PROTAC-Mediated Dual Degradation of BCL-xL and BCL-2 Is a Highly Effective Therapeutic Strategy in Small-Cell Lung Cancer.

BCL-xL and BCL-2 are validated therapeutic targets in small-cell lung cancer (SCLC). Targeting these proteins with navitoclax (formerly ABT263, a dual BCL-xL/2 inhibitor) induces dose-limiting thrombocytopenia through on-target BCL-xL inhibition in platelets. Therefore, platelet toxicity poses a barrier in advancing the clinical translation of navitoclax. We have developed a strategy to selectively target BCL-xL in tumors, while sparing platelets, by utilizing proteolysis-targeting chimeras (PROTACs) that hijack the cellular ubiquitin proteasome system for target ubiquitination and subsequent

Strategies to Reduce the On-Target Platelet Toxicity of Bcl-xL Inhibitors: PROTACs, SNIPERs and Prodrug-Based Approaches.

Apoptosis is a highly regulated cellular process. Aberration in apoptosis is a common characteristic of various disorders. Therefore, proteins involved in apoptosis are prime targets in multiple therapies. Bcl-xL is an antiapoptotic protein. Compared to other antiapoptotic proteins, the expression of Bcl-xL is common in solid tumors and, to an extent, in some leukemias and lymphomas. The overexpression of Bcl-xL is also linked to survival and chemoresistance in cancer and senescent cells. Therefore, Bcl-xL is a promising anticancer and senolytic target. Various nanomolar range Bcl-xL inhibitor

Chromosome 6q mediated loss of LATS1 tumour suppression as a targetable mechanism of ultra-high-risk posterior fossa A ependymoma.

Posterior Fossa A (PFA) is the most frequent and clinically aggressive ependymoma (EPN) subgroup, with relapse typically representing a terminal event despite intensive multimodal therapy. Ultra-high-risk PFA EPNs are characterised by chromosome 6q loss (6q-) with or without chromosome 1q gain (1q +). While the molecular mechanisms driving 1q+ have been elucidated, 6q- PFA pathobiology remains poorly understood. Here, we show that loss of 6q in PFA EPN results in deletion of LATS1 (Large Tumour Suppressor Kinase 1), thereby dysregulating the Hippo signalling pathway and enhancing YAP/TEAD acti

GPCR signaling systems facilitate precision interventions for multi-oncology therapy.

Precision oncology has undergone a transformative evolution from static genomic profiling toward dynamic, multi-omics frameworks capable of capturing tumor heterogeneity and adaptive resistance in real time. Central to this paradigm shift is the emerging recognition that G protein-coupled receptors (GPCRs) constitute a vastly underexploited axis of oncogenic signaling, tumor microenvironment (TME) modulation, and non-genetic adaptive resistance. Here we delineate the effective transition to GPCR-focused precision oncology, emphasizing the integrated deployment of next-generation sequencing (NG

Advances in JNK inhibitor development: therapeutic prospects in neurodegenerative diseases and fibrosis.

c-Jun N-terminal kinases (JNKs), a subfamily of mitogen-activated protein kinases (MAPKs), are key mediators of cellular responses to environmental stress, inflammation, and apoptotic signals. The three isoforms-JNK1, JNK2, and JNK3 exhibit both overlapping and isoform-specific functions. While JNK1 and JNK2 are broadly expressed across tissues and regulate immune signaling, cell proliferation, and apoptosis, JNK3 expression is largely restricted to the brain, heart, and testis, where it plays a crucial role in neuronal function and survival. Subtle structural variations among the isoforms, pa

Targeting the CDK6 Dependence of Ph+ Acute Lymphoblastic Leukemia.

Ph+ ALL is a poor-prognosis leukemia subtype driven by the BCR-ABL1 oncogene, either the p190- or the p210-BCR/ABL isoform in a 70:30 ratio. Tyrosine Kinase inhibitors (TKIs) are the drugs of choice in the therapy of Ph+ ALL. In combination with standard chemotherapy, TKIs have markedly improved the outcome of Ph+ ALL, in particular if this treatment is followed by bone marrow transplantation. However, resistance to TKIs develops with high frequency, causing leukemia relapse that results in <5-year overall survival. Thus, new therapies are needed to address relapsed/TKI-resistant Ph+ ALL. We h

Characterizing the conformational ensemble of PROTAC degraders in solutions via atomistic simulations.

