Dwarfism in Pinus taeda originates from independent somatic mutations co-localized in a shared genomic region.

Somatic mutations in long-lived conifers are rarely characterized yet offer a unique window into the spontaneous genetic forces that shape variation in plants. In Pinus taeda, dwarf phenotypes originate from abnormal branches, colloquially known as "witches' brooms", where progeny derived from the affected branch segregate for dwarfism in an apparent Mendelian 1:1 ratio. In this study, we genotyped six unrelated wind-pollinated families segregating for dwarfism using single-nucleotide polymorphism markers that had been previously positioned on a linkage map. Trait-loci association analyses ide

Effect of indoxacarb and commercial pesticide adjuvant on the honeybee (Apis mellifera) temperature preference, CO2 emission and expression of detoxification-related genes.

BACKGROUND: Plant Protection Products (PPPs) consist not solely of active ingredients. Some of the other ingredients of the finished formulations are intended to enhance the efficacy of the main active ingredient. The PPPs' effects impact honeybees that pollinate crops by exposing them to a complex of numerous chemical compounds. Depending on their composition, PPPs affect these insects in various ways. The present study estimated the effect of sublethal doses of insecticide (Active Ingredient (AI): indoxacarb) and commercial adjuvant (AI: alkylbenzene sulphonic acid sodium salt) applied indiv

Design and Synthesis of Indole-Containing Tetrahydrothiazolo[3,2-a]pyrimidine Mesoionics against Hemipteran Pests.

Mesoionic insecticides have emerged as promising pest nAChR modulators devoid of cross-resistance with neonicotinoids, demonstrating exceptional efficacy against sap-feeding insects. Nevertheless, the tetrahydrothiazolo[3,2-a]pyrimidine structure remains underexplored within this chemotype. Here, we report an indole-based scaffold hopping of tetrahydrothiazolo[3,2-a]pyrimidine, which leads to the development of a series of novel mesoionics. Their insecticidal potency is evaluated against the hemipteran pest Acyrthosiphon pisum (A. pisum) and Sogatella furcifera (S. furcifera). Derivative G1 ex

Whole-genome duplication enables rapid evolution of male-biased sex allocation.

Sex allocation in hermaphrodites involves a balance between fitness gained through male and female functions. Whole-genome duplication (WGD) might disrupt such a gradually evolved balance, if the relatively instantaneous effects of WGD on cell and organ sizes affect sexual functions differently. We studied sex allocation patterns in intraspecific populations of neo-autotetraploids and their diploid progenitors within Galax urceolata. Floral organ sizes and numbers were measured using light microscopy and genome sizes verified with flow cytometry. Relative to diploids, autotetraploids had propo

Avoiding the tragedies of parasite tolerance in Darwinian beekeeping.

Bee declines have been partly attributed to the impacts of invasive or emerging parasite outbreaks. For western honeybees, Apis mellifera, major losses are associated with the virus-vectoring mite, Varroa destructor. In response, beekeepers have focused breeding efforts aimed at conferring resistance to this key parasite. One method of many is survival-based beekeeping where colonies that survive despite significant Varroa infestations produce subsequent colonies. We argue that this 'hands-off' approach will not always lead to Varroa resistance evolving but rather tolerance. Tolerance minimize

RNAi-Based Bioinsecticides for Controlling Vector-Borne Diseases.

Vector-borne diseases account for 17% of all infectious diseases. The most effective strategies for controlling these diseases have focused on decreasing the vector population, primarily through the use of insecticides. Many insecticides have no specific targets, harming pollinators and beneficial insects. Additionally, the vector populations are developing resistance, reducing the effectiveness of these strategies and increasing ecological damage. Double-strand RNA (dsRNA) is widely used in insects to study gene function by knocking down their expression. Recently, this technology has been ap

Enhancing Honey Bee Health: Evaluating Pollen Substitute Diets in Field and Cage Experiments.

Honey bees (Apis mellifera L.) play vital roles as agricultural pollinators and honey producers. However, global colony losses are increasing due to multiple stressors, including malnutrition. Our study evaluated the effects of four pollen substitute diets (Diet 1, Diet 2, Diet 3, and Control) through field and cage experiments, analyzing 11 parameters and 21 amino acids. Notably, Diet 1 demonstrated significantly superior performance in the field experiment, including the number of honey bees, brood area, consumption, preference, colony weight, and honey production. In the cage experiment, Di

The Greasy Pole Syndrome in Alliaria petiolata (Brassicaceae): The Pubescence and Wax Coverage on Stems Reduce Invasion by Lasius niger Ants.

To reduce negative effects of floral visitation by ants, which do not serve as reliable cross-pollinators, some plants have developed a non-floral, stem-based defense mechanism called greasy pole syndrome. In the present study, we examined the effects of two surface features (trichomes and three-dimensional epicuticular wax coverage) on stems of Alliaria petiolata plants on visiting frequencies, travelled distances, and running velocities of Lasius niger ants. The experiments were performed with stem samples prepared from different (apical and basal) stem portions showing different surface mor

Naturally Occurring Triploidy in Cannabis.

Polyploidy is a significant evolutionary process in plants that involves the duplication of genomic content and has been recognized as a key mechanism driving plant diversification and adaptation. In natural populations, polyploids frequently arise from unreduced gametes, which subsequently fuse with reduced or unreduced gametes, resulting in triploid or tetraploid offspring, respectively. Cannabis sativa L. is a diploid species, but recent work using artificially induced polyploidy has demonstrated its potential advantages in an agricultural setting. Further, recent work has identified that s

Characterization of the Volatile Components of Essential Oils of Selected Plants in Kenya.

