Snake cytotoxins bind to membranes via interactions with phosphatidylserine head groups of lipids.

The major representatives of Elapidae snake venom, cytotoxins (CTs), share similar three-fingered fold and exert diverse range of biological activities against various cell types. CT-induced cell death starts from the membrane recognition process, whose molecular details remain unclear. It is known, however, that the presence of anionic lipids in cell membranes is one of the important factors determining CT-membrane binding. In this work, we therefore investigated specific interactions between one of the most abundant of such lipids, phosphatidylserine (PS), and CT 4 of Naja kaouthia using a c

Cobrotoxin alleviates lipopolysaccharide-induced depression-like behavior in mice.

Depression is a debilitating psychiatric disorder closely linked to neuroinflammation. Cobrotoxin (CoTX), an active component of Naja atra venom, exhibits multiple therapeutic effects, including analgesic, anti-inflammatory, and microcirculation-improving properties. However, its role in neuroinflammation-associated depression remains unexplored. This study investigated the effects of CoTX on lipopolysaccharide (LPS)-induced depression-like behavior in mice and underlying mechanisms. Our results demonstrated that CoTX treatment significantly ameliorated LPS-induced depression-like behaviors, a

A review and in silico screening of plant-derived snake venom/toxin inhibitors: ADMET, drug-likeness, and medicinal chemistry profiling.

Snakebite envenomation remains a pressing global health issue, contributing to high rates of illness and death. It also imposes significant socio-economic burdens on affected communities. Recent conservative estimates suggest that approximately 5.4 million snakebite incidents occur annually, leading to nearly 138,000 fatalities. In Africa alone, as many as 500,000 cases are documented each year. This research seeks to explore phytochemicals reported to exhibit in vitro and in vivo inhibitory effects on snake venom and toxin targets. A systematic review was conducted utilizing six electronic da

Bacterial Adaptation to Venom in Snakes and Arachnida.

Animal venoms are considered sterile sources of antimicrobial compounds with strong membrane-disrupting activity against multidrug-resistant bacteria. However, venomous bite wound infections are common in developing nations. Investigating the envenomation organ and venom microbiota of five snake and two spider species, we observed venom community structures that depend on the host venomous animal species and evidenced recovery of viable microorganisms from black-necked spitting cobra (Naja nigricollis) and Indian ornamental tarantula (Poecilotheria regalis) venoms. Among the bacterial isolates

Heparin-incorporated whey protein isolate-derived hydrogels with an intended dual function as snakebite wound dressings and drug delivery systems inhibit spitting cobra venom-induced cytotoxicity.

Snakebite envenoming affects millions of people annually, with current treatments limited to animal-derived antivenoms. Repurposed drug-inhibitors of toxin families offer an exploitable avenue to improve snakebite treatment, including heparins which can inhibit cytotoxic three-finger toxins. However, to be effective therapies in-the-field such treatments must be engineered into drug delivery devices capable of rapidly administering drug(s) to the envenomation site. Herein we introduce the concept of integrating heparins, specifically unfractionated heparin (H) and its low molecular weight hepa

Complete amino acid sequence of kaouthiagin, a novel cobra venom metalloproteinase with two disintegrin-like sequences.

The primary structure of kaouthiagin, a metalloproteinase from the venom of the cobra snake Naja kaouthia which specifically cleaves human von Willebrand factor (VWF), was determined by amino acid sequencing. Kaouthiagin is composed of 401 amino acid residues and one Asn-linked sugar chain. The sequence is highly similar to those of high-molecular mass snake venom metalloproteinases from viperid and crotalid venoms comprised of metalloproteinase, disintegrin-like, and Cys-rich domains. The metalloproteinase domain had a zinc-binding motif (HEXXHXXGXXH), which is highly conserved in the metzinc

Enzymatic properties of rat group IIA and V phospholipases A(2) compared.

Group IIA and V phospholipases A(2) (PLA(2)s) are known to play a role in inflammatory responses. We have constructed a bacterial expression vector for rat group IIA and V PLA(2)s, over-expressed, folded and purified the proteins with the aim to study and compare the properties of the enzymes in detail. For zwitterionic phospholipid micelles, both enzymes display optimum activity at pH 8. 0 and absolutely require Ca(2+) for enzymatic activity. In the presence of substrate, group V PLA(2) has a high affinity for Ca(2+) (K(Ca2+)=90 microM) while K(Ca2+) of group IIA PLA(2) was found to be 1.6 mM

Characterization by infrared spectroscopy of the interaction of a cardiotoxin with phosphatidic acid and with binary mixtures of phosphatidic acid and phosphatidylcholine.

