Polymer multilayers loaded with antifungal β-peptides kill planktonic Candida albicans and reduce formation of fungal biofilms on the surfaces of flexible catheter tubes.

Candida albicans is the most common fungal pathogen responsible for hospital-acquired infections. Most C. albicans infections are associated with the implantation of medical devices that act as points of entry for the pathogen and as substrates for the growth of fungal biofilms that are notoriously difficult to eliminate by systemic administration of conventional antifungal agents. In this study, we report a fill-and-purge approach to the layer-by-layer fabrication of biocompatible, nanoscale 'polyelectrolyte multilayers' (PEMs) on the luminal surfaces of flexible catheters, and an investigati

Encapsulation of essential oils within a polymeric liposomal formulation for enhancement of antimicrobial efficacy.

Essential oils and their constituents are known to possess antimicrobial activity; however, their inherent volatility is a limiting factor. In order to exploit the antimicrobial efficacy of essential oils, encapsulation within polymeric liposomal systems was undertaken. The liposomes were subsequently polymer-coated in order to further enhance the stability of the formulations. Essential oils distilled from Artemisia afra, Eucalyptus globulus and Melaleuca alternifolia were encapsulated into diastearoyl phosphatidylcholine and diastearoyl phosphatidylethanolamine liposomes employing a reverse

Biotinylation of TiO(2) nanoparticles and their conjugation with streptavidin.

Photoactive TiO(2) can be used to mediate a variety of disinfection processes. It was postulated that TiO(2) particles could be directed to specific targets of interest using biotin/streptavidin linkages. Biotinylated TiO(2) nanoparticles (anatase) were obtained by treating TiO(2) nanoparticles with 3-aminopropyltriethoxysilane (APTS) in anhydrous DMSO, followed by reaction with N-hydroxysuccinimidobiotin. 29Si CP-MAS NMR, 13C CP-MAS NMR, and FTIR spectra showed that biotin was covalently bound to the TiO(2) surface. Transmission electron microscopy (TEM) demonstrated that prolonging the silan

Glycoprotein C-dependent attachment of herpes simplex virus to susceptible cells leading to productive infection.

Herpes simplex viruses encode several glycoproteins dispensable for infection and replication in cell culture. Evidence is presented that there exist at least two pathways for viral attachment to cells, i.e., one mediated by the dispensable glycoprotein C (gC) and one independent of that glycoprotein. Thus, whereas the polycations neomycin and polylysine inhibit attachment but not entry of already attached herpes simplex virus 1 (HSV-1) into baby hamster kidney (BHK) cell line, they have no effect on HSV-2 attachment to the same cells (N. Non-Covalent Interaction Induced Supramolecular Precipitate with Hetero-Motif Polyionic Junction for Durable Antimicrobial Activity and Infected Wound Healing.

The advent of the COVID-19 pandemic has underscored the pressing demand for antimicrobial materials that offer both durability and efficacy. Herein, the successful design and fabrication of a "water-insoluble" supramolecular precipitate is reported through the "bottom-up" assembly of polyanion sodium alginate (SA) with the antimicrobial motifs A2G and Cu2+. This innovative hetero-motif polyionic junction leverages a network of hydrogen bonds aligning with electrostatic interactions, and hydrophobic effects to mitigate the rapid release of active components, providing exceptional long-term anti

Micelle-embedded coating with ebselen for nitric oxide generation.

Nitric oxide generation is considered to be a key factor to mimic endothelial function in terms of anti-coagulation and anti-hyperplasia. Herein, ebselen which could play the similar role as glutathion peroxidase-like was loaded into micelles and was further assembled into a layer-by-layer coating. The ability of nitric oxide generation and corresponding biological effect were investigated. Endothelial-mimetic surface has now attracted huge attention in blood-contacting materials, due to its inherent ability of secreting nitric oxide. Among those categories, nitric oxide generation surface is

Effect of humic acid on pyrene removal from water by polycation-clay mineral composites and activated carbon.

Pyrene removal by polycation-montmorillonite (MMT) composites and granulated activated carbon (GAC) in the presence of humic acid (HA) was examined. Pyrene, HA, and sorbent interactions were characterized by FTIR, fluorescence and zeta measurements, adsorption, and column filtration experiments. Pyrene binding coefficients to the macromolecules were in the order of PVPcoS (poly-4-vinylpiridine-co-styrene) > HA > PDADMAC (poly diallyl-dimethyl-ammonium-chloride), correlating to pyrene-macromolecules compatibility. Electrostatic interactions explained the high adsorption of HA to both composites

Polyanionic carbosilane dendrimers as a new adjuvant in combination with latency reversal agents for HIV treatment.

BACKGROUND: The major obstacle impeding human immunodeficiency virus-1 (HIV-1) eradication in antiretroviral treatment (ART) treated HIV-1 subjects is the establishment of long-lived latently infected resting CD4+ T cells. Due to the fact that no drug has been effective, the search for new drugs and combinations are a priority in the HIV cure. Treatments based on nanotechnology have emerged as an innovative and promising alternative to current and conventional therapies. In this respect, nanotechnology opens up a new door for eliminating latent HIV infection. We studied the role of G1-S4, G2-S

Chitosan based surface modulation of core-shell nanoparticles for oral delivery of exenatide via balancing mucus penetration and cellular uptake.

