and blood, and its contribution to the diagnosis of cause of death.

Gene expression has become an interesting research area in forensic pathology to investigate the process of death at the molecular level. The aims of this study were to analyze changes in gene expression patterns in relation to the cause of death, and to propose new molecular markers of myocardial ischemia of potential use for the postmortem diagnosis of early ischemic heart damage in cases of sudden cardiac death (SCD). We determined mRNA levels of five proteins related with ischemic myocardial damage and repair - TNNI3, MYL3, TGFB1, MMP9 and VEGFA - in specific sites of the myocardium, blood

Mutations in the SCN5A gene: evidence for a link between long QT syndrome and sudden death?

Mutations in cardiac ion channel genes leading to channel dysfunctions or changes in the gene expression may cause inherited arrhythmogenic diseases. These genetic diseases are important causes of sudden unexplained death (SUD). Ten cases of SUD, including six cases of sudden infant death syndrome (SIDS) and four cases of SUD from people in the age of 14-40 years were examined by postmortem molecular analysis. Genomic DNA was extracted from blood cells and two long QT syndrome relevant genes, SCN5A encoding the alpha-subunit of the voltage-gated sodium channel Nav1.5 and KCNH2 encoding the alp

A validated forensic STR profiling method for the Peregrine Falcon (Falco peregrinus) to identify individuals and investigate familial relationships.

Falconry, and the captive breeding of Peregrine Falcons (Falco peregrinus) and their hybrids is a thriving industry in both the UK and the Middle East, with thousands of birds traded internationally on an annual basis. Wild Peregrine Falcons, having suffered a population collapse in the 1960s, are protected by UK legislation and the Convention on International Trade in Endangered Species of Flora and Fauna (CITES) lists Peregrines on Appendix I, which restricts international trade in these birds. As such, in the UK, captive-bred birds must be registered within a few weeks of hatching and birds

Molecular autopsy identifies the NaV1.5 p.Leu96Pro variant causing sodium current loss-of-function in unexplained sudden cardiac death.

Molecular autopsy can identify candidate variants in unexplained sudden cardiac death (SUD), but functional evidence is often required to support pathogenic interpretation and family risk assessment. Here, we investigated the functional consequences of the SCN5A c.287 T > C (NaV1.5_p.Leu96Pro) variant, identified in a 29-year-old man who died of SUD. Cascade screening identified additional relatives carrying the variant, with variable clinical expression. Sodium current (INa) was analyzed in heterologously transfected human embryonic kidney (HEK) tsA201 cells and in human induced pluripotent s

Data-driven methods allow prediction of utility of DNA rework.

In forensic DNA casework, complex trace samples may benefit from rework procedures involving additional PCR replicates and lowered analytical thresholds. These procedures may strongly increase or decrease the likelihood ratio (LR), simplifying decisions to be taken downstream. However, the procedures are resource-intensive. Decisions to pursue rework currently rely on laboratory guidelines and expert judgment rather than empirical evidence. This study aims to develop a model that predicts whether rework will yield a more decisive LR, defined as crossing the laboratory's reporting thresholds. T

The EDNAP collaborative study on mtDNA heteroplasmy detection using Sanger and Massively Parallel Sequencing technologies.

This EDNAP (European DNA Profiling Group) collaborative study evaluated the detection and reporting of mitochondrial DNA (mtDNA) point heteroplasmy (PHP) and length heteroplasmy (LHP) across forensic laboratories using Sanger, Ion Torrent, and Illumina sequencing platforms. Standardized DNA extracts were centrally prepared and distributed to participating laboratories to assess inter-laboratory concordance and platform-specific (platform = instrument + software) effects. Raw sequence data showed high overall consistency across platforms, and observed mitotype discrepancies were due to transcri

Deconvolving DNA mixtures with Demixtify.

While there are many tools to detect DNA mixtures, few can deconvolve mixed genetic profiles at genomic scales. Demixtify is one such application. Demixtify characterizes and deconvolves DNA mixtures in whole genome sequencing data. Demixtify is also fully amenable to modern imputation strategies, which in turn allows samples to be accurately characterized even when the information content is limited (<1×). In the present study Demixtify is applied to two-person in silico DNA mixtures. Genotypes are often accurately inferred, with noticeable increases in performance when genotypes are also ref

Flexible reading of single-primer extension microhaplotypes improves degraded DNA mixture interpretation.

