Extracellular Matrix Protein Signatures of the Outer and Inner Zones of the Rat Adrenal Cortex.

This study analyzes the extracellular matrix (ECM) signatures of the outer (OF = capsule + subcapsular + zona glomerulosa cells) and inner fractions (IF = zona fasciculata cells) of the rat adrenal cortex, which comprise two distinct microenvironment niches. Proteomic profiles of decellularized OF and IF samples, male and female rats, identified 252 proteins, with 32 classified as ECM-component and ECM-related. Among these, 25 proteins were differentially regulated: 17 more abundant in OF, including Col1a1, Col1a2, Col6a1, Col6a2, Col6a3, Col12a1, Col14a1, Lama5, Lamb2, Lamc1, Eln, Emilin, Fbl

Development of human ectocervical tissue models with physiologic endocrine and paracrine signaling†.

There is a shortage of research models that adequately represent the unique mucosal environment of human ectocervix, limiting development of new therapies for treating infertility, infection, or cancer. We developed three microphysiologic human ectocervix models to study hormone action during homeostasis. First, we reconstructed ectocervix using decellularized extracellular matrix scaffolds, which supported cell integration and could be clinically useful. Secondly, we generated organotypic systems consisting of ectocervical explants co-cultured with murine ovaries or cycling exogenous hormones

Transplantation of Isl1+ cardiac progenitor cells in small intestinal submucosa improves infarcted heart function.

BACKGROUND: Application of cardiac stem cells combined with biomaterial scaffold is a promising therapeutic strategy for heart repair after myocardial infarction. However, the optimal cell types and biomaterials remain elusive. METHODS: In this study, we seeded Isl1+ embryonic cardiac progenitor cells (CPCs) into decellularized porcine small intestinal submucosa extracellular matrix (SIS-ECM) to assess the therapeutic potential of Isl1+ CPCs and the biocompatibility of SIS-ECM with these cells. RESULTS: We observed that SIS-ECM supported the viability and attachment of Isl1+ CPCs. Importantl

Extracellular vesicles in stem cell-based gonadal regeneration: mechanisms, therapeutic potential, and translational challenges.

Gonadal dysfunction resulting from conditions such as premature ovarian insufficiency, chemotherapy-induced damage, or genetic disorders often leads to infertility and hormone imbalance. Although assisted reproductive technologies and hormone replacement therapies mitigate clinical symptoms, they remain incapable of reinstating native gonadal architecture and physiological function. In recent years, stem cell-based therapies, particularly those employing mesenchymal stem cells (MSCs), have demonstrated regenerative potential. However, limitations including poor engraftment, potential tumorigen

Vascular smooth muscle cell senescence accelerates medin aggregation via small extracellular vesicle secretion and extracellular matrix reorganization.

Vascular amyloidosis, caused when peptide monomers aggregate into insoluble amyloid, is a prevalent age-associated pathology. Aortic medial amyloid (AMA) is the most common human amyloid and is composed of medin, a 50-amino acid peptide. Emerging evidence has implicated extracellular vesicles (EVs) as mediators of pathological amyloid accumulation in the extracellular matrix (ECM). To determine the mechanisms of AMA formation with age, we explored the impact of vascular smooth muscle cell (VSMC) senescence, EV secretion, and ECM remodeling on medin accumulation. Medin was detected in EVs secre

A detoxified exosome-ECM hydrogel depot for localized and sustained chemotherapy in prostate cancer.

Advanced prostate cancer often necessitates chemotherapy, yet conventional agents such as doxorubicin (DOX) suffer from poor tumor specificity, systemic toxicity, and acquired resistance. Here, we report a biomimetic, injectable hydrogel depot that integrates enzyme-detoxified tumor exosomes (Detox-EXOs) with a chitosan–decellularized prostate extracellular matrix (CS–dpECM) scaffold for localized and sustained chemotherapy. The Detox-EXOs, generated by enzymatic removal of oncogenic and immunogenic contents while preserving membrane integrins and targeting ligands, function as safe, tumor-hom

Ovulation: A consequence of acute inflammation cultivated by E2-induced reactive oxygen species and triggered by progesterone withdrawal.

