Advanced age decreases local calcium signaling in endothelium of mouse mesenteric arteries in vivo.

Aging is associated with vascular dysfunction that impairs tissue perfusion, physical activity, and the quality of life. Calcium signaling in endothelial cells (ECs) is integral to vasomotor control, exemplified by localized Ca(2+) signals within EC projections through holes in the internal elastic lamina (IEL). Within these microdomains, endothelium-derived hyperpolarization is integral to smooth muscle cell (SMC) relaxation via coupling through myoendothelial gap junctions. However, the effects of aging on local EC Ca(2+) signals (and thereby signaling between ECs and SMCs) remain unclear, a

Prolonged matrix metalloproteinase-3 high expression after cyclic compressive load on human synovial cells in three-dimensional cultured tissue.

Excessive mechanical stress is thought to be a factor in the development of joint disorders through the expression of matrix metalloproteinases (MMPs) and related cytokines. Although studies revealed that mechanical stress on the synovium induces MMP expression, it is still not known which MMPs prolonged high level expression. The authors focused on MMP-3, which is one of the major factors in joint disorders such as rheumatism and temporomandibular joint disorders. They examined mRNA and protein levels of MMP-3, other MMPs and related cytokines after loading stress. Human synovial cells were s

Barrier membranes used for ridge augmentation: is there an optimal pore size?

PURPOSE: To identify the optimal pore size of barrier membranes for successful alveolar ridge reconstruction procedures, to determine if cortical perforations have any effect on bone regeneration, and to reiterate that bone graft containment is an important parameter for successful regeneration. MATERIALS AND METHODS: This was a prospective, randomized, controlled study performed on hound dogs. Corticocancellous tibial bone grafting was performed to the lateral border of the mandible and protected with barrier membranes (meshes). The experiment analyzed three different pore sized meshes, comp

The use of porous prostheses in delayed reconstruction of total hip replacements that have failed because of infection.

Between March 1984 and March 1989, thirty-four patients who had an infection at the site of a cemented total hip prosthesis were managed with resection arthroplasty and delayed implantation of a porous total hip prosthesis without cement. The interval from the time of the resection arthroplasty to the implantation of another prosthesis averaged eight months (range, three to nineteen months). At an average of forty-seven months (range, twenty-four to seventy-two months) after the reimplantation, six patients (18 per cent) had recurrence of the infection. Patients who had rheumatoid arthritis we

Enhanced osteogenic proliferation and differentiation of human adipose-derived stem cells on a porous n-HA/PGS-M composite scaffold.

This study explored the applicability, cellular efficacy, and osteogenic activities of porous nano-hydroxyapatite/Poly (glycerol sebacate)-grafted maleic anhydride (n-HA/PGS-g-M) composite scaffolds. Nuclear magnetic resonance (NMR) analyses indicated that approximately 43% of the hydroxide radicals in PGS were displaced by maleic anhydride. Resonance bands at 1036 cm-1 occurred in scaffolds containing nHA powders, and peak areas increased when n-HA weight increased in PGS-M-n-HA-0.4, PGS-M-n-HA-0.5, and PGS-M-n-HA-0.6 scaffolds. The n-HA/PGS-g-M composite scaffolds exhibited porous microstruc

Synthesis of and in vitro and in vivo evaluation of a novel TGF-β1-SF-CS three-dimensional scaffold for bone tissue engineering.

The role of transforming growth factor-β1 (TGF-β1) in normal human fracture healing has been previously demonstrated. The objective of the present study was to examine the biocompatibility of TGF-β1-silk fibroin-chitosan (TGF-β1-SF-CS) three-dimensional (3D) scaffolds in order to construct an ideal scaffold for bone tissue engineering. We added TGF-β1 directly to the SF-CS scaffold to construct a 3D scaffold for the first time, to the best of our knowledge, and performed evaluations to determine whether it may have potential applications as a growth factor delivery device. Bone marrow-derived

Comparison of hydroxyapatite- and porous-coated stems in total hip replacement.

24 patients underwent bilateral hip replacement, using the SROM modular system (Depuy Johnson & Johnson, MA, USA) with the porous-coated sleeve in one hip and an HA-coated sleeve in the other. 20 patients were followed for at least 4 years with clinical and radiographic evaluations. We found no difference between the two sides as regards the time of disappearance of thigh pain and hip scores. Radiographs showed a buttress sign in 18 HA-coated and 15 porous coated stems. There were no differences in the serial bone-remodeling pattern around the proximal sleeve and measurement of wear volume bet

Impact Microindentation Evaluates Bone Strength, Bone Quality, and Fracture Susceptibility Across Skeletal Sites: A Cadaver Study.

BACKGROUND: Impact microindentation (IMI) is a minimally invasive technique for assessing bone material properties that can be performed clinically. In this study, we aimed to determine how measurements obtained by IMI (bone material strength index [BMSi]) relate to bone strength, fracture resistance, and susceptibility to fracture. QUESTIONS/PURPOSES: (1) Is BMSi associated with bone strength of the femur, radius, and vertebrae, and does incorporating BMSi alongside bone mineral density (BMD) improve the estimation of strength? (2) Does BMSi evaluate fracture toughness and mechanical propert

[Recent advances on bioaffinity fishing technology for discovery of active components from natural products].

Natural products have consistently served as an invaluable source for novel drug development, and the efficient and targeted screening of bioactive constituents from these complex mixtures is crucial for advancing drug discovery. Bioaffinity fishing techniques exploit the highly specific interactions between ligands and their biological targets-such as antigen-antibody, enzyme-substrate, and hormone-receptor interactions-to simultaneously separate and identify active compounds from complex natural product extracts. When coupled with liquid chromatography-mass spectrometry, bioaffinity fishing

Biomechanical effects of endoscopic ULBD in a reduced‑stiffness lumbar spine model: focus on endplate stress and hypothesis‑generating risk indicators (a finite element analysis).

This finite element study aimed to compare the biomechanical effects of endoscopic unilateral laminotomy for bilateral decompression (Endo-ULBD) on lumbar spinal stability and load distribution in both healthy and osteoporotic spine models, with a specific focus on identifying biomechanical changes in a reduced‑stiffness bone model that may serve as hypothesis‑generating risk indicators, recognizing that the model does not simulate full osteoporotic pathology. A validated L4-L5 finite element model was developed. Virtual Endo-ULBD was performed on both healthy and reduced‑stiffness (osteoporos

Current State-of-the-Art 3D MRI Sequences for Assessing Bone Morphology with Emphasis on Cranial and Spinal Imaging: A Narrative Review.

Traditionally, CT has been the go-to method for visualizing bone structures, while MRI has been preferred for assessing soft tissues, because structures containing tightly bound water molecules - such as bones, tendons, cartilage, and ligaments - produce a rapidly decaying T2* signal, which conventional MRI sequences fail to capture. To address this limitation, spoiled gradient echo sequences were refined, and short-TE sequences were introduced, enabling radiation-free bone imaging. This advance is particularly crucial for pediatric patients and in scenarios where an MRI-only approach is prefe
