Mechanistic investigation of calcium phosphate mineralization potentiating the cellular immune response of foot-and-mouth disease virus-like particle vaccines.

Virus-like particles (VLPs), composed of structural proteins, have emerged as a promising vaccine platform for preventing infectious diseases due to their outstanding advantages. Enhancing immune responses specific to VLPs, particularly cellular immunity, has emerged as a pivotal area of current immunological research. The nanovaccines for the foot-and-mouth disease virus (FMDV) composed of VLPs mineralized with calcium phosphate (VLPs-CaP), developed based on biomineralization principles, demonstrates superior immunogenicity. In this study, we elucidated the efficacy and mechanism of VLPs-CaP

Chromosome-level haplotype-resolved genome assembly of the linguliform brachiopod Discradisca antillarum (d'Orbigny, 1845).

Brachiopods are a group of shelled, filter-feeding marine invertebrates. Though they superficially resemble bivalve mollusks, brachiopods fall within the clade Lophophorata (Brachiopoda, Bryozoa, Phoronida) and possess an abundant and diverse fossil record spanning more than 500 million years. This long evolutionary history makes Brachiopoda particularly important for understanding the evolution of the morphologically disparate superphylum Lophotrochozoa. Brachiopods also stand to provide insight into the evolution of biomineralization, as most lineages produce skeletons of calcium carbonate w

Simultaneous ocean acidification and warming do not alter the lipid-associated biochemistry but induce enzyme activities in an asterinid starfish.

Ocean acidification and warming affect marine ecosystems from the molecular scale in organismal physiology to broad alterations of ecosystem functions. However, knowledge of their combined effects on tropical-subtropical intertidal species remains limited. Pushing the environmental range of marine species away from the optimum initiates stress impacting biochemical metabolic characteristics, with consequences on lipid-associated and enzyme biochemistry. This study investigates lipid-associated fatty acids (FAs) and enzyme activities involved in biomineralization of the tropical-subtropical sta

Was the massive increase in use of teratogenic agrichemicals in western states (USA) associated with declines in wild ruminant populations between 1994 and 2013?

Population declines were documented in multiple ruminant species in Montana and surrounding states starting in 1995. While weather, food sources, and predation certainly contributed, the declines were often attributed, at least partly, to unexplained factors. Use of teratogenic agrichemicals, notably neonicotinoid insecticides, fungicides, and glyphosate-based herbicides, massively increased regionally in 1994-96. The question explored in this review is whether this vastly increased use of these teratogenic pesticides might have contributed to observed population declines. We provide reference

Gelatin Methacryloyl (GelMA) - 45S5 Bioactive Glass (BG) Composites for Bone Tissue Engineering: 3D Extrusion Printability and Cytocompatibility Assessment Using Human Osteoblasts.

3D extrusion printing has been widely investigated for low-volume production of complex-shaped scaffolds for tissue regeneration. Gelatin methacryloyl (GelMA) is used as a baseline material for the synthesis of biomaterial inks, often with organic/inorganic fillers, to obtain a balance between good printability and biophysical properties. The present study demonstrates how 45S5 bioactive glass (BG) addition and GelMA concentrations can be tailored to develop GelMA composite scaffolds with good printability and buildability. The experimental results suggest that 45S5 BG addition consistently de

Ceria-based nanotheranostic agent for rheumatoid arthritis.

Rheumatoid arthritis (RA) is an autoimmune disease that affects 1-2% of the human population worldwide, and effective therapies with targeted delivery for local immune suppression have not been described. We address this problem by developing a novel theranostic nanoparticle for RA and assessed its therapeutic and targeting effects under image-guidance. Methods: Albumin-cerium oxide nanoparticles were synthesized by the biomineralization process and further conjugated with near-infrared, indocyanine green (ICG) dye. Enzymatic-like properties and reactive oxygen species (ROS) scavenging activit

Remineralized bone matrix as a scaffold for bone tissue engineering.

There is a need for improved biomaterials for use in treating non-healing bone defects. A number of natural and synthetic biomaterials have been used for the regeneration of bone tissue with mixed results. One approach is to modify native tissue via decellularization or other treatment for use as natural scaffolding for tissue repair. In this study, our goal was to improve on our previously published alternating solution immersion (ASI) method to fabricate a robust, biocompatible, and mechanically competent biomaterial from natural demineralized bone matrix (DBM). The improved method includes

An aquaglyceroporin governs cellular water and CO2 conductance relevant for vesicular mineral formation in a marine calcifier.

Many marine organisms generate biominerals by endocytotic uptake of seawater which is modified within calcifying cells to produce an amorphous mineral precursor required to form the mineralized shell or skeleton. In this process, water must be removed from the endocytotic vesicles and the cell to promote concentration of calcification substrates and production of a water-free mineral. Here, we demonstrate that in the sea urchin larva, an aquaglyceroporin (spAQP9) is involved in the cellular calcification process. spAQP9 is found exclusively in the calcifying primary mesenchyme cells (PMCs), an

A biomimetic urchin-like photothermal nanoplatform for the efficient eradication of Streptococcus mutans and biofilm dispersion.

BACKGROUND: Dental caries, primarily driven by Streptococcus mutans (S. mutans) biofilms, remains a formidable clinical challenge due to the protective extracellular polymeric substance (EPS) matrix and the limited efficacy of conventional antibiotics. METHODS: To address this, we report a biomimetic photothermal nanoplatform (FWA NPs) featuring a hierarchical, urchin-like multi-spiked architecture, synthesized via the co-assembly of ferrocene-tryptophan conjugates and in situ biomineralized gold nanoparticles. RESULTS: Benefiting from this unique topological structure, FWA NPs maximize inte

Thermophiles in nanosized biocalcification: a novel approach for heavy metal remediation.