Microhaplotypes (MHs) are informative forensic markers for degraded and mixed DNA because they combine short target regions, haplotypic diversity and stutter-free allele detection. However, degraded templates often generate reads that cover only part of an MH target. Discarding these partial reads may reduce the information available for mixture interpretation. We evaluated a single-primer extension-based MH massively parallel sequencing system (SPE-MH-MPS) for low-template DNA, simulated degraded DNA, two-person mixtures and degraded two-person mixtures. A 91-locus autosomal panel developed f

Touch DNA is predominantly transferred from leukocytes, not skin cells.

DNA extracted from touched surfaces is an important analyte in forensic casework, often leading to the identification of suspects. A common assumption underlying this approach is that touch DNA originates in skin cells of the touching individual. Here we test this assumption, using cell type-specific DNA methylation markers to determine the origins of touch DNA. Strikingly, hand swabs and touched objects contain only traces of skin DNA. The majority of hand swab and touch DNA originates in leukocytes (mostly neutrophils) and to a lesser extent in buccal cells, paralleling the composition of na

Improving accessibility of species identification for wildlife crime prosecution: Expanding WildSEQ for calcified and keratinised materials.

The growing need for simplified and accessible species identification methods is critical for supporting the prosecution of illegal wildlife trafficking. This requires the development of approaches suitable for implementation across both well-resourced and resource-limited jurisdictions. Here, we expand a previously developed targeted quantitative PCR (qPCR) and nanopore sequencing workflow (WildSEQ) to additional taxa including rhinoceros, hippopotamus, walrus, and water buffalo. Taxon-specific qPCR assays were developed to screen samples for multiple suspected species and this was combined w

Metagenomic profiling reveals lung multi-kingdom microbes as forensic markers for aquatic corpses investigation.

The forensic investigation of corpses recovered from aquatic environments presents a major practical challenge. Recent studies have demonstrated that the bacterial community in the lung serves as a valuable indicator for diagnosing drowning, determining the drowning medium and estimating postmortem submersion interval (PMSI). However, the application and significance of lung multi-kingdom microbiome (archaea, eukaryota, and viruses) remains inadequately characterized. Meanwhile, the insufficient sequencing depth of commonly employed techniques, such as amplicon sequencing, restricts our unders

Male DNA on underwear: Can Y- STR results help discriminate activities?

This research is based on a case in which the complainant alleged that the defendant had removed her underwear, while the defendant claimed they had only danced together. To assist in resolving this issue, DNA was collected from the sides of the complainant's underwear and Y-chromosome STR (Y-STR) analysis performed using the PowerPlex® Y23 System. As no published research were available on DNA recovered from the sides of underwear given the two alleged activities, 60 experiments were conducted to generate specific data on the expected results given underwear removal and given dancing. Time heals all wounds: Identification of RNA biomarkers for forensic wound age estimation in human skin tissue samples.

Estimating wound age can be crucial in forensic work for establishing a causal link between the injury and death, or for better characterizing the timing and circumstances of the injury. Conventional methods for estimating wound age are not always reliable and often fail to achieve sufficient accuracy. Animal studies have shown that RNA-based methods for wound age estimation can produce reliable and reproducible results. However, the transferability of wound markers identified in animal experiments to human samples is questionable, and a model based on human tissue needs to be developed. Twent

Construction of a succession model for the microbiome in water from submerged corpses based on single-molecule real-time sequencing.

Decomposition of corpses in aquatic environments is regulated by multiple factors, and traditional methods for estimating the postmortem submerged interval (PMSI) have clear limitations. To explore the microbial succession patterns in the water of submerged corpses and their potential application in PMSI estimation, this study submerged rat carcasses in both river water and tap water, collecting water samples on days 0, 2, 6, 9, 14, 20, 27, 40, 54, and 70 postmortem. The microbial succession dynamics in the water were analyzed using single-molecule real-time sequencing. The results indicated t

Comparison of DNA profiles from samples collected from underneath fingernails and hand deposits following everyday activity.

In instances of direct physical contact between individuals involved in criminal activity, body samples can provide significant and relevant information to aid in criminal investigations and court proceedings. Fingernails are one such forensically relevant body area that is capable of providing evidence of direct contact and potentially revealing whether the interaction involved was of a forceful kind. Several studies have investigated the prevalence of non-self DNA under fingernails after different crime-related scenarios; however, few have assessed the types of DNA profiles found after every