Targeted protein degradation has emerged as a promising strategy for developing novel therapeutics, particularly for "undruggable" disease-related proteins. One approach is the use of PROteolysis TArgeting Chimeras (PROTACs) degraders, which induce the formation of ternary complexes between the target protein and E3 ligase, leading to ubiquitination and degradation of the target protein. Understanding the conformational behavior of PROTACs in solutions and how it relates to their pharmacokinetic properties and membrane permeability is crucial for optimizing PROTAC design and efficacy. Due to t

CADD-engineered peptide protacs efficiently target PCSK9 for hypercholesterolemia in vivo.

BACKGROUND: Proprotein convertase subtilisin/kexin type 9 (PCSK9) promotes the degradation of low-density lipoprotein receptors (LDLR), leading to elevated plasma LDL cholesterol (LDL-C) and increased risk of hypercholesterolemia. Current therapeutic approaches, such as monoclonal antibodies and gene-editing tools, face significant challenges including high cost, safety issues, and limited ability to target intracellular PCSK9. METHODS: Using computer-aided drug design (CADD), we developed Cadd4, a novel peptide-based degrader targeting PCSK9. Molecular docking was employed to identify a high

SMYD3 protein degraders in HPV-negative head and neck squamous cell carcinoma.

Despite the advent of immunotherapy in the treatment of human-papilloma-virus (HPV)-negative head and neck squamous cell carcinoma, the overall prognosis still remains dismal. Novel therapeutic approaches are thus urgently needed. We recently reported that SET and MYND containing 3 (SMYD3), a protein methyltransferase that is known to activate the transcription of its target genes through H3K4me3, is overexpressed in ∼60% of HPV-negative HNSCC tumors, it drives proliferation and invasion, and induces resistance to immunotherapy in HPV-negative HNSCC. These phenotypes are mediated partially thr

Emerging Strategies Targeting the PI3K/AKT/mTOR Pathway in HR+/HER2- Advanced Breast Cancer.

Hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2-) breast cancer accounts for approximately 70% of breast cancer cases. Despite recent advances with cyclin-dependent kinase 4/6 inhibitors (CDK4/6i), resistance inevitably develops, often driven by activation of the phosphatidylinositol 3-kinase (PI3K)-AKT-mammalian target of rapamycin (mTOR) pathway. Genetic alterations such as PIK3CA mutations (present in ~ 45% of HR+/HER2- tumors), AKT1 mutations, and PTEN loss contribute to endocrine resistance and poor outcomes. This review summarizes emerging strateg

Beyond hypoxia and ccRCC: the expanding universe of VHL novel targets and next-generation therapeutics.

The von Hippel-Lindau (VHL) gene encodes the core substrate-recognition subunit of the Cullin2-RING E3 ubiquitin ligase complex, regulating cellular oxygen sensing and proteostasis. While targeting the canonical VHL-hypoxia-inducible factor (HIF) axis (e.g. belzutifan) has improved advanced clear cell renal cell carcinoma (ccRCC) management, intrinsic non-responsiveness and acquired resistance to HIF-2α inhibitors present major clinical limitations. Expanding beyond the HIF-centric paradigm reveals a broader network of non-canonical VHL targets, offering new opportunities for therapeutic devel

Targeting signaling pathways in lymphoma: From molecular mechanisms to clinical breakthroughs.

AbstractThe treatment paradigm for lymphoma, a highly heterogeneous group of hematologic malignancies, has been revolutionized by the development of therapies targeting oncogenic signaling pathways. This shift from conventional chemotherapy to precision medicine is driven by a deep molecular understanding of the pathways that govern lymphoma cell survival and proliferation. This review comprehensively surveys the landscape of these targeted therapies, from fundamental molecular mechanisms to clinical breakthroughs. We first dissect the molecular architecture of key oncogenic drivers, covering

Post-Translational Modifications in Traumatic Brain Injury: Decoding the Proteomic Landscape and Molecular Mechanisms of Secondary Injury.

