Populations of photoinactivated photosystem II reaction centers characterized by chlorophyll a fluorescence lifetime in vivo.

Photosystem (PS) II centers, which split water into oxygen, protons, and electrons during photosynthesis, require light but are paradoxically inactivated by it. Prolonged light exposure concomitantly decreased both the functional fraction of PSII reaction centers and the integral PSII chlorophyll (Chl) a fluorescence lifetime in leaf segments of Capsicum annuum L. Acceleration of photoinactivation of PSII by a pretreatment with the inhibitors/uncoupler lincomycin, DTT, or nigericin further reduced PSII Chl a fluorescence lifetimes. A global analysis of fluorescence lifetime distributions revea

Fucoxanthin regulates Nrf2 signaling to decrease oxidative stress and improves renal fibrosis depending on Sirt1 in HG-induced GMCs and STZ-induced diabetic rats.

Diabetic nephropathy (DN) is one of the major microvascular complications of diabetes. Nuclear factor erythroid 2-related factor 2 (Nrf2) is a crucial cellular defense factor to cope with oxidative stress. Silent information regulator T1 (Sirt1) is a deacetylase with antioxidative stress activity. Fucoxanthin is a marine-derived carotenoid. This study was conducted to investigate whether fucoxanthin could alleviate oxidative stress by activating Sirt1/Nrf2 signaling to alleviate DN. In streptozotocin-induced diabetic rats, fucoxanthin treatment effectively improved renal function, alleviated g

Astaxanthin attenuates alcoholic cardiomyopathy via inhibition of endoplasmic reticulum stress-mediated cardiac apoptosis.

Chronic excessive ethanol consumption is associated with a high incidence of mortality due to ethanol-induced dilated cardiomyopathy, known as alcoholic cardiomyopathy (ACM). Mechanistic studies have demonstrated that apoptosis is key to the pathogenesis of ACM, and endoplasmic reticulum (ER) stress-associated apoptosis contributes to various ethanol-related diseases. Astaxanthin (AST) is a natural carotenoid that exerts an anti-ER stress effect. Importantly, strong evidence has shown that AST induces beneficial effects in various cardiovascular diseases. The present study aimed to investigate

Depression can be prevented by astaxanthin through inhibition of hippocampal inflammation in diabetic mice.

The critical factor considered in a depression induced by diabetes is the inflammation eliciting hippocampal, amygdala and thalamic neuronal injury. Therefore, inhibiting inflammatory reactions in the brain and reducing neuronal injury can alleviate depression in rodents suffering from diabetes mellitus. The oral administration of astaxanthin has been employed in emotional disorders and diabetic complications due to its anti-depressant, anti-inflammatory and anti-apoptotic functions. However, it has not been reported whether astaxanthin can improve diabetes-related depression-like behavior, an

Astrakurkurone targets multidrug resistant Staphylococcus aureus biofilms through ROS-mediated cell membrane damage and β-lactamase inhibition.

Methicillin-resistant Staphylococcus aureus (MRSA) pose a formidable health threat owing to their multidrug resistance (MDR) nature, robust biofilms and persistence. Natural product-based medications are a highly viable strategy for challenging MDR and biofilm persistence with diverse therapeutic benefits. In this study, the triterpenoid Astrakurkurone, previously purified from a wild edible mushroom Astraeus hygrometricus, showed antibacterial efficacy against MRSA BAA1717 and a clinical isolate (MRSA CI-1) with MICs ranging from 150 to 300 μg/mL. 1/2 MIC concentration of Astrakurkurone also

Effect of Astaxanthin on Activation of Autophagy and Inhibition of Apoptosis in Helicobacter pylori-Infected Gastric Epithelial Cell Line AGS.

Helicobacter pylori (H. pylori) infection leads to the massive apoptosis of the gastric epithelial cells, causing gastric ulcers, gastritis, and gastric adenocarcinoma. Autophagy is a cellular recycling process that plays important roles in cell death decisions and can protect cells by preventing apoptosis. Upon the induction of autophagy, the level of the autophagy substrate p62 is reduced and the autophagy-related ratio of microtubule-associated proteins 1A/1B light chain 3B (LC3B)-II/LC3B-I is heightened. AMP-activated protein kinase (AMPK) and mammalian target of rapamycin (mTOR) are invol

β-Cryptoxanthin exerts greater cardioprotective effects on cardiac ischemia-reperfusion injury than astaxanthin by attenuating mitochondrial dysfunction in mice.