The effect of cardiotoxin IIa from Naja mossambica mossambica, a small basic protein extracted from snake venom, on dimyristoylphosphatidic acid (DMPA) and on equimolar mixtures of DMPA and dimyristoylphosphatidylcholine (DMPC) has been studied by Fourier transform infrared spectroscopy. The interaction of cardiotoxin with DMPA dispersions decreases both the cooperativity of the phase transition of the lipid and the molecular order of the lipid acyl chains in the gel phase. This effect increases with the proportion of the toxin in the complexes and leads to the total abolition of the phase tra

Evaluation of the lethal potency of scorpion and snake venoms and comparison between intraperitoneal and intravenous injection routes.

Scorpion stings and snake bites are major health hazards that lead to suffering of victims and high mortality. Thousands of injuries associated with such stings and bites of venomous animals occur every year worldwide. In North Africa, more than 100,000 scorpion stings and snake bites are reported annually. An appropriate determination of the 50% lethal doses (LD₅₀) of scorpion and snake venoms appears to be an important step to assess (and compare) venom toxic activity. Such LD₅₀ values are also commonly used to evaluate the neutralizing capacity of specific anti-venom batches. In the present

Structure-function relationships and mechanism of anticoagulant phospholipase A2 enzymes from snake venoms.

Phospholipase A(2) (PLA(2)) enzymes from snake venom are toxic and induce a wide spectrum of pharmacological effects, despite similarity in primary, secondary and tertiary structures and common catalytic properties. Thus, the structure-function relationships and the mechanism of this group of small proteins are subtle, complex and intriguing challenges. This review, taking the PLA(2) enzymes from spitting cobra (Naja nigricollis) venom as examples, describes the mechanism of anticoagulant effects. The strongly anticoagulant CM-IV inhibits both the extrinsic tenase and prothrombinase complexes,

Assessing the Binding of Venoms from Aquatic Elapids to the Nicotinic Acetylcholine Receptor Orthosteric Site of Different Prey Models.

The evolution of an aquatic lifestyle from land dwelling venomous elapids is a radical ecological modification, bringing about many evolutionary changes from morphology to diet. Diet is an important ecological facet which can play a key role in regulating functional traits such as venom composition and prey-specific targeting of venom. In addition to predating upon novel prey (e.g., fish, fish eggs and invertebrates), the venoms of aquatic elapids also face the challenge of increased prey-escape potential in the aquatic environment. Thus, despite the independent radiation into an aquatic niche

Natural and synthetic cathelicidin peptides with anti-microbial and anti-biofilm activity against Staphylococcus aureus.

BACKGROUND: Chronic, infected wounds typically contain multiple genera of bacteria, including Staphylococcus aureus, many of which are strong biofilm formers. Bacterial biofilms are thought to be a direct impediment to wound healing. New therapies that focus on a biofilm approach may improve the recovery and healing rate for infected wounds. In this study, cathelicidins and related short, synthetic peptides were tested for their anti-microbial effectiveness as well as their ability to inhibit the ability of S. aureus to form biofilms. RESULTS: The helical human cathelicidin LL-37 was tested a

Acetylcholinesterase from snake venom as a model for its nerve and muscle counterpart.

Acetylcholinesterase (AChE) plays a key role in cholinergic transmission. At the neuromuscular junction of vertebrates, for example, it allows a fine temporal control of muscle contraction. The presence of AChE in tissues devoid of cholinergic function is also well known and raises the question of its role. In particular, AChE is abundant in the venoms of Elapid snakes, except mambas. AChE purified from snake venom consists of soluble, hydrophilic monomers. Cloning of cDNA of the AChE from Bungarus fasciatus venom showed that its C-terminal peptide is very different from those of other AChEs.

Protective effects of clinical anticholinergic and anticholinesterase agents against Bungarus multicinctus venom and neurotoxin-rich snake venoms.

Bungarus multicinctus is one of the most venomous and lethal snake species in mainland China, with envenomation resulting in a mortality rate as high as 23%. Currently, antivenin against B. multicinctus remains the gold standard for treating bites from this species. However, in remote mountainous areas or rural regions of China, access to antivenin may be delayed or unavailable following a bite. The lethal components of B. multicinctus venom include α-, β-, γ- and κ-bungarotoxins, which act on either the presynaptic or postsynaptic membrane, thereby altering acetylcholine levels in the synapti