Traumatic brain injury (TBI) initiates a complex secondary injury cascade that significantly contributes to long-term neurological deficits, with post-translational modifications (PTMs) emerging as pivotal molecular regulators of this process. Unlike primary mechanical damage, secondary injury evolves over hours to years and involves intricate proteomic alterations that changes in gene expression alone cannot fully explain. PTMs-including phosphorylation, ubiquitination, acetylation, SUMOylation, glycosylation, and emerging modifications such as succinylation, lactylation, and nitrosylation-se

A PROTAC-inspired bifunctional small molecule recruits CDK6 to target EYA2 degradation and suppress breast tumor growth.

Cyclin-dependent kinase 6 (CDK6) has been reported to exert tumor-suppressive functions through its non-enzymatic activity and promotes the degradation of the oncogenic driver EYA2. This suggests a potential strategy for targeted protein degradation by exploiting endogenous protein-protein interactions. Here, we report the design and synthesis of a bifunctional small molecule, EC21, capable of simultaneously engaging CDK6 and EYA2. Structure-guided design based on the reported binding modes of palbociclib and the EYA2 inhibitor ETC-616 enabled the identification of EC21 as a lead compound. Bio

Discovery of Potent Dual PROTAC Degraders Targeting BET-Kinase To Overcome FLT3 Inhibitor Resistance.

FMS-like tyrosine kinase 3 (FLT3) is a validated therapeutic target in acute myeloid leukemia (AML). However, numerous single-agent FLT3 or multitarget inhibitors fail to achieve complete and sustained suppression of FLT3 signaling due to the development of drug resistance. Herein, we report the design, synthesis, and evaluation of a novel series of dual BET-kinase degraders targeting FLT3, JAK2, and BRD4. Optimization led to 13e, which efficiently degraded FLT3, JAK2, and BRD4 in MV4;11 cells with DC50 values of 5.23, 0.678, and 1.17 nM, respectively. Mechanistic studies confirmed cereblon- a

Down-regulation of AKT proteins slows the growth of mutant-KRAS pancreatic tumors.

Serine/threonine kinase AKT isoforms play a well-established role in cell metabolism and growth. Most pancreatic adenocarcinoma (PDAC) harbors activation mutations of KRAS, which activates the PI3K/AKT signaling pathway. However, AKT inhibitors are not effective in the treatment of pancreatic cancer. To better understand the role of AKT signaling in mutant-KRAS pancreatic tumors, this study utilizes proteolysis-targeting chimeras (PROTACs) and CRISPR-Cas9-genome editing to investigate AKT proteins. PROTAC down-regulation of AKT proteins markedly slowed the growth of three pancreatic tumor cell

Kinase-independent signaling by PIM1 promotes drug resistance by increasing mitophagy and reducing oxidative stress.

PIM kinases are overexpressed in castration resistant prostate cancer (CRPC) and many small molecule PIM kinase inhibitors (smPIM inhibitors) have been designed to block the catalytic activity of PIM. However, smPIM inhibitors have shown limited efficacy in solid tumors. Notably, all these inhibitors share the common property that they increase total PIM protein levels, which limits their efficacy because PIM1 has kinase-independent pro-survival effects. Here, we identify high mobility box group 1 (HMGB1) as a novel PIM1 binding partner. Stabilization of PIM1 by smPIM inhibitors increases the

Discovery of DCAF16-based PROTACs as SHP2 degraders.

The overexpression of SHP2 in various cancers establishes it as an attractive therapeutic target. Growing evidence indicates that the scaffold function of SHP2 contributes significantly to its oncogenic signaling and tumorigenesis. Unlike traditional inhibitors, PROTACs can simultaneously eliminate both the enzymatic and scaffold functions of the target protein, representing a more promising therapeutic strategy. Herein, we report the design and synthesis of SHP2 PROTACs that recruit the DCAF16 E3 ligase for targeted SHP2 degradation. Among which, SK8 exhibited the most potent anti-tumor activ