Essential oils are secondary metabolites that plants produce for protection from pests and predators, attraction of pollinators, and seed dispersal. The oils are made up of a mixture of compounds that give a characteristic flavour and odour. Currently, essential oils are receiving great attention in research for their phytochemical and antimicrobial activities. However, there is scanty information on the chemical composition of many plants. This study provides a detailed analysis of the chemical composition of essential oils of ginger, garlic, tick berry, and Mexican marigold in Kenya. The ess

Multiphase Porous Electrochemical Catalysts Derived from Iron-Based Metal-Organic Framework Compounds.

Herbicide use has attracted attention recently due to potential damage to human health and lethality to the honey bees and other pollinators. Fenton reagent treatment processes can be applied for the degradation of herbicidal contaminants from water. However, the need to carry out the normal Fenton reactions under acidic conditions often hinders their practical application for pollution control. Herein, we report on the synthesis and application of multiphasic porous electro-Fenton catalysts prepared from calcinated metal-organic framework compounds, CMOF@PCM, and their application for the min

Biopesticide spinosad induces transcriptional alterations in genes associated with energy production in honey bees (Apis mellifera) at sublethal concentrations.

Bees experience substantial colony losses, which are often associated with pesticides. Besides synthetic insecticides biological compounds such as spinosad are used in agriculture and organic farming against insect pests. However, potential adverse effect at sublethal concentrations to pollinators are poorly known. Here we aim to determine potential adverse outcome pathways of spinosad and to identify molecular effects by investigating transcriptional alterations in the brain of honey bees. We experimentally exposed bees to three sublethal concentrations of 0.05, 0.5 and 5 ng spinosad/bee, and

Nanotechnology-based recent approaches for sensing and remediation of pesticides.

Pesticides are meant to control and destroy the pests and weeds. They are classified into different categories on the basis their origin and type of pest they target. Chemical pesticides such as insecticides, herbicides and fungicides are commonly used in agricultural fields. However, the excessive use of these agrochemicals have adverse effects on environment such as reduced population of insect pollinators, threat to endangered species and habitat of birds. Upon consumption; chemical pesticides also cause various health issues such as skin, eye and nervous system related problems and cancer

Recent speciation of Capsella rubella from Capsella grandiflora, associated with loss of self-incompatibility and an extreme bottleneck.

Flowering plants often prevent selfing through mechanisms of self-incompatibility (S.I.). The loss of S.I. has occurred many times independently, because it provides short-term advantages in situations where pollinators or mates are rare. The genus Capsella, which is closely related to Arabidopsis, contains a pair of closely related diploid species, the self-incompatible Capsella grandiflora and the self-compatible Capsella rubella. To elucidate the transition to selfing and its relationship to speciation of C. rubella, we have made use of comparative sequence information. Our analyses indicat

Human factors facilitating the spread of a parasitic honey bee in South Africa.

Workers of the honey bee subspecies Apis mellifera capensis (Eschscholtz) produce female offspring by thelytokous parthenogenesis and can parasitize colonies of other subspecies. In 1990, translocation of 400 colonies of A. m. capensis into the distribution area of A. m. scutellata by a commercial beekeeper triggered a dramatic parasitic phenomenon. Parasitized colonies died within a few months of infestation, and this resulted in the loss of tens of thousands of colonies by commercial beekeepers in the A. m. scutellata range in South Africa. To deal with the problem and to identify methods th

Transcriptional and post-translational regulation of S-adenosyl-L-methionine: salicylic acid carboxyl methyltransferase (SAMT) during Stephanotis floribunda flower development.

Methyl salicylate (MeSA) and a number of other volatiles are primarily emitted in the evening/night by Stephanotis floribunda leading to attraction of night active pollinators. A second minor emission peak for MeSA occurs in the morning/day. To understand these emission patterns, we have studied in detail the temporal regulation of the last step of the biosynthetic pathway of MeSA, the convertion of salicylic acid (SA) to MeSA catalysed by S-adenosyl-L-methionine: salicylic acid carboxyl methyltransferase (SAMT). We observed that in young flowers a maximum in SAMT activity occurs in the night,

Honeydew microbial ecology: a neglected frontier in multitrophic networks.

Honeydew, the sugary excretion produced by sap-feeding Hemiptera, is one of the most common carbohydrate resources in many plant-based food webs. Honeydew supports a wide range of organisms, including ants, pollinators, (hyper-)parasitoids wasps, predatory insects, and microbes. As a sugar-rich resource, honeydew is frequently colonized by specific microbes (i.e. bacteria and fungi), which consume its sugars and other nutritional constituents. Recent research suggests that microbes within honeydew may modify its traits. Thereby driving microbial succession and acting as important 'hidden playe

Blind genomic tree scans identify loci underlying adaptive peaks in Antirrhinum.

A key question in evolutionary biology is how barriers to gene flow operate and arise between populations, leading to speciation. Barriers are typically identified by comparing populations distinguished by phenotype, geographical location and/or environment. However, such prior classifications may bias recovery towards features obvious to the human eye, influencing mechanistic interpretations. Here we develop a method blind to prior classifications. We apply it to 18 Antirrhinum populations occupying habitats from coastal to alpine. By scanning the genome for regions with deeply rooted similar