Bio deposition of minerals is a prevalent occurrence in the biological realm, facilitated by various organisms such as bacteria, fungi, protists, and plants. Calcium carbonate is one such mineral that precipitates naturally as a consequence of microbial metabolic processes. This study investigates an innovative approach for MICP- mediated heavy metal remediation, carbon dioxide (CO2) sequestration by utilizing thermophilic microorganisms isolated from such geographical area which is not yet been subjected to any systematic scientific study. Beyond the well-established urea hydrolysis pathway,

Mineralized Bacteria as a Potent Vaccine Against Staphylococcus aureus Infections.

Staphylococcus aureus (S. aureus) as common Gram-positive pathogenic bacteria, causes local and systemic infections, including sepsis and bacteremia. In particular, the high prevalence of drug-resistant S. aureus further complicates the post-infection treatment. Highly effective S. aureus vaccines are urgently desired. Herein, a novel S. aureus vaccine (MnO2@FS) is developed via biomineralizing manganese dioxide (MnO2) on formaldehyde-fixed S. aureus (FS). In such vaccine, with FS to induce bacteria-specific immune responses, MnO2 via releasing Mn2+ can activate the cyclic GMP-AMP synthase-sti

Simple and rapid formic acid sample treatment for the isolation of HgSe nanoparticles from animal tissues.

The present work explores for the first time the potential of formic acid on the extraction of tiemannite (HgSe) nanoparticles from seabird tissues, in particular giant petrels. Mercury (Hg) is considered one of the top ten chemicals of major public health concern. However, the fate and metabolic pathways of Hg in living organisms remain unknown. Methylmercury (MeHg), largely produced by microbial activity in the aquatic ecosystems is biomagnified in the trophic web. HgSe is considered the end-product of MeHg demethylation in biota and an increasing number of studies focuses on the characteriz

The effect of extracellular polymeric substances (EPS) of iron-oxidizing bacteria (Ochrobactrum EEELCW01) on mineral transformation and arsenic (As) fate.

Extracellular polymeric substances (EPS) are an important medium for communication and material exchange between iron-oxidizing bacteria and the external environment and could induce the iron (oxyhydr) oxides production which reduced arsenic (As) availability. The main component of EPS secreted by iron-oxidizing bacteria (Ochrobactrum EEELCW01) was composed of polysaccharides (150.76-165.33 mg/g DW) followed by considerably smaller amounts of proteins (12.98-16.12 mg/g DW). Low concentrations of As (100 or 500 µmol/L) promoted the amount of EPS secretion. FTIR results showed that EPS was compo

A silk-based high impact composite for the core decompression of the femoral head.

Core decompression of the femoral head is a recommended head-conserving strategy for early-stage osteonecrosis of the femoral head. However, no ideal filling material has been found so far. In this study, we fabricated a "solid core-porous coating" composite scaffold, which is a silk fibroin/hydroxypropyl methylcellulose (SF/HPMC) scaffold, by a "two-step" process. The solid core scaffold possesses a sufficient compression modulus (860 MPa) for support, while the porous coating scaffold with controllable pore size and porosity provides a suitable microenvironment for the osteoblast cell to adh

Antioxidant defense mechanisms and its dysfunctional regulation in the mitochondrial disease, Friedreich's ataxia.

Redox stress is associated with the pathogenesis of a wide variety of disease states. This can be amplified potentially through redox active iron deposits in oxidatively active organelles such as the mitochondrion. There are a number of disease states, including Friedreich's ataxia (FA) and sideroblastic anemia, where iron metabolism is dysregulated and leads to mitochondrial iron accumulation. Considering FA, which is due to the decreased expression of the mitochondrial protein, frataxin, this iron accumulation does not occur within protective storage proteins such as mitochondrial ferritin.

Hunter-Gatherers Harvested and Heated Microbial Biogenic Iron Oxides to Produce Rock Art Pigment.

Red mineral pigment use is recognized as a fundamental component of a series of traits associated with human evolutionary development, social interaction, and behavioral complexity. Iron-enriched mineral deposits have been collected and prepared as pigment for use in rock art, personal adornment, and mortuary practices for millennia, yet little is known about early developments in mineral processing techniques in North America. Microanalysis of rock art pigments from the North American Pacific Northwest reveals a sophisticated use of iron oxide produced by the biomineralizing bacterium Leptoth

Investigating the genetic and epigenetic basis of big biological questions with the parthenogenetic marbled crayfish: A review and perspectives.

In the last 15 years, considerable attempts have been undertaken to develop the obligately parthenogenetic marbled crayfish Procambarus virginalis as a new model in biology. Its main advantage is the production of large numbers of offspring that are genetically identical to the mother, making this crustacean particularly suitable for research in epigenetics. Now, a draft genome, transcriptome and genome-wide methylome are available opening new windows for research. In this article, I summarize the biological advantages and genomic and epigenetic features of marbled crayfish and, based on first

Mineral Surface-Templated Self-Assembling Systems: Case Studies from Nanoscience and Surface Science towards Origins of Life Research.

An increasing body of evidence relates the wide range of benefits mineral surfaces offer for the development of early living systems, including adsorption of small molecules from the aqueous phase, formation of monomeric subunits and their subsequent polymerization, and supramolecular assembly of biopolymers and other biomolecules. Each of these processes was likely a necessary stage in the emergence of life on Earth. Here, we compile evidence that templating and enhancement of prebiotically-relevant self-assembling systems by mineral surfaces offers a route to increased structural, functional
