Discrimination power of Investigator DIPplex loci in Finnish and Somali populations.

In this study, the suitability of the Investigator DIPplex insertion/deletion polymorphism (indel) kit for forensic casework was assessed through the genotyping of 151 Finns and 175 Somalis. Allele frequency and heterozygosity (H) of this 30-indel marker set were determined, and forensic efficacy was evaluated through estimation of discrimination power (DP), match probability (MP), typical paternity index (TPI), power of paternity exclusion (PE), and polymorphic information content (PIC). A high level of discrimination power was observed for the marker set in both sample groups (CDP>0.9999). E

Who packed the drugs? Improved DNA recovery and its impact on findings given activity-level propositions.

The interpretation of forensic DNA findings given activity-level propositions depends on the processes governing DNA transfer, persistence, and recovery, as well as on the laboratory methods used to recover DNA. This study investigated the effects of transfer type, substrate characteristics, and DNA extraction strategy on DNA yield, STR profiling performance, and resulting likelihood ratios given activity-level propositions. Direct and indirect transfer experiments were performed using personal bags (backpacks and purses) and zip-lock bags. Three analytical approaches were compared: Chelex ext

Mutation or uniparental disomy? Evaluating abnormal inheritance patterns using Object-Oriented Bayesian Networks.

Current methods of analysis in paternity testing are often based on the assumption that most incompatibilities observed between the child and the parents genetic profiles have a paternal origin. In addition, many inconsistencies are commonly treated as a product of slippage mutational events, and this premise is considered when evaluating the statistical result. Recent high-density genotype data suggest that uniparental disomy (UPD) has a higher frequency of occurrence than previously believed. Therefore, the possibility of UPD must be considered when likelihood ratios (LR) for paternity hypot

Influence of physiological activation on Touch DNA deposition and STR profile quality during competitive computer gaming: A forensic pilot study.

Touch DNA recovery from handled objects is influenced by donor, substrate, transfer, persistence, and collection-related factors; however, the role of short-term physiological activation during contact remains poorly understood. This pilot crossover study evaluated whether competitive gaming was associated with differences in Touch DNA deposition and STR profile quality on computer input devices compared with routine computer browsing. Twenty-four adult participants completed both 30-min activity conditions in randomized order, separated by a 30-min washout period. Heart rate and palmar moistu

Hyperplex PCR for forensic mRNA-based cell typing.

Identification of body fluids or cell types may strengthen the evidential value of biological crime scene stains by clarifying the connection between the evidence and a specific activity. Studying gene expression through mRNA analysis is an effective way to determine the cell type, due to the specific expression of some genes in certain cells. A method that allows for quick analysis of high numbers of mRNA markers spanning all relevant cell types and has a large dynamic range is therefore warranted. Hyperplex PCR (hpPCR) is a new technique that enables simultaneous detection of up to 100 RNA o

Observations and investigation of STR genotype inconsistencies due to mutations, silent alleles, and uniparental disomy in South African populations.

The fundamental concept of parentage and kinship testing is that a child inherits DNA in accordance with the principles of Mendelian inheritance. A child receives half of its DNA from each parent. However, sometimes the observed genotype patterns between a child and its purported parents do not fully align with Mendelian inheritance, which can result in a false conclusion regarding parentage. Mutations, silent alleles, and uniparental disomy (UPD) are examples of events that can produce non-Mendelian results. This study examines STR profiles from South African parentage testing data to assess

Optimizing hard cutoff quality control metrics for IBD segment inference and genotyping accuracy in low-coverage sequencing data.

Effective quality control (QC) is critical for reliable identity-by-descent (IBD) inference from low-pass whole-genome sequencing (LPWGS), where genotype uncertainty under low coverage can bias relatedness estimates. We conducted a comprehensive grid search to evaluate genotype posterior probability (GP) and Bayes factor (BF) thresholds across multiple coverage levels, assessing performance by false positive rate, false negative rate, and mean squared error (MSE) of total IBD sharing. At moderate-to-high coverage (> 0.10 ×), GP > 0.99 maintained stable performance and additional BF filtering p

Long-read, high-throughput sequencing of 10,000 human mitogenomes using the PacBio Sequel IIe.

Sequence analysis of the human mitochondrial genome (mitogenome) is of interest to the molecular anthropology, medical, and forensic communities. Quality mitogenome data is an essential component of haplotype search databases, serving as an important element of forensic investigations to ensure that weight estimates are reflective of accurate coincidental match probabilities. The European DNA Profiling group (EDNAP) Mitochondrial DNA Population database (EMPOP) is considered the gold standard for this purpose, serving as a reference database and a quality-control tool. The current study report

Species-origin identification of the deer in the Purtier Placenta Live Stem Cell Therapy by DNA typing.

Purtier Placenta Live Stem Cell Therapy is a food supplement by Riway Singapore Pte. Ltd., claimed to contain living deer placenta stem cells together with 11 other active ingredients. It is also promoted as maintaining health and youthfulness and as being supported by numerous patents and research trials. Individual ingredients are accompanied by many other statements implying that Purtier Placenta can alleviate or cure diseases such as diabetes, cancer, and neurodegenerative diseases. However, none of these claims is traceable in clinical trial registries, and the mechanism allowing stem cel

A multiplex bacterial assay for identifying vaginal fluid in various individual states and mixture stains.