Oral delivery of peptide and protein drugs (PDs) is hindered by the impermeable intestinal mucosa, which consists of both the mucus layer and the epithelium. Therefore, double-layer (mucus layer and epithelium) overcoming nanocarriers need to be designed to enhance the transporting efficiency of PDs. However, the requirements for surface properties to penetrate these two barriers are quite distinct. In this study, nanoparticles (NPs) with balanced mucus permeation and cellular uptake were developed by modulating surface properties to improve the endocytosis efficiency of exenatide (EXT). The E

The Synthesis of Green Palladium Catalysts Stabilized by Chitosan for Hydrogenation.

The proposed paper describes a simple and environmentally friendly method for the synthesis of three-component polymer-inorganic composites, which includes the modification of zinc oxide or montmorillonite (MMT) with chitosan (CS), followed by the immobilization of palladium on the resulting two-component composites. The structures and properties of the obtained composites were characterized by physicochemical methods (IRS, TEM, XPS, SEM, EDX, XRD, BET). Pd-CS species covered the surface of inorganic materials through two different mechanisms. The interaction of chitosan polyelectrolyte with z

Differential proteomic analysis of virus-enriched fractions obtained from plasma pools of patients with dengue fever or severe dengue.

BACKGROUND: Dengue is the most widespread mosquito-borne viral disease of public health concern. In some patients, endothelial cell and platelet dysfunction lead to life-threatening hemorrhagic dengue fever or dengue shock syndrome. Prognostication of disease severity is urgently required to improve patient management. The pathogenesis of severe dengue has not been fully elucidated, and the role of host proteins associated with viral particles has received little exploration. METHODS: The proteomes of virion-enriched fractions purified from plasma pools of patients with dengue fever or severe

Reversible hydration and dehydration of polyanionic brushes bearing carboxylate, phosphate and sulfonate side groups: a comparative AFM study.

The present work reports on a systematic study of the swelling/collapse transition of three anionic polymer brushes bearing carboxylate, phosphate and sulfonate side groups via AFM measurement. Time scale of conformation change process can be approximately evaluated directly. All the three brushes in their sodium salt forms stretch away from the surface in pure water, as a result of charge repulsion and uptake of water. The collapse of weak brushes has two ways: normal charge screening and precipitation (strong ion pairing), depending on the types of cations that have different coordination ca

Polyanionic microbicides modify Toll-like receptor-mediated cervicovaginal immune responses.

Topical microbicides are being developed as a preventative approach to reduce the sexual transmission of human immunodeficiency virus type 1 (HIV-1) and other infections. For them to be efficacious, it is believed that they should avoid inducing inflammation while allowing the vaginal epithelium to initiate protective Toll-like receptor (TLR)-mediated innate responses against pathogens. In this study, human cervical and vaginal epithelial cells were exposed to polyanionic HIV entry inhibitors and the following synthetic TLR ligands: (i) the bacterial lipoprotein Pam(3)CSK(4), binding cell surf

Design, synthesis, and antibacterial activities of neomycin-lipid conjugates: polycationic lipids with potent gram-positive activity.

Aminoglycoside antibiotics and cationic detergents constitute two classes of clinically important drugs and antiseptics. Their bacteriological and clinical efficacy, however, has decreased recently due to antibiotic resistance. We have synthesized aminoglycoside-lipid conjugates in which the aminoglycoside neomycin forms the cationic headgroup of a polycationic detergent. Our results show that neomycin-C16 and neomycin-C20 conjugates exhibit strong Gram-positive activity but reduced Gram-negative activity. The MIC of neomycin-C16 (C20) conjugates against methicillin-resistant Staphylococcus au

Protein/DNA polyplexes for gene therapy.

The development of molecular conjugates as components of Protein/DNA polyplexes has resulted in the creation of a simple, non-virus vector for the targeted delivery of nucleic acids into specific cell types. This vector has many of the positive attributes of viruses, on without the limitations that continue to plague recombinant viruses. The simplicity of this vector allows for quick analysis of nucleic acids, expression vectors, and therapeutic genes in vitro and potentially in vivo, because the time that would be involved in the generation of recombinant viral vectors is not present. Boosting oral bioavailability of taraxasterol via a nanocrystal-mucoadhesive micropatch platform for potent multi-mechanistic protection against drug-induced liver injury.

Drug-induced liver injury (DILI) is a serious adverse reaction with limited therapeutic options. Taraxasterol (TAS), a natural triterpenoid with hepatoprotective potential, suffers from poor aqueous solubility and low oral bioavailability, hindering its clinical application. Here, we develop an integrated oral delivery platform comprising TAS nanocrystals (TAS NCs) stabilized by D-α-tocopherol polyethylene glycol succinate (TPGS), embedded within chitosan-carbopol polyelectrolyte complex (EPC) micropatches, and encapsulated in enteric-coated capsules (TAS NCs@EPC/CAP). Chlorophyllin-immobilized chitosan/xanthan gum composite as an adsorbent and visible-light photosensitizer for the removal of genotoxic azo, anthraquinone and triphenylmethane dyes.

The discharge of genotoxic synthetic dyes, particularly from complex tannery wastewater, poses severe threats to aquatic ecosystems and public health. To overcome limitations of conventional single-mechanism treatments, a novel dual-action adsorbent and visible-light-responsive photosensitizer was synthesized by immobilizing sodium copper chlorophyllin (5.0% w/w) within a cross-linked chitosan/xanthan gum polyelectrolyte complex. Characterisation confirmed an amorphous, mesoporous framework with a specific surface area of 40.32 m2 g-1 (after lyophilization) and enhanced thermal stability up to