SCOPE: β-Cryptoxanthin and astaxanthin are antioxidant carotenoid pigments that inhibit lipid peroxidation as potently as vitamin E. We hypothesized that acute treatment with β-cryptoxanthin and astaxanthin causes similar reductions in the sizes of cardiac infarcts caused by ischemia-reperfusion (I/R) injury by attenuating oxidative stress and cardiac mitochondrial dysfunction. METHODS AND RESULTS: C57BL/6 mice (n = 36) were randomized to receive vehicle, β-cryptoxanthin, astaxanthin, or vitamin E at 50 mg/kg by gavage feeding prior to I/R injury. Cardiac I/R was induced by left anterior desc

Diet and lifestyle correlates of lutein in the blood and diet.

Observational studies have suggested an inverse relationship between dietary or serum lutein and risk for age-related macular degeneration and cataracts. This evidence has stimulated interest in the biological and other characteristics of lutein, and also zeaxanthin, a structurally similar carotenoid; together, they comprise the macular pigment. Accurate interpretation of data linking dietary intake or serum concentrations of lutein and zeaxanthin and risk for eye disease in epidemiologic and clinical studies requires knowledge of biological and nondietary factors influencing these intake data

Orally engineered liver-targeted garlic exosome-like nanovesicles for astaxanthin precise delivery against alcoholic liver disease in mice.

Orally targeted strategy of nutrients has attracted obvious attention for reducing side effects and enhancing the intervention efficiency of alcoholic liver disease (ALD). Herein, novel lactobionic acid-modified garlic exosome-like nanovesicles (LA-GELN) were designed to precise delivery astaxanthin (AXT) against alcohol-induced lipid metabolism disorders. The targeted modification of nanovesicles enhanced the encapsulation efficiency of AXT (80.03%). Meanwhile, the encapsulation strategy also improved the solubility and gastrointestinal stability of AXT. In the HepG2 cell model, LA-GELN-AXT d

Neuroprotective effect of apo-9'-fucoxanthinone against cerebral ischemia injury by targeting the PI3K/AKT/GSK-3β pathway.

Neuronal loss in cerebral ischemia primarily results from the combined effects of inflammatory responses and programmed cell death. Currently, there is an urgent need for potent neuroprotectants targeting both inflammatory and apoptotic responses for the treatment of ischemic stroke. Marine natural products are a vital source of novel drug candidates. Apo-9'-fucoxanthinone (APO-9'), a degradation product of fucoxanthin derived from marine brown algae, is known for its substantial anti-inflammatory effects, yet its neuroprotective action has not been clearly defined. In this study, the neuropro

The anti-apoptotic effect of fucoxanthin on carbon tetrachloride-induced hepatotoxicity.

This study evaluated the anti-apoptotic activity of fucoxanthin in carbon tetrachloride (CCl(4))-induced hepatotoxicity. An in vitro study using the 3-(4,5-dimethylthiazol-2-yl) 2,5-diphenyltetrazolium bromide (MTT) assay clearly demonstrated an attenuation of CCl(4)-induced hepatotoxicity with fucoxanthin. This effect was dose-dependent; 25 µM was more effective than 10 µM of fucoxanthin for attenuating the hepatotoxicity induced by 5 mM of CCl(4). Acute CCl(4)-hepatotoxicity in rats, with numerous cells positive for the terminal deoxynucleotidyl - transferase (TdT) -mediated deoxyuridine tri

Fucoxanthin Induces Ferroptosis in Hypopharyngeal Carcinoma Cells by Activating the p53/SLC7A11/GPX4 Axis.

Fucoxanthin, a marine carotenoid abundantly derived from brown algae, has been increasingly recognized for its broad-spectrum antitumor activities; however, its role in regulating ferroptosis remains insufficiently defined. Hypopharyngeal carcinoma is a highly aggressive head and neck malignancy with limited therapeutic options, highlighting the need for novel marine-derived anticancer agents. In this study, we investigated whether fucoxanthin induces ferroptosis in human hypopharyngeal carcinoma cells (Fadu) and elucidated the underlying molecular mechanisms. Astaxanthin alleviates DSS-induced ulcerative colitis in mice associated with Nrf2-mediated ferroptosis independently of gut microbiota modulation.

Astaxanthin, a natural carotenoid predominantly synthesized by marine microorganisms, has shown promise in attenuating inflammatory diseases, yet its role in colitis remains unclear. Here, we evaluated the therapeutic effects of astaxanthin in dextran sulfate sodium (DSS)-induced ulcerative colitis in mice. Our findings revealed that astaxanthin significantly ameliorated colitis symptoms, notably at the dose of 100 mg/kg, demonstrated by reduced Disease Activity Index (DAI), increased colon length, diminished colon histopathological damage, and enhanced goblet cell population.