Accurate identification of vaginal fluid is imperative for forensic investigations, particularly in resolving sexual assault cases. Body fluid-specific microbiota are considered potential indicators for identifying body fluids. In this study, we selected vaginal core bacteria based on species level and developed a multiplex PCR system for identifying vaginal fluid in various individual states and mixed stains. This system was validated using a set of 350 samples (245 vaginal samples and 105 other body fluid samples). The system demonstrated high sensitivity, stability against inhibitors, and s

Environmental microbiota from substrate may interfere with microbiome-based identification of forensically relevant body fluids: A pilot study.

The microbiome is a promising tool for identifying body fluids which can be deposited on various substrates at a crime scene. Body fluids collected from crime scenes are not entirely free from substrate microbes whose effects on the microbiome-based identification of body fluids are not well understood. In this study, five body fluids (peripheral blood, menstrual blood, nasal secretions, saliva, and semen) were deposited on sterile swabs, bedspreads, and floors under indoor exposure conditions for 7 days. The microbial communities in the samples were characterized using amplicon sequencing tar

Characterization and evaluation of nine Cannabis sativa chloroplast SNP markers for crop type determination and biogeographical origin on European samples.

Cannabis sativa can be classified in two main types, according to psychotropic cannabinoid ∆9-tetrahydrocannabinol (∆9-THC) content: the drug-type and the fiber-type. According to the European Monitoring Center for Drugs and Drug Addiction, most of the European Union countries consider the possession of cannabis, for personal use, a minor offense with possibility of incarceration. Despite of the model of legal supply (i.e., Spanish cannabis clubs, Netherlands coffee shops) or medical use (i.e., Italy), cannabis remains the most used and trafficked illicit plant in the European Union. DNA transfer to placed, stored, and handled drug packaging and knives in houses.

Forensic laboratories often sample weapons and clip-seal plastic bags (CSPB) used to package illicit material for the purpose of identifying the handler(s). However, there may be other explanations as to how a person's DNA was transferred to such items. This may include an individual storing the item among their personal belongings for somebody else or the item being stored among their belongings without their knowledge. Here we investigate the direct transfer of DNA to knives and CSPB during handling and explore two feasible alternative explanations related to the indirect transfer of DNA to

Serial estimation of gene expression of cardiac troponin I (cTnI) and autophagy gene HMGB1 to determine postmortem interval.

The estimation of accurate post mortem interval (PMI) is a crucial question in forensic medicine. Several approaches have been used to determine the PMI including physical, metabolic, autolytic, entomological, physiochemical and biochemical methods over time. For estimation of PMI, RNA degradation after death is reported to be an important tool. This study aimed to analyse the pattern of gene expression by serial estimation of cardiac specific cardiac troponin I (cTnI) gene and autophagy gene HMGB1 for determining PMI at room temperature by using housekeeping gene GAPDH. Persistence of blood (DNA/RNA) on shoe soles under varying casework related conditions.

Blunt force traumas by footwear can result in severe and even fatal head and upper body injuries. Oftentimes, footwear impressions are only partially available and evidential value is limited. DNA evidence on shoe soles could provide crucial evidence helping to solve crimes by linking target DNA to the activity of interest. Little is known about the persistence and detectability of biological material post such offenses and the interplay of factors affecting the analytical success. In this study, we assessed the persistence of blood on shoe soles under varying parameters such as blood location

The compatibility of immunolabeling with STR profiling.

Immunolabeling is a technique, which has recently been introduced to enhance the quality of developed fingermarks and subsequently strengthen the evidential value. The effect of this method on subsequent DNA analysis, however, has not been explored yet. Therefore, the current pilot study aimed to determine whether STR profiling is possible after immunolabeling. Since immunolabeling involves washing steps which could reduce DNA quantities, the use of different fixatives including methanol, formaldehyde and universal molecular fixative (UMFIX) were investigated. STR profiles from the (immunolabe

Technical in-depth comparison of two massive parallel DNA-sequencing methods for formalin-fixed paraffin-embedded tissue from victims of sudden cardiac death.

Sudden cardiac death (SCD) is a tragic and traumatic event. SCD is often associated with hereditary genetic disease and in such cases, sequencing of stored formalin fixed paraffin embedded (FFPE) tissue is often crucial in trying to find a causal genetic variant. This study was designed to compare two massive parallel sequencing assays for differences in sensitivity and precision regarding variants related to SCD in FFPE material. From eight cases of SCD where DNA from blood had been sequenced using HaloPlex, corresponding FFPE samples were collected six years later. DNA from FFPE samples were

The genital microbiome and its potential for detecting sexual assault.

Since its inception, the Human Microbiome Project (HMP) has provided key discoveries that can be applied to forensics, in addition to those of obvious medical value. Whether for postmortem interval estimation, geolocation, or human identification, there are many applications of the microbiome as an investigative lead for forensic casework. The human skin microbiome has shown great potential for use in studies of transfer and human identification, however there has been little focus on the genital microbiome, in particular penile skin which differs from other body sites. Our preliminary data on

Extraordinary claims require extraordinary evidence in asserted mtDNA biparental inheritance.

A breakthrough article published in PNAS by Luo et al. challenges a central dogma in biology which states that the mitochondrial DNA (mtDNA) in humans is inherited exclusively from the mother. We re-analyzed original FASTQ files and results reported by Luo et al. to investigate methodological issues (e.g. nuclear mitochondrial DNA or NUMTs, DNA rearrangements) that could lead to biological misinterpretations. A comprehensive analysis of their data reveals several methodological and analytical issues that must be carefully addressed before challenging the current paradigm. We first show that th