Ovulation is a complex biological process essential for female fertility, driven by the luteinizing hormone (LH) surge and involving a cascade of molecular events that lead to follicular rupture and oocyte release. This process shares characteristics with acute inflammation, including the generation of reactive oxygen species (ROS) by E2, immune cell recruitment, and tissue remodeling. E2 enhances mitochondrial ROS production through integrin-dependent signaling, regulating key ovulatory events such as cumulus expansion and extracellular matrix breakdown. The LH surge triggers follicular lutei

Characterization of glomerular extracellular matrix by proteomic analysis of laser-captured microdissected glomeruli.

Abnormal extracellular matrix (ECM) remodeling is a prominent feature of many glomerular diseases and is a final common pathway of glomerular injury. However, changes in ECM composition accompanying disease-related remodeling are unknown. The physical properties of ECM create challenges for characterization of composition using standard protein extraction techniques, as the insoluble components of ECM are frequently discarded and many ECM proteins are in low abundance compared to other cell proteins. Prior proteomic studies defining normal ECM composition used a large number of glomeruli isola

SINEUP-mediated enhancement of HNF4α improves metabolic function in Huh7-based bioengineered hepatic microtissues.

BACKGROUND/AIMS: Human liver cell-based drug-screening platforms require stable hepatocyte identity and metabolic competence, but Huh7 cells show reduced hepatic function. We investigated whether SINEUP-mediated enhancement of hepatocyte nuclear factor 4 alpha (HNF4α) could improve the metabolic performance of Huh7-based three-dimensional hepatic microtissues. METHODS: Decellularized liver extracellular matrix-derived microparticles were incorporated into three-dimensional microtissues containing Huh7 cells, human umbilical vein endothelial cells, and Wharton's jelly mesenchymal stem cells us

Glycosylated extracellular matrix drives immune suppression by modulating macrophage-T cell crosstalk in triple-negative breast cancer.

The tumor extracellular matrix (ECM) is increasingly recognized as a key driver of immune suppression and therapy resistance in cancer. However, the specific ECM components and mechanisms that create this immunosuppressive environment remain poorly understood, hindering the development of new therapies. Here, we use comprehensive multi omics profiling of triple-negative breast cancer (TNBC), an aggressive and treatment-resistant subtype, to investigate this issue. We report that ECM immunomodulation in TNBC is mediated by post-translational glycan modifications on ECM proteins. Using decellula

Meniscus Tissue Engineering Scaffolds: Biomaterials, Biofabrication, and Translation.

The meniscus is a fibrocartilaginous tissue essential for load transmission, shock absorption, joint stability, and cartilage protection in the knee. However, its intrinsic healing capacity is severely limited, particularly in the avascular region and in complex defects, often resulting in persistent symptoms, functional impairment, and progressive joint degeneration. Although current clinical interventions, including meniscal repair, partial meniscectomy, allograft transplantation, and scaffold-assisted meniscal substitution, can provide symptomatic and functional improvement in selected pati

Advances in Biomaterial-Based Cardiac Organoids Engineering for Modeling Development and Congenital Heart Defects.

Human cardiac organoids have emerged as transformative platforms that bridge the gap between reductionist 2D cultures and poorly predictive animal models. By recapitulating the three-dimensional tissue architecture, multicellular composition, and developmental dynamics of the human heart, these systems enable mechanistic studies of heart development and congenital heart disease. This review surveys the bioengineering strategies underlying cardiac organoid generation, with an emphasis on how biomaterial-based approaches enhance organoid fidelity, functional maturation, and experimental reproduc

Harmonizing Rigidity and Flexibility: Embedding COF Quantum Dots Into Extracellular Matrix Gel as Carbon Monoxide Depot for Acoustically Triggered Time-Programmable Anti-Infective Therapy.

Multidrug-resistant (MDR) bacterial infections persist as a critical threat due to the dual challenge of pathogen burden and challenging post-infection tissue repair. Herein, this work presents an acoustically triggered carbon monoxide (CO) depot based on rigid-flexible integrated polymer networks that are engineered for high-capacity storage and sustained release of CO for infection-to-regeneration. This depot is architected by embedding CO precursor-loaded metallized covalent organic framework quantum dots (COFQDs) into a decellularized extracellular matrix (ECM) gel network derived from sma

Artificial Cornea Substitute Based on Hydrogel Skeletons with Natural Stromal Hierarchical Structure and Extracellular Matrix for Sutureless Transplantation.

