Host range expansion of Leucobacter holotrichiae: first evidence of mammalian infection and comparative genomic insights.

Leucobacter holotrichiae has previously been identified exclusively in insect hosts, with no reports of its presence in mammals. This study is the first to report the isolation of three L. holotrichiae strains (LH23001, LH23002, and LH23003) from mixed cultures of bovine actinomycotic abscesses in a large-scale dairy farm in Xinjiang, China. Phenotypic analysis revealed that these Gram-positive bacilli are non-spore-forming, non-flagellated, non-hemolytic, non-motile, and capable of biofilm formation. The isolates formed transparent membranous white colonies on modified Gao's medium and could

Aneurinibacillus thermoaerophilus CCM 8960 is a thermophilic bacterium isolated from compost in Brno. The bacterium accumulates polyhydroxyalkanoates (PHAs), a biodegradable and renewable alternative to petrochemical polymers. The bacterium reveals several features that make it a very interesting candidate for the industrial production of PHA. At first, due to its thermophilic character, the bacterium can be utilized in agreement with the concept of next-generation industrial biotechnology (NGIB), which relies on extremophiles. Second, the bacterium is capable of producing PHA copolymers conta

Microbial diversity, ecological networks and functional traits associated to materials used in drinking water distribution systems.

Drinking water distribution systems host complex microbial communities as biofilms that interact continuously with delivered water. Understanding the diversity, behavioural and functional characteristics will be a requisite for developing future monitoring strategies and protection against water-borne health risks. To improve understanding, this study investigates mobilisation and accumulation behaviour, microbial community structure and functional variations of biofilms developing on different pipe materials from within an operational network. Samples were collected from four pipes during a r

Metagenomic Analysis of Antibiotic Resistance Genes in Dairy Cow Feces following Therapeutic Administration of Third Generation Cephalosporin.

Although dairy manure is widely applied to land, it is relatively understudied compared to other livestock as a potential source of antibiotic resistance genes (ARGs) to the environment and ultimately to human pathogens. Ceftiofur, the most widely used antibiotic used in U.S. dairy cows, is a 3rd generation cephalosporin, a critically important class of antibiotics to human health. The objective of this study was to evaluate the effect of typical ceftiofur antibiotic treatment on the prevalence of ARGs in the fecal microbiome of dairy cows using a metagenomics approach. β-lactam ARGs were foun

Novel Recombinant Endolysin LysMYM94 against Dry-Grind Corn Mash Bioethanol Fermentation Contaminant Limosilactobacillus fermentum.

Bacterial contamination by lactic acid bacteria particularly Limosilactobacillus fermentum poses a significant challenge to commercial bioethanol fermentation facilities, resulting in reduced productivity and yield. Here we characterize a novel bacteriophage endolysin, LysMYM94 isolated from a L. fermentum prophage, that selectively targets lactic acid bacteria contaminants isolated from bioethanol fermentation plants. Recombinant endolysin LysMYM94, a 35.9 kDa endolysin, consists of an N-acetylmuramidase GH25-like enzymatically active domain and a SH3b-like cell-binding domain. The endolysin

Beyond stool: sampling context and inferential scope in gut mechanistic studies.

SUMMARYHuman gut microbiome research still relies heavily on fecal sampling because stool is noninvasive, scalable, repeatable, and essential for population-level and longitudinal studies. These strengths have made stool central to human microbiome science, but fecal output is not a neutral representation of every intestinal compartment. It reflects material that has persisted through digestion, transport, microbial turnover, host absorption, and distal colonic processing. This distinction becomes important when studies move from detecting microbial associations to explaining localized activit

Comparative Genomic Analysis of Pseudomonas shahriarae Reveals Virulence Potential, Antimicrobial Resistance, and Environmental Adaptation.

Pseudomonas shahriarae is a recently identified member of the P. fluorescens group. Its ecological range and ability to cause disease are still mostly unknown, especially in aquaculture settings. This work presents the first genome sequence of P. shahriarae isolated from diseased Siberian sturgeon (Acipenser baerii). To obtain deeper understanding of its evolutionary history, pathogenicity, and capacity of antibiotic resistance, this genome was compared with seven other publicly available genomes. The draft genome of strain SK21 was 6.12 Mb size and had a GC content of 60.5%. Core genome analy

A comprehensive study on Salmonella enterica serovar Richmond in farmed fish Pangasianodon hypophthalmus: insights into zoonotic potential, virulence and antimicrobial resistance.

Salmonella enterica serovar Richmond is an emerging pathogen from contaminated animal food, posing a risk of global foodborne outbreaks. Few reports have documented this species in live aquatic organisms. In this study, Salmonella Richmond COFI-RLBCAU-I was isolated from live Pangasianodon hypophthalmus, and its pathogenicity was tested on the three most culturable fish species, P. hypophthalmus, Labeo rohita, and Oreochromis niloticus. The results showed that the bacteria are non-pathogenic to these three species. Techniques, including β-hemolytic activity, Gram staining, biochemical tests su

Defence systems encoded by core genomic islands of seventh pandemic Vibrio cholerae.