Corneal substitutes with structural and compositional characteristics resembling those of natural corneas have attracted considerable attention. However, biomimicking the complex hierarchical organization of corneal stroma is challenging. In this study, humanized corneal stroma-like adhesive patches (HCSPs) are prepared through a multi-step process. First, polyethylene glycol diacrylate is cast and cured within decellularized porcine cornea (DPC) templates. The DPCs are then enzymatically digested to obtain hydrogel skeletons, which are finally integrated with human corneal extracellular matri

In situ vascularization and epithelialization of segmental bioengineered trachea based on marrow-derived stem/progenitor cells.

The success of tracheal transplantation depends on the rapid establishment of vascularization and epithelialization to support functional tissue formation. This study presents an innovative approach for in situ transplantation of a biomimetic tracheal graft, integrating microvascularization and epithelialization. First, endothelial progenitor cells (EPCs) and mesenchymal stem cells (MSCs) were isolated and purified from bone marrow, serving as seed cells for graft vascularization and epithelialization. Next, 3D printing was employed to create a bilayered tracheal graft using poly(ε-caprolacton

Immunomodulatory extracellular matrix hydrogel induces tissue regeneration in a model of partial glossectomy.

While oropharyngeal cancer treatment regimens, including surgical resection, irradiation, and chemotherapy, are effective at removing tumors, they lead to muscle atrophy, denervation, and fibrosis, contributing to the pathogenesis of oropharyngeal dysphagia - difficulty swallowing. Current standard of care of rehabilitative tongue strengthening and swallowing exercises is ineffective. Here, we evaluate an alternative approach utilizing an acellular and injectable biomaterial to preserve muscle content and reduce fibrosis of the tongue after injury. A Multifunctional Tissue-Engineering Hydrogel Aimed to Regulate Bacterial Ferroptosis-Like Death and Overcoming Infection Toward Bone Remodeling.

Infection is the most common complication after orthopedic surgery and can result in prolonged ailments such as chronic wounds, enlarged bone defects, and osteomyelitis. Iron, which is essential for bacterial metabolism and immune cell functions, is extremely important. Bacteria harness iron from nearby cells to promote biofilm formation, ensuring their survival. Iron deficiency within the infection microenvironment (IME) consequently hampers macrophage function, enabling further dissemination of the infection and hindering macrophage polarization to the M2 phenotype. Therefore, a novel approa

Selective Extracellular Matrix Guided Mesenchymal Stem Cell Self-Aggregate Engineering for Replication of Meniscal Zonal Tissue Gradient in a Porcine Meniscectomy Model.

Degenerative meniscus tears (DMTs) are prevalent findings in osteoarthritic knees, yet current treatment is mostly limited to arthroscopic partial meniscectomy rather than regeneration, which further exacerbates arthritic changes. Translational research regarding meniscus regeneration is hindered by the complex, composite nature of the meniscus which exhibit a gradient from inner cartilage-like tissue to outer fibrous tissue, as well as engineering hurdles often requiring growth factors and cross-linking agents. Here, a meniscus zonal tissue gradient is proposed using zone-specific decellulari

3D-Bioprinted Difunctional Scaffold for In Situ Cartilage Regeneration Based on Aptamer-Directed Cell Recruitment and Growth Factor-Enhanced Cell Chondrogenesis.

Articular cartilage (AC) lesions are fairly common but remain an obstacle for clinicians and researchers due to their poor self-healing capacity. Recently, a promising therapy based on the recruitment of autologous mesenchymal stem cells (MSCs) has been developed for the regeneration of full-thickness cartilage defects in the knee joint. In this study, a 3D-bioprinted difunctional scaffold was developed based on aptamer HM69-mediated MSC-specific recruitment and growth factor-enhanced cell chondrogenesis. The aptamer, which can specifically recognize and recruit MSCs, was first chemically conj