Vibrio cholerae, the causative agent of cholera, has triggered seven pandemics, with the seventh pandemic emerging in 1961. The success of seventh pandemic El Tor (7PET) V. cholerae as a human pathogen is linked to its acquisition of mobile genetic elements (MGEs) like the CTXΦ prophage and Vibrio pathogenicity island 1 (VPI-1). Additional MGEs, including VPI-2 and the Vibrio seventh pandemic islands (VSP-I and VSP-II), are thought to have further enhanced the pathogen's virulence potential. However, recent research suggests that these MGEs serve as reservoirs for defence systems, which may re

PMCNA_RS00975 activates NF-κB and ERK1/2 through TLR2 and contributes to the virulence of Pasteurella multocida.

INTRODUCTION: Pasteurella multocida is a pathogenic bacterium known to cause hemorrhagic septicemia and pneumonia in poultry. Reports have indicated that certain proteins, either directly involved in or regulating iron metabolism, are important virulence factors of P. multocida. Therefore, understanding virulent factors and analyzing the role of pro-inflammatory cytokines can help us elucidate the underlying pathogenesis. METHODS: In this study, the PMCNA_RS00975 protein, a putative encapsuling protein encoded by a gene from a specific prophage island of the pathogenic strain C48-1 of P. mult

The influence of extrachromosomal elements in the anthrax "cross-over" strain Bacillus cereus G9241.

Bacillus cereus G9241 was isolated from a welder who survived a pulmonary anthrax-like disease. Strain G9241 carries two virulence plasmids, pBCX01 and pBC210, as well as an extrachromosomal prophage, pBFH_1. pBCX01 has 99.6% sequence identity to pXO1 carried by Bacillus anthracis and encodes the tripartite anthrax toxin genes and atxA, a mammalian virulence transcriptional regulator. This work looks at how the presence of pBCX01 and temperature may affect the lifestyle of B. cereus G9241 using a transcriptomic analysis and by studying spore formation, an important part of the B. anthracis lif

Physiological characteristics and virulence gene composition of selected serovars of seafood-borne Salmonella enterica.

BACKGROUND AND AIM: All serotypes of Salmonella enterica are considered potentially pathogenic. However, the non-typhoidal Salmonella (NTS) serotypes vary considerably in terms of pathogenicity and the severity of infections. Although diverse serotypes of NTS have been reported from tropical seafood, their sources, physiological characteristics, and virulence potentials are not well understood. This study aimed to compare the physiological characteristics of selected serovars of Salmonella from seafood and investigate possible variations in the distribution of known genes within the pathogenic

Control of resistance against bacteriophage killing by a metabolic regulator in meningitis-associated Escherichia coli.

Ecologically beneficial traits in bacteria are encoded by intrinsic and horizontally acquired genes. However, such traits are not universal, and the highly mosaic nature of bacterial genomes requires control at the transcriptional level to drive these processes. It has emerged that regulatory flexibility is widespread in the Escherichia coli species, whereby preexisting transcription factors can acquire new and unrelated roles in regulating beneficial traits. DsdC is the regulator of D-serine tolerance in E. coli, is essential for D-serine catabolism, and is often encoded by two copies in neon

Tyroviruses are a new group of temperate phages that infect Bacillus species in soil environments worldwide.

BACKGROUND: Bacteriophages are widely considered to be highly abundant and genetically diverse, with their role in the evolution and virulence of many pathogens becoming increasingly clear. Less attention has been paid on phages preying on Bacillus, despite the potential for some of its members, such as Bacillus anthracis, to cause serious human disease. RESULTS: We have isolated five phages infecting the causative agent of anthrax, Bacillus anthracis. Using modern phylogenetic approaches we place these five new Bacillus phages, as well as 21 similar phage genomes retrieved from publicly avai

The roles of antimicrobial resistance, phage diversity, isolation source and selection in shaping the genomic architecture of Bacillus anthracis.

Bacillus anthracis, the causative agent of anthrax disease, is a worldwide threat to livestock, wildlife and public health. While analyses of genetic data from across the globe have increased our understanding of this bacterium’s population genomic structure, the influence of selective pressures on this successful pathogen is not well understood. In this study, we investigate the effects of antimicrobial resistance, phage diversity, geography and isolation source in shaping population genomic structure. We also identify a suite of candidate genes potentially under selection, driving patterns o

Release mechanisms and molecular interactions of Pseudomonas aeruginosa extracellular DNA.

Pseudomonas aeruginosa infection is a significant threat for clinicians. Increasing incidents of resistant biofilm infection result in high mortality rates worldwide. There is a considerable current interest in the field of extracellular DNA (eDNA)-mediated P. aeruginosa biofilm formation. eDNA acts as a glue to make biofilm more stable. This review focuses on the diverse mechanisms and factors, which enhance the eDNA release into the extracellular milieu. Furthermore, eDNA-mediated molecular interactions within the biofilm are emphasized. In addition, drug resistance mechanisms due to the ver
